Abstract
本发明涉及生物医药技术领域,提供了女性阴道分泌物来源的细胞外囊泡的应用。所述女性阴道分泌物来源的细胞外囊泡从健康女性阴道分泌物中提取得到,其通过携带白介素10,一方面能够提高精子活力,另一方面能够改善Th17/Treg失衡,增加胚胎着床。

技术领域
本发明涉及生物医药技术领域,具体涉及女性阴道分泌物来源的细胞外囊泡的应用。
背景技术
子宫内膜异位症(Endometriosis,EMS)是一种常见的妇科疾病,其主要症状之一是痛经和不孕。本疾病导致子宫内膜异位生长,可能影响受精卵的着床。关于EMS的相关治疗包括手术、药物等治疗,但不能有效提高妊娠率。目前关于EMS引起不孕的相关机制仍不明确,因此探索EMS相关性不孕的具体机制是目前研究的重要方向。
细胞外囊泡(EVs)是细胞释放的小囊泡,内含有生物活性分子,它们在调节细胞之间的通信和信号传递中起到关键作用。最近的研究表明,细胞外囊泡可能在生殖系统中发挥重要作用,特别是在卵子和精子的相互作用中。这些囊泡可能携带影响受精和胚胎着床的信号分子,从而对生育能力产生影响。
精子作为男性生殖系统的核心组成部分,其数量、质量和活力对于成功受精至关重要。精子的活力决定了其是否能够成功穿越女性生殖道并与卵子结合。同时,胚胎是否能着床也是妊娠成功的关键。因此,精子活力、胚胎着床问题常常被认为是不孕的一个主要因素。
研究细胞外囊泡和精子在不孕问题中的作用有助于更好地理解不孕的病因,并有望为不孕患者提供更有效的治疗和辅助生殖技术。对于不孕的调查和治疗,细胞外囊泡和精子的研究都是重要的方向,有望改善不孕患者的生育机会。
发明内容
基于此,本发明提供女性阴道分泌物来源的细胞外囊泡的应用,至少解决现有技术中的一个问题。
第一方面,本发明提供女性阴道分泌物来源的细胞外囊泡在制备治疗不孕症药物中的应用,所述女性阴道分泌物来源的细胞外囊泡从健康女性阴道分泌物中提取得到。
其中,所述健康女性是指20-40岁、黄体生成素(luteinizing hormone,LH) 2-7U/L、月经周期正常、子宫腔正常的女性。
在一些优选的实施例中,所述女性阴道分泌物来源的细胞外囊泡的制备方法包括以下步骤:
将健康女性阴道分泌物离心处理,去除细胞和细胞碎片,得到上清液;
将所述上清液与Exo-spinTM沉淀剂混合,并在4℃孵育至少1 h,得到混合液;
将所述混合液离心处理,得到含有外泌体的沉淀,所述沉淀即为所述女性阴道分泌物来源的细胞外囊泡。
在一些优选的实施例中,所述上清液与Exo-spinTM沉淀剂的体积比例为2:1。
第二方面,本发明提供一种用于治疗不孕症的药物,所述药物的活性成分为女性阴道分泌物来源的细胞外囊泡,所述女性阴道分泌物来源的细胞外囊泡从健康女性阴道分泌物中提取得到。
在一些优选的实施例中,所述药物还包含药学上可接受的载体。
在一些优选的实施例中,所述药学上可接受的载体为稀释剂,例如PBS缓冲溶液。
在一些优选的实施例中,所述药物还包含抗炎活性物质,例如白细胞介素4(IL-4)、白细胞介素10(IL-10)、白细胞介素13(IL-13)。
由于采用了以上技术方案,本发明的实施例至少具有以下有益效果:本发明首先收集健康女性阴道分泌物,然后通过Exo-spinTM技术结合沉淀法和尺寸排除色谱法获得细胞外囊泡,该细胞外囊泡通过携带的IL-10提高精子活力和胚胎着床,为不孕提供了新的临床治疗方法及策略。

具体实施方式
以下将对本发明的构思及产生的技术效果进行清楚、完整的描述,以充分地阐述本发明的目的、方案和效果。
在以下实施例中,健康女性为于2021年10月20日始收取来自南昌大学第二附属医院门诊体检正常的5例育龄女性(健康组)。健康女性的纳入标准:黄体生成素(luteinizinghormone,LH) 2-7U/L、20-40岁、月经周期正常、子宫腔正常。健康女性的排除标准:(1)认知功能障碍或患有精神类疾病;(2)伴有心、肝、肾等脏器功能障碍;(3)存在手术禁忌证;(4)存在药物禁忌证;(5)无法满足随访条件。
在以下实施例中,内异症不孕患者女性为自2021年10月20日至2022年10月20日在南昌大学第二附属医院接受治疗的5例子宫内膜异位症合并不孕患者(不孕组)。内异症不孕患者女性的纳入标准:(1)符合子宫内膜异位症诊断标准;(2)在未采取避孕措施情况下,超过1年未怀孕;(3)排除明显的子宫性、内分泌性、免疫性、排卵性不孕等其他不孕原因;(4)参与本研究前3个月内未使用激素类药物;(5)年龄20-45岁;(6)夫妇双方排除生殖器官解剖结构异常,无淋球菌、支原体和衣原体等感染、无男性不孕。内异症不孕患者女性的排除标准:(1)认知功能障碍或患有精神类疾病;(2)伴有心、肝、肾等脏器功能障碍;(3)存在手术禁忌证;(4)存在药物禁忌证;(5)无法满足随访条件。
健康组和不孕组妇女都是采集月经黄体中期(嘱入选的研究对象从月经第5-7天开始,通过B超动态检测卵巢排卵情况,于排卵后的7-9天考虑取材时间,期间嘱咐患者勿同房)的阴道分泌物,将收集的阴道分泌物标本放入4℃无菌PBS中,1 h内带回实验室,并放入-80℃冻存。
在健康组和不孕组妇女同意参与之前,每个预期的参与者都得到了一份参与者信息表,并向其解释研究的基本原理,所有参与者均提供书面知情同意书。所有的生物样本都被分配匿名的参与者身份号码,这些号码与参与者的名字在一个安全存储的链接文件中进行匹配,记录参与者姓名、年龄、性别、联系方式、病理诊断、组织学类型、手术时间、手术名称、原发或复发等。
实施例1:制备H-EVs
按照以下步骤制备健康女性阴道分泌物来源的细胞外囊泡(H-EVs):
(1) 将1 mL的健康女性的阴道分泌物样本置入离心管,以300 x g离心10 min去除细胞和细胞碎片;
(2) 将离心后的上清液置入新的离心管,并以16000 x g离心30 min去除剩余的细胞碎片;
(3) 将离心后的上清液置入新离心管中,并以2:1的比例加入Exo-spinTM沉淀剂;
(4) 上下倒置离心管数次将溶液混匀,并在4℃孵育至少1 h;取出混合液以16000x g离心1 h,小心吸出并弃去上清液(注意不要让沉淀物干燥,以免对外泌体造成损害);
(5) 用100 µL PBS重悬含有外泌体的沉淀,所述沉淀即为H-EVs。
实施例2:制备E-EVs
按照以下步骤制备内异症不孕患者女性阴道分泌物来源的细胞外囊泡(E-EVs):
(1) 将1 mL的内异症不孕患者女性的阴道分泌物样本置入离心管,以300 x g离心10 min去除细胞和细胞碎片;
(2) 将离心后的上清液置入新的离心管,并以16000 x g离心30 min去除剩余的细胞碎片;
(3) 将离心后的上清液置入新离心管中,并以2:1的比例加入Exo-spinTM沉淀剂;
(4) 上下倒置离心管数次将溶液混匀,并在4℃孵育至少1 h;取出混合液以16000x g离心1 h,小心吸出并弃去上清液(注意不要让沉淀物干燥,以免对外泌体造成损害);
(5) 用100 µL PBS重悬含有外泌体的沉淀,所述沉淀即为E-EVs。
实施例3:EVs形态、大小验证
将实施例1和2得到的EVs样品分别在PBS中以1:1000稀释,以获得每帧60-100个颗粒的浓度。进行纳米颗粒跟踪分析(NTA),使用ZetaView软件(版本8.05.14 SP7)测定尺寸分布和颗粒浓度数据。用透射电子显微镜(TEM)观察EVs形态。将重悬的细胞外囊泡样品逐滴添加到200目网格上,然后孵育10分钟;随后,用2%磷钨酸对格栅进行阴性染色,总共3分钟;除去过量的溶液,对样品进行空气干燥并进行TEM分析。
如图1-3所示,NTA和TEM分析表明,EVs为圆形或椭圆形,直径为50-200 nm的囊性结构;蛋白免疫印迹(WB)分析证实了囊泡上存在外泌体特异性标记物TSG101和CD9。以上结果表明女性阴道分泌物释放的EVs大多为外泌体。
实施例4:EVs影响正常精子活力相关实验
在本实施例中,精液样本来源于健康成年男性,每份精液样本的体积、细胞数和精子活动率等均符合世界卫生组织的第五版标准(精液体积参考值下限为1.5ml;精液pH值的参考值下限为7.2;精子密度的参考值下限为15×106/ml;总运动精子比例的参考值下限为40%,前向运动精子比例的参考值下限为32%;活精子(细胞膜完整精子)比例的参考值下限为58%;正常形态精子比例的参考值下限为4%)。
将精子分别与稀释2倍、5倍、10倍的EVs(实施例1和2得到)共培养1、4、6小时,并设置精子与PBS混合物为对照组,进行计算机辅助精子分析(CASA)。
如图4所示,计算机辅助精子分析(CASA)显示,与对照组相比,H-EVs处理后的总活力(>50%)、进行性活力水平(>40%)和平均线速度(>40µm/s)显著增加;而E-EVs处理的精子活力呈剂量依赖性下降,稀释2倍的E-EVs使精子活力最大下降(总活力,29.03% vs54.47%;精子进行性活力,22.68% vs 47.61%;线速度,20.79µm/s vs 40.42µm/s;p<0.0001)。
利用计算机辅助精子分析技术(CASA)测定EVs在不同时间点(1小时、4小时和6小时)对人类精子活力的影响。如图5所示,与对照组(PBS混合物)相比,暴露于H-EVs 6小时后,精子活力显著增加(总活力,82.8% vs 47.1%;精子进行性活力,79.03% vs 41.06%;线速度,46.76µm/s vs 41.91µm/s;静止精子,17.19% vs 52.86%);而E-EVs处理的精子在不同时间点的活力下降,在6小时时效果最大(总运动率,26.46% vs 47.1%;精子进行性运动率,18.78% vs 41.06%;线速度,21.98µm/s vs 41.91µm/s;静止精子,73.54% vs 52.86%)。
实施例5:伊红染色及爬高实验
在本实施例中,精液样本来源于健康成年男性,每份精液样本的体积、细胞数和精子活动率等均符合世界卫生组织的第五版标准(精液体积参考值下限为1.5ml;精液pH值的参考值下限为7.2;精子密度的参考值下限为15×106/ml;总运动精子比例的参考值下限为40%,前向运动精子比例的参考值下限为32%;活精子(细胞膜完整精子)比例的参考值下限为58%;正常形态精子比例的参考值下限为4%)。
首先,通过伊红Y染色评估精子活性。将人精子与实施例1和2得到的EVs(稀释2倍、5倍和10倍)在HTF中孵育1、4和6小时,并使用伊红-黑色素染色试剂盒(中国有限公司欣然生物技术有限公司)评估精子活力。未存活精子的精子头被染成红色,而存活精子保持未染色。
如图6所示,与对照组(PBS混合物)相比,在不同时间点暴露于E-EVs后,精子活性均下降,其中6小时下降最明显,而经H-EVs处理的精子活性增加。
通过爬高实验进一步研究EVs对精子穿透粘液能力的影响。将甲基纤维素与HTF培养基仔细混合,形成溶液,将其引入内径为1.0mm、长度为7.5cm的扁平毛细管中。管子的一端用橡胶粘土牢牢密封。首先,将人类精子和实施例1和2得到的EVs在37℃的温度下,在5%CO2的存在下共同孵育3小时。接下来,将毛细管引入培养混合物中,并在相同条件下共孵育1小时,然后取出。然后使用Leica DM2500立式显微镜(德国)对样品进行观察。
在离试管底部1厘米和2厘米的距离处,对照组(PBS混合物)的平均精子数分别约为88和32。如图7所示,与对照组相比,孕酮(P4)和-EVs组在1和2cm处的平均精子数量分别增加了约40%和20%。然而,E-EVs使精子的平均数量减少了约75%,P4处理逆转了这一趋势。
实施例6:测定单细胞钙离子浓度、顶体反应实验
在本实施例中,精液样本来源于健康成年男性,每份精液样本的体积、细胞数和精子活动率等均符合世界卫生组织的第五版标准(精液体积参考值下限为1.5ml;精液pH值的参考值下限为7.2;精子密度的参考值下限为15×106/ml;总运动精子比例的参考值下限为40%,前向运动精子比例的参考值下限为32%;活精子(细胞膜完整精子)比例的参考值下限为58%;正常形态精子比例的参考值下限为4%)。
检测EVs对人体精子[Ca2+]i的影响,研究EVs对人类精子功能影响的机制。精子用5μM Fluo-4 AM和0.05% Pluronic F-127(Molecular Probes,USA)处理,并在37℃下孵育30分钟。随后,用HTF培养基洗涤精子。然后将净化后的精子放置在96孔板中,并与不同浓度的实施例1和2得到的EVs(稀释2倍、5倍和10倍)孵育30分钟。使用多色V型分光光度计(TILLPhotonics GmbH,德国)对精子[Ca2+]i信号进行成像,通过带通滤波器(HQ540/50,Chroma,USA)筛选来自488nm激发和525nm发射的所得信号,这种组合在1秒的时间间隔内被仔细检查。
如图8所示,与对照组(PBS混合物)相比,H-EVs在1分钟内增加了人类精子[Ca2+]i浓度,而E-EVs导致精子[Ca2+]i浓度在2分钟后突然下降。如图9所示,精子[Ca2+]i浓度具有EVs浓度依赖性,在稀释至2倍的EVs浓度下,精子[Ca2+]i变化最大。
进一步分析EVs对人类精子获能和AR的影响。在Leica DM2500显微镜下使用340-380 nm滤光片过滤的汞激发光束观察染色精子,并使用400 nm的二色镜观察荧光发射。简言之,将人类精子在37℃下与不同浓度的EVs或PBS孵育4小时,用金霉素(CTC)染色30秒,在载玻片上并在室温下风干过夜。通过高功率荧光显微镜评估完整顶体的存在。
如图10所示,与对照组(PBS混合物)相比,H-EVs显著提高了AR比率,而E-EV降低了AR比率。此外,如图11所示,AR比率随着H-EVs的增加而增加,但随着E-EV的减少而降低,在稀释到2倍的EV浓度下显著影响了AR比率。
实施例7:ELISA检测H-EVs和E-EVs中IL-10表达水平
为了明确H-EVs和E-EVs是否通过携带IL-10改善精子活力,利用ELISA测定H-EVs和E-EVs表达水平,具体操作如下:
(1) 从室温平衡 20min 后的铝箔袋中取出所需板条,剩余板条用自封袋密封放回 4℃;
(2) 设置标准品孔(由ELISA试剂盒提供,标准品(S0-S5)浓度依次为:0、3、6、12、24、48 pg/mL)和样本孔(H-EVs、E-EVs),标准品孔各加浓度为0、3、6、12、24、48 pg/mL的标准品 50μL;
(3) 样本孔先加待测样本(H-EVs、E-EVs) 10μL,再加样本稀释液 40μL;空白孔不加;
(4) 除空白孔外,标准品孔(S0-S5)和样本孔(H-EVs、E-EVs)中每孔加入辣根过氧化物酶(HRP)标记的检测抗体 100μL,用封板膜封住反应孔,37℃水浴锅或恒温箱温育60min;
(5) 弃去液体,吸水纸上拍干,每孔加满洗涤液,静置 1min,甩去洗涤液,吸水纸上拍干,如此重复洗板 5 次;
(6) 每孔加入底物 A、B 各 50μL,37℃避光孵育 15min;
(7) 每孔加入终止液 50μL,15min 内,在 450nm 波长处测定各孔的OD 值。
在 Excel 工作表中,以标准品浓度作横坐标,对应OD 值作纵坐标,绘制出标准品线性回归曲线,按曲线方程计算各样本浓度值。下表显示了测定结果,可以看出,与E-EVs相比,H-EVs中的IL-10的表达水平明显增加。

实施例8:动物实验
在本实施例中,所用的小鼠为雄性Bacl/c小鼠(鼠龄6-8周)和雌性Bacl/c小鼠(鼠龄6-8周)。保持温度、光照(12小时光照:12小时黑暗周期)和噪音、换气等条件控制在标准的实验室条件下,在实验期间小鼠自由进水和进食。动物饲养每天给予标准颗粒饲料,每日定时清洁。为保证实验结果的准确性,对它们进行了为期一周的适应性喂养。
收集雄性小鼠的精子,具体操作如下:解剖小鼠,取小鼠附睾尾部组织放于盛装High Solution(HS)培养液的35 mm培养皿中,用注射器针头挑碎,37℃孵育20min,1600rpm离心5 min沉淀精子,再用HS培养液wash两遍去除杂质。
为了探索EVs对小鼠精子的影响,在不同的时间点(1小时、4小时和6小时)将小鼠精子与实施例1得到的H-EVs或实施例2得到的E-EVs在精子获能液(HIF)中共培养(培养条件为37℃,5% CO2),并设置小鼠精子与PBS混合物为对照组。
如图12所示,CASA测量的运动参数显示,在6小时时,E-EVs组的精子总运动性、进行性运动性和线速度显著低于对照组,而H-EVs改善了精子运动。
为了进一步研究EVs对雌性生育能力的影响,建立子宫内膜异位症小鼠模型,该模型通过超声成像和HE染色进行了验证,图13显示了囊性子宫内膜异位病变的形成,随即将小鼠分组为空白对照组(NC)、模型组(M)、H-EVs阴道移植组(H-EVs-VT)、E-EVs阴道移植组(E-EVs-VT)。将H-EVs、E-EVs和PBS(对照组)分别移植到子宫内膜异位症模型小鼠的阴道中给药1个月和2个月。
1个月的结果如图14-16所示,与空白对照组(NC)相比,子宫内膜异位症小鼠模型组(M)的受孕率(100% vs 80%)和胚胎植入部位数量(10.3±2.41 vs 7.6±3.06)降低;E-EVs使内异症小鼠受孕率进一步降低约70%,平均植入部位数为4.5±3.21;用H-EVs治疗增加了子宫内膜异位症小鼠的平均植入位点数量,但没有统计学意义。2个月的结果与1个月的相似。
实施例9:流式细胞术及HE染色实验
通过收集和匀浆小鼠子宫组织,获得悬浮液并悬浮在1ml PBS中,然后通过200目筛网并以900rpm离心10分钟。为了消除红细胞,将沉淀物重悬在2ml红细胞裂解缓冲液(eBioscience,USA)中。然后,通过200目筛过滤细胞,并用荧光标记的抗体染色,包括抗CD4-FITC(eBioscience,11-0040-82,0.5μl/test)、抗CD25-APC(BioLegend,101909,0.5μl/test)、抗IL17A-PE(eBiocience,12-7177-81,0.5μl/test)和抗Foxp3-PE(4Abio,FMPFOX-01.050,10μl/test),然后用PBS洗涤。用PBS重悬样品,随后使用流式细胞仪(FACSAria II,BD,USA)进行检查。Th17细胞群通过计算IL-17A+/CD4+细胞的百分比来定量,调节性T细胞(Treg)群通过确定Foxp3+CD25+CD4+细胞的百分比进行评估。最后,计算IL-17A+/Foxp3+CD25+比率以指示Th17/Treg平衡,这反映了子宫组织的炎症状态与百分比的关系。使用CytExpert 2.5软件对数据进行分析。
如图17所示,HE染色结果显示,与空白对照组(NC)相比,模型组(M)或 E-EVs阴道移植组(E-EVs-VT)小鼠子宫组织中的炎症细胞浸润或纤维化。如图18所示,为了证实组织学分析的结果,将CD4+IL-17A+Th17细胞群与CD4+CD25+Foxp3+Treg细胞群进行了比较,以证实子宫中的炎症反应。如图19所示,与空白对照组(NC)相比,模型组(M)Th17/Treg细胞的比例增加了4倍,E-EVs通过增加CD4+IL-17A+Th17细胞的比例和减少CD4+CD25+Foxp3+Treg细胞的比例,进一步将Th17/Treg-细胞的比例提高了5倍,而H-EVs处理后使Th17/Treg细胞的比例降低了1.6倍。
实施例10:蛋白印迹法、qRT-pcr实验
为了在mRNA水平上研究EVs对子宫炎症细胞因子的影响,通过qRT-PCR测量相对mRNA表达。从子宫组织中提取总RNA,并使用市售的逆转录试剂盒(Yeasen,11141ES60,中国)进行逆转录。然后使用Hieff UNICON®Universal Blue qPCR SYBR Green Master Mix(Yeasen,11184ES08)对所得互补DNA(cDNA)进行qRT-PCR,以检测白细胞介素6(IL-6)、IL-10、IL-17A、转化生长因子β1(TGFβ1)、ROR-γt、Foxp3和3-磷酸甘油醛脱氢酶(GAPDH)。使用2-ΔΔCt方法评估和定量细胞因子表达水平。
如图20所示,与空白对照组(NC)相比,模型组(M)抗炎细胞因子IL-10的相对mRNA表达降低(p<0.05);E-EVs降低了IL-10的表达,并增加了促炎细胞因子的相对mRNA表达,包括TGF-1β、IL-6、ROR-γt和IL-17A。而用H-EVs治疗增加了IL-10和Foxp3的表达。
另外,通过蛋白质印迹检测了子宫中与Th17分化相关的关键蛋白的水平。使用含有蛋白酶抑制剂的RIPA裂解缓冲液(Solarbio,R0010,中国)从裂解的小鼠子宫组织样品中分离蛋白质,用SDS的聚丙烯酰胺凝胶电泳(PAGE)分离并转移到含氟聚合物滤膜上,并用脱脂乳封闭2小时。随后,将膜与适当的一级抗体在4℃的温度下进行通宵孵育。随后用稀释度为1:5000的第二抗体孵育60分钟。主要抗体包括抗信号转导子和转录激活子-3(STAT3)(细胞信号技术(CST),9139S,稀释1:1000)、抗磷酸化STAT3(p-STAT3),抗CD9(Abclone,A1703,以1:1000稀释)和抗TSG101(Abcam,ab125011,以1:5000稀释)。使用抗GAPDH(CST5174S,1:1000稀释)使蛋白质表达正常化。
如图21和22所示,与空白对照组(NC)相比,模型组(M)增加了IL-17A的蛋白质表达水平和p-STAT3/STAT3比率,并降低了IL-10的表达(p=0.0003);E-EVs使IL-17A和p-STAT3/STAT3比率进一步增加了4倍以上,而用H-EVs治疗使IL-10的表达增加了约2倍。
以上所述,只是本发明的较佳实施例而已,本发明并不局限于上述实施方式,只要其以相同或等同的手段达到本发明的技术效果,都应属于本发明的保护范围。在本发明的保护范围内,其技术方案和/或实施方式可以有各种不同的修改和变化。
Claims (9)
1.女性阴道分泌物来源的细胞外囊泡在制备治疗不孕症药物中的应用,所述女性阴道分泌物来源的细胞外囊泡从健康女性阴道分泌物中提取得到。
2.根据权利要求1所述的应用,其特征在于,所述女性阴道分泌物来源的细胞外囊泡的制备方法包括以下步骤:
将健康女性阴道分泌物离心处理,去除细胞和细胞碎片,得到上清液;
将所述上清液与Exo-spinTM沉淀剂混合,并在4℃孵育至少1 h,得到混合液;
将所述混合液离心处理,得到含有外泌体的沉淀,所述沉淀即为所述女性阴道分泌物来源的细胞外囊泡。
3.根据权利要求2所述的应用,其特征在于,所述上清液与Exo-spinTM沉淀剂的体积比例为2:1。
4.一种用于治疗不孕症的药物,所述药物的活性成分为女性阴道分泌物来源的细胞外囊泡,所述女性阴道分泌物来源的细胞外囊泡从健康女性阴道分泌物中提取得到。
5.根据权利要求4所述的药物,其特征在于,所述药物还包含药学上可接受的载体。
6.根据权利要求5所述的药物,其特征在于,所述药学上可接受的载体为稀释剂。
7.根据权利要求6所述的药物,其特征在于,所述药学上可接受的载体为PBS缓冲溶液。
8.根据权利要求4所述的药物,其特征在于,所述药物还包含抗炎活性物质。
9.根据权利要求8所述的药物,其特征在于,所述抗炎活性物质为白细胞介素4、白细胞介素10、白细胞介素13中的至少一种。
