Description

Vynález sa týká spdsobu izolácie povrchového antigénu 987-P (pilusového) bakteriíThe invention relates to a method for isolating a 987-P (pilus) bacterial surface antigen

Escherichia coli, which is used either alone to immunize sows or calf cows or as a supplement to other vaccines against inflammatory bowel disease and diarrhea.

Colinitis and constipation in newborn calves and pigs are caused by enteropathogenic bacteria Escherichia coli, which adhere to the intestinal wall of affected animals and then, by secreting enterotoxins, cause intestinal inflammation and diarrhea, as a result of which newborns often die. Overcoming this intestinal inflammation is a serious problem. The most successful are the immunization of animals with an adherence antigen, which prevents bacteria from adhering to the intestinal wall and thus applying their toxins in the necessary amounts, thus causing disease. So far, the most effective immunization of mothers before delivery has been parenteral, with a cellular or subcellular vaccine containing either whole, gently killed enteropathogenic bacteria carrying the appropriate antigen on their surface, or an isolated pilus antigen or a combination of both.

V poslednom čase sa stává aktuálnou otázka výroby vakcíny z povrchového antigénu 987-P. Ooteraz známe postupy izolácie tohoto povrchového pilusového antigénu E.coli majú komplikovaný postup a velké prácnosť. Při izolácii sa používá až devať operácií, z ktorých možno spemenéť centrifugáciu, ultracentrifugáciu, ionexovú chromatografiu, zrážanie, ultrafiltráciu, gélovú chromatografiu (Nagy, L.K., Me Kenzie, T., Painter, K.R.: Veterinary record (1985), 117, 408-413), pričom výťažok je relativné nízký.Recently, the production of a vaccine from surface antigen 987-P has become a topical issue. The methods known so far for isolating this E. coli surface pilus antigen have a complicated procedure and a great deal of work. Up to nine operations are used in isolation, including centrifugation, ultracentrifugation, ion exchange chromatography, precipitation, ultrafiltration, gel permeation chromatography (Nagy, LK, Me Kenzie, T., Painter, KR: Veterinary record (1985), 117, 408- 413), the yield being relatively low.

Uvedené nedostatky v podstatnej miere odstraňuje spůsob izolácie povrchového pilusového antigénu 987-P z baktérií Escherichia coli podlá vynálezu, ktorého podstata spočívá v tom, že sa pilusový antigén 987-P z povrchu těla enteropatogénnych baktérii E.coli separuje homogenizáciou a izoluje autofokusáciou do takej koncentrácie, že vykazuje jasné precipitačnú liniu pri difúzii v agare voči antiséru anti 987-P až do riedenia 1 : 128.The method of isolating the 987-P surface pilus antigen from Escherichia coli according to the invention substantially eliminates these drawbacks by separating the 987-P pilus antigen from the body surface of enteropathogenic E. coli bacteria by homogenization and isolating them by autofocusing to such a concentration. that it shows a clear precipitation line when diffused in agar against anti-987-P antiserum up to a dilution of 1: 128.

Výhodou izolácie povrchového pilusového antigénu podlá vynálezu sú nízké výrobně náklady, jednoduchá manipulácia pri imunizácii zvierat, jednoduchost’ vlastného spósobu izolácie antigénu, ktorý vylučuje trvalé obsluhu aparatury a potřebu akýchkoťvek chemikálií, protože scparácia prebieha len v apyrogénnej destilovanej vodo. The advantages of isolating the surface pilus antigen according to the invention are low production costs, easy handling in immunizing animals, simplicity of the antigen isolation method itself, which eliminates permanent operation of the apparatus and the need for any chemicals, as the separation takes place only in pyrogen-free distilled water.

Uvedený příklad ilustruje, ale neobmedzuje predmet vynálezu. The above example illustrates but does not limit the scope of the invention.

Jeho pil sa upraví na hodnotu 7 a koncentrácie sa upraví tak, že vykazuje pri sledovaní prucipitácie v agare jasné precipitačnú liniu voči antiséru anti 987-P až do riedenia 1 : 128.Enteropathogenic bacteria Escherichia coli 987-P, carrying on their surface the pilus antigen 987-P, r> n after normal cultivation, guaranteeing the production of said antigen, concentrates and the bacterium without supernatant is resuspended in sterile distilled water cooled to 4 ° C by homogenizer for 5 to 10 minutes at 3,000 rpm and warm to 4 ° C. This separates the piluses containing the protein component of the 987-P antigen. The homogenate is then centrifuged at 70,000 ms -2 for 20 minutes at a temperature up to 4 ° C. The supernatant, the conductivity of which is adjusted to values up to 800 .mu.l / cm -1 by the addition of distilled water, is applied to an autofocus apparatus and, after closing, a maximum direct current of 3V is applied and the solution is focused at a temperature of up to 4 DEG C. until to one tenth of its original value at a maximum voltage of 1,000 V. After focusing, the pH and antigen content of the 987-P antigen are measured in all working chambers with a titer. Pilus antigen 987-P focuses automatically to pH 4.2 to 4.8. Pilus antigen 987-P is collected from compartments with a pH in this range. Its pH was adjusted to 7 and the concentration was adjusted to show a clear precipitation line against anti-987-P antiserum up to a dilution of 1: 128 when monitoring prucipitation in agar.

After reaching this concentration, an oil adjuvant is added to the solution and administered parenterally to sows about 6 weeks before farrowing. This time is sufficient for said dose to produce antibodies to the 987-P antigen in the mothers, and after drinking the colostrum, these are transferred to the intestines of the newborn. These antibodies then prevent the enteropathogenic bacteria E. coli 987-P from adhering to the intestinal wall and thus causing colienteritis and echinoderms.
