Mozkový pentapeptid enkefalin byl izolován v roce 1975 (Hughes I. et.al., Nátuře 258, 577, 1975). V rámci stabilizace struktury peptidu byly připraveny analogy s řadou obměn zabraňujících např. znepřístupněním peptidových vazeb enzymovému štěpení insercí stéricky namáhané aminokyseliny (Pospíšek J. et al., AO 270617). Dalargin a jeho stabilisované analogy se staly také předmětem praktického zájmu např. v odchovu rybího plůdku jako antistresové působky (Kouřil J. et al., AO 272150 a patent č. 276 916. Analogy obecného vzorce I, se připraví tak že se peptidy synthetisují postupnou výstavbou řetězce na p-methylbenzhydroaminové pryskyřici z Boc - aminokyselin. Finální produkt se z pryskyřice odštěpí kyselinou trifluoroctovou. Surový produkt se přečistí RP HPLC. Boc-N-methyl-D-fenylalaninuBoc-N-methyl-D-phenylalanine
Do směsi 50,4 ml tetrahydrofuranu a 5,6 ml dimethylformamidu bylo přidáno 5,3 g Boc-D-fenylalaninu. Za míchání bylo přidáno 2,88 g olejové suspense hydridu sodného a po 10 minutách 10 ml methyljodidu. Celá reakční směs byla zahřívána na 80 C po dobu 24 hodin. Poté byla provedena hydrolysa a produkt vytřepán do diethyletheru. Etherický roztok byl vysušen bezvodým síranem horečnatým a po odfiltrování sušidla byl ether vakuově oddestilován. Ke zbytku bylo přidáno 20 ml dioxanu a 20 ml 2M NaOH. Reakční směs byla míchána 2 hodiny při teplotě místnosti, pak okyselena kyselinou citrónovou a produkt byl vytřepán do ethylacetátu. Ethylacetátový produkt byl vysušen bezvodým síranem hořečnatým a po odfiltrování sušidla bylo rozpouštědlo oddestilováno. Zbylý olej byl rozpuštěn v etheru a produkt vysrážen ve formě soli pomocí dicyklohexylaminu.To a mixture of 50.4 mL of tetrahydrofuran and 5.6 mL of dimethylformamide was added 5.3 g of Boc-D-phenylalanine. 2.88 g of an oil suspension of sodium hydride were added with stirring and after 10 minutes 10 ml of methyl iodide. Do směsi 75,6 ml tetrahydrofuranu a 8,4 ml dimethylformamidu bylo přidáno 8,0 g Boc-D-fenylalaninu. Za míchání bylo přidáno 4,32 g olejové suspense hydridu sodného a po 10 minutách 15 ml methyljodidu. Celá reakční směs byla zahřívána na 80 °C po dobu 24 hodin. Poté byla provedena hydrolysa a produkt vytřepán do diethyletheru. Etherický roztok byl vysušen bezvodým síranem hořečnatým a po odfiltrování sušidla byl ether vakuově oddestilován. Ke zbytku bylo přidáno 30 ml dioxanu a 30 ml 2M NaOH. Reakční směs byla míchána 2 hodiny při teplotě místnosti, pak okyselena kyselinou citrónovou a produkt byl vytřepán do ethylacetátu. Ethylacetátový produkt byl vysušen bezvodým síranem hořečnatým a po odfiltrování sušidla bylo rozpouštědlo oddestilováno. Aminokyseliny jednotlivých aminokyselin byly chráněny Boc-skupinou a quanidinová skupina argininu skupinou tosylovou a hydroxylová skupina tyrosinu skupinou 2,6-dichlorbenzylovou. Při synthese bylo použito trojnásobného přebytku jednotlivých aminokyselin. Z pryskyřice byl peptid zároveň s chránícími skupinami odštěpen bezvodým HF za přítomnosti anisolu. Po oddestilování HF byla reakční směs rozmíchána v 50 % kyselině octové, pryskyřice byla odfiltrována, promyta vodou a spojené filtráty byly lyofilisovány.
