Charakteristik des bekannten Standes der TechnikCharacteristic of the known state of the art
Zschr. gerichtl. Med., 1963,53,175-185; W.Klumpen, Diss. Marburg, 1967; A. Rambow, Diss. Mainz, 1978). Wiss., 1982,47,75-85; H.Wissel, W.Fregin, Krimin, for. Wiss., 1986,63/64,327).In addition to the microscopic detection of spermatozoa or of spermatozoa (H. Wisset, W. Fregin, Krimin, for. Wiss., 1986, 61 / 62,141-143), the immunological detection of sperm-specific antigens as a method of choice for the identification of sperm in Criminally relevant research material in the investigation and education of sexual offenses. For some time, attempts have been made to prepare anti-sperm sera which, on the one hand, react specifically with sperm antigens and, on the other hand, detect antigens present in semen also in dried, denatured material (H. Suyama, H. Savada, Gersch. , 1963, 53, 175-185, W. Klopump, Diss. Marburg, 1967, A. Rambow, Diss. Mainz, 1978). However, such polyclonal antisera give cross-reactions with other body materials, such as nasal secretions or saliva, which are due to the presence of ubiquitous proteins in human body fluids (H. Schmechta, B. Kopetz, H. Schlinger, Krimin, for. Wiss., 1982, 47, 75-85, H.Wissel, W.Fregin, Krimin, for .Wiss., 1986, 63/64, 337).
Sei., 1978,23,106-115). Im Kaninchen erzeugtes polyklonales Anti-P30 der Firma Seri (USA) ist in verschiedenen immunologischen Verfahren zum Nachweis von Sperma in kriminalistisch relevanten Spuren geeignet (J.Streeter, S.Sugarman, S.Swarner, J. For. Sei. Soc, 1983,23,324; N.A.Stubbings, PJ.Newall, J. For. Sei., 1983,30,604-614; H.Wissel, W. Fregin, Krimin, for. Wiss., 1989,73/74,157-160). Sei. Soc., 1989,29,233-241; A.Tsutsumi, Y. Yamamoto, H. Ishizu, Act. Crim. Japon., 1988, 54,239-246). An essential prerequisite for the generation of monospecific polyclonal antisera has been provided in the isolation of the sperm specific protein "P30", a prostatic gastric glycoprotein having a molecular weight of about 30000D (GFSensabaugh, J. For. Sei., 1978, 23, 106-115 Rabbit-generated polyclonal anti-P30 from Seri (USA) is suitable in various immunological methods for the detection of semen in criminally relevant lanes (J. Streeter, S.Sugarman, S.Swarner, J. For. 1983, 23, 344; NAStubbings, PJ Newall, J. For. Sci., 1983, 30, 604-614; H. Wissel, W. Fregin, Krimin, for. Wiss., 1989, 73/74, 157-160) Monoclonal antibodies against various sperm antigens have already been obtained and used in particular in tumor research. Also, their principal suitability could also be confirmed for cognitive investigations (L. Kamenev, M. Leclerq, Ch. Francois-Gerard, J. Fors. Sei. Soc. 1989, 29, 233-241; A. Tsut sumi, Y. Yamamoto, H. Ishizu, Act. Crim. Japon., 1988, 54, 239-246). Besides the problem of the antigen-antibody specificity known from the polyclonal system, additional effects must be considered when using monoclonal antibodies. These are due to epitope-paratope interactions, which may also cause a previously unknown "method specificity" and / or unexpected cross-reactivity, resulting in the need to use appropriate and precisely matched antibody combinations in the construction of enzyme immunoassays.
In the investigation of criminally relevant biological specimens further, the presence of endogenous peroxidase activities is generally to be expected, which requires the blockage of this activity when using peroxidase labeling for visualization. In individual cases, this blockade reaction may impair the binding of the first antibody to the target antigen (epitope). For the labeling of the secondary antibody, therefore, another enzyme is required. A commercially available test system for detecting sperm specific antigens in the molecular weight range around 3OkDa from kriminalistisch relevant biological traces using the generally accepted advantages of monoclonal antibodies for detection, such as high epitope specificity and unlimited availability as a prerequisite for a method standardization is not yet known.
