Abstract

Die Erfindung betrifft ein auf einem Träger immobilisiertes Netzwerk zum Nachweis von Analyten sowie ein Verfahren zur Herstellung eines auf einem Träger immobilisierten Netzwerkes. Im analytischen Bereich erfolgt die Bindung von aktiven Biomolekülen an festen Materialien bzw. Oberflächen oft adsorptiv über elektrostatische Wechselwirkungen, Wasserstoffbrückenbindungen oder dispersive Kräfte und gilt als schwer kontrollierbar und zum Teil nur begrenzt stabil. Sowohl die große räumliche Nähe zur Oberfläche als auch der Adsorptionsprozess an sich kann dabei insbesondere die aktiven Stellen des Proteins negativ beeinflussen und somit die biologische Aktivität eines Biomoleküls wie z. B. eines Proteins herabsetzen. (
Alternativ erfolgt eine reaktive Immobilisierung durch kovalente Bindungsknüpfung zwischen Trägermaterial und Biomolekül. Dieser Weg verspricht eine größere Robustheit und Kontrollmöglichkeit. Gleichzeitig gilt auch hier, dass eine Beladung der Oberfläche durch den maximalen Bedeckungsgrad begrenzt ist und einer statistischen Verteilung der Proteinorientierungen auf der Oberfläche unterliegt. Da die Präsentation der aktiven Stellen bzw. der Bindungstaschen des Biomoleküls für die molekulare Erkennung wichtig ist und daher für die Assay-Performance oder auch für die Zelladhäsion entscheidend ist, sollten die jeweiligen Biomoleküle dabei idealerweise gerichtet, bzw. orientiert auf der Oberfläche gebunden sein. Die korrekte Orientierung der immobilisierten Biomoleküle auf der Oberfläche stellt daher stets eine Herausforderung dar. (
Für eine höhere Beladung mit Biomolekülen kommen 3-dimensional beschichtete Oberflächen zum Einsatz. Hierbei kann zwischen dem Einsatz von dendritischen Polymeren oder Gelen wie z. B. Polyacrylamid-Gelen unterschieden werden. Allerdings stellen hier die zu große Diffusionsbarriere und damit verbundene zu lange Inkubationszeiten bis das thermodynamische Gleichgewischt eintritt oftmals das größte Hindernis dar. Des Weiteren kommt es auch hier in der Regel zu einer kovalenten oder nicht-kovalenten Wechselwirkung zum Biomolekül um es im Gel zu halten und ein Hinausdiffundieren zu verhindern. (
Insbesondere bei der Immobilisierung von großen Biomolekülen auf Oberflächen mit funktionalisierten dendritischen Polymeren können außerdem nur wenige der funktionellen Gruppen für eine chemische Reaktion erreicht werden und verbleiben nicht abreagiert in der Matrix. So wurde z. B. im Fall der kovalenten Immobilisierung von Streptavidin an eine Polyacryl-säure-Oberfläche gezeigt, dass nur 0,1 % der Carboxylgruppen erreicht werden. Durch Einführung von Copolymeren konnte dieser Wert um den Faktor 5 gesteigert werden, was jedoch noch immer sehr wenig ist. (
Ein weiteres Problem stellt der eigentliche Prozess der Oberflächenbeschichtung bei vorausgegangener separater Herstellung eines Hydrogels dar: Mit dieser Technik sind zwar definierte Hydrogele herstellbar, das Aufbringen auf die Oberfläche des Trägers (z. B. mittels Spincoating) gestaltet sich aber oftmals schwierig, da Schichtdickenschwankungen kaum zu vermeiden sind. Zudem sind viele Geometrien und Formate wie z. B. Mikrotiterplatten so nur schwer zugänglich. Letztendlich kann die Haftung der Hydrogele durch kovalente Verankerung (grafting to) am Träger nur durch separate Aktivierung des Trägers selbst, beispielsweise durch eine Plasmabehandlung erfolgen.
