Abstract
The invention relates to a method for the storage of microorganisms for the production of protein from C1 compounds, in particular methane-oxidizing bacteria. The invention belongs to the field of the microbiological industry. The invention aims to develop a process for the storage of microorganisms that does not require additional equipment, processing steps and chemicals in large quantities, whereby the microorganisms may not lose their activity during storage. It has been found that this is achieved by adding to the biomass suspension from the fermentor 3 to 10% by weight of methanol (based on biomass dry matter). Subsequently, the thus treated biomass suspension is stored in closed containers at 4 to 10C. The stored biomass suspension can be used directly for a new cultivation as a seed material.

Verfahren zur Lagerung von MikroorganismenMethod for storage of microorganisms
Die Erfindung gehört in das Gebiet der mikrobiologischen Industrie,.The invention belongs to the field of the microbiological industry.
The invention can be used especially for the storage of microorganisms for protein production from C. compounds, in particular for methane-oxidizing bacteria.
Die Kohlenwasserstoffe wirken quasi alsMethods for storage of microorganisms are known, in particular they are preserved by cooling. It is shown in the original SU 505 308 that the conventional cooling methods are disadvantageous in that large cooling capacities are required and the possible storage time is minimal. Therefore, it is suggested that various yeasts and bacteria spec, from cultivation with petroleum distillate, be concentrated to 5 to 20 % Dry matter and with a content of hydrocarbons - 10%, preferably 10 to 50%, be stored at 0 to + 25 0 G. The hydrocarbons act as a kind of
Konservierungsmittel. Microbiol. 6i_ (1970), 203 - 218). Der Nachteil des Verfahrens besteht in derPreservative. Methane oxidizing bacteria are known to have a process of regular inoculation (once every two weeks) of the microorganisms on solid and liquid nutrient media in a methane / air atmosphere (AN GRIGORJAN, J. Gen. Microbiol. 1970), 203 - 218). The disadvantage of the method consists in the
notwendigen häufigen Überimpfung und in der Aufrechterhaltung eines bestimmten Methan/Luft-Gemisches.necessary frequent inoculation and in the maintenance of a certain methane / air mixture.
Methanoxidiereηde Mikroorganismen werden auch aufbewahrt in Porm von Mikroorganismenassoziationen (Methylotrophe und Heterotrophe) auf festen Agarböden (Ju, R. MALASEMO, Y. A. RGMANOVSKAJA, Ju. A-. Methane oxidates of microorganisms are also conserved in the form of microorganism associations (methylotrophs and heterotrophs) on solid agar soils (Ju, R. MALASEMO, YA RGMANOVSKAJA, Ju. A.- ÜROCENKO, "Methane Oxidizing Microorganisms", Nauka Publishers, Moscow 1978). The difficulty of this method is based on the fact that for each methane-oxidizing bacterial strain individually a heterotrophic companion strain must be selected. According to another method, methane-oxidizing bacteria are stored in a liquid mineral medium under a methane / air atmosphere under pressure (copyright SU N 570 640). The disadvantage of this is that special containers for a pressure up to 200 atm are necessary. In another method, the methane oxidizing microorganisms in the form of a paste with a humidity of 70 to 80 % in a mixture with silica gel (weight ratio 1: 1 to 3: 1) must be kept (copyright SU N 759 018). The disadvantage of this method is the necessity of concentrating the biomass and producing a defined mixture with silica gel.
According to the copyright SU N 707 325, methane oxidizing microorganisms are kept in a concentrated solution of the nutrient salts. This method requires a great deal of work because a concentrated salt solution consisting of 6 to 15 salts must be prepared. In addition, according to this storage mode, longer storage phases were observed from 24 to 30 h as the culture grew *
Another possibility for the preservation of methane-oxidizing microorganisms consists in the addition of 3 to 5 wt .-% ammonium sulfate to biomass suspension (copyright SU N 938 616). This procedure requires.
Although an extension of the possible storage time and a shortening of the lag phase (compared with the above-mentioned use of concentrated-salt solutions), but it is relatively laborious because the microorganisms must be centrifuged or decanted and washed before re-cultivation.
The object of the invention is to develop a process for the storage of microorganisms which does not require any additional equipment, processing steps and chemicals in large quantities, whereby the microorganisms must not lose their activity during storage.
The invention is therefore based on the object of preserving the biomass suspension from the per- mentor in the original state by adding minimal amounts of a suitable substance and thereby to maintain the activity of the microorganisms during storage. A measure of the activity is the length of the lag phase on growth during post-storage KuI activation.
According to the invention, the object is achieved in that the biomass suspension from the per- mentor 3 to 10 wt .-% methanol (based on biomass dry matter) are added. Subsequently, the thus treated biomass suspension is stored in closed containers at + 4 to + 10 0 C. The stored bacterial suspension can be used directly for a new cultivation as a seed material.
In nachfolgenden Beispielen wird die Erfindung erläutert. In the following examples, the invention will be explained.
