Verfahren zur Herstellung der Restriktase H ind IIIProcess for the preparation of the restrictase H ind III
Die Erfindung betrifft ein Verfahren zur Gewinnung der Restriktase H ind III.The invention relates to a method for obtaining the restrictase H ind III.
Areas of application of the invention are microbiology and genetic engineering.
Microorganisms that produce the endonuclease H ind III are Haemophilus influenzae and Bordetella pertussis. The major difficulties in working with these strains are that they are pathogenic to humans, make special demands on the nutrient medium, form relatively small amounts of bacteria per nutrient unit, combine several endonucleases in one strain, thereby greatly improving the isolation and recovery of pure pergolas difficult.
The aim of the invention is to obtain the restrictase H ind III from a bacterial strain which selectively produces only this type of restrictase, is not pathogenic for humans and does not place any special demands on the culture media.
The inventive method for obtaining the Restriktase H ind III is characterized in that the bacterial strain Bordetella bronchiseptica W 543 (ZIMET 4404J ) is grown and killed in a conventional manner, the Restraktase H ind III is obtained from the bacterial mass according to a known method e
Haemophilus influenzae erreicht. The use of the strain ZIMET Ί4ό1ψ according to the invention allows to provide large amounts of bacteria for the recovery of pure restrictase H ind III · The strain ZIMET MOlf grows on low-cost Uährmedien under easy-to-handle breeding conditions and has a short YMehrehrungszeit · Since the strain ZIMET 4404+ only the Restrictase H ind III forms, complex purification processes are not necessary. This is a significant advantage over the previously used bacterial strains, such as Haemophilus influenzae achieved.

AlsThe strain Bordetella bronchiseptica ZIMET ήΊΟ ^ Ψ is characterized in that the bacteria are morphologically small gram-negative cocoa rods of 0.3 to 0.5 σ 0.8 to 2.0 / um, which have electronic microscopic peritrich flagella »Auf Bordet-Gengou Agar makes the trunk grayish-white smooth convex colonies with a smooth margin reaching 0.8 to 1.0 mm diameter after 24 hours. On charcoal casein agar, the stem forms grayish-yellowish, smooth-edged colonies that are easy to lift off and rub, and have 0.8 to 1.0 mm diameter after 24 hours. The stem does not form a pigment. On peptone agar "Difco" colonies of yellowish color, which reach a diameter of 1.5 to 2.0 mm after 24 h, can be easily lifted off and rubbed. In Cohen-Wheeler liquid agar, the strain grows without pigmentation under aerobic conditions by agitation in the shaker or by aeration in the fermentor. The bacterial suspension is greyish-white in color. The optimal growth temperature is 33 to 36 ° C. As

For this, lysozyme is 100 μg / ml, phenylmethylsulfonyl fluoride 2.5 μg / μg / ml and 1 mM. Ua-azide added · The mixture is crushed by ultrasound. The cell debris are centrifuged at 100,000 rpm for 60 min at +2 0 C. The supernatant is brought to about 0.1 M HaCl solution and placed on a column of phosphocellulose (P II-Whatman). The column is previously washed with a 2 to 3 times column volume of the above buffer to which a 0.1 M UaCl solution has been added. The elution is carried out in a linear gradient of 0.1 to 1.0 M NaCl solution in above buffer and is collected in fractions. Equal amounts of a 3 nil of each fraction are added 1 h. at 37 ° C. with 1 μg of phage lambda DNA in the buffer containing 50 mM Iris HCl-pH 8.0, 10 mM MgCl 2 , 20 mM UaCl, and then analyzed electrophoretically in an agarose gel. The ferment can be eluted between 0.2 and 0.3 M NaCl solution. Fractions of specific activity are pooled, dialyzed against the above buffer, and passed through a Mono-Q column of FPLC (Pharmacia). The substance having specific activity free of nonspecific nucleases passes through the column and is concentrated by dialysis against the above buffer containing 50 % glycerol and 50 mM NaCl. The resulting concentrated ferment, referred to as oil, has a specific activity of 1000 U / ml. 1 unit corresponds to the activity of the minimum amount of enzyme needed to produce 1 / ug phage DUS within

Bestimmung des gewonnenen Ferments als Restriktase H ind III.Determination of the recovered enzyme as a restrictase H ind III.
Rosenvold, A, Honigmann, W. Szybalski: Gene 10 (1980) 357-370).To determine the oil recognizable by the enzyme B sequence of the nucleotide at the H site n is the hydrolysis of various DNA was performed. The DHS of the phages lambda and <j) X 174 El, the virus SY 40 and the plasmid pBR 322 were investigated. The electrophoresis of the hydrolyzed DUS showed that the B fermentation oil acts at the point AACCTT and thus has the specific effect of the H ind III restriction enzyme (Fuchs, C, E, C. Rosenvold, A, Honigmann, W. Szybalski: Gene 10 (FIG. 1980) 357-370).
