Abstract

本发明为室内空气污染物二异氰酸甲苯酯的致毒机理研究。旨在研究二异氰酸甲苯酯在动物体内吸收、分布、排泄状况及代谢机制、途径和代谢产物,主要代谢靶器官,并应用RT-PCR方法,结合激光共聚焦扫描技术及数字图像处理系统研究二异氰酸甲苯酯及代谢产物对靶细胞受体、基因表达的影响,从分子水平揭示该化合物的致毒机理,为生态风险性评定、环境执法、确定防治对策提科供科学理的理论依据。

室内空气污染物二异氰酸甲苯酯的致毒机理研究
(一)技术领域
本发明属于环境毒理学实验方法研究,具体的说是对低毒类有机化合物二异氰酸甲苯酯(TDI)产生毒性的作用方式、途径及机理的研究。
(二)背景技术
在经济发展的今天,室内空气的污染程度日趋严重,本专利所进行的有关二异氰酸甲苯酯的基础性研究,为该类化合物的解毒机制提供第一手资料,为环境安全性研究提供坚实的理论基础,对于建立我国有毒化学污染物环境安全的理论与技术支持体系,建立污染预防与控制的技术及方法学体系等具有重要的学术意义。
本发明是对于了解该类的其他化合物的毒性和致毒机制,预测该类化合物族对人体健康的危害就十分必要。
(三)发明内容
本发明研究了二异氰酸甲苯酯产生毒性的作用方式、途径及机理,对于确定置于环境中的安全量或容许量,进行健康危险性评价,为生态风险性评定和环境执法、为阻断污染途径和确定防治对策提供科学理论依据。
本发明主要目的通过以下途径实现:
1.呼吸道染毒途径、三种染毒剂量的二异氰酸甲苯酯在大鼠体内吸收、分布、排泄及代谢动力学;
2.二异氰酸甲苯酯的体外代谢实验:体外实验作为体内实验的重要补充,通过反应的酶解动力学研究,从细胞、亚细胞水平进一步揭示二异氰酸甲苯酯的主要代谢器官、代谢产物,二者相互配合更深入揭示二异氰酸甲苯酯化合物的代谢机制及途径;
3.应用LSCM(激光共聚焦扫描)技术研究二异氰酸甲苯酯及代谢产物对细胞形态及细胞核DNA结构的影响;
4.采用RT-PCR方法(反转录酶-多聚酶链反应法)研究二异氰酸甲苯酯及代谢产物对靶细胞基因表达的影响。
本发明的优点在于:
1.体内实验代谢与体外代谢实验相辅相成,从细胞、亚细胞水平共同揭示二异氰酸甲苯酯在动物体内的吸收、分布、排泄状况及代谢途径和机制,以确定主要代谢的靶器官。使该化合物在机体内的毒作用代谢机理表达得更趋完善,为明确该化合物对人体健康影响提供坚实的理论基础和数据资料,同时填补国内在此方面研究的空白点。
2.应用RT-PCR、聚焦激光扫描显微镜和数字图象处理等现代技术工具,研究二异氰酸甲苯酯及代谢产物对大鼠靶细胞基因表达调整的影响,并结合化合物的新陈代谢特点,共同揭示二异氰酸甲苯酯及其代谢产物对靶器官内细胞因子表达的影响,从分子水平揭示该化合物的致毒作用机制。
3.二异氰酸甲苯酯在体内活性代谢物的确定,为寻找该化合物的生物标志物,为环境污染的提早预防提供第一手资料,并通过代谢物对靶细胞基因表达的反应性,更深入阐明其致毒机理。
4.本项目研究的整个过程,选用几个剂量研究,为环境安全性的评价、确定置于环境中的安全量或容许量、生态风险性评定、环境执法及其阻断污染径和确定防治对策提供科学理论依据。
具体实施方式
本发明的实验过程,实验对象选取大鼠,先进行二异氰酸甲苯酯在大鼠体内的代谢动力学参数的测定利用HPLC、GC或同位素标记技术,考察二异氰酸甲苯酯及代谢产物在血浆中随时间而变化的动态趋势。并以上述代谢动力学模型中的有关参数与实际测定值的模型嵌合状况。然后进行二异氰酸甲苯酯的体外代谢实验,应用LSCM(激光共聚焦扫描)技术研究二异氰酸甲苯酯及代谢产物对细胞形态及细胞核DNA结构的影响,采用RT-PCR方法(反转录酶-多聚酶链反应法)研究二异氰酸甲苯酯及代谢产物对靶细胞基因表达的影响。

1.室内空气污染物的致毒机理研究,其特征是:它是空气污染物中二异氰酸甲苯酯的致毒机理研究,从细胞、亚细胞水平共同揭示二异氰酸甲苯酯在动物体内的吸收、分布、排泄状况及代谢途径和机制,以确定主要代谢的靶器官。
2.根据权利要求1所述的空气污染物中二异氰酸甲苯酯的致毒机理研究的代谢途径,其特征是:通过呼吸道染毒途径、三种染毒剂量的二异氰酸甲苯酯在大鼠体内吸收、分布、排泄及代谢动力学来研究的。
