Description
The invention relates to a device for preparing tissue samples for the production of paraffin blocks for histological evaluation, in particular of animal or human organs, preferably of elongated, small-lumen animal or human organs. The invention also relates to a method for preparing tissue samples. One area of application is, for example, the processing of murine intestines (rodents), e.g. for the production of so-called Swiss rolls.
For example, embedding cassettes are known from the prior art, in which tissue cut to size is fixed and embedded in paraffin. The cassettes are cast vertically aligned in one block. Layers can then be cut from the block using a microtome and examined using a microscope.
The problem with this type of embedding cassette is that both the size of the installation cassette (inside: 30mm x 25mm, height: 5 or 10mm) and the height of the base mold and thus the height of the paraffin blocks that can be produced is limited, e.g. to 1 cm. Furthermore, the production of a paraffin block with a vertically aligned piece of tissue with a length L<1 cm is not possible with the embedding cassettes known from the prior art. For the histological evaluation of elongate tissues or organs with a length of several centimeters, it is therefore necessary to produce a large number of paraffin blocks.
When preparing the tissue for evaluation using step sections, the required number of sections are made, trimmed in shape and size, the excess material is discarded and then the next step is cut. The disadvantage here is that when creating step cuts, tissue lying between two steps is lost.
For example, the processing of human punch biopsies, for example mammary, gland, or muscle punches, is known, with three to five pieces of tissue being removed from the affected organ. A well-known disadvantage is that only one cutting plane can be viewed and the remaining tissue is archived, since the small sample size means that it is not possible to process the tissue in sections without tools.
The processing of murine intestines using the Swiss Roll method is also known from the prior art. The intestine is opened and rolled in a snail shape with the help of medical instruments (e.g. tweezers) and blocked out horizontally. The aim of the procedure is to provide an overview of the complete longitudinal section of the intestine.
The problem with the processing of murine intestines is that artefacts can arise as a result of the opening of the intestines and the contents of the intestines are lost as a result. Due to its nature, the casing can slip during the blocking-out process, which makes it difficult to produce a section for an overview of the complete longitudinal section of the casing. Another disadvantage is that it is not possible to precisely display the same plane for different preparations.
It is therefore an object of the invention to provide a device that makes it possible to produce standardized and optimized stabilized tissue samples, preferably with the length L>1 cm, in particular also Swiss rolls, and to facilitate the preparation of tissue samples.
This object is achieved in that, in a device according to the invention, on a plate element, preferably on a plate element with four sides, in particular two of which are parallel opposite one another, at least one side of the edge, preferably two opposite sides of the edge, are adjacent, preferably has recesses which are next to one another in the direction in which the edge extends and are separated by separating webs, into which the tissue sample can be placed.
In particular, the separating webs extend in a direction away from the edge, in or parallel to a plane in which the plate element lies. The separating webs can be aligned parallel to one another and perpendicular to the edge extension direction on this side.
However, the separating webs can also run at an angle other than 90 degrees to the extent of the edge; in particular, in such an embodiment, two adjacent separating webs can be inclined towards one another. In a region between the separating webs, a respective recess is open to the outside, ie open in a direction away from the edge.
In the method, the object is achieved in that the tissue sample is placed in the recesses of such a device according to the invention, in particular by wrapping the device the tissue sample is successively placed in the recesses on opposite edge sides. The method and the device enable the tissue to be stabilized sample in that it is held in the recesses, in particular between the separating webs of a respective recess.
In contrast to the prior art, it is possible with such a device, within a paraffin block in which the device is cast, in particular together with a holding aid for a microtome cutting device, to display a complete overview in cross section and longitudinal section of the tissue sample to be examined.
In particular, there is the possibility of processing the fabric in stages in a precise and standardized manner and thus avoiding waste/waste. This optimizes the processing of all human and animal tissues and organs that have an elongated shape, e.g. intestines, arteries, veins, nerves, in particular murine intestines or human blood vessels.
A panel element according to the invention is preferably a flat body with four sides, each with two parallel opposite panel surfaces, in particular the two largest panel surfaces of the panel element, which are spaced apart in the thickness direction of the panel element.
The plate element preferably has an extension of 2 to 6 cm, preferably 4 to 5 cm, in its longitudinal direction, 1 to 3 cm, preferably 2 to 3 cm, in its width direction, and 0.1 to 0.5 cm, preferably 0.1 to 0.1 cm, in its thickness direction .3 cm up.
The plate element according to the invention is preferably made entirely of an elastomer, in particular made of silicone with a Shore A hardness of 15 to 95, preferably 15 to 30, preferably 20 to 25. In particular, production from such a material, in particular silicone, has the advantage that silicone can be easily cut, in particular in comparison to the tissue sample and has comparable cuttability to the enclosing paraffin, and has a low material price.
The plate element according to the invention has on at least one side of the edge, preferably on two opposite sides of the edge, adjacent recesses into which a tissue sample can be placed. In the preferred arrangement, the recesses arranged on two opposite sides are offset from one another, alternatively arranged exactly opposite one another.
The invention provides for spacing adjacent recesses from each other by means of a separating web. The separating webs are preferably formed with a wall thickness of 0.5 to 2 mm, preferably 1 mm, viewed in the direction of the edge extension, and more preferably parallel to one another.
In a preferred embodiment, the openings of the recesses can each be reduced in relation to the maximum recess cross section by at least one roof web which, starting from one of the two separating webs (in particular its end) surrounding one recess, extends towards the other separating web. A respective opening is preferably reduced by two roof webs compared to the maximum cross section of the recess, each of which, starting from one of the two separating webs (in particular starting from the end of the separating web) surrounding a recess, are directed toward one another.
The free ends of the roof webs facing each other can contact each other and thereby form a closed but openable opening area of the recess. Alternatively, the free ends of the roof webs lie opposite one another at a distance and as a result form an open, preferably even further openable, opening region of the recess.
Preferably, especially if the plate element is not made entirely of a flexible material, at least the at least one roof ridge, preferably at least the roof ridges and/or dividing ridges are made of a flexible material, in particular the respective roof ridges can be molded onto the dividing webs. Due to the flexibility of at least the roof ridges, a major advantage of the roof ridges is the possibility of deflecting the roof ridges in the plane of the plate and/or perpendicular to the plane of the plate, which allows the tissue sample to be easily introduced into the recess by retracting the roof ridges. Further advantages of the roof bars result from their functionality as barbs, which ensures that the tissue sample is stabilized in position and enables precise preparation for histological evaluation.
Between them, the free ends of the roof webs form the open or at least openable opening region of the recess, through which the tissue sample can be introduced into the recess.
In general, recesses have a reduction in their recess cross section, in particular the width in an opening area. An opening area is preferably an area designed with an undercut, preferably an open or closed but openable area rer area through which the tissue sample can be inserted into the respective recess.
The cross section, in particular the width of the recesses viewed in the direction in which the edges extend, is 2 to 5 mm, preferably 3 to 4 mm, and thus limits the cross section of the fabric that can be placed in the recesses.
In the method according to the invention for preparing a tissue sample, the tissue samples are placed one after the other in the recesses of the plate element, in particular by wrapping the device in the recesses on opposite edge sides.
Alternatively, the tissue sample can only be inserted into the recesses on one edge side of the plate element, resulting in meandering tissue samples/tissue spirals that are particularly suitable for producing Swiss rolls.
In the preferred embodiment with roof bars, the opposite free ends of roof bars are spread apart when the tissue sample is inserted into the recess and automatically move towards one another when the tissue sample is placed in the recess. The tissue sample is then securely enclosed in a recess by the separating webs and the roof webs.
A major advantage of the method of the invention is that it is easier to operate, so that the device can be used without prior medical or histological knowledge. Accordingly, a high degree of standardization, reproducibility and precision can also be achieved and enables comparability between the samples.
In a further preferred embodiment, at least one cover element, preferably two cover elements, can be placed on opposite sides over the longitudinally extending sides of the edge of the plate element to fix the fabric spiral produced using the method according to the invention and to prevent the fabric from slipping.
In a preferred embodiment, two cover elements are pushed over each of the recesses on one side of the edge and then have a spaced area between them. With the help of the spacer area, it is possible to make a cut in the spacer area, in particular to sever the plate element, preferably in a direction parallel to the edge sides that have the recesses.
Precise positioning of the incision, due to the arrangement of the cover element, is advantageous since the loss of tissue can be reduced in this way. A cut through the tissue sample, in particular through the plate element, is also possible without the cover elements by positioning the cut over the recesses.
