Description
The invention relates to a method for cell analysis, wherein the cells to be analyzed are adhered to a carrier and stained by means of a first colorant and wherein a first digital image of the stained cells applied to the carrier is generated and stored.
These treatments include, in particular, the so-called "staining" of the cells, in which the cells are brought into contact with a chemical substance which, depending on their properties, triggers different chemical reactions in the cells, usually resulting in a change in the optically perceptible properties of the cells precipitates and changes the transmission, absorption and / or luminescence behavior of the cells. In the simplest case, the cells change their color depending on their properties, so that the cells can be classified according to their color into different categories, in particular normal / abnormal.
It should be emphasized at this point that in the following in this application, although always only a "staining" by means of a "colorant" is spoken, but that is to be understood as all types of treatments adhered to a carrier cells, the emission -, Transmission and / or absorption behavior of the cells with respect to electromagnetic waves, in particular with respect to electromagnetic waves with wavelengths in the visible range change. "Staining" in this sense can therefore also mean in particular that the cells not a chemical, but a physical treatment such. B. exposure to radiation or a heat treatment.

Depending on the type and nature of the cells to be analyzed, it may be that the staining of the cells with only a single colorant does not bring out different properties of the cells in the clarity necessary for the analysis, in particular for the reliable diagnosis of certain diseases.
However, there is often the problem that cells from one and the same sample develop differently on different carriers. Each analysis thus provides only information about the particular stained cells, while it would be desirable to gain more information about those cells already having information from an initial analysis.
Based on this, the object of the invention is to provide a method for cell analysis, which makes it possible in a particularly simple manner to obtain different, complementary statements about one and the same cells.
The object is achieved by a method of the type mentioned above with the features of claim 1. Thus, the invention is based on the surprising finding that even in routine operation already treated with a staining process cells can be subjected to further staining reactions, whereby new, important knowledge about the cells studied can be found.
It has been found that the method steps treating the same cells on the same carrier in particular with a colorant for the purpose of changing optically measurable properties of the cells and generating and storing a digital image of the applied on the carrier cells also several times each with different treatment methods of the cells, in particular with different Colorants can be repeated, because when using conventional glass or plastic carriers, the cells adhere very well to the carrier and survive without significant losses and several of their optically measurable properties changing treatments. Since the cell analysis methods of this invention typically deposit far more than 1000 cells on a support, it would not alter the significance of the analysis if some cells were to detach from the support in successive treatments.
The cells to be analyzed will usually be of human origin and z. B. smears, fine needle punctures or any body fluids originate. For the analysis of such cells, the following group of methods of staining has been particularly useful: May-Grünwald-Giemsa stain or Papanicolaou stain, Feulgen stain, silver stain, peroxidase stain, supravital stain.
In the case of human cells, it is particularly useful for tumor diagnosis to subject the cells applied to the support to first a May-Grünwald-Giemsa stain or Papanicolaou stain, then a Feulgen stain, then a silver stain in which, after each staining, a digital image of the cells is generated and stored.
The digital images can be further processed and evaluated in any known manner. In particular, they can be subjected to an automatic image analysis, it being advantageous for a plurality of digital images of the same cells to be combined after different treatments, in particular for different colors, and in particular to be superimposed. Classification algorithms can then be applied to the combined and / or superimposed images thus produced, which classify the cells according to predefinable criteria, count and, if appropriate, output results of the analysis after querying a preferably self-learning database.
The images taken after each different treatment of the cells images can be "matched" by software, so that when displaying different images of the same cells on an output device in each image displayed the same cells in the same relative position in the image shown, which in particular a human observer, the comparative analysis eases the pictures. However, since the cells practically do not change their position on the carrier, it is also possible to ensure, by aligning the carrier before producing different digital images of the same cells, that the same cells are each in the same relative position in the images.
Es zeigen:Further details and advantages of the invention will become apparent from the following purely exemplary and non-limiting description in conjunction with the drawings. Show it:
For example, an operator may view the carrier in an enlarged view on a screen and identify and mark certain regions (ROIs) that appear to be of particular significance and whose change is to be observed in the subsequent treatments. Depending on the type of cells to be examined and the expertise available in computerized form, such ROIs can, of course, also be defined automatically.
It will be understood by those skilled in the art that the images taken after the individual treatments of the cells can be combined and correlated in a variety of ways, both for automatic image analysis and for displaying the images or an image formed therefrom on an output device such as a screen or a printer.
In the context of the inventive concept, numerous modifications and developments are possible, the z. B. refer to the processing and evaluation of the digital images, it being emphasized at this point that the images are indeed generated directly in digitized form, for example by means of an array of CCD cells, that it is quite possible, first an analog Take a picture and then digitize it for automatic image analysis. The corresponding cell analysis devices can also be designed in a wide variety of forms and thus be adapted advantageously to the respective intended use and place of use. For example, small mobile devices are conceivable as well as facilities designed for the analysis of many thousands of samples per day for use in professional analysis laboratories. In any case, a basic idea according to the invention is to successively subject the cells applied to a carrier to different, optically measurable properties of the cell-changing treatments and to correlate the images of the cells obtained after each treatment.
