Unter den Bedingungen des Beispiels 1 werden unter Verwendung des Stammes Fusarium oxysporum f. vasinfectum ATCC 22,5g 4-Androsten-3,17-dion in 24 Stunden Kontaktzeit zu-19,36g 15a-Hydroxy-4-androsten-3,17-dion vom Schmelzpunkt 197°C umgewandeltUnder the conditions of Example 1, using the strain Fusarium oxysporum f. vasinfectum ATCC 22.5g of 4-androstene-3,17-dione converted to 19.36g of 15a-hydroxy-4-androstene-3,17-dione of melting point 197 ° C in 24 hours of contact time
B. Beispiel zurVerwendung des 15a-Hydroxy-4-androsten-3,17-dions Example of using the 15a-hydroxy-4-androstene-3,17-dione
Es werden 53g rohes 15a-Benzoyloxy-4-androsten-3,17-dion als Öl erhalten.a) A solution of 40 g of 15a-hydroxy-4-androstene-3,17-dione in 200 ml of pyridine is cooled in an ice bath and then treated dropwise with 25.7 ml of benzoyl chloride. After stirring for 30 minutes with ice-bath cooling, 3.2 ml of water are added and the mixture is stirred for a further 30 minutes. The reaction solution is then stirred into ice-water, and the resulting precipitate is filtered off with suction and washed with water. In this case, the precipitate starts to become oily, it is therefore taken up in methylene chloride. The organic phase is washed with water, dried over sodium sulfate and evaporated. There are obtained 53 g of crude 15a-benzoyloxy-4-androstene-3,17-dione as an oil.
Man eluiert 22,7g 15,8,16/3-Methylen-4-androsten-3,17-dion. Eine aus Diisopropylether umkristallisierte Probe schmilzt bei 185-186,5°C.b) 101.5 g of trimethylsulfoxonium iodide are stirred vigorously in 11 dimethylsulfoxide with 17.88 g of NaOH for 1.5 hours at room temperature, the NaOH is then largely gone into solution. 53 g of T5a-benzoyloxy-4-androstene-3,17-dione are then dissolved in 200 ml of dimethyl sulfoxide under argon and water cooling, added to the reaction mixture over the course of 5 minutes, which is subsequently stirred for 1.5 hours. The reaction solution is then, with the addition of about 2 kg NaCl, stirred into 10 times the amount of ice water. The precipitated precipitate is washed with water and dried in vacuo at 5O 0 C. 31 g of the crude methylene compound 15β, 16/3-methylene-4-androstene-3,17-dione are obtained. The entire aqueous phase is extracted with diethyl ether, the ether phase is washed with saturated NaCl solution, dried over Na 2 SO 4 and evaporated. A further 6 g of crude methylene compound 15 / 3,16 / 3-methylene-4-androstene-3,17-dione are obtained. Precipitate and ether extract are combined and chromatographed on silica gel with an acetone-hexane gradient (201.0-25% acetone). 22.7 g of 15,8,16 / 3-methylene-4-androstene-3,17-dione are eluted. A sample recrystallized from diisopropyl ether melts at 185-186.5 ° C.
Then 34.1 ml of propargyl alcohol are added dropwise within 5 minutes. The reaction mixture is stirred for a further 15 minutes and stirred into 10 times the amount of ice water. It is neutralized with dilute sulfuric acid, any excess is removed with NaHCO 3 and saturated with sodium chloride. The precipitate is filtered off, washed with water and dried in vacuo at 6O 0 C. This gives 30.2 g, which are recrystallized from ethyl acetate with activated charcoal.
