Vynález se týká nového mikroorganizmu Trichoderma reesei M 6 CCF 1856, ktorý produku-je vo zvýSenom výtažku extracelulárny celulolytický enzymový komplex.
Imperfektná mikroskopická huba Trichoderma reesei produkuje pri povrchovej a submerznejkultivácii na vhodnom lignocelulózovom substráte celulózový enzýmový komplex schopný hydro-lyzovať natívnu celulózu, to znamená zmes polysacharidov typu celulóz a hemicelulóz na mono-a oligosacharidy. NakoTko kmene nachádzajúce sa v prírode produkujú len veTmi nízké hladinycelulóz, přikročilo sa v niektorých laboratóriach vo svete k mutagenéze prírodných kmeňovza úřelom zvýgenia ich schopnosti produkovat celulazu /Mandels M. , Weber <J., Parizek R. :

Aulanko, Finland, (1975)/ Takto sa z prírodného kmeňa Tricho-derma viride QM 6a podařilo připravit kmen QM 9414, pomenovaný Trichoderma reesei, ktorývykazoval v priemere átvornásobne vygšiu produkciu celulolytických enzýmov ako materský kmen.Při submerznej kultivácii kmeňa QM 9414 na mikrokryštalickej celulóze sa dosahuje výťažokokolo 1-2 FP-jednotiek aktivity na 1 ml média (Mandels M., Andreotti R., Roche C.: Biotech-nol. Bioeng. Symp. 6, 21 /1976/). Přibližné dvojnásobná produkciu pri rovnakých podmienkach vykazuje kmen Trichodermareesei M 6 CCF 1856, ktorý je predffietom tohto vynálezu. Mikroorganizmus je uložený v zbier-ke Katedry botaniky Univerzity Karlovy, Praha 2, Benátská 2.
Kmen sa získal ozařováním konídií kmeňa Trichoderma viride QM 9414 získaného z Ameri-can Culture Collection of fungi (QM) University of Massachusetts, Amherst, MassachusettsUSA, UF-svetlom počas 5 minát. Hlavnou výhodou nového kmena je zvýšená produkcia celulóz, 226613
Claims (1)
- 226613 2 ktorá dosahuje při submernej kultivácii na 1 % mikrokryštalickej celulóze hodnoty okolo4 FP-jednotiek aktivity na 1 ml. Morfologie: vláknité, často vetvené, nepravidelné septované mycélium. Priemer hýf1,8 až 3,0 A1®· Konídionosiče dlhé, niektoré nepravidelné, kratšie a rozvětvené, fialidyzvyčajne jednotlivé, striedavé, alebo protistojace. Konídiá hojné, zeleno až zelenohnedopigmentované, elipticko-ovoidné, priemer 2,3 x 3,0 yum, alebo niekedy elipsoidné, priemer2,5 až 3,0 x 3,8 až 4>5 /un. Při submerznej kultivácii občasné chlamydospóry, sférické,priemer do 8,0 /um. Konidiácia indukovateTná svetlom. V bohatej pdde produkuje žitý, vo vo-dě rozpustný pigment. Mikroorganizmus raste na: D-glukóze, D-galaktóze, D-manóze, laktóze,melibióze, D-fruktóze, D-xylóze, L-arabinóze, D-arabinóze, D-ribóze, celobióze, eskulíne,trehalóze, škrobe, laminaríne, etanole, glycerole, D-erytritole, D-ribitole, D-manitole,D-sorbitole, dulcitole, m-inozitole, acetáte, sukcináte, fumaráte, maláte, citráte, gluku-ronáte, celulóze, slabo rastie na: maltóze, rafinóze, L-sorbóze, arbutine, salicíne, inu-líne, lektáte, tartráte a malonáte, nerastie na: sacharóze, alfa-metyl-D-glukozide, mele-zitóze, L-ramnóze. Příklad Kmen Trichoderma reesei M 6 sa kultivuje vo fermentore v médiu nasledovného zloženia(v gramoch na liter): močovina 0,3; síran amonný 1,4; dihydrogénfosforečnan draselný 2,0;heptahydrát síranu horečnatého 0,3; chlorid vápenatý 0,3; kvasničný extrakt 0,1; mikrokryš-talická celulóza 10,0; Tween 80 2,0; peptón 1,0; síran železnátý 5.10”hydrát síranumanganatého 1,4.10”chlorid zinečnatý 1,7.10”^; chlorid kobaltnatý 2.10-°. Teplota kultivácie je 28 °C, prevzdušňovanie 0,15 až 0,30 v/v/min, rýchlosť otáčokmiešadla 300 až 500 ot./min v závislosti na konštrukciu fermentora. Ako inokulum sa použi-je baničková kultúra v logaritmickéj fáze rastu vypěstovaná submerzne z konídií v rovnakommédiu, aké je použité vo fermentorovej kultivácii. Objem inokula je 10 % z objemu fermentač-nej zmesi. Kultivácia prebieha počas 5 až 7 dní, pričom pH kultúry sa udržiava v hraniciach4 až 4,5. V priebehu kultivácie sa sleduje aktivita celuláz v živnom médiu metodou podl’a(Mandels M., Andreotti R., Roche C.: Biotechnol. Bioeng. Symp. 6, 21 /1976/). Keď celulá-zová aktivita dosiahne maximálněj hodnoty, okolo 4 FP j/ml, médium sa scentrifuguje a čírysupernatant sa použije ako zdroj celulolytických enzýmov. Celulolytické enzýmy sa dajú využit pri sacharifikácii celulózy a lignocelulózovýchsubstrátov, pri zvyšovaní kvality siláže, při zlepšení extrakcie létok z rastlinných suro-vin napr. v potravinárstve, farmácii, pivovarníctve a vinárstve, a všade tam, kde je po-třebné rozložit celulózový podiel obsiahnutý v rastlinnom materiáli. Gelulázy sa ďalej da-jú využit napr. v priemysle papiera a celulózy na výroby materiálov s modifikovanými vzhle-dovými a mechanickými vlastnosťami.
