Abstract
The invention relates to a preparation method of a platelet-rich plasma gel compound for facial filling, which comprises the following steps: placing the collected blood in a container under aseptic conditions; placing the container containing blood in a centrifuge for centrifugal separation; after centrifugal separation, the blood in the container is divided into three layers, namely a plasma layer, a platelet layer, a leucocyte layer, a fibrin layer and a erythrocyte layer from top to bottom; extracting the plasma layer, blood platelet, white cell and fibrin layer with a syringe respectively, heating the syringe containing the plasma layer in water bath, and cooling the plasma layer to form gel; mixing the gel with blood platelet, leucocyte and fibrin layer via medical three-way pipe to obtain the blood platelet-rich plasma gel compound. The platelet-rich plasma gel compound prepared by the invention is rich in platelets, white blood cells and fibrin, can be injected into various layers of skin, and plays a role in filling.
Description
Technical Field
The invention relates to the technical field of biomedical materials, in particular to a preparation method of a platelet-rich plasma gel compound for facial filling.
Background
Platelet-rich plasma (PRP) is a platelet concentrate obtained by extracting autologous venous blood and adopting a density gradient centrifugation method, is rich in various bioactive substances such as growth factors, cytokines, antibacterial peptides and the like, and has the effects of promoting cell proliferation, differentiation, matrix synthesis, tissue regeneration, repair and the like.
Currently, platelet rich plasma is mostly extracted by high-speed centrifugation. Such extraction methods are prone to platelet destruction. Furthermore, platelet rich plasma extracted according to such methods has the following drawbacks: (1) is almost free of white blood cells and fibrin; (2) The growth factors in the platelet-rich plasma are released quickly, the release time is 12-24 hours, and the slow release effect is relatively poor; (3) Only liquid smearing or injection can be carried out, and the injection layer is only a dermis layer, so that the filling effect cannot be achieved.
And secondly, the white blood cells can remove harmful substances and necrotic tissues, resist invasion of pathogenic agents and participate in repair and regeneration of the tissues. Fibrin can form a scaffold that loads platelet rich plasma, provides facial filling support and allows slow release of growth factors within the platelet rich plasma. The platelet-rich plasma prepared by the high-speed centrifugal separation method lacks white blood cells and fibrin, and cannot obtain the beneficial effects of the white blood cells and the fibrin.
Therefore, there is an urgent need to provide a method for preparing a platelet-rich plasma gel complex useful for facial filling, having a slow release of growth factors in platelet-rich plasma.
Disclosure of Invention
The present invention has been made in view of the above problems, and has as its object to provide a method for preparing a platelet rich plasma gel-complex for facial filling which overcomes or at least partially solves the above problems.
In a first aspect, embodiments of the present invention provide a method for preparing a platelet rich plasma gel-composite for facial filling, comprising the steps of:
step 1), placing collected blood in a container under a sterile condition;
step 2), placing the container containing blood in a centrifugal machine for centrifugal separation;
step 3), after centrifugal separation, the blood in the container is divided into three layers, namely a plasma layer, a platelet layer, a leucocyte layer, a fibrin layer and a erythrocyte layer from top to bottom;
step 4), respectively extracting the plasma layer, the platelet, the leucocyte and the fibrin layer by using a syringe, heating the syringe containing the plasma layer in water bath, and cooling the plasma layer to form gel;
Step 5), mixing the gel with the platelet, the white blood cell and the fibrin layer through a medical three-way tube to obtain the platelet-rich plasma gel compound.
Preferably, the container is a centrifuge tube or a blood collection tube without anticoagulant.
Preferably, the blood is autologous venous blood of the person seeking beauty.
Preferably, the centrifugal force of the centrifugal machine in the step 2) is 150 Xg-300 Xg, and the centrifugal time is 4-11 minutes.
Preferably, the centrifugal force of the centrifuge in step 2) is 200 Xg and the centrifugation time is 8 minutes.

The platelet-rich plasma gel compound prepared by the method provided by the embodiment of the invention is rich in platelets, white blood cells and fibrin, can be injected into various layers of skin, and plays a role in filling; secondly, the release time of the growth factors in the platelet-rich plasma in the compound can reach 90 days, and the slow release time of the growth factors is longer than the release time of the growth factors in the platelet-rich plasma prepared by a high-speed centrifugation method; the compound can be used in combination with collagen, recombinant collagen, hyaluronic acid, etc. for facial filling.

Detailed Description
The present invention will be further described with reference to examples for the convenience of those skilled in the art.
In a first aspect, embodiments of the present invention provide a method for preparing a platelet rich plasma gel-composite for facial filling, comprising the steps of:
step 1), placing collected blood in a container under a sterile condition;
step 2), placing the container containing blood in a centrifugal machine for centrifugal separation;
step 3), after centrifugal separation, the blood in the container is divided into three layers, namely a plasma layer, a platelet layer, a leucocyte layer, a fibrin layer and a erythrocyte layer from top to bottom;
step 4), respectively extracting the plasma layer, the platelet, the leucocyte and the fibrin layer by using a syringe, heating the syringe containing the plasma layer in water bath, and cooling the plasma layer to form gel;
Step 5), mixing the gel with the platelet, the white blood cell and the fibrin layer through a medical three-way tube to obtain the platelet-rich plasma gel compound.
In one embodiment, the container is a centrifuge tube or a blood collection tube without anticoagulant.
In one embodiment, the blood is autologous venous blood of the subject.
In one embodiment, the centrifugal force of the centrifuge in step 2) is 150×g to 300×g, and the centrifugation time is 4 to 11 minutes.
In one embodiment, the centrifuge of step 2) has a centrifugal force of 200 Xg and a centrifugation time of 8 minutes.
In one embodiment, the temperature of the water bath heating of step 4) is 60-80 ℃; the heating time is 8-10 minutes.

Examples

step 3), after centrifugal separation, the blood in the container is divided into three layers, namely a plasma layer, a platelet layer, a leucocyte layer, a fibrin layer and a erythrocyte layer from top to bottom;
Step 4), respectively extracting the plasma layer, the blood platelets, the white blood cells and the fibrin layer by using a syringe, heating the syringe containing the plasma layer in a water bath at the temperature of 70 ℃ for 9 minutes, and cooling the plasma layer to form gel;
Step 5), mixing the gel with the platelet, the white blood cell and the fibrin layer according to the volume ratio of 4:1 through a medical three-way tube to obtain the platelet-rich plasma gel compound.
Injecting the prepared platelet-rich plasma gel compound into the lacrimal duct and apple muscle parts of the face of the person to be beautified; the face photograph of the person who looks after the injection (fig. 2) was taken before the operation (fig. 1), and the face photograph of the person who looks after the operation (fig. 3) was taken for 4 days after the operation as a control.

step 4), respectively extracting the plasma layer, the platelet, the leucocyte and the fibrin layer by using a syringe, heating the syringe containing the plasma layer in water bath, and cooling the plasma layer to form gel;
Step 5), mixing the gel with the platelet, the white blood cell and the fibrin layer through a medical three-way tube to obtain the platelet-rich plasma gel compound.
2. The method of preparing a platelet rich plasma gel composition for facial filling according to claim 1, wherein: the container is a centrifuge tube or a blood collection tube without anticoagulant.
3. The method of preparing a platelet rich plasma gel composition for facial filling according to claim 1, wherein: the blood is autologous venous blood of the person for beauty.
