Abstract

转基因棉花抗虫检测法,属检测方法。它分为检测试剂的制备和操作方法两部分。试剂由对照液(Bt毒蛋白)、提取液(Tris缓冲液)、封闭剂(胎牛血清蛋白)、酶标抗体(HRP-兔抗Bt抗体)、显色液(二氨基联苯胺)和支持膜(硝酸纤维素膜)组成。操作方法分为快诊膜制备、检测和显色判别三个步骤。该方法适于检测转基因棉花对棉铃虫的抗性。也可用于检测其它转基因植物对一些昆虫的抗性。因此,种子单位可用来检测转基因种子抗虫性,育种单位可用于选择抗虫单株。

Description

转基因棉花抗虫检测法
本发明属检测方法。
对转基因棉花抗虫性的检测,目前国内外采取两种方法,即分子检测和生物学测定。分子检测要使用一些精密仪器和同位素,操作复杂,一般由分子生物学研究人员完成。生物学测定,一方面需要饲养棉铃虫,另一方面需种子长成植株后才能鉴定,另外环境气候还造成检测结果不准确。种子经营和基层育种单位一般不具备分子检测的条件,他们虽然具备生物学测定的条件,但种子单位无法在购种时检测种子是否抗虫,育种单位无法在选择单株时确定所选种子是否抗虫。因此,目前的方法不能满足科研生产单位对转基因棉花抗虫的检测。
本发明的目的就是为种子和育种单位提供一套快速检测转基因棉花抗虫检测试剂及检测方法。
本发明包括试剂盒、试剂制备方法和检测方法。试剂盒由4支5ml离心管、1支10ml离心管和支持膜组成,可以放置在一个塑料包装袋内,也可以组成一个盒。这5个离心管和支持膜的组成成份如下。
1.对照液,体积1ml,将0.875mg纯化毒蛋白溶于0.5mol/L磷酸缓冲液中。毒蛋白通过苏云金杆菌HD-1菌株发酵制备并提纯。
2.提取液,体积10ml,使用时蒸馏水稀释10倍。其成份内含5g聚山梨醇脂20,0.5mol/LTris-HCl,pH值7.9,1.5mol/L NaCl。
3.支持膜,5张,每张82mm2,为国产或进口硝酸纤维素膜。
4.封闭剂,体积5ml,用时蒸馏水稀释10倍。其成份跟提取液基本相同。再加入7.5g胎牛血清蛋白。
5.酶标抗体,体积0.5ml,使用时用提取液稀释500倍。制备方法为,用提纯抗原免疫日本大耳白兔,纯化高效价IgG抗体,用过碘酸法标记辣根过氧化物酶,再加入适量牛血清蛋白作保护剂制备而成。
6.显色液,体积5ml,用时用蒸馏水稀释10倍,内含25mg,3'3二氨基联苯胺、0.5mol/L Tris-HCl,用前加30%双氧水。
操作方法分为快诊膜制备、检测和显色判别三个步骤。
本方法具有操作方便、检测灵敏、成本低廉、重复性好、不需复杂仪器等优点。最主要的特点是可以在种子期(或幼苗期)取样,6个小时内即获得检测结果。经过对目前推广的转基因抗虫棉检测验证。符合率达100%。

本发明最佳实施例:
实施本发明的基本原料均已组配在试剂盒中,实施过程中还需要加样器、蒸馏水和40℃左右的温水。操作方法分为以下三步:
(1)快诊膜的制备,取1g种子(或叶片),加1ml提取液,研细,静置1小时。用加样器取5μl上清液加在支持膜上,并加1μl对照,自然干燥。可以立即检测。也可以长期保存。(2)检测。将快诊膜放入10ml封闭剂中,在37℃下封闭1小时,取出后放置在10ml酶标抗体的稀释液中,在37℃温育1小时。(3)显色判别,取出快诊膜、置入10ml稀释显色液中,待对照出现黄色斑点时,取出干燥。凡能显示黄色斑点的为抗虫种子,否则不抗虫。
本方法在种子经营和育种单位具有较广阔的使用前途。检测结果灵敏、方法简单。

Claims (3)

1.一种转基因棉花抗虫检测法,检测试剂主要由对照液、提取液、支持膜、封闭剂、酶标抗体和显色液组成,其特征在于将种子或叶片提取液加在支持膜上,在封闭剂和酶标抗体液中温育后,再由显色液显色,以测定棉花是否抗虫。
2.根据权利要求1所述的转基因棉花抗虫检测法,其特征在于支持膜为硝酸纤维素膜。
3.根据权利要求1所述的转基因棉花抗虫检测法,其特征在于酶标抗体为兔抗Bt抗体。
