Description
The invention relates to a device for deflecting a light beam a unit rotatable about a first axis, the two fixed to each other Reflection surfaces - namely a first and a second reflection surface - includes, and which receives a beam of light and to a third Reflecting surface forwards about a second axis that is perpendicular to the first axis of rotation.
Furthermore, the invention relates to a scanning microscope with a light source emitted a beam of light for illuminating a sample and with a Device for deflecting the light beam, one about a first axis has a rotatable unit that has two mutually stationary reflection surfaces - namely a first and a second reflecting surface - includes, the rotatable unit receives a beam of light and sends it to a third Reflecting surface forwards about a second axis that is perpendicular to the extends first axis of rotation, is rotatable.
An optical arrangement for scanning a beam in two essentially axes perpendicular to each other, especially for use with confocal laser scanning microscopes, with two using one drive each axes perpendicular to each other - x-axis and y-axis - rotatable Mirroring is known from German Patent 196 54 210 C2. The Optical arrangement is to avoid serious imaging errors characterized in that one of the two mirrors is another mirror is rotatably associated in a predetermined angular position, so that the mutually associated mirrors - first and second mirrors - around together rotate the y-axis while rotating the beam around a pivot point that points to the axis of rotation - x-axis - of the third mirror rotating alone.
German patent application 100 33 549.7 discloses an optical one Arrangement for deflecting a light beam in particular in two substantially perpendicular directions, preferably to Use in confocal scanning microscopes, with two using one each Rotary drive around mutually perpendicular axes - x-axis and y- Axis - rotating mirrors, one of the two mirrors another Mirror is assigned non-rotatably in a predetermined angular position so that the associated mirrors - first and second mirrors - together rotate around the y-axis while rotating the light beam around a pivot point, which lies on the axis of rotation - x-axis - of the third mirror. To minimize and ideally to eliminate those generated by the arrangement The optical arrangement is characterized by that the mirrors are arranged such that the optical axis of the always running between the second and the third mirror light beam essentially in a direction encompassing the x-axis, perpendicular to the y-axis standing level. The patent application also proposes that with a view to flexible use, the individual components of the Arrangement are modular and interchangeable.
In scanning microscopy, a sample is illuminated with a light beam observe the reflection or fluorescent light emitted by the sample. The focus of an illuminating light beam is controlled using a controllable one Beam deflection device, generally by tilting two mirrors, in an object plane moves, the deflection axes mostly perpendicular stand on each other, so that one mirror in the x-, the other in the y-direction distracting. The mirror can be tilted, for example, with the help of Galvanometer actuators accomplished. The performance of the object coming light is dependent on the position of the scanning beam measured. The control elements are usually used with sensors Determination of the current mirror position equipped.
Especially in confocal scanning microscopy, an object with the focus of a light beam scanned in three dimensions.
A confocal scanning microscope generally includes a light source focusing optics with which the light from the source is directed onto a pinhole - the so-called excitation diaphragm - is focused, a beam splitter, a Beam deflection device for beam control, microscope optics, a Detection aperture and the detectors for the detection of the detection or Fluorescent light. The illuminating light is through a beam splitter coupled. The fluorescent or reflection light coming from the object passes back to the beam splitter via the beam deflection device, passes this, in order to then be focused on the detection aperture, behind which the detectors are located. Detection light that does not directly come from comes from the focus region, takes a different light path and passes through it Detection aperture is not, so that you get a point information by sequential scanning of the object leads to a three-dimensional image. A three-dimensional image is usually created by layer-by-layer image data acquisition achieved, the path of the scanning light beam on or in the object more ideal Way describes a meander. (Scan a line in the x direction at constant y-position, then stop x-scanning and use y- Swivel the adjustment to the next line to be scanned and then, at constant y position, scan this line in the negative x direction u. s. w.). Around The sample table or will enable layered image data acquisition the lens moved after scanning one layer and so the next layer to be scanned brought into the focal plane of the lens.
The devices disclosed in the cited documents have the disadvantage that that the interchangeability of the mirrors or their motor drives through the high demands on the adjustment accuracy in particular is restricted. A complex readjustment of the optical components is usually unavoidable.
The invention is therefore based on the object of a device for Deflection of a light beam to indicate a simple, straightforward and quick replacement of the individual elements deflecting the light beam allows.
The above object is achieved by a device for deflecting a light beam solved, which is characterized in that the rotatable unit one to the first and to the second reflection surface stationary further reflection surface has, the first and the further reflection surface perpendicular to the second reflection surface are arranged.
It is also an object of the invention to provide a scanning microscope which simple, uncomplicated and quick exchange of the light beam allows deflecting individual elements and in particular a simple one and easy switching between a resonant and a non-resonant drive allowed.
This task is solved by a scanning microscope is characterized in that the rotatable unit one to the first and to the second reflection surface has a fixed further reflection surface, wherein the first and the further reflection surface perpendicular to the second Reflecting surface are arranged.
Another object of the invention is a method for investigation to disclose a sample that is an improved, largely complete and error-free, high-resolution recording of a wavelength spectrum from allows a sample of outgoing detection light.
The invention has the advantage that it is simple, uncomplicated and quick Exchange of the reflection surfaces and especially the motor Drives is possible without a complex, time-consuming and ultimately expensive adjustment is required.
The device according to the invention has the decisive advantage that the Light beam at right angles on each reflective surface of the rotatable unit is reflected, making this arrangement very insensitive to Is misalignments; because the incident light beam occurs according to the Cat's eyes (retroreflector effect) always largely perpendicular from the rotatable unit.
The reflecting surfaces can be used as mirror surfaces or as totally reflecting surfaces Surface of a vitreous body.
The vitreous body preferably comprises at least one prism. In a preferred embodiment, the rotatable unit consists of two prisms are preferably cemented together. It is also possible the whole to produce a rotatable unit monolithically from one body. The rotatable Unit preferably has entry and exit windows, these for Avoidance of interfering interference are preferably arranged that the angle of incidence of the light beam on the entrance window from the Difference angle of exit from the exit window. In the case of right angles Deflection of the light beam by the rotatable unit is advantageous if the entrance window and the exit window are in a slightly 90 degree deviate from each other.
In a very particularly preferred embodiment, the rotatable unit is rotatable about the first axis together with the third reflection surface, whereby in imaging scanning systems, such as scanning microscopes, image rotation is possible.
Both the rotatable unit and the third reflecting surface is is preferably driven by a motor. All common drives are used Motors, such as stepper motors, can be used. In one prefer Design includes the drive a galvanometer. especially the third reflection plane is preferably as one of a resonant Galvanometer driven oscillating mirror designed.
In a very particularly preferred embodiment, the scanning microscope has a third reflection surface designed as a mirror, which one does not resonant galvanometer is driven. Furthermore, a fourth Reflective surface provided with the same optical properties that is powered by a fast, resonant galvanometer. In this Design is the third reflective surface, along with its drive against it fourth reflecting surface interchangeable. There are guide rails for this provided along which the reflection surfaces can be moved. Farther stop elements are provided, the working position of each Define selected reflection surface and its drive. The Scanning microscope thus offers all the advantages of a non-resonant deflection, such as. B the possibility of the light beam on a selectable point of the sample position and "park" there, as well as all the advantages of a resonant Distraction, which is particularly characterized by speed and high frame rates distinguished.

The third reflection surface 29 is evaporated on a mirror substrate 43 . By rotating the rotatable unit 35 about the axis 27 , the light beam 5 is deflected perpendicular to the plane of the paper. The rotation of the third reflection surface 35 about the axis 37 causes a deflection of the light beam 5 perpendicular to the plane of the drawing. The arrangement is inexpensive and can be manufactured particularly precisely. It is particularly advantageous if the entrance window 59 is not exactly perpendicular to the exit window 61 of the rotatable unit, as a result of which disturbing interference is avoided.
FIG. 4 shows a further embodiment of a device for deflecting a light beam, which essentially corresponds to the arrangement shown in FIG. 2. In addition, a deflection mirror 13 is provided, which reflects the light beam 5 onto the first axis of rotation 27 . The rotatable unit 29 is driven by a stepper motor 63 .
Fig. 5 shows an apparatus for deflecting a light beam in a perspective view. The device includes a unit 29 which can be rotated about a first axis 27 and which contains two reflection surfaces 31 , 33 which are stationary relative to one another - namely a first reflection surface 31 and a second reflection surface 33 - and which receives the light beam 5 and forwards it to a third reflection surface 35 which is rotatable about a second axis 37 , which runs perpendicular to the first axis of rotation 27 . The rotary unit 29 has a fixed to the first reflecting surface 31 and the second reflection surface 33 additional reflection surface 39 which receives the light beam from the first reflective surface 31 and reflected to the second reflection surface 33rd The rotatable unit 29 is driven by a galvanometer 41 via a drive shaft 69 . The third reflection surface 35 is evaporated onto a third mirror substrate 43 ; the substrate is driven by a resonant galvanometer 65, not shown in the figure, via a further drive shaft 67 . The first reflection surface 31 is vapor-deposited on a first mirror substrate 45 , the second reflection surface 33 on a second mirror substrate 47 and the further reflection surface 39 on a further mirror substrate 49 . The rotatable unit 29 has a housing 51 which encloses the reflection surfaces and protects them from contamination. In addition, a deflection mirror 13 is provided, which reflects the light beam 5 onto the first axis of rotation 27 . The third reflection surface 35 , the mirror substrate 43 and the resonant galvanometer 65 are combined to form a module 71 , which is compared to a further module 73 , which has a fourth reflection surface 75 on a mirror substrate 77 , which has the same optical properties as the third reflection surface 35 , and includes a non-resonant galvanometer 79 as a drive.
FIG. 6 shows a further embodiment of a device for deflecting a light beam, which essentially corresponds to the arrangement shown in FIG. 5. In addition, a guide rail 81 is provided in this embodiment, which enables the modules 71 , 73 to be replaced simply and precisely. Furthermore, stop elements 83 , 85 are provided which precisely define the respective working position of the modules 71 , 73 in such a way that the desired reflection surface can be positioned exactly at the intersection of the axis 27 and the second axis 37 .
