Abstract
Die vorliegende Erfindung betrifft eine pharmazeutische Zusammensetzung umfassend einen Träger und einen Überzug, dadurch gekennzeichnet, dass der Überzug wenigstens eine Lipase umfasst. Die Lipase ist bevorzugt Burlulipase. Sie betrifft ferner eine flüssige pharmazeutische Zusammensetzung, die durch Einbringen einer erfindungsgemäßen pharmazeutischen Zusammensetzung in eine Flüssigkeit hergestellt wird, wobei es sich um Lösungen in Wasser oder anderen Getränken handeln kann. Sie betrifft ferner Arzneimittel zur Vorbeugung und/oder Behandlung von Lipasemangelerkrankungen, die solche pharmazeutischen Zusammensetzungen oder flüssigen pharmazeutischen Zusammensetzungen enthalten oder daraus bestehen. Insbesondere betrifft sie solche Arzneimittel, die zur Vorbeugung und/oder Behandlung von Verdauungsproblemen, insbesondere exokriner Pankreasinsuffizienz geeignet sind. Ferner soll ein Verfahren bereitgestellt werden, das es erlaubt ein solches Arzneimittel herzustellen, ohne dass die Aktivität der eingesetzten Lipase während des Herstellungsprozesses stark abnimmt. Zusätzlich soll das Arzneimittel die Herstellung von flüssigen Zubereitungsformen der Lipase und insbesondere der Burlulipase ermöglichen, bei denen die Aktivität der Lipase gegenüber der Aktivität der ursprünglich zur Herstellung des Arzneimittels eingesetzten Lipase nur geringfügig oder gar nicht herabgesetzt ist. Das Arzneimittel soll die Verabreichung der Lipase in flüssigen Zubereitungsformen, Nahrungsmitteln und/oder als Bestandteil einer künstlichen Ernährung ermöglichen. Besonders bevorzugt handelt es sich um eine mikrobielle Lipase. Mikrobielle Lipasen lassen sich leicht großtechnisch herstellen, und es kann sichergestellt werden, dass sie keine für den Menschen schädlichen mikrobiellen Verunreinigungen enthalten. Ganz besonders bevorzugt ist die wenigstens eine Lipase eine bakterielle Lipase. Die Herstellung fester Zubereitungsformen der Burlulipase, wie etwa Tabletten, geht in der Regel mit einem großen Verlust der Aktivität der Burlulipase einher. Typischerweise verbleiben nur etwa 60 % der ursprünglichen Aktivität (gemessen in TBU Einheiten) der eingesetzten Burlulipase in den Tabletten und oft auch weniger als 60 %. Das Ausmaß des Aktivitätsverlustes hängt auch von den Verfahrensbedingungen ab. Gleichzeitig scheint Burlulipase empfindlich gegen einige der in Tabletten eingesetzten Hilfsstoffe zu sein. Der Aktivitätsverlust ist so groß, dass die Verwendung solcher Tabletten als Arzneimittel in der Regel nicht möglich ist.
