Abstract

A method of wet deposition of a microbial culture to a solid surface intended for the taxonomic identification of cultures by means of desorption-ionization techniques of mass spectrometry characterized by the fact that a deposition medium containing water or a mixture of water and a water-miscible polar solvent, and further containing a lysing agent selected from the group consisting of oxalic acid, formic acid, acetic acid, is applied to the solid surface of the solid substrate intended for desorption-ionization techniques of mass spectrometry, wherein the concentration of the lysing agent in the deposition medium is in the range of 30 to 70% by weight, after which a microbial culture is introduced into the medium and resuspended therein, and the resulting suspension is allowed to dry to form a homogeneous film.
