Description
The present invention belongs to a new process for producing sugar by using corn starch primary pulp as raw material and adopting fermentation process.
In the prior art, the common method for fermenting and producing the corn starch is to use refined corn starch to prepare sugar and then add molasses, corn steep liquor and various inorganic salts for fermenting and producing, the process is mature, the fermentation cost is high, the control precision is high, if the fermentation production is abnormal, the factors are more, the analysis is difficult and difficult to judge, the sources of the molasses and the corn steep liquor are different, the production places are different, and the biotin content is different. The content of molasses and corn steep liquor in each batch is different, and is a factor influencing the fermentation production process. The process flow is as follows: corn → soaking → crushing → separating embryo bud → grinding → removing dregs of semen persicae → protoplasm → centrifuging protein → starch slurry → adjusting pH with alkali → liquefying → saccharifying → pressure filtering → sugar solution + corn slurry + molasses + inorganic salt (potassium salt, phosphorus salt, magnesium salt, manganese salt) → digestion → fermentation production → glutamic acid. The fermentation production by using the crude raw materials is unsuccessful, and the method is failed in that the dried corn is degermed, crushed and then used as sugar for fermentation. The defects of the method are that the waste water amount is large, the production process cannot be cleaned, the cost is high, the investment of equipment and manpower is large, and the fermentation process is unstable.
The invention aims to overcome the defects and design a novel process for producing sugar by fermenting corn starch protoplasm, which adopts the protoplasm as the sugar production process, adjusts the pH value of the sugar to 5.0-7.0 by lime, hydrolyzes the sugar liquid by adding protease, does not add corn steep liquor and molasses in the fermentation formula, reduces the dosage of phosphorus salt by 20-50%, improves the sugar yield by 5-8%, reduces the discharge of waste water, has clean process production, reduces the production cost, has less equipment and manual investment and stable fermentation production process, adopts water for washing embryo buds and powder residues to circulate to flush corn, and adopts a double-enzyme method for spraying and liquefying twice to produce sugar.
The invention relates to a novel fermentation production process by using corn starch raw pulp as sugar, which is realized by adopting the following technical scheme that the production process of raw pulp starch is adopted, the pH value of the raw pulp starch is adjusted to be 5.0-7.0 by using lime; the sugar liquor is hydrolyzed by adding protease, no corn steep liquor and molasses are added in the fermentation formula, the dosage of the phosphorus salt is reduced by 20-50%, the sugar yield is improved by 5-8%, the discharge of waste water is reduced, the production process is clean, the production cost is reduced, the equipment and manual investment is low, and the fermentation production process is stable. The process flow comprises the following steps: corn → soaking → crushing → separating germ → grinding → filtering with fructus forsythiae → protoplasm → adjusting pH value → liquefying → saccharifying → pressing filtering → sugar solution → adjusting pH value to neutral → adding protease for hydrolysis for 2 hours → adding magnesium salt, phosphorus salt, potassium salt and manganese salt → continuous disinfection → fermentation production.
The invention soaks the corn into 0.25-0.5% of sulfurous acid water, soaks 72-120 hours, produces the mixture of the starch of magma and yellow powder directly, produces the magmaThe water for washing germ and powder residue is recycled to be used as corn washing water, lime is used for adjusting the pH value of primary pulp to 5.0-7.0, and the reaction equation is as follows:
the sugar is prepared by adopting a double-enzyme method of twice jetting and liquefying, and the filtered sugar solution is hydrolyzed by adopting protease. The protease hydrolysis is carried out by adjusting pH to 7.0-8.0, adjusting temperature to 45-55 deg.C, adding protease in an amount of 3-10 units/g protein, and reacting for 2 hr. In the fermentation formula of the invention, molasses and corn steep liquor are not added, and a little phosphate is added.
The method has the effects that no corn steep liquor and molasses are added in the fermentation formula, the use amount of the phosphorus salt is reduced by 20-50%, the process flow is short, the operation is convenient, the cost is low, the fermentation production process is stable, the discharge of waste water is reduced, the production process is clean, the investment of equipment and labor is reduced, the sugar yield is improved by 5-8%, the pH value is adjusted by lime to adjust the pH value to 5.0-7.0 during sugar making, and a double-enzyme method of twice spraying liquefaction is adopted for sugar making.
The present invention will be described in further detail with reference to examples. The process flow comprises the steps of corn → soaking → crushing → embryo separation → grinding → sieving → plasmogen → pH value adjustment → liquefaction → saccharification → pressure filtration → sugar liquor → pH value adjustment to neutral → protease hydrolysis for 2 hours → magnesium salt, phosphorus salt, potassium salt and manganese salt → continuous disinfection → fermentation production.
The invention adopts sulfurous acid to soak the corn, the concentration of sulfurous acid water is 0.25-0.5%, the soaking is carried out for 72-120 hours, a large amount of biotin is soaked out and enters the soaking solution, the separation of starch, germ and fiber is more thorough, the load of the next procedure is reduced, the content of biotin in sugar solution is reduced, and favorable conditions are provided for the control of fermentation production.
The starch production process of the invention omits a centrifugal separation part and a yellow powder precipitation part, directly produces the primary pulp, namely the mixture of the starch and the yellow powder, reduces energy and labor intensity and reduces production cost. Reduce the discharge of waste water, and recycle the water for washing the germs and the powder residues to the corn washing water. When the pH value of the raw pulp is adjusted, adjusting the pH value to be 5.0-7.0 by adopting lime, adjusting the pH value by using lime powder aims to neutralize sulfite in the raw pulp and the lime to generate calcium sulfite, and removing the calcium sulfite during filtering, wherein the fermentation production is influenced by excessive sulfurous acid, and the reaction equation is as follows:
the method for making sugar adopts a double-enzyme method of twice injection liquefaction to make sugar. The protein in the filtered sugar solution is hydrolyzed into various amino acids by protease under the process conditions of pH value of 7.0-8.0, temperature of 45-55 ℃, protease addition of 3-10 units/g protein and reaction time of 2 hours. The fermentation formula of the invention does not add molasses and corn steep liquor, reduces the dosage of the phosphorussalt by 20-50%, saves energy by adopting the new process, improves the sugar yield by 5-8%, and reduces the cost.
