Als eine der beiden Basismethoden zur Festphasensynthese von Oligonucleotiden ist das sogenannte Phosphattriesterverfahren (S.A. Narang et al.. Methods Enzymol., Acad. Press 1980, 65,610) bekannt und weitgehend eingeführt. In diesem Verfahren werden als Träger, d.h. als Verbindungen, die in den verwendeten Lösungsmittelsystemen unlöslich sind und an denen die wachsende Oligonucleotidkette aufgebaut wird, wobei letztere durch einfache Filtration aus dem Reaktionsgemisch abgetrennt werden kann, bisher unter anderem substituierte Acrylamid-Copolymerisats (M. J. Gait, S. G. Popov et al., Nucleic Acids Rea., Symp. Ser. No7,1980,242) sowie vernetzte Polystyrene (K. Miyoshi, T. Miyake et al.. Nucleic Acids Res. 1980,8,5473) eingesetzt. Wird jedoch einer der Träger aus diesen Verbindungsklassen herangezogen, so ergeben sich für das Phosphattriesterverfahren Effektivitätsverluste, denn diese Träger weisen gegenüber den einzusetzenden verschiedenen Lösungsmitteln unterschiedliche Quellbarkeit sowie bei wachsender Oligonucleotidkette eine Änderung im chemischen Verhalten auf; außerdem kann bei ihnen die Zugänglichkeit der auf dem Träger befindlichen Ankergruppen noch nicht befriedigen. Weiterhin tritt beim Kettenaufbau in 3'-5'-Richtung Depurinisierung des Desoxyadenosins bei Abspaltung der 5'-Tritylschutzgruppe im sauren pH-Bereich ein. In Verbindung mit dem Bemühungen, diese Nachteile des Trägerverfahrens zu überwinden, ist weiterhin über die Herstellung und den Einsatz von auf Polytetrafluorethylen-Pulver (PTFE-Pulver) gepfropften funktionalisierten Polystyrenen als Träger in der Festphasensynthese von Oligonucleotiden berichtet worden (V. Durch das unvollständige Eindringen des Styrens in die Hohlräume bzw. Poren der Partikel des PTFE-Pulvers wird außerdem nur eine ungleichmäßige Oberflächen-Pfropfung «rreicht. Dies hatzurfolge, daß derGrad der Zugänglichkeit der Ankergruppen bzw. derwachsenden Oligonucleotidketten nicht genügend hoch sowie in Abhängigkeit von den Herstellungsbedingungen ungleichmäßig und nicht gut reproduzierbar ist. Beim Phosphattriesterverfahren unter Einsatz von nach o.g. Verfahren gepfropften Trägern resultieren damit in der Regel vergleichsweise niedrige Oligonucleotidausbeuten, wie Übersicht 1 zeigt.However, this method does not produce uniform, reproducible products, ie. In diesem Reaktor, der gleichzeitig der energiereichen ionisierenden Strahlung einer y^oder Elektronen-Bestrahlungsanlage ausgesetzt ist (Synchronbestrahlung), findet bei einer Einsatztemperatur von 400C bis 800C und bei einem (Rest-)Sauerstoffgehalt unter 200 ppm die Pfropfreaktion zwischen dem PTFE-PuIver und dem kontinuierlich zugeführten dampfförmigen Styren statt, wobei das Pulver zweckmäßigerweise durch Wirbeln oder Rütteln bewegt wird.
