技术领域
本实用新型涉及一种培养皿,特别是一种皿底四周设有垂直刻度标尺及皿底底部设有以荧光颜料刻划的刻度标尺的培养皿。
背景技术
现有的培养皿都是平板透明圆柱形的,皿体上没有任何定量刻度标记,通常进行细菌培养时,需向培养皿里倾倒一定量的培养基,当样本量较大时,对每个培养皿中倾倒培养基的量的控制,是一件很繁琐的工作,而仅靠肉眼直接识别,难以达到准确定量的效果,另外,所有样本培养皿中培养基铺设是否均匀一致,没有一个定量的平衡标准,这势必导致培养基定量不准、营养分布不均,从而产生试验误差;另外,在细菌培养试验完成后,普通培养皿难以实现对菌落扩散区域大小的直接确定工作,若采用外部手段测定,势必增加工作量。
实用新型内容
本实用新型所要解决的技术问题是提供一种皿底四周有垂直刻度标尺,皿底底部有荧光颜料刻划的同心圆及方孔刻度标尺的培养皿,其能准确定量倾倒入培养皿内的培养基,并作为培养基铺设均匀性评价的参考标准,同时,通过皿底底部荧光颜料刻划的刻度标尺可以对菌落扩散区域大小进行直接确定。
为解决上述技术问题,本实用新型是按如下的方式来实现的:本实用新型所述的荧光定量培养皿,由皿底和皿盖组成;所述皿底为圆柱形,其四周环绕平均间隔为90度的4个垂直刻度标尺,皿底底部设有以荧光颜料刻划,并参照皿底圆心的扩散同心圆及边长均等的方孔刻度标尺;皿盖为圆柱形,皿盖外径大于皿底内径,培养细菌时,皿盖盖在皿底上面。
所述皿盖和皿底的材料均为医用级聚苯乙烯;皿底底部刻划刻度标尺的颜料为稀土有机配合物荧光颜料。
本实用新型的积极效果在于:皿底四周环绕平均间隔为90度的4个垂直刻度标尺,可以此刻度标尺测量、控制倾倒入培养皿中培养基的量,并以对称的任意两个(或更多)标尺刻度作为培养基铺设平衡的判断依据,以此考察培养基铺设的均匀性;此外,皿底底部设有以荧光颜料刻划,并参照皿底圆心的扩散同心圆及边长均等的方孔刻度标尺,在紫外光照射下,皿底底部的荧光刻度标尺可透过培养基,以便试验人员直接以肉眼对菌落扩散区域进行定位及面积估算;本培养皿使用成本低,减少了试验工作量,虽然结构简单,但其应用普遍,具有一定的经济效益和社会效益。

如图1、图2、图3所示,本实用新型所述的荧光定量培养皿由皿底1和皿盖2组成;所述皿底1为圆柱形,其四周环绕平均间隔为90度的4个垂直刻度标尺3,皿底1底部设有以荧光颜料刻划,并参照皿底圆心的扩散同心圆及边长均等的方孔刻度标尺4;皿盖2为圆柱形,皿盖2外径大于皿底1内径,培养细菌时,皿盖2盖在皿底1上面。
所述皿底1和皿盖2的材料均为医用级聚苯乙烯;皿底1底部刻划刻度标尺4的颜料为稀土有机配合物荧光颜料。
本实用新型的使用方法是:在皿底内倒入培养基时,以四周任意互相对称的2个垂直刻度标示为参照,确定注入培养基的量,在倒入一定量的培养基后,对培养皿稍事抖动,对比不同位置的刻度标尺,以确定培养基是否铺设均匀。菌落培养完成后,将培养皿置于紫外光下照射,底部荧光刻度标示即可呈现出来,试验人员直接以肉眼按照荧光同心圆及其角度标示对菌落扩散区域进行定位,并按照荧光方孔数目对其面积进行估算。
本实用新型的皿底和皿盖的形状不限于圆柱形,所有刻度标示方式不限于同心圆、方孔、垂直标尺,不论出现形状各异、大小各异、高低各异、标尺方式各异的培养皿,均在本实用新型的保护范围之内。
