Description
The invention relates to a process for the production of penicillin in an iron apparatus, wherein the toxic effect of iron by the addition of a specific Titriplex is eliminated.
Chelatones can theoretically eliminate this effect; unfortunately, detoxifying agents usually inhibit growth, making them unusable, or their in vivo efficacy is insufficient. Iron only inhibits penicillin biosynthesis, not growth. This claim can be demonstrated by the following results obtained with Penicillium chrysogenum 65/41 (in all model experiments, iron was added in the form of ferrous sulphate). *
S kmenem 65/41 je inhibice penicilinu následující:The intensity of penicillin biosynthesis inhibition depends on the iron concentration in the soil fermentation. Up to an iron concentration of 50 gamma / ml soil fermentation, the relationship between iron concentration and inhibition of biosynthesis is linear, above this concentration the decrease in production is slower. However, an iron concentration of 50 gamma / ml is precisely the amount that is eliminated in penicillin fermentations in iron tanks in about 120 hours. With strain 65/41 the inhibition of penicillin is as follows:
+^Mocenství železa neovlivňuje intenzitu inhibice. + ^ Iron strength does not affect the intensity of inhibition.
Citlivost produkčních kmenů na železo je různá, u většiny kmenů však inhibice nepřesahuje při 50 ‘ gama železa na ml fermentační půdy 30%, u vysokoprodukčního kmene Penicillium chrysogenum ; 65/41 je inhibice 70%.The sensitivity of the production strains to iron varies, but in most strains the inhibition does not exceed 30% at 50 .mu.g of iron per ml of fermentation broth, in the high-production strain Penicillium chrysogenum; 65/41 inhibition is 70%.
Iron penetrates the cell membrane and is deposited in the mycelium. The higher the concentration of iron in the soil, the more it accumulates in the mycelium.
The concentration of iron in the mycelium does not change by its age, only with the onset of autolysis the iron is released from the mycelium again into the culture medium.
Jelikož se jedná při inhibičním působení železa o poškození enzymů syntetizujících penicilín, je vliv železa výraznější v době, kdy se tvoří syntetizující aparát:Since the inhibitory action of iron is a damage to penicillin synthesizing enzymes, the effect of iron is more pronounced at the time the synthesizing apparatus is formed:
Jako potenciální detoxifikační látky přicházejí v úvahu komplexony a titriplexy. Výsledky s použitými látkami v optimálních účinných koncentracích jsou následující:Possible detoxifying agents are complexons and titriplexes. The results with the substances used in optimal effective concentrations are as follows:
Chemické složení použitých látek:Chemical composition of used substances:
Navrhovaný způsob eliminace toxického vlivu železa odstraňuje i nepříznivý vliv nekvalitních surovin. However, the source of iron during fermentations in tanks is not only unsuitable construction material, but also raw materials, especially peanut flour, ammonium sulphate, etc. The proposed method of eliminating the toxic influence of iron removes also the unfavorable effect of poor quality raw materials.
