Abstract
本发明公开了一种月桂酸甲酯在增强番茄植株抗旱性中的应用,本发明提供了月桂酸甲酯灌根处理促进干旱胁迫下番茄开花结果的方法,以开花结果期的番茄为试验材料,外源月桂酸甲酯施加后干旱胁迫下的番茄生长情况良好,相比对照植株,处理植株叶片萎蔫程度明显降低,叶片失水速度降低、坐果率显著提高,说明月桂酸甲酯具有提高番茄对干旱逆境胁迫耐受性的功能。本发明对番茄抗干旱分子机制的研究具有重要意义,为在生产中保障番茄在旱地种植时的开花结果提供了实用技术,对农业发展具重大意义。

技术领域
本发明涉及干旱胁迫下番茄生长研究技术领域,涉及一种化合物在植物抗旱中的应用;尤其是一种月桂酸甲酯在增强番茄抗旱性中的应用。
背景技术
番茄因富含维生素、营养价值高、风味独特而广受人们喜爱,是重要的农业作物,具有很高的经济价值。在栽培过程中番茄需面对和适应各种不良环境中的非生物胁迫,其中干旱是对番茄生长发育最有害的非生物胁迫之一,会导致生长量、产量及品质的下降,甚至导致植株的死亡,严重影响番茄的经济效益。因此,提高番茄的抗旱性是农业生产的研究热点。
开花是植物从营养生长转为生殖发育的重要发育节点事件,番茄在开花结果期对水分十分敏感,土壤由湿转干或植株短时间内失水过多,不仅对生长发育造成影响,还会引发花粉不育等现象进而造成落花落果和果实产量的显著下降,严重影响番茄的经济效益。因此,提高番茄在开花结果期的抗旱性是当今农业的研究热点。
现有技术中公开了多种提高番茄抗旱性的方法,如CN115807017A公开了利用番茄基因SlMAPK12提高番茄抗旱性的育种方法,其通过测定番茄植株在干旱胁迫下的一系列生理生化指标,结合干旱胁迫下的植株表型,发现SlMAPK12的RNA干涉番茄转基因植株相比于野生型番茄植株抗旱性提高,明确了SlMAPK12基因负调控番茄抗旱性,为番茄抗旱品种选育以及抗旱性机理研究提供了重要的种质资源;CN110771447A公开了一种提高持续干旱条件番茄抗旱性的栽培方法,其通过在番茄移栽前的土壤中填埋块状农用岩棉,施加基肥后进行水分灌溉,显著提高了番茄的抗旱性能及在持续干旱胁迫下番茄作物的产量;CN109111299A公开了一种番茄专用叶面肥配方,其通过配制含海藻提取物、豆渣提取液、腐植酸、甘蔗糖蜜、中微量元素、杀菌剂、膨润土的番茄专用叶面肥,有效地提高了番茄抗旱的能力。上述技术虽提高了番茄抗旱性,但也都存在成本高、工艺复杂、周期较长等缺点。
作为重要的细胞组分和能源物质,脂肪酸(Fatty acids,FA)及其衍生物在植物生长发育和应对非生物、生物胁迫等过程中均发挥着关键作用。根据碳原子数量的不同,脂肪酸可以分为短链(小于6个碳原子)、中链(6-12 个碳原子)、长链(13-24个碳原子)。重要的中链脂肪酸月桂酸含12个碳原子,可从椰子油、山苍子核油、棕榈核油、山胡椒核油、樟树子油等果核油中直接提取,作为表面活性剂和杀菌消毒剂被广泛使用。
以月桂酸和甲醇为原料,可经酯化反应生成月桂酸甲酯(Methyl Laurate)。月桂酸甲酯又称十二酸甲酯、十二烷酸甲酯,化学式为C13H26O2,为无色澄清油状液体,具有花香及酒香样气味,无毒性。作为一种绿色安全的天然脂肪酸衍生物,目前月桂酸甲酯已被广泛用于化学用料、有机合成中间体、食用香精、日化香料等,但月桂酸甲酯在激活植物抗旱性等逆境胁迫的应用还未有报道,将月桂酸甲酯应用于提高番茄抗旱性,对提高逆境条件下番茄的产量具有重要意义。
发明内容
针对上述番茄在其生长过程中易受干旱胁迫危害,影响其正常生长导致产量降低的问题,为了弥补现有技术中存在的不足,本发明的目的在于提供一种月桂酸甲酯在增强番茄抗旱能力方面的应用。
本发明的目的通过下述技术方案实现:
一种月桂酸甲酯在增强番茄植株抗旱性中的应用。
所述的应用,是在人工气候室中,使用月桂酸甲酯对开花结果期的番茄植株灌根处理后暗培养8-12 h,然后再将所述番茄植株在人工气候室中进行干旱处理培养;其中,
所述灌根处理,使用浓度为250- 500 mg/L的月桂酸甲酯溶液对番茄植株的根部直接进行浇灌;
所述人工气候室中,其温度设置为日间25℃、夜间20℃;
所述干旱处理培养,其时间为7-15d。
经由上述的技术方案可知,与现有技术相比,本发明的有益效果如下:
本发明提供了月桂酸甲酯在抗旱胁迫番茄生产中的应用。抗旱性试验表明,相对于未施加月桂酸甲酯处理的对照组植株,适当浓度月桂酸甲酯灌根处理能够减缓干旱胁迫对番茄叶片生长及开花结实造成的影响,显著提高成苗对干旱胁迫的抗性。
2、250-500 mg/L月桂酸甲酯可显著增强番茄抗旱性,500 mg/L月桂酸甲酯的提高效果最佳。与对照相比,500 mg/L月桂酸甲酯灌根处理后干旱处理15天的番茄叶片失水率显著下降。500 mg/L月桂酸甲酯灌根处理缓解了干旱胁迫对番茄叶片生长的抑制效应,在干旱胁迫下月桂酸甲酯灌根处理后的番茄地上部分相对含水量较对照处理组显著增加。
3、在番茄生殖发育过程中,灌施适宜浓度的月桂酸甲酯显著提高了干旱胁迫下番茄的果实大小和重量,降低干旱胁迫对产量的影响,提高番茄生殖期对干旱胁迫的抵御能力。
由本发明的技术方案可知,月桂酸甲酯可抑制干旱胁迫下番茄叶片的失水,显著提高干旱胁迫下番茄的果实鲜重。上述结果表明,外源施加月桂酸甲酯在调节番茄抗旱性方面具有重要作用。本发明提供的外施脂类化合物提高番茄抗旱性的诱导方法,有效解决了番茄在干旱胁迫条件下生长不良和果实产量下降的问题。对精细调控番茄的干旱响应,改善番茄对干旱胁迫的抗性的同时保持产量,有着重要意义,为其它重要粮食、经济作物的抗旱提供了研究基础和科学依据。

下面结合具体实施例,进一步阐述本发明。应理解,这些实施例仅用于说明本发明而不用于限制本发明的范围。下面将对本发明实施例中的技术方案进行清楚、完整地描述,显然,所描述的实施例仅仅是本发明一部分实施例,而不是全部的实施例。基于本发明中的实施例,本领域普通技术人员在没有做出创造性劳动前提下所获得的所有其他实施例,都属于本发明保护的范围。实施例中采用Micro-Tom为试验材料,月桂酸甲酯纯度为99%,购自阿拉丁试剂(上海)有限公司。
文中所使用的所有专业与科学用语与本领域熟练人员所熟悉的意义相同。此外,任何与所记载内容相似或均等的方法及材料皆可应用于本发明中。文中所述的较佳实施方法与材料仅作示范之用。
本发明提供了一种月桂酸甲酯在增强番茄抗旱性中的应用。
所述的应用,包括以下步骤:
1)选择长势一致、无病虫害的番茄品种两周龄幼苗进行栽培,种植于塑料方盆中;
2)在植物培养型人工气候室内,通过正常水分管理将番茄植株培养至八周大,进入开花结果期;
3)使用月桂酸甲酯溶液对番茄进行灌根处理,然后再将上述外源施加月桂酸甲酯处理后的番茄植株在人工气候室中进行黑暗培养,次日倒去拖盘内残留溶液,得到具有抗旱性的番茄。
优选的,所述步骤3)中使用浓度为250-500 mg/L的月桂酸甲酯进行灌根处理。
进一步优选的,所述步骤3中使用浓度为500 mg/L的月桂酸甲酯进行灌根处理。
优选的,所述步骤2和步骤3中人工气候室的光周期设置为16 h光照/8 h黑暗,温度设置为日间25 ℃/夜间20 ℃。
优选的,所述步骤3中黑暗培养的时间为8-12 h。
优选的,所述番茄品种为任意品种的番茄。
进一步优选的,所述番茄品种为Micro-Tom。
步骤一:选择长势一致、无病虫害的两周龄番茄‘Micro-Tom’植株栽种于底部有孔的塑料花盆中,基质为黑土、蛭石、珍珠岩混合物,每盆栽种1株。
步骤二:栽后移至人工气候培养室内养护至8周龄大进入开花结果期,进行正常水分管理,防止水分亏缺,同时预防病虫害的发生。
步骤三:采用整株灌根法,以蒸馏水作为对照(0 mg/L月桂酸甲酯),使用250 mg/L及500 mg/L月桂酸甲酯对番茄进行诱导处理,对照组设置20个重复,其余每个处理组各设置10个重复。
步骤四:黑暗培养下月桂酸甲酯处理番茄8后,倒去托盘内残余溶液,留10棵对照正常浇水,对剩余植株进行连续15天的干旱胁迫处理,期间每天进行表型特征变化的观测分析,根据‘Micro-Tom’番茄表型特征的变化,判断植株的萎蔫程度,确定其干旱胁迫的程度。
步骤五:在干旱处理后的第15天分别对不同处理的番茄植株进行拍照,记录植株整体形态特征,每组4个生物学重复。
结果如图1中显示,与对照组相比,干旱胁迫15天后,经外源施加月桂酸甲酯灌根处理的番茄植株整体更为鲜绿,且叶片萎蔫程度显著减轻。进一步比较可发现,相较于250mg/L月桂酸甲酯处理组,500 mg/L月桂酸甲酯处理下的番茄的抗旱性有更大程度的提升,表明适当浓度的外源月桂酸甲酯处理可以提高番茄对干旱胁迫的耐受性。

结果如图2中显示,统计比较干旱处理前后番茄植株的地上部分(叶)的相对含水率(RWC),对照干旱处理组的番茄在干旱胁迫后的地上部分相对含水量出现显著下降,外源施加250 mg/L月桂酸甲酯处理后的番茄在干旱胁迫后的含水量较处理前下降的幅度比对照组显著降低,外源施加500 mg/L月桂酸甲酯处理后的番茄在干旱胁迫后的含水量较处理前几乎没有明显的降低,展现出了良好的保水性。表明适当浓度的外源月桂酸甲酯处理显著增强了番茄植株抵抗干旱胁迫的能力。

步骤五:在干旱处理15天后分别对对照处理组及500 mg/L月桂酸甲酯处理的番茄植株叶片进行取样称重,每组3个生物学重复,每个重复中各取10片叶片,在人工气候培养室内25 ℃、55 %湿度条件下,每隔1 h对每个叶片用分析天平进行称重,连续记录9小时。
步骤六:以离体叶片失水前的重量为W1,以叶片失水后的重量为W2。
分别计算出番茄各处理组植株的离体叶片在失水前及失水1、 2、 3、4、5、6、7、8、9h后的叶片失水率—[ (W1-W2)/W1]×100%,绘制离体叶片失水率的柱形图,分析比较不同处理组番茄离体叶片的失水情况。
结果如图3中显示,统计比较干旱处理前后番茄植株的离体叶片失水率,对照组的番茄叶片在离体后的0-9 h内出现失水率的逐步下降,外源施加500 mg/L月桂酸甲酯处理后的番茄叶片在离体后的0-9 h内失水率的下降幅度显著减小,下降速率减缓。表明适当浓度的外源月桂酸甲酯处理显著增强了番茄植株叶片保持水分的能力。
步骤一:选择长势一致、无病虫害的两周龄番茄‘Micro-Tom’植株栽种于底部有孔的塑料花盆中,基质为黑土、蛭石、珍珠岩混合物,每盆栽种1株。
步骤二:栽后移至人工气候培养室内养护至8周龄大进入开花结果期,进行正常水分管理,防止水分亏缺,同时预防病虫害的发生。
步骤三:采用整株灌根法,以蒸馏水作为对照(0 mg/L月桂酸甲酯),使用250 mg/L及500 mg/L月桂酸甲酯对番茄进行诱导处理,对照组设置20个重复,其余每个处理组各设置10个重复。
步骤四:黑暗培养下月桂酸甲酯处理番茄12 h后,倒去托盘内残余溶液,留10棵对照正常浇水,对剩余植株进行连续15天的干旱胁迫处理,期间每天进行表型特征变化的观测分析,根据‘Micro-Tom’番茄表型特征的变化,判断植株的萎蔫程度,确定其干旱胁迫的程度。
步骤五:对干旱处理15天后不同处理组的番茄植株所结的果实进行取样拍照,每组30个生物学重复。
结果如图4中显示,与正常浇水对照组相比,干旱处理下各组的番茄果实发育都受到了显著抑制,但与对照干旱处理组相比,干旱胁迫15天后,经外源施加月桂酸甲酯灌根处理的番茄植株所结的果实更大。以上结果表明,适当浓度的外源月桂酸甲酯处理能够减缓花果期的番茄由于干旱导致果实发育受抑制的现象。

表1为不同浓度月桂酸甲酯处理后的番茄在15天干旱后的果实大小、重量、横向直径和纵向直径统计结果。如表1中显示,与正常浇水对照组相比,干旱处理下各组的番茄单株的结实数量、单株果实总重、单颗果实重均出现显著下降。但与对照干旱处理组相比,干旱胁迫15天后,经外源施加月桂酸甲酯灌根处理的番茄植株所结的果实单粒重有明显增加,且横向直径和纵向直径数值也有明显增大。500 mg/L月桂酸甲酯灌根处理组的果实单粒重及直径数值较250 mg/L处理组有进一步增加。以上结果表明,适当浓度的外源月桂酸甲酯处理能够显著提升番茄在干旱胁迫下的果实产量。

1.一种月桂酸甲酯在增强番茄植株抗旱性中的应用。
2.根据权利要求1所述的应用,其特征在于,在人工气候室中,使用月桂酸甲酯对开花结果期的番茄植株灌根处理后暗培养8-12 h,然后再将所述番茄植株在人工气候室中进行干旱处理培养;其中,
所述灌根处理,使用浓度为250- 500 mg/L的月桂酸甲酯溶液对番茄植株的根部直接进行浇灌;
所述人工气候室中,其温度设置为日间25℃、夜间20℃;
所述干旱处理培养,其时间为7-15d。
