Die Aufnahme von chemischen Wirkstoffen kann allerdings das Verhalten und die Vitalität von Zellen verändern, weshalb die Zellen für weitere Experimente nur noch eingeschränkt verwendbar sind. Es gibt zwar Publikationen mit experimentellen Werten, aus denen sich aber keine aussagekräftigen Informationen über den Aufbau und die Struktur der Probe entnehmen lassen. Ein wesentlicher Grund besteht darin, dass in einer typischen biologischen Probe das Impedanzsignal von den unterschiedlichen chemischen Verbindungen und Substanzen in der Probe erzeugt wird, die jede für sich eine unterschiedliche Impedanz aufweisen. Das gemessene Impedanzsignal besteht deshalb aus der Superposition dieser einzelnen Signale. Eine Aufteilung des Superpositionssignals in seine einzelnen Komponenten ist bisher nicht möglich.
Es ist bekannt, dass bei niedrigen Frequenzen die sog. Alpha-Dispersion auftritt, die von den Gewebeeigenschaften abhängt. Für die Alpha-Dispersion wird z. B. ein Frequenzbereich von 10 Hz bis zu einigen kHz angegeben. Man muss hier allerdings noch verschiedene Bereiche der Alpha-Dispersion unterscheiden, die durch die mechanische Auslenkung der polarisierten Moleküle in einem elektrischen Feld auftreten und insbesondere bei sehr niedrigen Frequenzen sichtbar werden. Je nach Struktur, Größe und Polarisation der Moleküle sind ihre Auslenkung und ihr Zeitverhalten unterschiedlich. Diese Effekte treten bis zu sehr kleinen Frequenzen, die kleiner als 1 Hz sein können, auf. So können bei sehr kleinen Frequenzen die Moleküle dem elektrischen Feld nachfolgen, so dass es zu einem charakteristischen Verlauf der Impedanz kommt. Die Erfindung stellt eine Anordnung zur elektrischen Charakterisierung von biologischem Material bereit, bestehend aus:
- a) Einer Probenkammer mit drei Paaren von Elektroden mit jeweils unterschiedlichen elektrischen Funktionen. Das erste Elektrodenpaar dient der Einspeisung und Ausleitung eines Wechselstromes durch das biologische Material (Arbeitselektroden). Mit dem zweiten Elektrodenpaar wird der Spannungsabfall abgeleitet, der beim Durchgang des Messstromes durch das biologische Material an diesem auftritt, und einem Auswertegerät zugeführt, das die Impedanz und die Phasenverschiebung, die an dem biologischen Material auftreten, bestimmt (Messelektroden). Mit dem dritten Elektrodenpaar wird ein elektrisches Feld an das biologische Material angelegt (Feldelektroden). Dieses elektrische Feld kann zeitlich konstant sein, aber auch zeitlich variabel. Dies gilt sowohl für die Stärke des elektrischen Feldes als auch für seine Richtung. - c) Aus einer gesamten Messanordnung, die so aufgebaut ist, dass an das erste Elektrodenpaar ein Pulsgenerator mit den beschriebenen Eigenschaften angeschlossen wird. An das dritte Elektrodenpaar wird ebenfalls ein Pulsgenerator mit den beschriebenen Eigenschaften angeschlossen. Beide Pulsgeneratoren sind über eine Kontrolleinrichtung an das Messsystem zur Bestimmung der Impedanz angeschlossen.
- d) Einer Ablaufsteuerung, die so aufgebaut ist, dass z. B. die Impedanzmessung mit einem bestimmten gepulsten Strom beginnt, dessen Frequenz in einem weiten Bereich variiert wird. Anschließend wird ein Puls mit unterschiedlicher Weite an das biologische Material angelegt und die zeitabhängige Variation der Impedanz gemessen, woraus sich z. B. die Relaxationszeit der biologischen Probe entnehmen lässt. Diese Relaxationszeiten sind spezifisch für die jeweiligen chemischen Verbindungen, aus denen die Probe aufgebaut ist. Diese Abhängigkeiten der Impedanz ergeben sich auch bei der Variation der anderen Pulsparameter, d. h. der Höhe der Pulsspannung oder des Pulsstromes, der Frequenz und des zeitlichen Verlaufs der ansteigenden und abfallenden Pulsflanke. Aufgrund der hohen Variabilität der verwendbaren Pulsparameter lässt sich nicht nur die Impedanz und die Phasenverschiebung bei einem gegebenen Wechselspannungs- oder Stromsignal bestimmen, sondern auch die Einschwingzeit, mit der die angeregten Moleküle einer schnellen Änderung der angelegten Spannung oder des angelegten Stromes folgen können. Durch die Feldelektroden, deren zeitabhängige Spannung in Abhängigkeit von der an die Arbeitselektroden angelegten Spannung oder Strom gesteuert wird, lässt sich unabhängig von dem Strom oder der Spannung an den Arbeitselektroden das elektrische Verhalten des biologischen Materials in definierter Form verändern, so dass ein zusätzlicher Parameter für die Charakterisierung des biologischen Materials zur Verfügung steht. Dies kann z. B. vorteilhaft verwendet werden, um zu verifizieren, ob durch die an die Arbeitselektroden angelegte Spannung oder Strom eine Bewegung der Moleküle erfolgt oder ob es sich um eine stationäre Rotation handelt. Weiterhin lässt sich durch Variation der Verzögerungszeit zwischen den Pulsen die Intensität der Bewegung und der Rotation ermitteln. Daraus lassen sich genaue Rückschlüsse auf die Struktur und Größe der Moleküle ziehen. Weiterhin können so durch die Pulsvariation an den Feldelektroden, durch die unterschiedliche Impedanzsignale erzeugt werden, verschiedene Substanzen voneinander unterschieden werden.
