zwischen den homologen Bereichen beider möglicher Rekombinationsorte,
wird nur die Interposonkassette mit dem Gm-Resistenzgen in das chromosomale
recA-Gen inseriert. Der Rest des Plasmides einschließlich
der Kanamycin-Resistenz geht jedoch verloren. Die rekombinanten
R. capsulatus-Stämme, die aufgrund des Antibiogramms als
putativ positive R. capsulatus B10S-T7 Stämme identifiziert
werden konnten, wurden anschließend eingehender charakterisiert.

8 Spuren 4, 6, 11 und 13;9 +I) in signifikanten Mengen, wohingegen die PKm-abhängige Expression von YFP, wie erwartet, konstitutiv und somit ohne die Zugabe eines Induktors erfolgt. - (iii) Durch Zugabe von 8 mM Fruktose kann eine vollständige Induktion der T7-Polymerase-abhängigen Expression des Reportergens erzielt werden (Daten nicht gezeigt). (iv) Zwei bis vier Stunden nach der Induktion wird in R. capsulatus der maximale Expressionslevel erreicht (Daten nicht gezeigt).

Zur Isolierung von Membranvesikeln wurden photoheterotrophen Litertestkulturen von R. capsulatus fraktioniert. Fraktion 2 wurde für die Saccharosedichtgradientenzentrifugation verwendet. Die eingesetzte Zellzahl entsprach ungefähr einer optischen Dichte von 1000 bei 660 nm pro Dichtegradient. Mit Hilfe einer Peristaltic Pump P-1 (Pharmacia Fine Chemicals) und eines 21A Multiphor II Gradientenmischers (LKB) wurden kontinuierliche Saccharosegradienten von 10 bis 60% Saccharose gegossen. Die Membranfraktion wurde anschließend auf den Gradienten aufgetragen und bei 175000 g und 4°C für 17 Stunden ultrazentrifugiert. Nach abgeschlossener Zentrifugation wurde der Dichtegradient in 1 ml Aliquots abgenommen. Anschließend wurde die Saccharosekonzentration in einem Refraktometer (Optech Abbe-Refraktometer) sowie die Proteinkonzentration nach Bradford analysiert. Ab einer gemessenen Absorption von 0,4 wurde die Probe adäquat verdünnt.
