Abstract
本发明提供LDHD抑制剂及其在制备治疗和/或预防食管鳞状细胞癌药物中的用途。该LDHD抑制剂为核酸抑制剂,具体为核酶、反义分子、寡核苷酸抑制剂、适体、microRNA或siRNA。本发明提供的LDHD抑制剂能够靶向食管鳞状细胞癌肿瘤干细胞进行特异性杀伤,从根本上抑制了食管鳞状细胞癌的起始、复发、转移和耐药问题,同时规避了传统的放化疗治疗食管鳞状细胞癌给患者带来的副反应。

技术领域
本发明属于生物医药领域,具体涉及LDHD抑制剂及其在治疗和/或预防食管鳞状细胞癌的用途。
背景技术
食管癌是全球第六大致死性癌症、第八大常见肿瘤类型,参见Cao,W.等人(2020)“Multi-faceted epigenetic dysregulation of gene expression promotesesophageal squamous cell carcinoma”Nat Commun 11(1):3675。食管癌包括食管鳞状细胞癌和食管腺癌两种组织亚型,大部分病例发生在东亚和欧洲,其中50%以上的新发病例在中国,参见Xu,W.W.等人(2020)“Direct Targeting of CREB1with ImperatorinInhibits TGFbeta2-ERK Signaling to Suppress Esophageal Cancer Metastasis.”AdvSci,7(16):2000925。尽管食管腺癌在西方国家的发生率呈上升趋势,食管鳞状细胞癌依然是食管癌的主要亚型,约占食管癌总病例数的90%,参见Yang,H.等人(2020)“CCL2-CCR2axis recruits tumor associated macrophages to induce immune evasion throughPD-1signaling in esophageal carcinogenesis”Mol Cancer 19(1):41。由于缺乏早期诊断和治疗的生物靶标,食管鳞状细胞癌患者的预后较差。目前对于晚期食管鳞状细胞癌患者的治疗是以铂类为主的联合化疗方案,但由于化疗耐药、肿瘤复发等原因,食管鳞状细胞癌患者的5年生存率不足20%,参见Uhlenhopp,D.J.等人(2020)
“Epidemiology of esophageal cancer:update in global trends,etiologyand risk factors”Clinical Journal of Gastroenterology 13(6):1010-1021。近年来,分子靶向治疗成为研究热点,并在部分恶性肿瘤治疗中取得突破性进展。
因此,开发更为有效的靶点分子成为攻克食管鳞状细胞癌的关键所在。
发明内容
为了解决上述问题,本发明的目的在于提供一种LDHD抑制剂,该LDHD抑制剂能够靶向食管鳞状细胞癌肿瘤干细胞进行特异性杀伤,从根本上抑制了食管鳞状细胞癌的起始、复发、转移和耐药问题。
本发明的另一目的在于提供LDHD抑制剂在制备治疗和/或预防食管鳞状细胞癌药物中的用途。
为了实现上述目的,本发明提供一种LDHD抑制剂,该LDHD抑制剂为核酸抑制剂。
优选地,所述核酸抑制剂为核酶、反义分子、寡核苷酸抑制剂、适体、microRNA、siRNA或shRNA。
更优选地,所述LDHD抑制剂为siRNA。
具体地,所述LDHD抑制剂为siRNA-1、siRNA-2或siRNA-3,其中,siRNA-1的正向序列如Seq ID No.1所示,反向序列如Seq ID No.2所示,siRNA-2的正向序列如Seq ID No.3所示,siRNA-2的反向序列如Seq ID No.4所示,siRNA-3的正向序列如Seq ID No.5所示,siRNA-3的反向序列如Seq ID No.6所示。
优选地,所述LDHD抑制剂为shRNA。
具体地,所述LDHD抑制剂为shRNA-2、或shRNA-3,其中,shRNA-2为如Seq ID No.7所示序列、shRNA-3为Seq ID No.8所示序列。
本发明还提供上述的LDHD抑制剂在制备治疗和/或预防食管鳞状细胞癌药物中的用途。
优选地,所述食管鳞状细胞癌来自哺乳动物。
具体地,所述哺乳动物为人。
本发明提供治疗和/或预防哺乳动物食管鳞状细胞癌的方法,包括向遭受所述癌症症状的患者给药治疗有效量的LDHD抑制剂。
本发明的有益效果在于:
本发明提供LDHD抑制剂在制备治疗和/或预防食管鳞状细胞癌药物中的用途。该LDHD抑制剂能够靶向食管鳞状细胞癌肿瘤干细胞进行特异性杀伤,从根本上抑制了食管鳞状细胞癌的起始、复发、转移和耐药问题,同时规避了传统的放化疗治疗食管鳞状细胞癌给患者带来的副反应。另一方面,本发明提供的LDHD抑制剂为双链RNA的核酸分子,经济成本较低、治疗效果较好,具有可观的临床应用前景。

下面将对本发明的实施例进行详细、完善的描述,以使本发明的优点和特征能更易于被本领域技术人员理解,从而对本发明的保护范围做出更为清楚明确的界定。
肿瘤干细胞(cancer stem cell,CSC),又称“肿瘤起始细胞(tumor initiatingcell,TIC)”,是肿瘤组织中唯一具有自我更新能力,并且能够分裂、分化形成其他特定类型肿瘤细胞的极少数肿瘤细胞,占比肿瘤总体积不到1%,参见Visweswaran,M.等人(2020)“Aberrant lipid metabolism as an emerging therapeutic strategy to targetcancer stem cells”Stem Cells 38(1):6-14。肿瘤干细胞能够对称地分裂成两个肿瘤干细胞或者不对称分裂成一个肿瘤干细胞和一个子代细胞,其自我更新能力是肿瘤发生的直接原因,参见Yang,L.等人(2020)“Targeting cancer stem cell pathways for cancertherapy”Signal Transduct Target Ther 5:8。此外,肿瘤干细胞能够停滞在G0期来逃避化疗药物和放射治疗的杀伤进而产生新的肿瘤,参见Chen,W.等人(2016)“Cancer StemCell Quiescence and Plasticity as Major Challenges in Cancer Therapy”StemCells Int 2016:1740936。因此,肿瘤干细胞是肿瘤起始、转移、复发和耐药的原动力,寻找靶向肿瘤干细胞杀伤食管鳞状细胞癌的潜在靶点或可为食管鳞状细胞癌患者的临床治疗和预后评判提供理论依据。
乳酸脱氢酶(lactate dehydrogenase,LDH)为一个四聚体酶,属于二羟基酸氧化还原酶家族。人类基因组包含4个LDH基因:LDHA、LDHB、LDHC和LDHD(参见Wang,Y.等人(2018)“Prognostic value of D-lactate dehydrogenase in patients with clearcell renal cell carcinoma”Oncol Lett 16(1):866-874)。LDH包括L-LDH和D-LDH两种存在形式,大量研究表明L-LDH由LDH-A、LDH-B和LDH-C 3种亚基组成,其结构和功能明确(参见Song,K.J.等人(2018)“Expression and prognostic value of lactatedehydrogenase-A and-D subunits in human uterine myoma and uterine sarcoma”Medicine(Baltimore)
97(14):e0268)。LDHD定位于16号染色体长臂2区3带,其蛋白含有507个氨基酸。LDHD在NCBI上的登录号为Gene ID:197257,其功能是催化甲基乙二醛途径的终产物D-乳酸在线粒体内与FAD/FMN反应生成丙酮酸、FADH2和FMNH2(参见de Bari,L.等人(2019)“Synthesis and metabolism of methylglyoxal,S-D-lactoylglutathione and D-lactate in cancer and Alzheimer's disease”Ageing Res Rev 53:100915)。已有研究报道LDHD表达下调与透明细胞型肾细胞癌患者预后较差相关。关于LDHD在酵母和植物线粒体中的研究已有报道,但是关于其在人体中的研究极少,LDHD在肿瘤发生发展进程中的作用未曾报道。
“核酸抑制剂”用于文中,指如适体的核酸分子,其通过以类似于对上述抗体所描述的方式结合多肽来抑制LDHD多肽活性,或者指与编码LDHD多肽的多核苷酸互补而结合所述多核苷酸的核酸分子,其抑制多核苷酸的转录或翻译。例如,抑制性核酸可通过干扰LDHD基因的正确转录作为三螺旋形成寡核苷酸发挥作用。而且,抑制性核酸可以是核酶,其特异地结合并降解LDHD转录物。
可替换地,其可以是能够结合、降解转录物或至少抑制其有效翻译的反义(核酸)、siRNA、microRNA或shRNA。后一类型的抑制性核酸的特征在于其通常包含于LDHD转录物中的序列互补的核酸序列。该互补序列应该足够长,且应当包含足够数目的匹配核苷酸,从而允许与细胞中转录物的特异杂交。
根据本发明的“核酶”是包含与LDHD转录物互补的序列的RNA分子。核酶技术是本领域公知的,本领域技术人员能够设计和应用合适的核酶,而无需周折;参见,例如,Khan2006,Clin.Chim.Acta367(1-2):20-27;Kalota 2004,Cancer Biology&Therapy 3(1):4-12。
“反义分子”用于文中,指与LDHD转录物互补的治疗性反义RNA或能够结合LDHD转录物的吗啉代寡核苷酸。包括吗啉代寡核苷酸应用的反义技术是本领域公知的,参见,例如Kalota 2004,Cancer Biology&Therapy 3(1):4-12;
Morcos 2007,Biochem Biophys Res Commun 358(2):521-7。
抑制性寡核苷酸用于文中,优选地,指能够结合靶标基因组DNA特定的区域,从而实现基因沉默(所谓的三螺旋形成寡核苷酸)的小双链DNA分子,或者指作为诱饵发挥作用,而阻隔靶标基因转录特异地需要的转录因子的寡核苷酸。这些技术已经成功地用于体内,而且在某种程度上在治疗中已经获得结果(也参见Kalota 2004,Cancer Biology&Therapy3(1):4-12)。
术语“适体”用于文中,指特异地结合LDHD多肽的核酸适体。通过使用,例如通过指数富集的配体系统进化(SELEX)技术,可生成适体的库(pool)。能将选择步骤用于那些特异地结合LDHD多肽的适体。在特异地结合的适体中,阻断配体结合那些适体,或阻断相互作用结构域的那些适体,因而能被鉴定为本发明意义上适合的适体。用于生成适体的技术是本领域公知的,参见,例如Tuerk 1990,Science.Aug 3;249(4968):505-10;Ellington 1990,Nature.Aug 30;346(6287):818-822。
在本发明意义上的“microRNA”指单链RNA分子,其与LDHD转录物中包含的核酸序列至少部分互补。microRNA通常具有大约19至26个核苷酸长度。microRNA作为前体合成,即所谓的pri-microRNA,所述pri-microRNA具有发夹结构和形成发夹茎的两个互补的自互补区域。
术语“小干扰RNA(siRNA)”指为双链RNA剂的核酸分子,其与LDHD转录物的一部分互补,并能够碱基配对。siRNA通过特异地指导宿主细胞中的酶而发挥作用,从而切割靶标RNA。凭借siRNA序列的特异性,以及其与RNA靶标的同源性,siRNA能够引起靶标RNA链的切割,从而失活靶标RNA分子。优选地,足以调节RNAi的siRNA包含如下核酸序列,其含有靶标基因反向重复片段和目标基因编码区域(或部分)。siRNA的互补区域允许siRNA足以与靶标RNA杂交,从而调节RNAi。在哺乳动物中,siRNA是大约19-25个核苷酸的长度。
在下述实施例中,若非特意表明,所用的试剂均为分析纯,所用试剂均可从商业渠道获得。除非另外指明,本发明实施例中涉及的western blot等操作均按照“分子克隆实验指南(第三版)”(科学出版社,2002[美]J.萨姆布鲁克D.W拉塞尔著,黄培堂等译)及厂商说明书进行,细胞培养、细胞传代、细胞复苏及冻存、细胞转染等操作均按照“动物细胞培养基本技术指南(第四版)”(科学出版社,2000,[英]弗雷谢尼(R.I.)著,章静波等译)及厂商说明书进行操作。
细胞、质粒、慢病毒、siRNA、动物模型
1.人源食管鳞状细胞癌细胞系KYSE150、KYSE410、KYSE450均为商业化细胞系,由日本京都大学的YutakaShimada教授馈赠,也可以到商业网站进行购买。
2.人食管鳞状细胞癌组织芯片购买自上海芯超生物科技有限公司,产品型号HEso-Squ180Sur-03,该芯片上带有86对食管鳞状细胞癌组织和配对的癌旁组织(癌旁组织指食管鳞状细胞癌组织2cm处的组织),通过蜡块包埋切片得到组织切片。
3.GV492-Vector、GV492-LDHD、GV493-shRNA non-coding control(shRNA-NC)、GV493-shLDHD(shRNA-2、shRNA-3)质粒和慢病毒由上海吉凯基因科技有限公司构建和包装。
4.siRNA-1、siRNA-2、siRNA-3委托广州市锐博生物科技有限公司合成。

实施例1:LDHD在贴壁的食管鳞状细胞癌细胞和食管鳞状细胞癌细胞球中的表达情况
利用Western blot检测贴壁的KYSE410和KYSE450细胞及其对应的3D悬浮生长的细胞球中LDHD的表达情况。
1.3D悬浮细胞球培养
(1)2×成球实验培养基的配制:将一包DMEM/F12粉末溶解于500ml双蒸水,加碳酸氢钠与Hepes粉末,调节pH至7.4后利用0.22μm滤器过滤。使用前加入无菌的细胞因子B27(25:1),EGF(20ng/ml),FGF(20ng/ml)。
(2)2%甲基纤维素的配置:2g甲基纤维素粉末中加双蒸水定容至100ml,置于水平摇床上,完全溶解后高压蒸汽灭菌。
(3)在超净工作台中,将生长速率处于对数期的KYSE410和KYSE450细胞制成单细胞悬液。
(4)取9μl的单细胞悬液与1μl的台盼蓝染色液混合,取10μl混合液加入细胞计数板中。
(5)将细胞计数板放入细胞计数仪中,读取数据。
(6)2×成球实验培养基重悬细胞,再与2%甲基纤维素按1:1进行混匀,取2ml细胞重悬液加入超低黏附6孔板中。
(7)置于培养箱内,培养环境为37℃,95%湿度,5%CO2,隔日补加500μl1×成球实验培养基。培养14天后,分别收集3D悬浮生长的KYSE410细胞球和KYSE450细胞球。
2.细胞总蛋白的提取
(1)分别取按常规方法培养的贴壁生长状态良好的细胞KYSE410细胞和KYSE450细胞,弃去培养基,用冰预冷的1×PBS清洗三遍;然后刮取细胞,3000rpm,4℃,离心5min,收集KYSE410细胞和KYSE450细胞的细胞沉淀;
(2)分别取步骤1)培养的3D悬浮生长的KYSE410细胞球和KYSE450细胞球,3000rpm,4℃,离心5min,收集KYSE410细胞球和KYSE450细胞球的沉淀;
(3)在细胞沉淀和细胞球沉淀中加入适量蛋白裂解液(1ml RIPA/107细胞),冰上裂解30min,每10min涡旋震荡一次。
(4)12,000rpm,4℃,离心20min。
(5)上清即为细胞总蛋白,于-80℃冰箱中保存备用。
3.蛋白质浓度的测定
按照BCA蛋白浓度测定试剂盒说明书进行操作。
(1)配制BCA工作液:根据标准品和样品数量,按50体积试剂A,1体积试剂B配制适量BCA工作液,充分混匀。
(2)将蛋白标准品按0、3.125、6.25、12.5、25μl加到96孔板的蛋白标准品孔中,加去离子水补足到25μl;取2μl待测样品加入96孔板,加去离子水补足到25μl。
(3)向待测样品孔和蛋白标准品孔中加入200μl BCA工作液(即样品与工作液的体积比为1:20)混匀。
(4)37℃温浴30min。
(5)酶标仪570nm波长下测定吸光度。
(6)制作标准曲线,从标准曲线中求出样品蛋白浓度。
4.Western Blot
十二烷基硫酸钠-聚丙烯酰胺(SDS-PAGE)蛋白电泳
(1)制胶:按蛋白分子量要求制备相应浓度的SDS-PAGE分离胶和5%的积层胶,待积层胶层凝固后,小心地拔出梳子,反复冲洗上样孔,将凝胶玻璃板固定于电泳装置上,加入1×Tris-甘氨酸电泳缓冲液,加样孔用电泳缓冲液冲洗后上样。
(2)上样:取等量蛋白,加入等体积上样缓冲液,100℃加热10min变性后上样。
(3)电泳条件:80V,直至染料到达最下沿。
恒流湿转
(1)提前配制好转膜缓冲液并预冷。
(2)裁下合适大小PVDF膜,在甲醇中激活,40s-60s。
(3)取出PAGE胶,去除积层胶,将分离胶放入转膜液中,按照“白板-海绵-滤纸-膜-胶-滤纸-海绵-黑板”注意不能有气泡,海绵板装好后,连接装置。黑对黑,红对白。
(4)电泳条件:恒流0.35A,90min。
蛋白免疫印迹
(1)将PVDF膜放入封闭液(1×PBS+5%脱脂奶粉)中,室温封闭1h。
(2)用封闭液按一定比例稀释一抗,装入杂交袋中,37℃孵育2h或4℃过夜。
(3)用PBST室温漂洗3次,每次5-10min。
(4)用封闭液按一定比例稀释二抗,装入杂交袋中,室温孵育1h。
(5)将Western Blot Chemiluminescence Luminol Kit的A液和B液等量混合,滴在膜上,使之完全覆盖PVDF膜。室温作用1min。曝光,结果如图1所示,其中,Ad为贴壁细胞组,Sph为3D悬浮细胞球组。
从图1可以看出,以β-actin的表达量(即条带颜色深浅)为标准,与贴壁的KYSE410细胞和KYSE450细胞(均属于分化的非食管鳞状细胞癌干细胞)相比,LDHD在3D悬浮生长的KYSE410细胞球和KYSE450细胞球(均属于食管鳞状细胞癌干细胞)中的表达显著上调,说明LDHD驱动食管鳞状细胞癌细胞获得干性特征的能力较强,促进肿瘤起始细胞形成能力较强,从而说明LDHD在食管鳞状细胞癌发生过程中发挥重要功能。
实施例2:检测LDHD在86对食管鳞状细胞癌和配对癌旁组织中的表达情况
1.免疫组织化学实验方法检测LDHD在人食管鳞状细胞癌组织芯片中的表达
免疫组织化学实验所用试剂盒为山羊二步法检测试剂盒(山羊增强聚合物法检测系统),其中的内源性过氧化物酶阻断剂用羊血清代替以达到更好的封闭效果,其余试剂均为试剂盒所带试剂,操作按试剂盒说明书进行。
(1)将组织玻片放在烘箱中60℃烘烤2h。
(2)将玻片放入二甲苯Ⅰ中脱蜡30min,再放入二甲苯Ⅱ30min,再将片子放入无水乙醇Ⅰ5min,再放入无水乙醇Ⅱ5min,90%乙醇5min,75%乙醇5min,60%乙醇5min。
(3)抗原修复,将组织切片放入EDTA抗原修复液(pH 9.0)中,用高压锅加热3min,自然冷却至室温。
(4)用1×PBS洗3次,每次5min。
(5)将适量的3% H2O2溶液滴加在切片上,置于湿盒中孵育10min,室温。
(6)用1×PBS洗3次,每次5min。
(7)切片上滴加适量的山羊血清,置于湿盒中孵育30min,室温,弃血清。
(8)滴加适量稀释后的一抗在切片上,置于湿盒中,4℃孵育过夜。
(9)次日,用1×PBS洗3次,每次5min。
(10)滴加合适浓度的生物素标记的二抗在切片上,将切片置于湿盒中,室温孵育30min。
(11)用1×PBS洗3次,每次5min。
(12)将现配好的DAB显色液滴加适量到切片上,显微镜下观察,直至显示满意的染色程度。
(13)将玻片放置在双蒸水中,终止DAB显色,并用双蒸水洗10min。
(14)滴加苏木素到片子上进行细胞核染色,孵育2min,再用1%的盐酸酒精分化,在流动的自来水下冲洗10min;
(15)酒精脱水,将片子分别置于60%乙醇,75%乙醇,90%乙醇,无水乙醇中脱水,每次2min;
(16)将片子置于二甲苯中透明5min,置于通风橱中晾干。
(17)滴加中性树胶后,用盖玻片封片。
(18)用显微镜观察,收集图片并保存,典型照片如图2A所示。
2.数据收集及处理
组织芯片染色结果表明LDHD在86对人食管鳞状细胞癌组织样本中表达显著高于其配对的癌旁组织样本,统计学上具有显著差异(P<0.05),统计结果如图2B所示。进一步根据染色强度和阳性率进行判读打分,分为LDHD低表达组和LDHD高表达组。根据组织芯片所带病人病理资料,结合染色结果分别统计LDHD高表达组和LDHD低表达组食管鳞状细胞癌样本在生存期大于5年组和生存期小于/等于5年组中的分布数量并进行卡方检验,结果如图2C所示。LDHD低表达和高表达两组病例的生存曲线采用Kaplan-Meier法分析,两组病例的生存率差别采用Log-rank检验,结果如图2D所示,P﹤0.05表示差异具有统计学意义。从图2C和图2D可以看出,LDHD低表达组食管鳞状细胞癌患者的生存期明显长于LDHD高表达组。说明LDHD可以作为靶向食管鳞状细胞癌干细胞的靶点分子。
实施例3:过表达LDHD对食管鳞状细胞癌细胞克隆形成能力的影响。
将LDHD基因(NM_153486)的CDS序列插入到过表达慢病毒载体GV492中制成过表达质粒GV492-LDHD,并以空载体GV492-Vector作为空白对照,并将GV492-LDHD和GV492-Vector分别进行慢病毒包装,上述质粒构建和慢病毒包装委托上海吉凯基因科技有限公司完成,包装GV492-LDHD的慢病毒的滴度为1E+9TU/ml,包装GV492-Vector的慢病毒的滴度为5E+8TU/ml。

1.稳定细胞株构建
分别将1-5×105KYSE150细胞、KYSE410细胞和KYSE450细胞铺至六孔板中,待细胞贴壁后弃去原培养基,用1×PBS洗涤细胞,弃掉PBS。每孔加入1ml完全培养基和1ml病毒浓缩液(GV492-LDHD慢病毒或GV492-Vector慢病毒),混匀后放入培养箱继续培养24h,加入2μg/ml的Puromycin筛选稳定株,并设置对照组(无处理组)。待对照组细胞全部被杀死,继续用1μg/ml的Puromycin对稳转细胞株进行培养,冻存KYSE150细胞、KYSE410细胞和KYSE450细胞的稳转细胞株保种。
2.平板克隆形成实验
(1)在超净台内分别消化稳转细胞株KYSE150、KYSE410和KYSE450并制成单细胞悬液。
(2)用培养基将细胞悬液的密度调整为1250个/ml。取4ml细胞重悬液加入到6cm细胞培养皿中,轻轻混匀后放入细胞培养箱中培养。
(2)当细胞克隆长到肉眼可见时,停止细胞培养。
(3)弃原培养基,用1×PBS洗涤细胞,弃1×PBS。
(4)加入1ml甲醇溶液固定细胞,室温15min,弃甲醇。
(5)再加入1ml结晶紫染液,室温染色30min。
(6)回收结晶紫染液,小心洗去多余的染液,室温干燥。
(7)使用Image J软件统计所形成的细胞克隆数,并拍照保存,结果如图3A所示,其中,LDHD为GV492-LDHD慢病毒稳转细胞组,Vector为GV492-Vector慢病毒稳转细胞组(对照组)。
3.数据分析
利用t-test进行统计,P<0.05认为具有统计学差异,结果如图3B所示。
从图3A和图3B可以看出,过表达LDHD后KYSE150、KYSE410和KYSE450克隆形成能力显著增强,说明LDHD可以促进食管鳞状细胞癌细胞增殖。
实施例4:过表达LDHD对食管鳞状细胞癌细胞侵袭能力的影响
1.Transwell细胞侵袭实验
(1)基质胶铺板:提前一晚将基质胶放入4℃过夜融化,用无血清培养基稀释Matrigel基质胶(冰上操作,枪头预冷),上室加100ul(20-30μg/孔),放入37℃培养箱中待其凝固,出现“白色层”。
(2)在超净台内分别消化GV492-LDHD慢病毒稳转的KYSE150细胞、KYSE450细胞,和GV492-Vector慢病毒稳转的KYSE150细胞、KYSE450细胞,然后用不含血清的培养基分别将细胞重悬,并调整细胞密度为10000个/ml。
(3)分别取200μl细胞悬液加入到上室中,下室中加入1ml含20%血清的细胞培养基,放入细胞培养箱中培养24h。
(4)弃去上下室内的培养基,并在下室加入1ml甲醇溶液固定细胞,固定15min,弃甲醇。
(5)在下室内加入1ml结晶紫染液,室温染色30min。
(6)回收结晶紫染液,小心洗去多余的染液,室温干燥。
(7)用Image J软件统计穿过小室的细胞数,并拍照保存,结果如图4A所示。
2.数据分析
利用t-test进行统计,P<0.05认为具有统计学差异,结果如图4B所示。
从图4A和图4B可以看出,过表达LDHD后,KYSE150和KYSE450的侵袭能力显著增强,说明LDHD可以促进食管鳞状细胞癌细胞转移。
实施例5:过表达LDHD对食管鳞状细胞癌细胞迁移能力的影响
1.细胞划痕实验
(1)培养板划线标记:用marker笔在6孔板背后画横线,每孔至少穿过5条线,每条线均匀且平行。
(2)细胞铺板:对GV492-LDHD慢病毒稳转的KYSE150细胞、KYSE450细胞和GV492-Vector慢病毒稳转的KYSE150细胞、KYSE450细胞根据细胞的生长快慢孔内接种5-10×105个细胞,确保铺匀,过夜后细胞能够长满。
(3)细胞划线:第二天用20μl灭菌枪头或牙签,垂直孔板背后的黑线划痕,使划痕与标记线相交。
(4)洗细胞,去除划下的细胞:划线完成后,使用无菌PBS洗细胞2-3次,去除划下的细胞,使留下的间隙肉眼即清晰可见,然后更换新鲜无血清或低血清(<2%)的培养基。
(5)细胞培养与观察:将细胞放入37℃,5% CO2培养箱中培养。48小时后取出细胞,在显微镜下观察并拍照。
(6)数据分析:使用Image J软件打开图片后,随机划取6至8条水平线,计算细胞间距离的均值,结果如图5A所示。
2.数据分析
利用t-test进行统计,P<0.05认为具有统计学差异,结果如图5B所示。
从图5A和图5B可以看出,过表达LDHD可促进KYSE150和KYSE450的迁移能力,说明LDHD或可作为预防和/或治疗食管鳞状细胞癌患者转移的重要靶分子。
实施例6:干扰LDHD表达对食管鳞状细胞癌细胞克隆形成能力的影响
设计靶向LDHD的siRNA序列siRNA-1、siRNA-2、siRNA-3,其中,siRNA-1的正向序列如Seq ID No.1所示:5’-GCGUUAACCUGACGCAUAU-3’,反向序列如Seq ID No.2所示:5’-AUAUGCGUCAGGUUAACGC-3’,siRNA-2的正向序列如Seq ID No.3所示:5’-GAAGCAUUGUCGGGCAUGU-3’,siRNA-2的反向序列如Seq ID No.4所示:5’-ACAUGCCCGACAAUGCUUC-3’,siRNA-3的正向序列如Seq ID No.5所示:5’-GUGUGCCCAUCAUCCCAUU-3’,siRNA-3的反向序列如Seq ID No.6所示:5’-AAUGGGAUGAUGGGCACAC-3’。
委托广州市锐博生物科技有限公司合成siRNA-1、siRNA-2、siRNA-3,合成时在3’端增加dTdT以增加siRNA稳定性。公司同时提供siRNA-NC作为对照。
使用Lipofectamine 2000Reagent说明书进行操作将siRNA-1、siRNA-2、siRNA-3、siRNA-NC分别转染到KYSE150和KYSE450细胞中。然后进行平板克隆形成实验,具体操作同实施例3,结果如图6A所示,统计结果如图6B所示。
从图6A和6B可以看出,与对照组相比,本发明提供的siRNA-1、siRNA-2、siRNA-3通过干扰LDHD表达后KYSE150和KYSE450的平板克隆形成能力显著下调,说明本发明提供的siRNA-1、siRNA-2、siRNA-3可抑制LDHD表达,从而有效阻断食管鳞状细胞癌细胞的恶性增殖。
实施例7:干扰LDHD表达对食管鳞状细胞癌细胞侵袭能力的影响
检测实施例6制备的瞬时转染siRNA和对照组siRNA的KYSE150细胞和KYSE450细胞的细胞侵袭能力,具体操作同实施例4,结果如图7A所示,统计结果如图7B所示。
从图7A和图7B可以看出,与对照组相比,本发明提供的siRNA-1、siRNA-2、siRNA-3干扰LDHD表达后KYSE150和KYSE450的侵袭能力被显著抑制,说明本发明提供的siRNA-1、siRNA-2、siRNA-3通过下调LDHD表达可以有效抑制食管鳞状细胞癌细胞的复发转移。
实施例8:干扰LDHD表达对食管鳞状细胞癌细胞迁移能力的影响。
将上述靶向LDHD的siRNA和对照组siRNA瞬时转染到KYSE150和KYSE450细胞后进行细胞划痕实验来检测细胞迁移能力的改变,具体操作同实施例5,结果如图8A和图8B所示,统计结果如图8C所示。
从图8A、图8B和图8C可以看出,本发明提供的siRNA-1、siRNA-2、siRNA-3可干扰LDHD表达,从而显著抑制KYSE150和KYSE450的迁移能力。
实施例9:过表达LDHD对食管鳞状细胞癌细胞干性相关基因表达的影响
ABCG2、SOX2和NANOG为肿瘤干细胞调控相关基因,也称干性相关基因,这些基因的异常表达在肿瘤干细胞驱动的肿瘤起始、转移、复发和耐药等过程中发挥重要作用。
利用Western blot技术检测实施例3制备的稳定过表达LDHD后KYSE150细胞、KYSE410细胞和KYSE450细胞中干性相关基因表达的改变,具体操作同实施例1,结果如图9所示。
从图9可以看出,与对照组相比,过表达LDHD后,KYSE150、KYSE410和KYSE450细胞中干性相关基因ABCG2、SOX2和NANOG表达均显著上调,说明LDHD可以通过增强这些干性相关基因的表达来维持食管鳞状细胞癌细胞的干性特征。因此,抑制LDHD表达或可从根源上解决食管鳞状细胞癌患者术后复发转移和治疗过程中出现的耐药问题,进一步改善患者的治疗效果和生存质量。
实施例10:过表达LDHD对食管鳞状细胞癌细胞体外球体形成能力的影响
1.体外球体形成实验
(1)成球培养基配制:将2%甲基纤维素与2×DMEM/F12培养基(已加入生长因子)等比例混合均匀,即为1×成球培养基。(参见实施例1步骤1)
(2)消化细胞:将实施例2制备的稳定过表达LDHD的KYSE150细胞、KYSE410细胞和KYSE450细胞,弃掉旧培养基,加入5ml无菌1×PBS清洗细胞2次,加入2ml胰蛋白酶,37℃消化3-5min(消化时间视不同细胞系而定),显微镜下观察细胞皱缩变圆,伪足消失,细胞之间分开,部分细胞贴壁能力减弱,脱离皿底。加入2ml完全培养基重悬细胞,并将细胞悬液稀释10倍,吹打均匀成单细胞悬液后备用。
(3)细胞计数:取10μl稀释后的单细胞悬液与10μl台盼蓝混合均匀,室温静置3min,加入到细胞计数板中,利用细胞自动计数仪,计算出活细胞浓度。
(4)铺板:计算所需活细胞总量(1个细胞/μl成球培养基)。以96孔低吸附细胞培养板为例:100个细胞/100μl成球培养基/孔×(6个副孔+3个损失备用孔)=900个细胞(活细胞浓度)=所需单细胞悬液体积(样品)。
(5)37℃、5% CO2培养箱中过夜培养,次日显微镜下计数,培养箱中继续培养,7天后补加100μl成球培养基/孔,约两周可在镜下观察到细胞球(直径≥100μm),如图10A所示。
2.数据分析
利用t-test进行统计,P<0.05认为具有统计学差异,如图10B所示。
从图10A和图10B可以看出,过表达LDHD后,KYSE150、KYSE410和KYSE450的体外球体形成能力显著增强,说明LDHD可以驱动食管鳞状细胞癌的起始过程,抑制LDHD表达有望从根源上控制食管鳞状细胞癌的发生。
实施例11:过表达LDHD对食管鳞状细胞癌细胞体内成瘤能力的影响
将实施例2制备的稳定过表达LDHD的KYSE150细胞和对照组细胞消化下来,细胞计数后,各取100和1000个细胞重悬于50μl无血清的RPMI-1640培养基中,各加入50μl基质胶与细胞悬液等比例混匀后置于冰上放置。利用胰岛素注射器吸取100μl细胞基质胶混悬液接种于NOD/SCID小鼠皮下。每组5只老鼠。接种2周后开始观察,待小鼠皮下最大肿瘤直径处于10mm-15mm范围内,将对照组与LDHD过表达组小鼠同时处死,解剖后取出皮下肿瘤,并拍照保存,结果如图11A所示。另一方面,利用基于极限稀释分析的网页工具,如http://bioinf.wehi.edu.au/software/elda/计算致瘤细胞频率,P<0.05认为有统计学差异,结果如图11B所示。
从图11A和图11B可以看出,LDHD过表达可显著上调KYSE50细胞的体内致瘤能力和致瘤细胞频率,说明LDHD对于食管鳞状细胞癌干细胞干性特征的维持过程是至关重要的。
实施例12:干扰LDHD表达对食管鳞状细胞癌细胞体外球体形成能力的影响
委托上海吉凯基因科技有限公司将shRNA-2、shRNA-3序列对应的DNA序列分别插入到公司提供的GV493载体中,制成敲降质粒GV493-shRNA-2、GV493-shRNA-3并进行慢病毒包装,公司同时提供GV493-shRNA-NC质粒及包装慢病毒作为对照。
shRNA-2序列如Seq ID No.7所示:
5’-CAGACCAAGGAGGAUCUGAAUCUCGAGAUUCAGAUCCUCCUUGGUCUG-3’;shRNA-3序列如Seq ID No.8所示:
5’-CAACAGGUACAGCAAGCUGAACUCGAGUUCAGCUUGCUGUACCUGUUG-3’,其中下划线为loop。
1.稳定细胞株构建,将GV493-shRNA-2慢病毒(滴度2E+9TU/ml)、GV493-shRNA-3慢病毒(滴度2E+9TU/ml)和GV493-shRNA-NC慢病毒(滴度2.5E+9TU/ml)分别与KYSE410细胞和KYSE450细胞混合,制备干扰LDHD表达的稳定细胞株KYSE410细胞和KYSE450细胞,具体操作同实施例3。
2.细胞体外球体形成实验,将干扰LDHD表达的稳定细胞株KYSE410细胞和KYSE450细胞制备成KYSE410细胞球和KYSE450细胞球,具体操作同实施例10,结果如图12A所示。
3.数据分析
利用t-test进行统计,P<0.05认为具有统计学差异,结果如图12B所示。
从图12A和12B可以看出,与对照组相比,干扰LDHD表达可显著抑制KYSE410和KYSE450体外球体形成能力,说明LDHD对于食管鳞状细胞癌干细胞干性特征的获得是必不可少的。阻断LDHD表达可成为靶向食管鳞状细胞癌干细胞根治食管鳞状细胞癌的关键所在。
实施例13:干扰LDHD表达对食管鳞状细胞癌细胞干性相关基因表达的影响
采用实施例12的方法制备干扰LDHD表达后KYSE150细胞,利用Western blot技术检测干扰LDHD表达后KYSE150、KYSE410和KYSE450细胞中干性相关基因表达的改变,具体操作同实施例1,结果如图13所示,其中,ABCG2、SOX2和NANOG为肿瘤干细胞干性调控相关基因。
从图13可以看出,与对照组相比,干扰LDHD表达后,KYSE150、KYSE410和KYSE450细胞中干性相关基因ABCG2、SOX2和NANOG表达均显著下调,说明LDHD对于干性相关基因的表达是不可或缺的。
实施例14:干扰LDHD表达对食管鳞状细胞癌细胞体内成瘤能力的影响
将100和1000个对照组(shRNA-NC)和LDHD敲低组(shRNA-2、shRNA-3)KYSE150细胞皮下接种到NOD/SCID小鼠体内,检测抑制LDHD表达对KYSE150细胞体内成瘤能力的影响,具体操作同实施例11,结果如图14A所示,统计结果如图14B所示。
从图14A和图14B可以看出,干扰LDHD表达后,KYSE150的体内成瘤能力和致瘤细胞频率均显著下调,本发明提供的shRNA-2、shRNA-3均可下调LDHD表达,从而有效抑制动物体内食管鳞状细胞癌的发生。
从上述实施例可以看出,本发明提供的LDHD抑制剂,其中siRNA序列可以有效抑制食管鳞状细胞癌的克隆形成、侵袭和迁移能力,shRNA序列可以显著干扰食管鳞状细胞癌细胞中干性相关基因表达、体外球体形成和体内致瘤能力。
以上所述实施例仅表达了本发明的几种实施方式,其描述较为具体和详细,但并不能因此而理解为对本发明专利范围的限制。应当指出的是,对于本领域的普通技术人员来说,在不脱离本发明构思的前提下,还可以做出若干变形和改进,这些都属于本发明的保护范围。因此,本发明专利的保护范围应以所附权利要求为准。
Claims (3)
1. LDHD抑制剂在制备治疗和/或预防食管鳞状细胞癌药物中的用途,其特征在于,该LDHD抑制剂为核酸抑制剂;
其中,该核酸抑制剂为siRNA或shRNA;
该siRNA为siRNA-1、siRNA-2或siRNA-3,其中,siRNA-1的正向序列如Seq ID No.1所示,反向序列如Seq ID No.2所示,siRNA-2的正向序列如Seq ID No.3所示,siRNA-2的反向序列如Seq ID No.4所示,siRNA-3的正向序列如Seq ID No.5所示,siRNA-3的反向序列如Seq ID No.6所示;
该shRNA为shRNA-2、或shRNA-3,其中,shRNA-2如Seq ID No.7所示序列、shRNA-3如SeqID No.8所示序列。
2.如权利要求1所述的用途,其特征在于,所述食管鳞状细胞癌来自哺乳动物。
3.如权利要求2所述的用途,其特征在于,所述哺乳动物为人。
