Description
The invention relates to a saliva test and a method for the detection of pathogens by analyzing such saliva tests.
In the prior art it is known to carry out so-called saliva tests to detect pathogens in the saliva of test persons. The most common procedure is to take a sample with a swab, which is then inserted into a tube filled with liquid. The tube is labeled, sealed and sent to the test laboratory in a suitable container, typically together with several other tubes in a cardboard box. The tube itself is packaged in order to meet the transport regulations for biological substances of category B (UN3373). However, this procedure is only suitable to a limited extent for use as a self-test or rapid test, since proper sampling can actually only be ensured by trained personnel and since packaging and shipping are comparatively complex.
In order to be able to use saliva tests as a self-test or rapid test for the detection of pathogens, the use of test cards made of filter paper has already been considered in the prior art, which can be spat on, packaged dry and sent to a laboratory (see e.g. Smit, PW, et al., "An Overview of the Clinical Use of Filter Paper in the Diagnosis of Tropical Diseases", Am J Trop Med Hyg. 2014 Feb 5; 90 (2): 195-210). The known solutions of this type have the disadvantage, on the one hand, that saliva is colorless and the spit-on area can no longer be seen, especially after the saliva has dried. This makes an efficient and reliable analysis difficult. Furthermore, compliance with the transport regulations for category B biological substances (UN3373) is problematic without resorting to processes that are as complicated as those for swabs.
The object of the invention is to provide a saliva test and a method for detecting pathogens with which these disadvantages are overcome.
Against this background, the invention relates to a saliva test suitable for a method for detecting pathogens in the saliva of a test person, the saliva test comprising a leaflet made of an absorbent material, with both a water-soluble dye and a disinfectant being distributed on the absorbent material of the leaflet .
By using the dye, the spit on area is made recognizable for both the test person and the laboratory. The dye dissolves in the saliva and, due to capillary forces, migrates from the center of the spit-on and therefore moistened area to the edges. After the saliva has dried, the spit-on area is discolored and is surrounded by a more strongly colored peripheral edge.
The disinfectant ensures that even drying-resistant pathogens on the leaflet are no longer infectious. This greatly facilitates safe transport and safe handling in the laboratory.
In particular, the disinfectant can be a virucide, so that the saliva test is suitable for a method for detecting viruses in the saliva of test persons. In this context, the use of a virucide which acts by disrupting the virus envelope is also preferred. In this variant, the saliva test is suitable for detecting enveloped viruses. The virus envelope usually encloses a capsid in which the viral nucleic acid is packaged. Enveloped viruses are usually more dangerous to humans than non-enveloped viruses due to evolutionary advantages, so that tests for the detection of enveloped viruses cover most of the practical applications. For example, the HI virus, SARS coronaviruses 1 and 2, influenza virus, Ebola virus and West Nile virus are enveloped viruses. After removing the virus envelope, the virus is inactive. Nevertheless, the capsid and the viral nucleic acid are not damaged by a virucide, which works by removing lipids in the virus envelope, and they can be analyzed in the laboratory without restrictions.
For example, it can be provided in the context mentioned that the virucide is a surfactant, preferably a nonionic surfactant. Examples of suitable surfactants include octoxinol 9, the reduced variant polyoxyethylene (10) isooctylcyclohexyl ether, polysorbate 20, generally surfactants with polyalkylene glycol chains, salts of bile acids such as cholate or deoxycholate, digitonin, dodecyl maltoside or octyl glucoside.
In connection with the dye, good water solubility and a low affinity for the material of the leaflet are particularly important, for example a low affinity for cellulose, since these factors enable or promote the migration of the dye from the center of the spit on point at its edges will. Furthermore, the dye should be biotolerant so that there is no interaction with the constituents of the sample and also no danger to the user exists when the leaflet is touched with lips or fingers.
In one embodiment, the dye has a solubility in water at 20 ° C. of more than 30 g / l, more than 50 g / l or more than 100 g / l.
Suitable dyes include, for example, dyes from the classes of the methine dyes or the thiazine dyes. An example of a suitable methine dye is fluorescein sodium / uranine. An example of a suitable thiazine dye is methylene blue.
The leaflet of the saliva test can, for example, be cut into the shape of a rectangular card. It is preferably made of filter paper. Filter paper is a flat material that essentially consists of fibers of vegetable origin, especially cellulose fibers. Compared to normal papers, filter paper is distinguished by its porosity and the associated absorbency. Filter paper types suitable for the saliva test according to the invention can, in one embodiment, have a weight per unit area of 50 to 150 g / m 2 and a thickness of 100 to 250 μm. Preferably the fibers are bleached and white.
In one embodiment, the saliva test also has a covering made of a plastic film which is dimensioned such that it can completely cover both surfaces of the leaflet. When the saliva test is delivered, the envelope can already be connected to the leaflet, for example glued, or it can be provided separately.
The wrapping is used to seal the leaflet after use. The leaflet can, for example, be glued with an underside on one half of the envelope or placed on one half of the envelope and the other half of the envelope can then be folded over and placed completely over the upper side of the leaflet. The leaflet sealed in this way can be sent to a laboratory in an envelope, for example. The sealed leaf can be punched in the laboratory, whereby the two pieces of film punched out as a by-product are discarded while the punched-out part of the leaf can be analyzed.
In the case of a gluing of the leaflets and the cover, the adhesive surface is composed in a preferred variant of one or more points or strips, which has the advantage over a flat adhesive surface of better ventilation and consequently drying of the saliva. The leaflet is preferably glued to one half of the wrapper so that the other half of the wrapper can be folded over and placed completely over the top of the leaflet.
If a cover is present, it can be provided in particular that the plastic film of the cover has an adhesive surface which runs in such a way that a folded half of the plastic film can be bonded to the other half of the adhesive film in such a way that an area enclosed between the halves has a uninterrupted adhesive seam is completely separated from the environment. The adhesive surface is, for example, an adhesive strip which extends over at least half the circumference or possibly also the entire circumference in the area of the circumferential contour of the plastic film. The adhesive surface is preferably formed by a pressure-sensitive adhesive applied to the plastic film. In particular, it can be provided that the adhesive surface is covered with a protective film which can be peeled off in use after spitting on the leaflet in order to then place the cover around the leaflet and close it.
Geeignete Materialien umfassen beispielsweise PE oder PP.In particular, it can be provided that the plastic film is transparent. The spit-on leaflet and in particular the color contour, which marks the position of the dried saliva residues, are visible from the outside. In the laboratory, the enclosed leaflet can therefore be punched in the right place. The plastic should also be biotolerant and contain few additives such as plasticizers, etc. Suitable materials include, for example, PE or PP.
The leaflet and / or the casing can be provided with a surface or marking for writing on the saliva test.
Against the background mentioned at the beginning, the invention further relates to a method for the detection of pathogens in the saliva of a test person, comprising the following steps: (a) providing a saliva test according to one of claims 1 to 10; (b) applying saliva to the leaflet of the saliva test and allowing the saliva to dry on the leaflet; and (c) redissolving the saliva residues and analyzing the dissolved saliva residues to identify the pathogens. The pathogens can be identified directly or indirectly through the detection of antibodies against the pathogen.
Step (b) can be performed by a user at home or in a rapid test facility. Step (c) typically takes place in a laboratory. Between steps (b) and (c), the leaflet with the dried saliva residues can be sent to a laboratory, for example by post. Before that, between the Steps (b) and (c) the sealing of the leaflet take place in an envelope, as has been described in connection with the saliva test according to the invention.
As part of step (c), the leaflet can be punched for sample preparation and the saliva residues can be detached from the punched-out part of the leaflet. The solution can then be analyzed with the aid of known analysis techniques. The preferred solvents are water or aqueous alcohol solutions. In the case of a leaflet sealed with an envelope in the form of a plastic film, two punched-out pieces of film are produced as a by-product, but these are discarded or can also remain in the analysis because they are chemically inert and do not interfere with the analysis. The punch should be made on the part of the leaflet on which the dried-up saliva remains. This can be recognized by the dye also through a transparent cover and the correct position can therefore be recognized manually or by machine.
The method according to the invention can in particular be a method for recognizing viruses, preferably enveloped viruses, in the test person's saliva. The viruses can be identified directly or indirectly by detecting antibodies against the viruses. The method according to the invention can serve as a rapid or self-test method for different viral diseases, for example for the detection of HI viruses, SARS coronaviruses 1 and 2, influenza viruses, the Ebola virus or West Nile viruses. It is currently of particular importance to use it as a rapid or self-test procedure to detect an infection with the SARS coronavirus 2.
