ESTO SE LOGRA TRANSFORMANDO LA LEVADURA DEL GENERO PICHIA PASTORIS MUTANTE HIS- CON UN PLASMIDO PREPARADO POR INGENIERIA GENETICA PARA ESTE FIN, EN EL CUAL SE ENCUENTRA EL GEN ALFA AMILASA DE BACILLUS LICHENIFORMIS, ACOPLADO A LA SECUENCIA SEÑAL DEL GEN SACAROSA INVERTASA Y REGULADO POR EL PROMOTOR DEL GEN ALCOHOL OXIDASA DE LA LEVADURA. ESTA LEVADURA TRANSFORMADA, ES CAPAZ DE PRODUCIR ALTOS NIVELES DE ALTA AMILASA BAJO DETERMINADAS CONDICIONES EN U N PROCESO DE FERMENTACION-INDUCCION. THE TECHNICAL OBJECTIVE THAT IS PURSUED IS THE CONSTRUCTION OF A RECOMBINANT YEAST STRAIN CAPABLE OF PRODUCING THE THERMOSTABLE ALPHA ALPHAMYLASE ENZYME, WHICH PRESENTS A VERY EFFICIENT SYSTEM FOR REGULATION OF THE DESIRED GENE PRODUCT IN LA SCIENTIST EXCRETARIUM. OF CULTIVATION. THIS IS ACHIEVED BY TRANSFORMING THE YEAST OF THE GENUS PICHIA PASTORIS MUTANTE HIS- WITH A PLASMID PREPARED BY GENETIC ENGINEERING FOR THIS PURPOSE, IN WHICH THE ALPHA AMYLASE GENE OF BACILLUS REGULAR SEA SIGNAL IS FOUND, LINKED TO THE SIGNAL SEQUAL REGULATION PROMOTER OF THE GENE ALCOHOL OXIDASE OF YEAST.
