Abstract
本发明公开了一种顺序释放药物的纳米递送系统及制备方法和用途,属于医药领域。该纳米递送系统以叶酸修饰的脂质体包覆聚合物胶束,并在聚合物胶束和脂质体内均独立负载有药物。本发明通过聚合物胶束和脂质体内包覆的两种药物的顺序输送、特异性去除CAF以及增加药物渗透的方式,有效抑制了TNBC的肿瘤生长和转移,为TNBC的治疗提供了新策略。并有望将此方法推广至多种恶性肿瘤的治疗。
Description
技术领域
本发明属于医药领域,具体涉及一种顺序释放药物的纳米递送系统及制备方法和用途。
背景技术
乳腺癌是女性中最常见的癌症,也是全球第二常见的新诊断癌症,其中,三阴性乳腺癌(TNBC)是乳腺癌中最致命的亚型。由于其异质性和通常具有侵袭性的行为,乳腺癌仍然是一个治疗挑战。虽然开发治疗方法的努力已经针对肿瘤细胞本身,但最近的研究发现,肿瘤微环境可以强烈影响肿瘤的生长、转移和对治疗的反应。特别是肿瘤周围基质中最丰富的癌症相关成纤维细胞(CAFs),通过向肿瘤发送信号并改变其周围的细胞外基质来调节肿瘤的发展。乳腺癌CAFs中α-平滑肌肌动蛋白(α-SMA)的过表达可在血管周围产生密集的胶原分泌,形成物理阻断,阻止药物递送至肿瘤细胞,这可能导致耐药性和转移。
单纯靶向癌细胞的传统治疗方法不足以抑制CAF活化,从而导致乳腺癌复发和转移。为了提高抗肿瘤疗效,联合治疗越来越受到重视,即使用两种或两种以上具有不同治疗机制的药物,通过协同作用来增强抗肿瘤疗效。游离药物组合的疗效通常会受到生物利用度不足的影响,而纳米载体共递送系统在联合治疗中具有潜在优势。迄今为止,许多纳米系统已被用于将两种或两种以上具有协同药理活性的分子递送到肿瘤微环境中的不同细胞靶点。因此,我们致力于开发一种高效的纳米递送治疗体系。
发明内容
针对现有技术中的上述不足,本发明提供一种顺序释放药物的纳米递送系统及制备方法和用途,本发明构建的递送系统在进入体内后,首先是脂质体破裂,释放雷公藤红素和包覆有桦木酸的胶束,然后桦木酸再从胶束中释放出来,通过顺序释放的方式,对肿瘤起到双重治疗功效。
为实现上述目的,本发明解决其技术问题所采用的技术方案是:
一种顺序释放药物的纳米递送系统,以脂质体包覆聚合物胶束,并在聚合物胶束和脂质体内均独立负载有药物。
进一步地,该脂质体为叶酸修饰的脂质体。
进一步地,聚合物胶束包封于脂质体的磷脂双分子层中的亲水层中。
进一步地,聚合物胶束为TPGS。
进一步地,脂质体内和聚合物胶束内负载的药物类型不同。
上述纳米递送系统的制备方法,采用薄膜分散法制备上述纳米递送系统。
上述纳米递送系统在制备治疗肿瘤的药物制剂中的用途。

本发明的有益效果:
本发明制备了一种能够在体内靶向肿瘤并顺序释放的新型多功能纳米制剂。经系统给药后,该制剂首先在受FA引导的间充质富集的肿瘤部位特异性聚集,释放CEL和BM。释放的CEL在间质中保留,通过下调CAF中诸如α-SMA、胶原蛋白I等蛋白的表达来抑制纤维化,为随后释放的抗肿瘤药物进入癌细胞清除了道路。同时,共载纳米制剂比单独使用任何一种药物都能更有效的抑制肿瘤生长,并且显著减少了小鼠肺转移的数量。
本发明通过两种药物的顺序输送、特异性去除CAF以及增加药物渗透的方式,有效抑制了TNBC的肿瘤生长和转移,为TNBC的治疗提供了新策略,并有望将此方法推广至多种恶性肿瘤的治疗。

具体实施方式
下面对本发明的具体实施方式进行描述,以便于本技术领域的技术人员理解本发明,但应该清楚,本发明不限于具体实施方式的范围,对本技术领域的普通技术人员来讲,只要各种变化在所附的权利要求限定和确定的本发明的精神和范围内,这些变化是显而易见的,一切利用本发明构思的发明创造均在保护之列。
本发明中使用的小鼠乳腺肿瘤细胞株4T1和小鼠胚胎成纤维细胞细胞株NIH 3T3购自中国科学院细胞库(中国上海)。
实施例1F/CL@BM的制备和表征
一、制备
1、制备BM
将TPGS和桦木酸按照10:1的重量比混合在乙醇中(5mL)。混合溶液在50℃的旋转蒸发仪上蒸发,形成脂质膜,随后在PBS(5mL)中水化10min。
2、制备CL@BM
将大豆卵磷脂和胆固醇按照10:2重量比混合在二氯甲烷中(5mL),加入雷公藤红素(1mg),然后在35℃的旋转蒸发仪上蒸发以形成脂质膜。膜在前述制备的BM(10mL)中水化30min,随后在冰水中使用65W的探头超声波处理10min。
3、制备F/CL@BM
将CL@BM和DSPE-PEG2000-叶酸(2mg)在37℃下孵育60min。
在实验中使用前,所有这些制剂都通过220nm的过滤器过滤,以去除未包封的药物。
二、表征
1、采用马尔文粒径仪来评估粒径和zeta电位。采用透射电子显微镜观察形态。在将样品用磷酸盐缓冲液(PBS)稀释至2mg/mL后,我们将稀释液均匀分散于铜网上。随后,使用10μL的1%(体积比)磷钨酸(PBS溶液中)对样品进行染色处理,并让其在空气中自然干燥。通过在220nm和425nm波长下使用高效液相色谱法,分别测量了桦木酸和雷公藤红素的载荷量和包封效率。包封效率(EE)的计算公式为:EE(%)=(F/CL@BM中药物的重量/投料药物的总重量)×100%。
如图1所示,透射电子显微镜显示BM和F/BM@CL呈均匀球形,胶束-脂质体显示出预期的双层结构(图1A)。各自的zeta电位分别为-3.05和-15.33mV(图1B),这意味着它们之间的相互排斥将防止它们在溶液或体内自聚集。两种纳米制剂的直径分别为25.6和121.3nm左右(图1C)。F/CL@BM的多分散性指数(0.257)明显低于BM(0.360)(图1D),这表明BM由于包封在脂质体中所以更加稳定。它们的包封率(EE)和载药量(LC)见表1。F/CL@BM的LC分别为8.46±0.20%(BA)和8.17±0.05%(CEL),EE分别为82.77±0.06%(BA)和88.34±0.05%(CEL)。实际上,F/CL@BM的大小和多分散性在磷酸盐缓冲盐水(PBS)或培养基中4℃静置时没有改变(图1E)。F/CL@BM在10%胎牛血清(FBS)中的直径比在PBS溶液中稍大。这种尺寸差异归因于在存在FBS时纳米颗粒表面形成的蛋白质冠层。
表1包封率(EE)和载药量(LC)
2、使用FRET实验确认桦木酸和雷公藤红素的共载,其中雷公藤红素被DiO替代以作为能量供体,桦木酸被DiI替代以作为能量受体。作为对比,含有DiO的脂质体(L-DiO)和含有DiI的胶束(M-DiI)以1:1的质量比简单混合,形成M-DiI@L-DiO。在470nm激发后,使用Synergy H1 Microplate Reader(BioTek Instruments,Winooski,VT,USA)记录荧光发射光谱。FRET比率被量化为568nm处的荧光发射与568nm和503nm处发射之和的比率。采用透析法测量BM@CL体外释放桦木酸和雷公藤红素。BM@CL溶解在1mL PBS中(两种药物浓度均为1mg/mL)置于分子量截止为10kDa的透析袋中,浸入含有0.5% Tween 80的20mL PBS中(pH调至6.5),并以100转/min搅拌。在指定时间取出1mL样品,并补充相同体积的新鲜PBS。使用高效液相色谱法测量桦木酸和雷公藤红素的含量。在14天内,每隔一天用动态光散射测量其尺寸和多分散性指数来评估F/CL@BM在PBS和DMEM培养基中的稳定性。
荧光光谱(图1G)显示,两种药物在F/CL@BM中的FRET比率明显高于它们在简单物理混合物中的比率(56.53%vs.28.40%,p<0.01;图1F),证实了它们在F/CL@BM中的共载。
体外释放实验显示,在最初的4h内,两种药物都没有显著释放,此后释放量增加,雷公藤红素的72h累积释放量为94.60%,而桦木酸为64.00%(图1H)。在相同的透析条件下,桦木酸从BM中的释放速度比从F/CL@BM中更慢。因此,F/CL@BM的胶束-脂质体结构导致CEL先于BA释放。
实施例2F/CL@BM对细胞的影响
1、对体外细胞活性的影响
将4T1细胞单独或与NIH 3T3成纤维细胞(每孔1×104细胞,NIH 3T3与4T1细胞的比例为1:1)一起接种在96孔板中,并孵育48h。然后,用F/CL@BM处理细胞24h,CEL和BA的浓度分别为0、0.025、0.125、0.25、0.5和1μg/mL。每孔加入CCK-8试剂10μL,然后在37℃下孵育30min。在450nm处测量吸光度。作为细胞活性的补充评估,4T1细胞接种在6孔板中(每孔5×105细胞),孵育24h,然后暴露于含有CEL和BA的F/CL@BM溶液中4h,浓度为0.1μg/L。细胞用PBS清洗,用钙黄绿-AM和碘化丙啶染色,然后在荧光显微镜下分析。
为了评估F/CL@BM的治疗效果,我们首先测试了它们对4T1小鼠乳腺癌细胞的杀伤能力,以及它们与NIH 3T3小鼠胚胎成纤维细胞共培养时的效果。所有的药物制剂都显示出随着浓度增加而增强的细胞杀伤效果(参见图2A和B)。特别是F/CL@BM,其效果甚至优于未经叶酸修饰的相同类型脂质体(CL@BM),这与其靶向肿瘤细胞的能力是一致的。另外,通过活/死细胞染色实验同样得出相似的结论(见图2C),这表明与NIH3T3细胞的共培养可能使肿瘤细胞对药物产生了更强的抵抗力。相比单独使用BA,BA与CEL的联合应用展现出更强的细胞毒性,这说明CEL可能有助于提升4T1细胞对BA的敏感性。
2、细胞摄取
4T1和NIH 3T3细胞按1:1比例混合后,接种于12孔板中,每孔大约有4万细胞。孵育过夜后,更换含有香豆素-6(C6)的新鲜培养基,浓度为1微克/毫升。这些培养基包括自由C6、C6胶束(MC)、C6脂质体(LC)、C6胶束-脂质体复合物(MC@LC)和表面覆盖叶酸的胶束-脂质体复合物(F/MC@LC)。两小时后,细胞进行固定和DAPI染色,随后在共聚焦激光扫描显微镜(TCS SP2,Leica)下观察。流式细胞分析实验中,收集的细胞用PBS重悬,使用流式细胞仪进行细胞内物质摄取的量化分析。图3显示,F/CL@BM的内化程度最高,甚至超过了CL@BM(见图3A)。这可能是叶酸受体介导的内化作用所致,因为在过量自由叶酸存在时,F/CL@BM的内化程度被抑制。这一现象在使用共聚焦显微镜的实验中得到了证实,并通过流式细胞仪进一步确认(图3B)。与共聚焦显微镜观察到的定性细胞摄取结果一致,如图3C所示,流式细胞仪的结果表明,改性叶酸的纳米载体对C6的摄取量超过了其他组,分别比C6组和CL@BM组高2.3倍和1.4倍(p<0.0001)。
3、对细胞迁移的影响
在六孔板上分别单独培养4T1细胞和与NIH 3T3细胞按1:1的比例混合培养,每孔接种细胞数量为10万。孵育24小时后,用无菌吸管头轻轻划破细胞层,再用PBS溶液清洗掉划破处的细胞。随后对剩下粘附的细胞进行不同处理,包括应用游离桦木酸、游离雷公藤红素、游离药物的混合物、CL、CL@BM或F/CL@BM。我们使用显微镜立即测量划痕宽度,并在24小时后再次测量。
图4展示了我们对4T1细胞进行的划痕实验,以评估不同药物制剂的抗迁移效果(见图4A)。实验结果显示,与对照组相比,所有药物制剂都能有效地抑制伤口愈合。白桦酸和雷公藤素联合使用的组别以及CL@BM处理组的抑制效果略优于单独使用雷公藤素的组别。经过配体改性的制剂处理组表现出了显著增强的抗迁移能力。如图4C所示,在F/CL@BM处理组中,细胞迁移率仅为15.92%,显著低于其他组别,表明该组合具有更强的抗迁移效果。
在肿瘤微环境中,肿瘤细胞和间质细胞之间的互动对于肿瘤的扩散起着关键作用。特别是间质细胞中的癌相关成纤维细胞(CAFs),它们能分泌促进肿瘤迁移和转移的分子。为了研究在CAFs存在的条件下不同药物制剂的抗迁移效果,我们在共培养的划痕实验模型中测试了这些药物制剂对细胞迁移的影响。如图4B和D所示,不论在哪种治疗条件下,4T1细胞在单独培养时的迁移能力都超过了共培养的情况,这说明CAFs分泌的细胞因子促进了肿瘤的迁移和转移。
4、对细胞内活性氧物质产生的影响
将4T1细胞接种在6孔板(每孔1×105细胞)或96孔板(每孔1×104细胞),孵育24h,用PBS清洗三次,然后用游离桦木酸、游离雷公藤红素、游离药物的混合物、BM、CL、CL@BM或F/CL@BM处理24h。细胞再用PBS清洗三次,用2',7'-二氯二氢荧光素二乙酸酯(DCFH-DA)孵育30min,清洗去除未结合的DCFH-DA。然后用荧光显微镜观察6孔板中荧光产物ROS,而在96孔板上使用酶标仪定量荧光产物ROS的水平(激发488nm,发射525nm)。
5、对线粒体膜电位的影响
将4T1细胞接种在六孔板中(每孔5×104细胞),孵育24h,然后用游离桦木酸、游离雷公藤红素、游离药物的混合物、CL、CL@BM或F/CL@BM处理30min。培养物用浓度为5μg/mL的JC-1在1640培养基中孵育20min,用PBS清洗,然后在荧光显微镜下观察。
BA可以穿透线粒体膜,破坏其作为物理屏障的功能,诱导产生细胞毒性的活性氧物质(ROS)。通过上述实验证实了F/CL@BM在单独培养或与NIH 3T3成纤维细胞共培养的4T1肿瘤细胞中诱导ROS产生,其效果大于任何一种药物单独使用(图5a-b)。同时,还进一步证实了这种ROS产生使线粒体膜去极化,F/CL@BM的效果强于任何一种药物单独使用(图5c)。
6、相关蛋白的表达
4T1细胞单独或与NIH 3T3细胞共同接种在6孔板中(每孔1×105细胞,NIH 3T3:4T1细胞比例为1:1),孵育24h,然后用各种药物制剂处理24h。单独培养的4T1细胞以相同方式处理作为对照。收集细胞,用RIPA缓冲液裂解,离心后取上清液通过10%的十二烷基硫酸钠-聚丙烯酰胺凝胶电泳分离,并转移到PVDF膜上。膜孵育12h,与α-SMA、Bcl-2、P-gp或GAPDH的一抗兔抗体一起孵育,然后在25℃下与山羊抗兔二抗偶联辣根过氧化物酶孵育1h。使用凝胶成像系统4.00进行成像。
如图6所示,当与4T1肿瘤细胞共培养时,NIH 3T3成纤维细胞产生了大量的α-SMA,这可能反映了CAFs的激活,而药物制剂,尤其是F/CL@BM,对此进行了下调(图6)。
同样,与成纤维细胞共培养的4T1细胞上调了P-糖蛋白1和Bcl-2,而药物制剂,特别是F/CL@BM,对此进行了下调。
实施例3F/CL@BM的靶向肿瘤疗效
1、在体内的生物分布
将携带有4T1/CAFs诱导的肿瘤(体积约200mm)的小鼠通过尾静脉注射了载DiD的胶束(BM)、载DiD的胶束-脂质体(CL@BM)和载DiD的改性叶酸胶束-脂质体(F/CL@BM)(每种条件下3只动物)。DiD的剂量为0.1mg/kg。在指定时间点,麻醉小鼠并进行红外成像。注射后24h,切除肿瘤和主要器官进行成像。
根据图7所示,虽然BM在肿瘤组织中较快消散,但在注射24小时后,肿瘤内部仍可检测到大量的F/CL@BM(图7A)。这一结果在对单独组织样本进行的离体检查中得到了验证(图7B),其中发现肿瘤组织中的F/CL@BM浓度是其他制剂的三倍(图7C)。另一方面,与肿瘤相比,CL@BM在肝脏中的积聚更为显著(图7D)。这些结果表明,叶酸修饰的脂质体在针对肿瘤的靶向中发挥了作用。
2、抗肿瘤功效
将携带有4T1/CAFs诱导的肿瘤(体积约80mm)的小鼠通过尾静脉注射了PBS(对照)、自由桦木酸、自由雷公藤红素、两种药物的物理混合物、BM、CL、CL@BM或F/CL@BM。桦木酸和雷公藤红素的剂量均为2mg/kg。
实验期间测量肿瘤体积和体重。注射后第14天,处死小鼠,切除肿瘤和器官,采集血液样本。切除的肿瘤称重、拍照后切片进行TUNEL染色。同时,肿瘤样本用10%福尔马林固定,嵌入石蜡,切片后在4℃下用α-SMA或胶原蛋白I的抗体染色过夜,然后与偶联辣根过氧化物酶的二抗孵育。通过孵育切片检测抗体结合。器官切片后进行苏木精-伊红染色。血液样本用于测量肌酐、AST和ALT的水平。
如图8所示,肿瘤体积测量显示,不同的制剂显著抑制了肿瘤生长(图8A)。然而,与其他组相比,F/CL@BM对肿瘤生长的抑制作用最强。这可能是由于BA和CEL的协同效应。切除肿瘤的代表性图像(图8B)大致符合图8C所示的肿瘤重量变化趋势。此外,F/CL@BM组的肿瘤生长抑制效果优于CL@BM,这可能是由于叶酸介导的内吞作用(p<0.05)。如图8C所示,与肿瘤体积的趋势一致,F/CL@BM组的肿瘤重量最低。此外,TUNEL染色显示,在PBS处理组中几乎没有凋亡细胞,而F/CL@BM处理组显示出最大的凋亡区域,其他组诱导了不同程度的肿瘤细胞凋亡,与上述结果一致(图8D)。
如图9所示,对F/CL@BM进行了肺转移疗效评估。如图9A所示,H&E染色的肺部图像显示除F/CL@BM组外,其他组肺部出现了不同程度的病变,表明F/CL@BM显著抑制了4T1肿瘤的肺转移。图9B的结果显示,肺部观察到大量转移结节,表明肿瘤成功侵入肺部,形成远处转移灶。联合治疗组与单药治疗相比,在肺部表面结节数量减少,表明联合治疗改善了抗转移效果。F/CL@BM组与其他药物治疗组相比,结节数量较少,表明抑制了肺转移。这一发现的可能原因是F/CL@BM能够通过减少肿瘤屏障,有效地将更多药物输送到转移区域,从而抑制肺结节的形成。因此,我们得出结论,F/CL@BM能够有效减少肿瘤组织中CAFs的激活。基于此,CAFs介导的物理屏障被打破,化疗药物的抗癌效果得到增强。
此外,图10A显示,与对照组相比,治疗组的心脏、肝脏、脾脏和肾脏无显著差异,表明纳米材料是抗肿瘤药物的安全输送载体。如图10B所示,所有组的小鼠体重没有显著变化或差异。检测的血清生化值包括ALT和AST,也没有发现差异。因此,这一发现表明,重复静脉注射制备的纳米颗粒不会导致严重的肝脏或肾脏功能障碍或并发症。因此,F/CL@BM是输送抗肿瘤药物的安全载体。
最后应说明的是,以上具体实施方式仅用以说明本发明的技术方案而非限制,尽管参照实例对本发明进行了详细说明,本领域的普通技术人员应当理解,可以对本发明的技术方案进行修改或者等同替换,而不脱离本发明技术方案的精神和范围,其均应涵盖在本发明的权利要求范围当中。
