Abstract
Die Aufgabe der Erfindung besteht darin, den Verklonungsprozess von Chamomilla-recutita-Hochleistungspflanzen zu vereinfachen und oekonomischer zu gestalten. Erfindungsgemaess werden dazu die in vitro vermehrten Sprosse in feuchtes Perlit pikiert und unter anfaenglich hoher, spaeter verringerter und schwankender Luftfeuchtigkeit an die Bedingungen des Gewaechshauses adaptiert. Dabei kommt es gleichzeitig zur Bewurzelung, so dass nach 10 Tagen Pflanzen erhalten werden, die im Gewaechshaus weiter kultivierbar sind.The object of the invention is to simplify and economize on the cloning process of high yield Chamomilla recutita plants. Dio Erfindung ermöglicht die Rationalisierung der Züchtung neuer, besserer Kamillensorten, ausgehend von ausgelesenen Hochleistungspflanzen. Die nach Patentschrift DD 250949 durchgeführte Vermehrung von ausgelesenen Hochleistungspflanzen von Chamomilla recutita umfaßt die Stufen: Anlage von Sproßkulturen, Vermehrung von Sprossen, Bewurzelung der Sprosse und Adaptation der in vitro erzeugten Pflanzen an die Bedingungen des Gewächshauses. Dabei nehmen die Phasen der Bewurzelung und der Adaptation an die Beengungen des Gewächshauses eine relativ lange Zeit in Anspruch und binden Kultivationsraum, Arbeitskräfte und Chemikalien. Bei verschiedenen Verfahren zur In-vitro-Vermehrung von Pflanzen ist deshalb versucht worden, die Phase der Bewurzelung der in vitro erzeugten Sprosse und die Adaptationsphass gleichzeitig ablaufen zu lassen (vgl. George, E. F., Sherrington, P. D.: Plant Propagation by Tissue Culture. Exegetics Ltd. Eversley, Basingstoke, Hants, England, pp. 370-371, 1984; Evans, D A., Sharp, W. R., Ammirato, P. V., Yamada, Y.: Handbook of Plant Cell Culture. Macmillan Publishing Co., New York, p. 211,1983; Debergh, P. C, Maene, L.: Scientia Hort. 14,335-345 (1981)). Für Kamille lag die Anwendung dieses Verfahrens jedoch nicht nahe, da aufgrund der geringen Sproß- und Blattgröße und einer damit verbundenen niedrigen Überlebensfähigkeit Schwierigkeiten bei der Überführung nach Patentschrift DD 250949 auftraten. Weiterhin lagen auf diesem Gebiet negative Ergebnisse bei anderen Arzneipflanzen (Digitalis lanata. Rauwolfia serpentina, Foeniculum vulgäre) vor. Aus Vorversuchen zur Freilandadaptation mit unverletzten und bewurzelten Chamomilla-recutita-Sprossen unter Verwendung von Bechergläsern zum Luftabschluß und Erde bzw. Erde-Sand-Gemischen ging hervor, daß die Wurzelregeneration und die Adaptation stark von Infektionen beeinträchtigt werden. Die Gefahr der Kontamination von verletztem pfi jnzlichem Gewebe und dem daraus resultierenden Absterben der betroffenen Sprosse war weitaus höher als bei intakten Chamomilla-reculita-Pflanzen. Das Problem wird erfindungsgemäß dadurch gelöst, daß Bewurzelung und Überführung der Sprosse in Perlit und unter Foliezelten erfolg!. Durch eine hohe Bepflanzungsdichte (Abstand der Sprosse zueinander 2cm) in den Pikierkästen zur Ausbildung eines stabilen Mikroklimas konnten Erfolgsraten von nahezu 100% erreicht werden.The propagation of high performance plants of Chamomilla recutita, carried out according to patent DD 250949, comprises the steps of planting shoots, propagating shoots, rooting the shoots and adapting the in vitro produced plants to the conditions of the greenhouse. Zur Gewährleistung einer anfänglich hohen und konstanten Umgebungsfeuchtigkeit werden die Pikierkästen mit den Sprossen in Foliezeiten aufgestellt. Das Pflanzenmaterial wird in periodischen Abständen mit Wasser besprüht und beginnend am zweiten Tag zunehmend lange belüftet. Die Belüftungsdauer richtet sich dabei jeweils nach dem Zustand der Sprosse und sollte spätestens dann beendet werden, wenn die Blätter zu welken beginnen. Um den Akklimatisationsprozeß zu beschleunigen, wird nach etwa 5 Tagen mehrmals täglich belüftet. Innerhalb von 10 Tagen haben sich die Sprossen an die Umweltverhältnisse des Gewächshauses angepaßt, und das Foliezelt kann entfernt werden. Nach einmonatigem Wachstum bei 20 + 2°C und Zusatzbelichtung sind die entstandenen Pflänzchen-zur Überführung auf Erde und eine weitere Kultivation ohne Vorsichtsmaßregeln geeignet. Die Zufuhr von Mineralsalzen wird während der Phase der Adaptation und Bewurzelung durch wöchentliches Gießen mit einer 0,1%igen WopilR-Nährlösung gesichert.Single shoots with a minimum size of 1 crn from the propagation cycle are pikiert on hand moist perlite (distance of the shoots to each other about 2cm).
