It can be used in the pharmaceutical industry to manufacture drugs for psychiatry and neurobiology.
Charakteristik des bekannten Standes der TechnikCharacteristic of the known state of the art
It is known that various higher species of fungi of the genera Psilocybe, Pluteus, Inocybe and Panaeolus form neurotropic indole alkaloids such as psilocybin and psilocin in the fruiting bodies.
A method (DE 1087321) describes the extraction of the active ingredients also from substrate mycelium and sclerotia, which have been grown by culturing mushroom species of the genera Psilocybe and Stropharia in surface cultures using malt extract solutions of 4 to 8% with addition of various mineral salts.
The disadvantage of the method is that using this natural, complex substrate only a content of.
Dry mycelia to psilocybin of 0.28% maximum results. Furthermore, the batch-to-batch composition of this substrate also makes it possible to detect strongly fluctuating amounts of indole alkaloids, so that often only traces of alkaloids are found in the mycelia.
In these methods must be fermented even at constant temperatures, which requires additional equipment expenses.

An addition of 0.2% agar allows a rapid spread of the fungus hyphae on the surface. The procedure is such that a fine suspension of mycelium flakes to the sterilized nutrient solution under aseptic conditions in suitable vessels such. B. penicillin or Fernbachkolben and incubate the solutions at temperatures between 16 and 32 0 C for 30 to 50 days. After filtering off, washing the mycelia with water, drying at 20 to 50 ° C, the mycelium is extracted by known methods and the active ingredients separated by means of chromatographic methods. By this procedure advantageously large yields of biomass can be obtained. The resulting mycelia have accumulated a high percentage of indole alkaloids, maintaining a constant fermentation temperature is not necessary. By the following examples, the invention will be explained in more detail.
One sets of basic diospores of the subtropical species Psilocybe cubensis (Earle) Sing a primary culture on 6% malt agar (agar: 1.5%). After 3 weeks aseptically a myceis piece of about 2 χ 2cm is removed and shaken in a 100ml sterile flask containing 20g saddle filling and 30ml of water for 10 minutes, the fillers crushing the mycelium to form a fine suspension of mycelium flakes.
Each 5 ml of the suspension is used to inoculate one liter of nutrient solution, which has the following composition and was prepared in 2.4-liter penicillin flasks.
