Vynález rieši sposob přípravy diagnostic-kého antigénu z cytomegalického virusu prereakciu vazby komplementu, ktorým sa mádiagnostikovat onemocnenie cytomegalickýmvírusom.
Na přípravu antigénu pre reakciu vazbykomplementu bolo popísaných niekotko me-téd. Niektorí autoři používali celú bunkovúkultúru a snažili sa uvolnit antigén z bunieksonifikáciou, alebo zmrazením a rozmraže-ním kultúry. Takýto antigén má však malúkapacitu a bývá antikomplementárny, pre-tože na pomnoženie virusu je třeba 7 až 11dní a za tento čas narastie v bunkovej kul-tuře antikomplementarita. Iní autoři koncen-trovali antigén po rozbití buniek ultracen-trifúgou. Niektorí autoři nebrali v úvahu ex-tracelulárny antigén a koncentrovali len an-tigén intracelulárny tak, že zliali médium zbunkovej kultúry, buňky zoškriabali zo sklaalebo uvolnili zmrazením a rozmražením kul-túry a čisté buňky rozbili zmrazovaním arozmrazováním alebo sonifikáciou. Buňkové antigény majú slabú stránku vnespecifických reakciách. Vyššie uvedenénedostatky sú odstránené postupom přípra-vy podl'a vynálezu, ktorého podstatou je, žek suspenzii cytomegalického virusu po roz-bití buniek sa přidá alkohol. Po zhluknutívirusu respektive virusového antigénu sus-penzia sa scentrifuguje a sediment resus-penduje. Resuspendovaný sediment předsta-vuje antigén. Tým sa antigén zbaví antikom-plementárnej zložky, využije sa intracelulár-ny i extracelulárny antigén a vyhne sa příliškoncentrovanému buňkovému materiálu, pre-tože tento robí nespecifické reakcie. Tátometoda je velmi rentabilná z hfadiska pro-dukcie, pretože pri jej použití sa získá z ví- 224010
Claims (1)
- 224010 rusovej suspenzie všetok antigén pomocouvelkokapacitných l'ahko dostupných centri-fúg. Takto sa m.óže rýchle připravit relativ-né velké množstvo hodnotného antigénu.Koncentráciu antigénu je možné regulovatriedením sedimentu. Příklad prevedenia Virusová suspenzia z buňkových kultúr sa ozvučí ultrazvukom za účelom uvolnenia vi-rusu z buniek. Potom sa k nej přidá alko-hol v množstve od 10 do 80 °/o. Táto zmes sanechá stát pokial' sa antigénne částice ne-zhluknú a scentrifugnje sa. Supernatant saodstráni a sediment sa suspenduje v množ-stve podlá potřeby, aby sa dostala žiadanákoncentrácia antigénu. PREDMET Sposob přípravy (jiagnostického antigénuz cytomegalického virusu pre reakciu vazbykomplementu vyznačujúci sa tým, že k víru-sovej suspenzii sa přidá alkohol v množstve vynalezu 10 až 80 %, po zhluknutí antigénu sa sus-penzia scentrifugnje a získá sa resuspendo-vaný sediment diagnostického antigénu.
