Abstract
Die Erfindung betrifft ein Verfahren zur Herstellung von immobilisierten, in ihrer biologischen Aktivitaet stabilisierten und in ihren pharmakologischen Eigenschaften modifizierten Peptidderivaten, vorzugsweise fuer die Gewinnung von parenteral oder enteral anwendbarer Retardformen. Es wird ein Verfahren zur Herstellung von Praeparaten biologisch aktiver Peptide, vorzugsweise Peptidhormone durch Komplexbildung mit Polyaminosaeuren unterschiedlichen Strukturtypes und unterschiedlicher mittlerer Molmasse beschrieben, das infolge Bindung an die Polyaminosaeure zur verzoegerten Abgabe und Langzeitstabilisierung des Peptidwirkstoffes fuehrt und eine grosse Variationsbreite in den pharmakologischen Eigenschaften sowie Moeglichkeiten fuer eine Dosisverminderung zulaesst.The invention relates to a process for the preparation of immobilized, in their biological activity stabilized and modified in their pharmacological properties peptide derivatives, preferably for the recovery of parenterally or enterally applicable sustained release forms. Erfindungsgemäß wird die Aufgabe dadurch pelöst, daß als Zusatzstoffe Polyaminosäuren (zukünftig PAS genannt) eines geeigneten Strukturtypes und zweckentsprechender mittlerer Molmasse eingesetzt werden. Wie in den Beispielen näher ausgeführt ist, sind Polyaminosäuren nicht nur untereinander bzw. mit Proteinen,wie z. B. Insulin zur Komplexbildung befähigt, sondern zeigen diese Eigenschaft auch gegenüber wesentlich einfacheren peptidischen Strukturen oben genannten Types. Dieser Effekt war nicht ohne weiteres zu erwarten, da niedrigmolekulare Peptide im Vergleich zu Insulin in weit geringerem Umfang zu zwischenmolekularen Wechselwirkungen befähigt sind.According to the invention, the object is achieved by using polyamino acids (referred to in the following as PAS) of a suitable structure type and suitable average molecular weight as additives. Erfindungsgemäß werden Komplexe mit den genannten Eigenschaften dadurch gewonnen, daß beide Komponenten unter Bedingungen gelöst werden, unter denen das Peptidhormon nicht denatuiert, die PAS jedoch teilweise oder ganz konformativ entfaltet wird. Derart wird bei nachfolgender gemeinsamer Ausfällung unter genau definierten P Bdingungen eine stabile Komplex- bzw.
