Abstract
Meßvorrichtung und Verfahren zur quantitativen Bestimmung von Substraten von Biokatalysatoren. Anwendung ist in der Chemie, Biochemie, Pharmazie, Medizin und im Umweltschutz. Die rechnergestützte Meßvorrichtung besteht aus einem ein Biosensorsystem enthaltenden, kalibrierten Meßsystem, das über einen AD-Wandler mit einer ebenfalls über den Rechner gesteuerten Steuer- und Auswerteeinheit verbunden ist, einer Dateneingabeeinheit zum Dialog mit der Meßvorrichtung und einer Anzeigevorrichtung beziehungsweise einem Dokumentationsgerät zur Meßergebnisausgabe. Mit dem Verfahren werden mittels der Meßvorrichtung Konzentrationen von Substraten der Biokatalysatoren in Gegenwart von die Meßergebnisse verfälschenden Störsubstanzen in Meßproben unterschiedlichster Herkunft schnell und mit guter Präzision bei Wegfall zeitaufwendiger, manueller Arbeitsschritte bestimmt.Measuring device and method for the quantitative determination of substrates of biocatalysts. Analysenverfahren zur quantitativen Bestimmung chemischer Substanzen unter Verwendung von biologisch aktiven Verbindungen als Analysereagenz sind aus der modernen Analytik nicht mehr wegzudenken. Von herausragender Bedeutung sind dabei Enzyme und in neuerer Zeit auch höher integrierte Systeme, wie z. B. Mikroorganismen. Als Reagentien in löslichen Analysensystemen werden diese Biokatalysatoren bzw. Biokatalysatorsysteme schon seit vielen Jahren angewendet. Dabei zeigt sich aber, daß in vielen Fällen Enzyme und Mikroorganismen trotz ihrer unbestreitbaren Vorteile, wie hoher Spezifität, in löslichen Analysensystemen wegen ihrer nur einmaligen Verwendbarkeit viel zu teure Reagentien sind, um eine breite und umfangreiche Anwendung zu finden. Deshalb wird auch seit vielen Jahren nach Analysenmethoden und Vorrichtungen gesucht, bei denen die teuren Biokatalysatoren mehrfach eingesetzt werden können. In den Biosensorsystemen hat man mit Sicherheit Vorrichtungen entwickelt, bei denen diese Forderungen erfüllt sind („Biosensors: Fundamentals and Applications" Oxford Univ. Press, 1987, Ed. by A. P. F. Turner, I. Karube and G. S. Die als erhebliche Nachteile auftretenden Einflüsse von Störsubstanzen auf ein Meßergebnis treten aber nicht nur bei Biosensorsystemen auf, die auf dem potentiometrischen Meßprinzip beruhen. Man findet sie auch bei den weitverbreiteten und erhebliche Bedeutung besitzenden amperometrischen Biosensorsystemen. Diese Störeinflüsse auf das Meßergebnis bei amperometrischen Biosensorsystemen können bis zu einem gewissen Grade durch Immobilisierung zusätzlicher Anti-Interferenzschichten beseitigt werden, weil diese den Zutritt der Störsubstanzen bis zur die gewünschte biokatalytisnhe Reaktion katalysierenden Biokatalysatorschicht bzw. bis zum elektrochemischen Transduktor verhindern (R. Renneberg, F. Schubert, F. Scheller, TIBS, 1986,11,216-220). Diese Vorgehensweise hat aber auch Nachteile, da zum einen schwierig herstellbare Multienzymmembranen verwendet werden müssen und zum anderen die abschirmende Wirkung der Anti-Interferenzschichten nur für vergleichsweise geringe Konzentrationen der Störsubstanzen wirksam sind. Diese Ausschlußkonzentrationen liegen z. Die erfindungsgemäße Meßvorrichtung besteht t us einem kalibrierten, mittels eines programmierbaren Rechners) stems gesteuerten Meßsystem, das über einen Analog-L <gital-Wandler mit einer Steuer- und Auswerteeinheit verbunden ist, einer Dateneingabeeinheit zu seiner Bedienung sowie ei.iem Anzeigegerät und/oder einem Dokumentiergerät. Das Meßsystem'enthält als wesentlichen Bestandteil ein Biosensorsystem. Das Biosensorsystem ist in einer thermostatierbaren Durchflußmeßzelle angeordnet und über seine elektrischen Leitungen und über den Analog-Digital-Wandler mit der Steuer- und Auswerteeinheit verbunden. Die Durchflußmeßzelle ist so konstruiert, daß in sie für einen Meßvorgang über eine Meßzuflußleitung mit durch die Steuer- und Auswerteeinheit steuerbarem Ventil die Meßlösung mit Hilfe einer Dosierpumpe aus einem Vorratsbehälter dosiert wird. Die Meßlösung wird durch einen Rührer gleichmäßig durchmischt. Die Meßlösung ist auf eine Pufferkonzentration von 2mol/l bis 10~6mol/l mit einem pH-Wert in der Nähe des pH-Optimums des jeweils immobilisierten Biokatalysators eingestellt. Nach einem Meßvorgang wird die Meßlösung mit Hilfe einer Absaugvorrichtung über eine Meßzellenabflußleitung mit durch die Steuer- und Auswerteeinheit steuerbarem, geöffnetem Ventil entfernt. Das Biosensorsystem enthält erfindungsgemäß mindestens drei elektrochemische Sensoren. Dabei dient der Analog-Digital-Wandler der Umwandlung der anfallenden Signale in die durch das Rechnersystem nur verarbeitbaren digitalen Signale. Ein Sensor ist in jedem Falle ein mit mindestens einem Biokatalysator beschichteter, elektrochemischer Se· sor. Mit einem Biokatalysator beschichtet bedeutet, daß Enzyme, Mikroorganismen, tierische, pflanzliche oder humane Zellen, Zellorganellen und andere biokatalytisch aktive Materialien, z. B. mit diesen Biokatalysatoren modifizierte Antikörper, Antigene, Haptene, Hormone oder Gemische der genannten Biokatalysatoren, auf den sensitiven Oberflächen des Sensors immobilisiert sind. Sein Vorhandensein ist jedoch für das Funktionieren des Biosensorsystems eine unabdingbare Voraussetzung. Die elektrochemische Transduktoren für die Sensoren können unterschiedlichster Art sein, so daß verschiedenartigste, durch die biokatalytische Reaktion erzeugte, physikalische Meßgrößen durch die Meßvorrichtung quantitativ bestimmt werden können. Erfindungsgemäß werden für die Entwicklung der Meßsensoren des Biosensorsystems ionenselektive Sensoren, z. B. pH-Elektroden bzw. pH-sensitive, ionenselektive Feldeffekttransistoren oder Sauerstoffelektroden vom Typ der Clark-Elektroden bzw. wie diese gleiche Eigenschaften besitzende und mit Mediatoren modifizierte Elektroden oder Leitfähigkeitselektroden verwendet. Bei Biosensorsystemen zur Bestimmung von Biokatalysatorsubstraten wie Harnstoff, Penicillin, Kreatinin, Aminosäuren, Acylcholinen, Peptiden oder einigen Sacchariden sind auf den pH-sensitiven Oberflächen der ionenselektiven Sensoren oder Leilfähigkeitselektroden des ersten Sensors solche Biokatalysatoren immobilisiert, bei deren biokatalytischer Wirkung nach Zugabe obengenannter Biokatalysatorsubstrate zur Meßlösung pH-Änderungen oder Änderungen anderer lonenkonzentrationen auftreten. Solche Biokatalysatoren sind z. B. Enzyme wie Urease, Penicillinase, Penicillinacylase, Kreatininase, Deaminasen, Decarboxylasen, Proteasen und einige Kohlehydrate umsetzenden Enzyme, z. B. Glukoseoxidase, aber auch höher integrierte biokatalytisch aktive Systeme wie Mikroorganismen, Zellen unterschiedlicher Herkunft, Zellorganellen, Organschnitte, die diese Enzymaktivitäten besitzen und mit diesen genannten Biokatalysatoren modifizierte Antikörper, Antigene, Heptene, Hormone und andere biologisch aktive Verbindungen. Biosensorsysteme mit auf diese beschriebene Weise beschichtetem erstem Sensor besitzen im allgemeinen einen unbeschichteten zweiten Sensor und einen Referenzsensor, wie er aus der Elektrochemie für pH-Werte messende Sensorsysteme bekannt ist. In einer anderen Variante ist das Biosensorsystem der Meßvorrichtung so aufgebaut, daß bei Verwendung von Sauerstoffkonzentration oder Konzentrationen seiner Reduktionsprodukte messenden Elektroden bzw. gleiche Eigenschaften besitzenden und mit elektrisch leitenden Verbindungen, den sogenannten Mediatoren, modifizierten Elektroden als Sensoren auf der sensitiven Oberfläche des ersten Sensors Biokatalysatoren immobilisiert sind, bei deren biokatalytischer Wirkung nach Zugabe ihrer Substrate zur Meßlösung Sauerstoff verbraucht oder gebildet wird.
