Vynález se týká nového produkčního kmene Corynebacterium glutamicum CCM 4023, který produkuje L-lysi'rf,.írta syntetických a semisyntetlckých fermentačních médiích. Kultivace probíhá v tekutých živných médiích, které obsahují jako hlavní zdroj uhlíku sachorózu nebo jiné sacharidy, jako zdroj dusíků amonné soli, kukuřičný extrakt, vitamíny, např. biotin a minerální soli. Fermentační média neobsahují komplexní zdroje dusíku, jako jsou hydrolyzáty bílkovinných substrátů, např. arašídové mouky apod.The present invention relates to a novel production strain of Corynebacterium glutamicum CCM 4023, which produces L-lysine, a variety of synthetic and semi-synthetic fermentation media. Výhodou výše uvedeného produkčního kmene je dále vysoká konverze uhlíkatého zdroje, která se pohybuje mezi 33 až 35 %. Produkční kmen Corynebacterium glutamicum CCM 4023 produkuje L-lysin na jednoduchých fermentačních médiích bez komplexních zdrojů dusíku; tím se zlevňuje a ekonomizuje fermentační proces přípravy L-lysinu. Nový kmen byl získán mutagenezí a selekcí z původního kmene Corynebacterium glutamicum ATCC 13 032 a vyznačuje se rezistencí na analogy aminokyselin, některá antibiotika a analogy biologicky aktivních látek.Further, the advantage of the above production strain is the high conversion of the carbon source, which is between 33 and 35%. Kmen optimálně roste za aerobních podmínek při pH 7,0 a teplotě 30 °C. Baňky se dále kultivují na rotační třepačce (frekvence 220 ot. min. ) při teplotě 28 až 30 °C po dobu 18 až 24 hodin. Vyrostlou kulturou se v množství 10 % obj. zaočkují 500ml varné baňky, které obsahují 20 ml fermentačního média o tomto složení: sacharóza těch. 100 g, biotin 50 pg/1, sírám amonný kryst. 25 g, hydrogenfosforečnan draselný 1 g, síran hořečnatý kryst. 0,1 g, uhličitan vápenatý mletý 35 g, voda dest. ad 1 000 ml; pH média 6,8 až 7,0; kultivace probíhá za aerobních podmínek na rotační třepačce při teplotě 28 až 30 °C a pH média se udržuje v rozmezí 6,8 až 7,0 pomocí roztoku 10 % obj. hydroxidu amonného.
