In the chemical industry, processes in which the two basic operations - reaction and separation - are integrated (so-called hybrid processes) have been used successfully for some time. Reactive rectification is an established process with many possible applications. With regard to the reaction taking place during reactive rectification, three types of process can be distinguished: reactive rectification with homogeneous catalysis, autocatalysis and heterogeneous catalysis. The implementation of the apparatus thus differs in the introduction of the catalyst into the column, with heterogeneous catalysis being the most widespread.
The first commercially available internals for fixing the catalyst were the catalyst-filled wire mesh casings listed in Table 1, which are also known as “tea bags”. The disadvantages of these internals are the poor transport properties, high pressure losses and slight column flooding. Mesh troughs and pipes have the disadvantage of a low separation capacity.
Eng. Science, 1999, 54 (10), S. 1413-1418). In Beers et.al. Another possibility is to coat the column internals themselves with a layer containing the catalyst. This has been demonstrated using zeolite catalysts as an example (Oudshoorn, O., Janissen, M., Van Kooten, W., Jansen, J., Van Bekkum, H., Van den Bleek, C., et al., Chem. Eng.Science, 1999, 54 (10), pp. 1413-1418). In Beers et al. (Applied Catalysis A: general 243, 2003, 237-250) a stainless steel packing is coated with a layer of silica by immersing the packing in a mixture of colloidal silica particles, water and additives. The zeolite layer is then applied. Column installation is proposed for use in chemical reactors. The application for reactive rectification is not described. A similar coating of column internals with catalysts other than zeolites is not known.
In all of the reactive rectification processes described above, only chemical catalysts are used; biocatalysts, on the other hand, were not used. However, the development of biocatalysts has made enormous progress in recent years.
Mitkowski et al. (Model-based reaction separation-process design. Improved properties in terms of both temperature stability and solvent resistance allow biocatalysts to be used in reactive rectification. However, there are very few examples of this. Paiva et al. (Journal of Molecular Catalysis B: Enzymatic, 1997, pp. 99-109; and Biotechnol. Prog. 2003, 19, pp. 750-754) report the successful transesterification of 1-butanol and ethyl butyrate to ethanol and butyl butyrate catalyzed with Lipozyme RM IM (commercially available Rhizomucor miehei lipase immobilized on an acidic ion exchange resin). Both processes were carried out in a laboratory facility, with the reaction and the separation taking place in a glass column containing a catalyst. No random or structured packings were used. Mitkowski et al. (Model-based reaction separation-process design. In V. Plesu, & P. Agachi (eds.), 2007, 17th European Symposium on computer-aided process engineering, pp. 395-400, Amsterdam: Elsevier) show in a theoretical case study how the esterification of oleic acid and cetyl alcohol catalyzed with Novozym 435 (commercially available Candida antarctica lipase B immobilized on acrylic resin) is designed by means of reactive rectification.
Schmidt-Traub et al. (Integrated Reaction and Separation Operations, Springer Verlag Berlin Heidelberg 2006, pp. 96 to 103) relates to statements on catalytic distillation and describes the use of catalytically active structures. Among other things, column internals in the form of structured packings for use in catalytic distillation are described. These packings can be coated with various catalysts, including enzymatic ones.
However, the implementation of a reactive rectification with immobilized biocatalysts in a column equipped with random or structured packing is not currently known in the literature.
- • The biocatalysts have sufficient catalytic activity for a sufficient process time.
- • The biocatalysts are fixed in the apparatus in such a way that there is no significant discharge during the process.
It is therefore an object of the present invention to provide a coated column installation that can be used in reactive rectification, the coating comprising a heterogeneous catalyst, in particular in the form of an inorganic catalyst and a biocatalyst, and the coating also being resistant at high temperatures and pressures, in particular under boiling conditions , is stable.

The biocatalyst is advantageously selected from the enzyme class EC1 to EC6, preferably from the classes EC1, EC3 and EC4 and particularly preferably from the enzyme class EC3 of the hydrolases, which in particular hydrolyzing enzymes such as lipases such as SP 523 lipase or lipase CAL B, esterases, amylases, pullulanases, glucosidases. Alternatively, it is also possible to use enzymes from the enzyme class EC1 of the oxidoreductases containing oxidizing and/or reducing enzymes, in particular oxidases such as peroxidases or laccases. The enzymes can either be of natural origin or in recombinant form.
The subject matter of the present invention is likewise a column internal which can be used in reactive rectification and which is coated with such a composition. According to the invention, a column installation such as a structured packing or a random packing for the rectification is covered directly with a gel layer containing the catalyst, in particular the biocatalyst. The gel can be applied by dipping, spraying, brushing, fluidizing, or other coating methods known to those skilled in the art. The catalyst is enclosed or immobilized in a gel matrix. It is advantageous that the catalysis can thus take place with the largest possible surface area while avoiding mass transport limitations. The column internals coated in this way are outstandingly suitable for use in a reactive rectification process.
Also advantageous are: (1) sufficient adhesion of the covering or coating to the column installation at relevant process temperatures, preferably for a period of at least 8 hours, and (2) sufficient activity of the catalyst immobilized in the coating layer under the reaction conditions, preferably above a period of at least 10 hours.

An aqueous hydroxide solution, in particular NH 4 OH, a suitable buffer system, in particular phosphate buffer (pH=5.8), and polyethylene glycol are then mixed to form a second solution. The concentration of the NH 4 OH solution can be varied, although the concentration of 25% by weight has proven particularly advantageous. At least one biocatalyst in the form of a solution or as a solid is added to this second solution. Both solutions are then combined and mixed. The concentrations of the individual components can be varied.
- a) preparing a liquid mixture comprising at least one composition according to any one of
claims 1 to 7, - b) coating of at least one column fixture, in particular by immersion in the mixture prepared in step a) for a period of 0.1 to 10 minutes, preferably 10 to 60 seconds,
- c) optionally repeating step b) for 1 to 20 times, preferably 3 to 5 times, and
- d) drying of the coated column internals for a period of 1 hour to 7 days, preferably 2 to 4 days.
The immersion of the column installation in step b) preferably takes place at the time when gelation of the mixture prepared in step a) begins. The gel coating on the column internals is completed by immersing the column internals in the resulting liquid mixture. All materials that ensure adequate adhesion of the coating, such as steel, ceramics, glass, plastics, etc., are suitable as packing or random packing material. The method can also be used for random packing.
Beispiel 5).When the solutions are combined, the viscosity of the resulting mixture increases after a few minutes as a result of the reaction. At this moment, the column structure is immersed in the mixture for a short time. In principle, immersion times of 0.1 to 10 minutes are suitable here. The immersion time between 10 and 60 seconds is particularly advantageous. The immersion is repeated several times until a comprehensive and uniform coating is obtained on the column installation or the gelation has progressed so far that it is no longer possible to achieve a uniform coating. The number of dips ranges from 1 to 20, although 3 to 5 times is particularly advantageous. Once the column internals have been coated, they are dried under ambient conditions for a period of 1 hour to 7 days (a time of 2-4 days is particularly advantageous) and can then be stored at various temperatures until further use. Temperatures between -10°C and 100°C are suitable, and a storage temperature of 4°C is particularly advantageous. The durability of the gel layer on the column structure is checked under separation conditions over a period of at least 8 hours (cf. Example 4). The activity of the catalyst is also measured (cf. Example 5).
The resulting coated column installation can then a reactive rectification, such as installed in a column and in a method of reactive rectification, in particular for Hydrolysis, esterification or transesterification and further enzymatic reactions with subsequent separation can be used. The separation preferably takes place under the boiling conditions (temperature, pressure) of the mixtures used. The reactive rectification can include both a batch and a counter-reactive rectification.
- - The enzymes are fixed in a gel layer directly on a column installation, so there is no discharge of the catalyst (enzyme) during the reactive rectification.
- - The gel adheres directly to a finished column internals, so that a subsequent application of a gel layer is possible and no changes in the manufacturing process of the column internals themselves are necessary.
- - The coated column internals can be introduced into the columns as usual, so no changes to the equipment are necessary.
- - The high porosity of the gel allows efficient transport of the products and educts of the reaction.
- - The effective catalyst surface is increased by the immobilization in the gel, thus increasing the efficiency of the catalyst.
- - The separating performance of the column is not significantly increased by the coating of the column internals.
Beispiel 3: Beschichtung der PackungExample 3: Coating of the packing

Zusammensetzung A:Composition A:
The coated pack or plate was kept in a boiling equimolar mixture of 1-butanol and ethyl butyrate at 70°C for 8 hours. The gel remained stably adhered to the plate. There was no visual difference from the initial state. Furthermore, no detached gel particles were observed in the solution. Thus, composition A is well suited as a coating for use in short chain organic solvents.
Zusammensetzung B:Composition B:
The coated panel was kept in boiling water at ambient pressure for 8 hours. The gel remained stably adhered to the plate. However, when coated plates were placed in 1-butanol, the gel film detached from the plate after just a few seconds. The gel itself was not decomposed. If longer-chain molecules (from C 6 groups) were used, the gel film hardly came off. Only small pieces were detached. If the molecules were also branched, the detachment was even lower. Composition B is thus suitable for use in aqueous systems and long chain organic solvents.
Beispiel 5: Nachweis der Aktivität des Katalysators in der BeschichtungExample 5: Detection of the activity of the catalyst in the coating
The transesterification reaction between 1-butanol and ethyl butyrate was used to determine the enzyme activity. Both starting materials were added in an equimolar ratio, stirred and heated to 60°C. A piece of column installation (10×5 cm) with the coating of composition A containing the enzyme lipase CAL B was then added. Samples were taken at defined time intervals. After each sampling, the sample was added to a defined amount of acetonitrile to stop the reaction and adjust the dilution ratio for gas chromatography. After adding the sample to the acetonitrile in a prepared 1.5 mL vessel, the mixture was briefly mixed on a vortexer and then centrifuged for one minute at 13200 rpm in order to separate any gel fragments or enzyme components that had been carried out. A portion of the supernatant was then analyzed by gas chromatography.
