Abstract
Die Erfindung betrifft einen Schnelltest zum Nachweis von H2O2-bildenden substratspezifischen Oxidasen, die als neue, spezifische Enzyme eine zunehmende Bedeutung bei der analytischen Erfassung von Substraten und Enzymen bei chemischen und biotechnologischen Prozessen, vor allem aber in der klinischen Diagnostik spielen. Erfindungsgemaess wird zu Kulturloesungen bzw. Zellextrakten von verschiedenen Mikroorganismen ein Gemisch, das ein Chromogen, Peroxidase und das spezielle Substrat der Oxidase enthaelt, zugesetzt. Aus ökonomischen Gründen erfolgt die Isolierung der Enzyme zunehmend und bevorzugt aus Mikroorganismen. Das Screening nach H]O]-bildenden Oxidasen gestaltet sich hierbei arbeitsaufwendig und langwierig. Durch ampe:ometrische Messung des Sauerstoffverbrauchs bzw. der H2O2-Bildung beschrieben KAMEI et al. (Chem.Pharm.Bull. 31,1307 (1963)) ein Screening nach einer L-Glutamatoxidase, bei dem etwa 500 Actinomycetales-Stamme untersucht wurden. Geleichfalls werden analytische Verfahren bei der Such« nach HjOj-bildenden Oxid« sen beschrieben, bei denen im quantitativen kolorimetrischen Test nach EMERSON (J. org. Chem. 8,41711943)) und TRINDER (Ann. Clin. Biochem. 6,24 (1969)) Oxidasen nochgewiesen werden können. Der Erfindung liegt die Aufgabe zugrunde, einen analytischen Schnelltest zu entwickeln, der es gestattet, HjOj-blldende Oxidasen, vorzugsweise mikrobielle, extra- bzw. intrazellulär lokalisierte, nachzuweisen. Erfindungsgemäß wird die Aufgabe dadurch gelöst, daß man durch einen Mikrotitertest zu Kulturlösungen bzw. Zellextrakten verschiedener Mikroorganismen ein Chromogengemisch gibt. Bei Verwendung von Mikrotiterplatten wird nach Inkubation der durch die Reaktion der Oxidase entstehende Farbstoff vi uell oder quantitativ mit Hilfe eines Auswertegerltes bestimmt. Das eingesetzte Chromogengemisch enthält Chromogene, Per« tidase, das spezielle Substrat der nachzuweisenden Oxidase und einen geeigneten Puffer. Der relativ einfache Mikrotitertest, vorwiegend für bakteriologische und immunologische Untersuchungen in der bakteriologischen und medizinischen Praxis und Forschung eingesetzt, gestattet allein auf einer Mikrotiterplatte die Analyse von 8 χ 12 Proben in 45 s. Der erfindungsgemlfte Test zum Nachweis von Oxidaten ist billig, schnell, effektiv und besonders für Serien- bzw. Screeninguntersuchungen geeignet.
