Agonist/Aktivator von NR2F6 zur Verwendung in der Behandlung einer Krankheit, welche mit einer erhöhten Immunreaktion verbunden ist.
Claims (19)
- Agonist/Aktivator von NR2F6 zur Verwendung in der Behandlung einer Krankheit, welche mit einer erhöhten Immunreaktion verbunden ist.
- Agonist/Aktivator nach Anspruch 1, wobei der Agonist/Aktivator von NR2F6 ausgewählt ist aus der Gruppe bestehend aus kleinen NR2F6-Bindemolekülen, NR2F6-Liganden, intrazellulären Bindepartnern, Aptameren und Intrameren.
- Agonist/Aktivator nach Anspruch 1 oder 2, wobei die Krankheit, welche mit einer erhöhten Immunreaktion verbunden ist, eine akute oder chronische Transplantatabstoßung, Hauterkrankung, T- und B-Zellen-vermittelte entzündliche Erkrankung, Transplantat-gegen-Wirt-Reaktion oder Autoimmunerkrankung ist.
- Agonist/Aktivator nach Anspruch 3, wobei die Hauterkrankung Schuppenflechte, atopische Dermatitis oder Kontaktallergie ist, wobei die T- und B-Zellen vermittelte entzündliche Erkrankung Asthma oder chronisch-obstruktive Lungenerkrankung (COPD) ist, oder wobei die Autoimmunerkrankung Multiple Sklerose, entzündliche Darmerkrankung, Lupus erythematodes und rheumatoide Arthritis ist.
- Pharmazeutische Zusammensetzung, umfassend den Agonist/Aktivator von NR2F6 nach einem der Ansprüche 1 bis 4 und einen pharmazeutischen Träger.
- Verfahren zur Identifizierung von Immunsuppressiva umfassend: (a) Inkontaktbringen einer Zelle, eines Gewebes oder eines nicht-menschlichen Tiers umfassend ein Reporterkonstrukt für NR2F6-Aktivierung mit einem Kandidatenmolekül; (b) Messen des Reportersignals; und (c) Auswählen eines Kandidatenmoleküls, welches das Reportersignal ändert.
- Verfahren nach Anspruch 6, wobei das Reporterkonstrukt für NR2F6-Aktivierung ausgewählt ist aus der Gruppe bestehend aus einem Ligandenvermittelten Reportergenexpressionskonstukt, einem Ligandenverdrängungskonstrukt, einem fluoreszierenden, zellulären Sensorfusionsmutantenkonstrukt und einem Liganden-induzierten homo- und/oder heterodimeren Konstrukt.
- Verfahren nach Anspruch 7, wobei das Liganden-vermittelte Reportergenexpressionskonstrukt einen NR2F6-Promotor-Reporter und nachfolgend exprimiertes NR2F6 umfasst, wobei eine Änderung in der Ligandenbindung an den NR2F6-Promotor-Reporter zu einer Änderung im Reportersignal führt, wobei das Ligandenverdrängungskonstrukt eine Ligandenbindedomäne von NR2F6 und einen nukleären Rezeptorliganden oder ein Co-Rezeptor-Protein umfasst, wobei die Verdrängung des nukleären Rezeptorliganden oder des Co-Rezeptor-Proteins von der Ligandenbindedomäne von NR2F6 zu einem Fluoreszenzpolarisations(FP)- oder Szintillationsnäherungs(SP)-Signal führt, wobei das fluoreszierende, zelluläre Sensorfusionsmutantenkonstrukt ein Reporterprotein umfasst, welches an eine Ligandenbindedomäne von NR2F6 fusioniert ist, wobei die Bindung eines Liganden an die Ligandenbindedomäne von NR2F6 zu einer Änderung im Reportersignal führt, oder wobei das Liganden-induzierte homo- und/oder heterodimere Konstrukt NR2F6 oder einen zur Dimerisierung befähigten Teil davon und einen Dimerisierungspartner von NR2F6 umfasst, wobei Dimerisierung von NR2F6 oder dem Teil davon und dem Dimerisierungspartner von NR2F6 zu einem Fluoreszenz-Energie-Resonanzenergietransfer(FRET)-Signal führt.
- Verfahren nach Anspruch 8, wobei der Dimerisierungspartner von NR2F6 ausgewählt ist aus der Gruppe bestehend aus PPAR, RXR, RAR, VDR, T3R, NF-AT, AP-1 und Nur77.
- Verfahren zur Beurteilung der Aktivität eines Kandidatenmoleküls, welches mutmaßlich ein Agonist von NR2F6 ist, umfassend die Schritte: (a) Inkontaktbringen einer Zelle, eines Gewebes oder eines nicht-menschlichen Tiers umfassend NR2F6 mit dem Kandidatenmolekül; (b) Messen einer Zunahme in der NR2F6-Aktivität oder einer Abnahme in der Aktivität der Komponenten einer NR2F6-abhängigen intrazellulären Signalkaskade; und (c) Auswählen eines Kandidatenmoleküls, welches die NR2F6-Aktivität steigert oder die Aktivität von Komponenten einer NR2F6-abhängigen intrazellulären Signalkaskade verringert; wobei eine Zunahme in der NR2F6-Aktivität oder eine Abnahme in der Aktivität von Komponenten der NR2F6-abhängigen intrazelluläre Signalkaskade auf die Fähigkeit der ausgewählten Moleküle hindeutet, eine erhöhte Immunreaktion zu unterdrücken.
- Verfahren nach Anspruch 10, wobei das Kandidatenmolekül, welches mutmaßliche ein Agonist von NR2F6 ist, ein Immunsuppressivum ist, das gemäß dem Verfahren nach den Ansprüchen 6 bis 9 identifiziert wird.
- Verfahren nach Anspruch 10 oder 11, wobei die Komponenten ausgewählt sind aus der Gruppe bestehend aus: NFAT, AP-1, IL2, IL17 und IFN-gamma.
- Verfahren nach einem der Ansprüche 6 bis 12, wobei die Zellen Jurkat T-Zellen sind.
- Verfahren nach einem der Ansprüche 6 bis 13, wobei das nicht-menschliche Tier ein transgenes nicht-menschliches Tier ist.
- Transgenes nicht-menschliches Tier wie in Anspruch 14 definiert oder Zellen oder Gewebe, die davon abgeleitet sind.
- Liganden-vermitteltes Reportergenexpressionskonstrukt, Ligandenverdrängungskonstrukt, oder fluoreszierendes, zelluläres Sensorfusionsmutantenkonstrukt wie in Anspruch 8 definiert und/oder Liganden-induziertes homo- und/oder heterodimeres Konstrukt wie in Anspruch 8 oder 9 definiert.
- Verwendung einer Zelle, eines Gewebes oder eines nicht-menschlichen Tiers wie in einem der Ansprüche 6 bis 15 definiert zum Screenen und/oder Validieren einer Verbindung, welche mutmaßlich ein Agonist von NR2F6 oder ein funktionelles Fragment davon ist.
- Kit, nützlich zum Ausführen des Verfahrens nach einem der Ansprüche 10 bis 14, umfassend Polynucleotide und/oder Antikörper, welche fähig sind, die Aktivität von NR2F6 nachzuweisen, oder fähig sind, eine Änderung in der Aktivität von Komponenten der NR2F6-abhängigen intrazellulären Signalkaskade wie in einem der Ansprüche 10 bis 14 definiert nachzuweisen.
- Kit nach Anspruch 18 gegebenenfalls eine Zelle wie in einem der Ansprüche 6 bis 15 definiert umfassend.
