Abstract
ESTA RELACIONADA CON LA BIOTECNOLOGIA, ESPECIALMENTE CON LA RAMA DE LA INGENIERIA GENETICA Y ESTA ENCAMINADA A LA OBTENCION DE UNA NUEVA CEPA DE LEVADURA SUPERPRODUCTORA DE QUIMOSINA POR VIA RECOMBINANTE. EL OBJETIVO TECNICO ES LOGRAR QUE A PARTIR DE UNA LEVADURA MANIPULADA GENETICAMENTE, SE OBTENGA DICHA ENZIMA EN EL MEDIO DE CULTIVO Y EN PROPORCIONES ELEVADAS POR VIA FERMENTATIVA. CON EL EMPLEO DE TECNICAS CONVENCIONALES DE INGENIERIA GENETICA, SE INTRODUJO EL GEN DE QUIMOSINA BOVINA BAJO EL PROMOTOR DE LA ALCOHOL OXIDASA 1 DE PICHIA PASTORIS Y LA SEÑAL DE SECRECION DE LA SUCROSA INVERTASA, Y SEGUIDO DE LA SECUENCIA TERMINADORA DE LA TRANSCRIPCION DE LA GLICERALDEHIDO 3 FOSFATO DESHIDROGENASA, AMBOS DE SACHAROIMYCES CEREVISIAE, CLONAJE REALIZADO EN UN VECTOR DE INTEGRACION LOGRANDOSE ASI A PARTIR DE UN MUTANTE HIS-3 DE LA LEVADURA METILOTROFA PICHIA PASTORIS UNA NUEVA CEPA CARACTERIZADA POR LA PRODUCCION DE ESTA ENZIMA PROTEOLITICA, PROPIA DEL TRACTO DIGESTIVO DE TERNEROS. LA IN VENCION ES APLICABLE EN LA OBTENCION DE UNA ENZIMA DE GRAN UTILIDAD EN LA INDUSTRIA LACTEA PARA LA ELABORACION DE QUESOS.\!IT IS RELATED TO BIOTECHNOLOGY, ESPECIALLY WITH THE BRANCH OF GENETIC ENGINEERING AND IS ON THE WAY TO OBTAIN A NEW SUPER-PRODUCTION YEAST STRAIN OF CHEMOSINE BY RECOMBINANT VIA. THE TECHNICAL OBJECTIVE IS TO ACHIEVE THAT, FROM A GENETICALLY MANIPULATED YEAST, SUCH ENZYME IS OBTAINED IN THE CULTIVATION MEDIUM AND IN PROPORTIONS ELEVATED BY FERMENTATIVE WAY. WITH THE USE OF CONVENTIONAL TECHNIQUES OF GENETIC ENGINEERING, THE GENE OF BOVINE CHEMOSINE WAS INTRODUCED UNDER THE PROMOTER OF ALCOHOL OXIDASE 1 FROM PICHIA PASTORIS AND THE SECURITY OF THE SUCROSA INVERTASA SECURITY, AND FOLLOWED BY THE SECURITY LAKE SEQUENCE. 3 PHOSPHATE DEHYDROGENASE, BOTH OF SACHAROIMYCES CEREVISIAE, CLONING MADE IN A VECTOR OF INTEGRATION, ACHIEVING THROUGH A MUTANT HIS-3 OF THE METILOTROFA PASTORI PASTORI DEVIATION BY LA PROOTA . THE INVENTION IS APPLICABLE IN THE OBTAINING OF A HIGHLY USEFUL ENZYME IN THE DAIRY INDUSTRY FOR THE PRODUCTION OF CHEESES. \!
