Abstract
用于前列腺癌诊断、治疗以及预后评估的标志物及其应用。本发明对前列腺癌的发病、发展机制进行了深入研究,发现MIR210HG的表达水平是一个与前列腺癌高度关联的因素;通过抑制MIR210HG的表达能够对前列腺癌细胞产生显著的抑制作用,进而对前列腺癌的发生、发展、转移等起到显著的抑制活性。本发明对于前列腺癌的发生发展机制进行了全新的阐明,为前列腺癌的疾病进展及靶向治疗提供了充分的理论依据,为后期相关药物开发以及临床诊断和治疗以及预后评估、疗效评价提供了全新的思路,具有极大的社会意义及市场前景。
Description
技术领域
本发明属于生物医学技术领域,涉及一种用于前列腺癌诊断、治疗以及预后评估的标志物及其应用。
背景技术
前列腺癌(prostate cancer,PCa)是全球范围内常见的恶性肿瘤,是男性第2大常见恶性肿瘤。每年前列腺癌新发病患者以及因前列腺癌而死亡的患者占所有肿瘤类型的7.3%和3.8%。前列腺癌不仅对男性健康产生了极大的威胁,同时也给社会造成了巨大的经济负担。虽然目前临床上前列腺癌的治疗方式较多,如前列腺根治疗法(RP)、雄激素剥夺疗法、放射治疗、内分泌治疗和免疫治疗等,但由于前列腺癌的高度异质性,当前的临床指标如血清前列腺特异性抗原(PSA)、格林森评分(Gleason Score)和肿瘤分期等难以满足现代医学中个体化治疗的需要,这往往容易导致治疗不足或过度治疗。因此,迫切需要对前列腺癌异质性进行更深入地研究。
前列腺癌由于早期症状不够显著,且缺乏可靠的能够予以提前进行诊断的生物标志物,从而使得多数前列腺癌患者就诊时已处于进展期,尽管临床上对此期前列腺癌患者进行了积极系统的治疗,但5年总生存率仍不够理想。因此,迫切需要进一步探索前列腺癌发生发展的确切机制,以期寻找新的前列腺癌诊断、预后判定的生物标志物和潜在的治疗靶点。长链非编码RNA(Long non-coding RNA,LncRNA)是一类长度大于200个核苷酸的非编码RNA。大量研究发现,LncRNA的表达或功能异常与人类疾病的发生密切相关,其中就包括癌症、退行性神经疾病在内的多种严重危害人类健康的重大疾病,具体表现为LncRNA在序列和空间结构上的异常、表达水平的异常、与结合蛋白相互作用的异常等。LncRNAs中的m6A修饰水平较高,并且LncRNAs中的m6A修饰相关研究已在多种肿瘤中被广泛报道,例如有研究人员通过m6A-seq发现与正常脑组织相比,胶质母细胞瘤组织中m6A修饰的LncRNA的丰度明显更高,且根据实验发现m6A修饰的LncRNA WEE2-AS1通过阻止cul2介导的RPN2 K322泛素化促进RPN2蛋白稳定,从而通过激活AKT信号通路促进胶质母细胞瘤恶性进展。
由于现有技术中关于前列腺癌的发病因素尚不够明确和完善,而对于LncRNA在前列腺癌的预防、治疗以及预后过程中的相关作用及影响更是少有研究,临床治疗缺乏有效的治疗手段。通过对前列腺癌患者与健康人群体内LncRNA进行分析,寻找与前列腺癌发生、发展相关的关键mRNA靶点具有至关重要的意义,有助于为临床上进行前列腺癌的预防、诊断、治疗以及预后评估提供切实的理论依据和基础。
发明内容
本发明的目的在于解决现有技术中所存在的前列腺癌发病因素不明确、难以进行提前预防的技术问题,从而提供了一种与前列腺癌发生、发展高度相关的LncRNA靶点,即MIR210HG。通过以MIR210HG作为标志物,能够用于对前列腺癌疾病的发生发展过程进行有效预测,从而筛选出前列腺癌的高发病人群,以进行合理的提前预防,显著降低疾病发生的概率及严重程度,减少对人体健康的损害。
为了解决上述技术问题,本发明是通过如下技术方案得以实现的。
本发明第一方面提供了MIR210HG抑制剂在制备用于预防和/或治疗前列腺癌的药物中的应用。
应理解的是,在没有特别说明的情况下,在本发明上下文中,所述MIR210HG抑制剂是指能够特异性下调MIR210HG表达水平和/或其成熟mRNA的转录水平和/或MIR210HG蛋白表达水平或活性的物质,例如采用反义寡核苷酸、siRNA、shRNA、sgRNA、antagomiRs、miRNA海绵、miRNA Erasers、Target Masking和/或多靶点等方法以下调MIR210HG表达水平和/活性,只要是能够实现MIR210HG的水平和/或活性降低均可。
作为优选地,所述MIR210HG抑制剂选自基于MIR210HG基因设计的siRNA。
作为优选地,所述基于MIR210HG基因设计的siRNA选自si-1、si-2、si-3中的一种或多种,其中si-1的序列为CAACACAGTTCACAATATA,si-2的序列为GAGCTAACTTACTGCCAGA,si-3的序列为GAAATAACCAAGCCGAGTT。
本发明第二方面提供了检测MIR210HG表达水平的试剂在制备用于前列腺癌辅助诊断和/或预后评估的产品中的应用。

应理解的是,在没有特别说明的情况下,在本发明上下文中,所述引物和/或引物对是指用于在PCR中合成MIR210HG基因cDNA链的PCR引物,从而用于检测MIR210HG基因mRNA的表达水平。除本发明所列出的引物和/或引物对外,本领域技术人员完全有能力根据MIR210HG的基因序列采用包括但不限于分子生物学等本领域的常规方法手段进行相应引物和/或引物对的设计,并通过常规实验手段对所设计的引物和/或引物对进行筛选,只要能够实现特异性检测MIR210HG表达水平即可。

应理解的是,在没有特别说明的情况下,本发明上下文中所述的“模型”即为临床预测模型,本领域技术人员能够理解所述的“模型”为一种产品。临床上通过使用参数/半参数/非参数数学模型来评估受试者当前患有某种疾病的概率或将来发生某种结局的可能性。通过该模型,利用已知特征来计算未知结局发生的概率。临床预测模型一般采用各种回归分析方法建模,回归分析的统计本质是寻找一种定量因果关系,可用于对疾病进行诊断和/或预后评估,从而在诊断治疗决策、患者预后管理、公共卫生资源配置等方面发挥作用。
本发明通过收集来自患者的原发性前列腺癌和转移性前列腺癌样本,并利用MeRIP-seq来定位m6a甲基化的LncRNAs。随后于3个多中心队列中构建并验证了一种基于9个甲基化差异LncRNAs的前列腺癌预后指标mLs。结果表明,m6A修饰的差异与m6A修饰的Lncrna表达的改变呈正相关,mLs高得分组患者的生化复发发生时间更早。基于该9个LncRNA进行深入分析发现,MIR210HG是与前列腺癌相关的风险因素最高的标志物之一,其在前列腺癌患者体内表达异常表达。
MIR210HG目前已被证实与多种肿瘤预后相关,如肝细胞癌、宫颈癌和多形性胶质母细胞瘤等。然而,目前尚缺乏MIR210HG在前列腺癌中的具体研究。因此以MIR210HG作为靶点开展了一系列的体内外实验。结果显示MIR210HG在前列腺癌组织中表达显著升高,且与患者生存预后、BCR等密切相关。且MIR210HG作为一个促癌因子,可以促进前列腺癌增殖、克隆形成、迁移和侵袭;而当对MIR210HG的表达进行抑制后,则能够显著抑制前列腺癌的增殖、克隆形成、迁移和侵袭能力。体内实验发现,在对MIR210HG进行抑制后,无论是肿瘤的生长速度还是肿瘤的大小明显得到有效的控制;表明MIR210HG能够促进裸鼠前列腺癌细胞异种移植瘤生长,当抑制MIR210HG的表达时,则能够显著抑制肿瘤的生长。进一步的分析发现,MIR210HG的高表达与TNFA signaling via NFKB、inflammatory response、allograftrejection、complement、KRAS signaling和UV response等通路相关。MIR210HG低表达则主要与E2F targets、MYC targets、oxidative phosphorylation、DNA repair和interferonalpha等通路相关。且MIR210HG的异常表达与KIRC、CESC、LUSC、UCEC、LIHC、KICH、UVM和THCA癌症的生存情况和预后相关。
本发明相对于现有技术具有如下技术效果:
(1)本发明对前列腺癌的发病、发展机制进行了深入研究,发现MIR210HG的表达水平是一个与前列腺癌高度关联的因素;进而,通过对受试者体内MIR210HG进行检测,获得其表达水平指标,从而可以有效对受试者罹患前列腺癌的几率进行合理预测,即MIR210HG的表达水平可以作为临床辅助诊断前列腺癌疾病的生物标志物,当MIR210HG表达水平明显增高时,则可以明确受试者为前列腺癌患者或罹患前列腺癌的高风险人群,有效防止疾病的迅速发展和恶化对患者造成不可以的健康损害。
(2)通过对前列腺癌患者体内MIR210HG表达水平进行检测,可以判断对患者体内肿瘤分化程度进行合理预测,从而提供个性化的治疗方案,以改善临床治疗效果;也可对患者的预后进行合理评估,为治疗及康复提供合理有效的指导作用。
(3)本发明明确了通过抑制MIR210HG的表达能够对前列腺癌细胞产生显著的抑制作用,进而对前列腺癌的发生、发展、转移等起到显著的抑制活性,对于前列腺癌的发生发展机制进行了全新的阐明,为前列腺癌的疾病进展及靶向治疗提供了充分的理论依据,为后期相关药物开发以及临床诊断和治疗以及预后评估、疗效评价提供了全新的思路,具有极大的社会意义及市场前景。

具体实施方式
为使本发明的目的、技术方案及效果更加清楚、明确,以下参照实施例对本发明作进一步详细说明。应当理解,此处所描述的具体实施例仅用以解释本发明,并不用于限定本发明。
在无特别说明的情况下,本发明上下文中所使用的试剂中,均通过市售获得。前列腺癌细胞系PC-3、22Rv1、C4-2和C4-2B均购自于BeNa Culture Collection。22Rv1、C4-2和C4-2B在RPMI-1640培养基中培养;PC-3在DMEM培养基中培养。两种培养基中均添加了10%胎牛血清和1%双抗生素(青霉素和链霉素)。所有细胞系均在37℃恒温和5% CO2的培养箱中培养。对于临床样本的获得,均来自于广州医科大学附属第一医院,临床样本使用,均与患者签署了知情同意书,相关程序及方法符合医学伦理学要求以及药物临床试验质量管理规范。本发明所使用的实验方法,例如分子生物学实验、细胞实验、模型构建、生物信息学分析、免疫组化等均为本领域的常规方法和技术。
生物学实验重复中选择具有代表性的结果呈现在上下文附图中,数据按照图示中规定的以mean±SD和mean±SEM展示。所有实验至少重复三次。数据的统计分析和可视化由R软件(version 4.1.0)和GraphPad Prism 8.0完成。Pearson和Spearman相关性分析用于计算相关系数。使用Wilcoxon秩和检验或Student’s t-tests比较分析两个连续变量之间的差异,卡方检验或Fisher精确检验用于分类变量间的比较。p<0.05被认为是一个显著的差异。
实施例1
(1)获取原发性前列腺癌患者肿瘤组织样本(primary组)和转移性前列腺癌患者肿瘤组织样本(metastatic组),使用Trizol从样品中提取总RNA,并使用NanoDrop ND-1000(Thermo Fisher Scientific)评估质量,随后分别利用MeRIP-seq和RNA-seq分析对其主要成分(PCA)进行分析,并获得各组中的m6A峰及LncRNA数量。其中RNA-seq、MeRIP-seq均按照本领域的常规方法进行,具体地,对于RNA-seq,首先使用rRNA Depletion Kit(New England Biolabs)去除rRNA;随后,使用TruSeq Stranded Total RNA Library PrepKit(Illumina)构建RNA文库;最后于Illumina NovaSeq 6000上进行测序。对于MeRIP-seq,首先使用/>m6A-IP kit(GenSeq)免疫沉淀RNA;随后使用/>UltraIIDirectional RNA Library Prep Kit(New England Biolabs)构建RNA库;最后于NovaSeq平台(Illumina)上进行测序。
结果如图1-2所示。结果显示,primary组和metastasis组在PCA方面基本没有重叠(参见图1)。在primary组中,共鉴定出11313个m6A峰,对应7931个LncRNA;在metastasis组中,共鉴定出10971个m6A峰,对应7802个LncRNA。在这两组中,重叠的m6A peaks和LncRNA数量分别为7696和6013,而primary组特有的m6A峰和LncRNA数量分别为3617和1918,metastasis组特有的m6A peaks和LncRNA数量分别为3275,1789(参见图2)。
(2)对primary组和metastasis组中重叠的m6A峰和LncRNA进行分析,并按照如下条件对所获得的重叠的m6A峰及LncRNA进行差异表达识别,以获得两组中具有显著差异丰度的m6A峰和差异表达的LncRNA:
p<0.05且|fold change|>2。
分析结果如图3-4所示。结果显示,primary组和metastasis组的7696个m6A重叠峰中有3712个具有显著差异丰度,其中有1764个显著低甲基化(hypo-methylated)的m6A峰和1948个显著高甲基化(hyper-methylated)的m6A峰(参见图3)。此外,两组共有的6013个LncRNA中有92个存在差异表达,其中包括54个表达下调的LncRNAs和38个表达上调的LncRNAs(参见图4)。
(3)为了研究m6A methylation与lncRNA表达量之间的关系,进一步对步骤(2)所获得的primary组和metastasis组中具有差异丰度的m6A峰和差异表达的LncRNA进行交叉分析(cross-analysis),获得同时具有甲基化差异以及表达水平差异的LncRNA。
结果如图5-6所示。结果显示,primary组和metastasis组之间存在21个同时具有甲基化差异以及表达水平差异的LncRNA,其中10个高甲基化的LncRNA显著上调,11个低甲基化的LncRNA显著下调(参见图5)。通过对MeRIP-seq和RNA-seq数据的相关性分析进一步证明了m6A峰差异丰度与LncRNA差异表达量之间呈现明显的正相关性(参见图6)。
(4)利用PCaDB工具(http://bioinfo.jialab-ucr.org/PCaDB/)中“Transcriptome Analysis-Prognostic Model”模块的“CoxLasso”方法的对步骤(3)获得的同时具有甲基化差异以及表达水平差异的LncRNA进行分析,筛选得到9个与前列腺癌具有显著相关性的LncRNA,分别为COL1A1、MIR210HG、AL158071.5、SNHG8、AL390719.2、LINC00920、PCA3、LINC01088和AF165147.1。
(5)基于步骤(4)中9个与前列腺癌具有显著相关性的LncRNA按照如下公式进行mLs的计算:
其中N代表所检测的标志物的数量,本实施例中N=9;Coefi表示所检测的各标志物的系数,Expression level ofRNAi表示所检测的各标志物RNA的相对表达水平,并利用Survminer包(version 0.4.9)确定cut-off值;当mLs评分未超出预设范围时,则说明受试者预后风险较低;当mLs评分超出预设范围时,则说明前列腺癌预后风险较高,且mLs评分与风险呈正比,即mLs评分越高,风险越高。
本实施例中,各标志物的Coefii如下表1所示。

为了评估mLs的预后价值,以TCGA-PRAD作为训练集,以Taylor和GSE54460作为验证集,采用Kaplan-Meier进行生存分析。分析结果如图7-8所示(图7-8)。结果显示,在TCGA-PRAD训练集和2个验证集中,mLs高得分组患者的生存率状况明显不如mLs低得分组患者,且mLs高得分组患者的BCR发生时间均明显早于mLs低得分组患者。
此外,在包括mLs、前列腺癌格利森评分(Gleason Score)、年龄、TNM分期和前列腺特异性抗原(Prostate-Specific Antigen(PSA))等临床因素在内的单、多因素Cox回归分析表明,在3个长期队列中,mLs均可作为前列腺癌BCR的独立危险因素(p<0.05,HR>1)(参见图9)。通过在3个队列中对各个临床因素变量的C指数进行计算比较发现,mLs的表现与其他几个临床指标相当甚至优于其他临床指标,尤其在TCGA-PRAD队列中(参见图10)。这表明mLs在大多数长期队列中具有可靠稳定的预测性能。
实施例2
前述实施例中筛选了与前列腺癌相关的9个LncRNA标志物,并基于该9个LncRNA建立了用于前列腺癌预测的mLs模型,通过检测该9个LncRNA在前列腺癌患者体内的相对表达水平,从而可以计算出对应的mLs得分,进而对患者的疾病发展及预后等作出合理预测及评估。为了获取对前列腺癌具有显著影响的关键靶点,通过对上述9个LncRNA进行深入分析发现,MIR210HG是与前列腺癌相关的风险因素最高的标志物之一,对此选择MIR210HG进行相关功能性验证。
首先,采用RT-qPCR对其中前列腺癌细胞中MIR210HG的表达水平进行检测,具体步骤如下:
(1)将前列腺癌细胞(PC-3、22Rv1、C4-2和C4-2B)分别培养于6孔板中,待细胞生长至对数期时分别收集1×106个前列腺癌细胞。
(2)向细胞液中加入1mL Trizol溶液,吹打混匀使细胞充分裂解,静置5min;加入200μL氯仿,剧烈振荡混匀30s,使水相和有机相充分解除,室温静置2min。
(3)4℃下12000g离心10min,可见分为三层,其中RNA在上层水相中,转移至新的RNase-free EP管中;加入等体积异丙醇,轻柔地充分混匀,于-20℃静置10min。
(4)4℃下12000g离心10min,收集RNA沉淀,取上清,用75%乙醇洗涤两次,超净台风干,加入20-60μL DEPC水溶解沉淀;总RNA的纯度控制OD260/OD280值在1.8-2.2,同时用1%琼脂糖凝胶电泳检测RNA的完整性,RNA置于-80℃保存。
(5)取1μg总RNA作为模板RNA,采用EasyScript First-Strand cDNA SynthesisSuperMix(TransGen,货号AE301-02)进行逆转录,逆转录反应体系按照试剂盒说明书或者本领域的常规体系进行,反应条件为:25℃保温10min;50℃保温30min;85℃保温5min。
(6)采用RT-qPCR检测前列腺癌细胞中MIR210HG表达水平,所采用的序列如下:MIR210HG-F:GGCAGATTTAGTGGACGCCT;MIR210HG-R:GTTAGCTCTGCAGGTGTGGA;qRT-PCR反应过程按照Power SYBR Green PCR Master Mix说明书操作;通过融解曲线分析和电泳确定目的条带,ΔΔCT法进行相对定量。
检测结果如图11所示。结果显示,在前列腺癌细胞中MIR210HG表达水平明显升高(*p<0.05,**p<0.01,***p<0.001,****p<0.0001,以正常前列腺上皮细胞系中MIR210HG表达水平作为对照)。
随后,收集前列腺癌患者的肿瘤组织进行FISH分析,具体步骤如下:将前列腺癌细胞于15mm的共聚焦培养皿(JET BIOFIL)中进行培养,针对homo-MIR210HG进行探针序列设计(Ribo Bio),按照RiboTM荧光原位杂交试剂盒的说明书进行操作和分析,样品使用Olympus激光扫描显微镜(FV3000)进行检测。检测结果如图12所示,结果显示,MIR210HG主要定位于前列腺癌细胞的细胞质中。
考虑到MIR210HG的表达与mLs模型中的m6A修饰密切相关,进一步开展了MeRIP-qPCR assay,具体步骤如下:收集前列腺癌细胞(22Rv1、PC-3),使用Trizol从样品中提取总RNA,并使用NanoDrop ND-1000(Thermo Fisher Scientific)评估质量,将RNA片段化并按一定比例作为输入对照。针对MeRIP-seq结果鉴定的甲基化位点设计引物(MIR210HG-F:GGCAGATTTAGTGGACGCCT;MIR210HG-R:GTTAGCTCTGCAGGTGTGGA),利用上述引物进行qPCR,进一步分析m6A的富集情况。通过将扩增周期值归一化到相应的输入部分,计算MIR210HG基因上m6A的相对富集程度。分析结果如图13所示,结果显示,在PC-3和22Rv1细胞系中抗m6A抗体可以显著富集MIR210HG,这表明MIR210HG可受到m6A直接修饰。
实施例3
为了验证MIR210HG对前列腺癌的作用,基于MIR210HG序列设计并合成了三种siRNA,其序列分别为:si-1:CAACACAGTTCACAATATA;si-2:GAGCTAACTTACTGCCAGA;si-3:GAAATAACCAAGCCGAGTT)。将其分别转染至PC-3和22Rv1细胞中,通过上述RT-qPCR对其中MIR210HG表达情况进行检测。结果显示,si-1和si-3均能够对MIR210HG的表达产生有效抑制作用,显著下调PC-3和22Rv1细胞中MIR210HG的表达水平(参见图14)。
随后,分别选择对MIR210HG具有显著抑制活性的si-1和si-3开展体外细胞功能性实验,首先进行细胞增殖实验研究,具体步骤如下:
(1)分别取对数生长期的转染si-1或si-3及阴性对照质粒(si-NC)的PC-3和22Rv1细胞,胰酶消化并计数,按各种细胞的倍增时间选择合适的细胞密度,接种到96孔板中(3个重复)。
(2)于37℃培养箱中进行培养,并分别于24h、48h、72h、96h、120h收集细胞,每孔加10μL CCK-8,将培养板在培养箱内孵育1-4h,于450nm测定吸光度,评估细胞的增殖状况。
实验结果如图15所示。结果显示,相较于空白对照组(si-NC),在利用抑制剂对MIR210HG的表达水平进行抑制后,能够显著降低前列腺癌细胞的增殖,差异具有统计学意义。
随后,通过集落形成实验研究MIR210HG对前列腺癌细胞的影响,具体步骤如下:
(1)取对数生长期的转染有si-1、si-3以及空白载体(si-NC)的PC-3和22Rv1细胞,分别用0.25%胰蛋白酶消化并吹打成单个细胞,并把细胞悬浮在10%胎牛血清的DMEM培养液中备用。
(2)将细胞悬液后稀释,按1000/孔的密度接种于含10mL 37℃预温培养液的皿中,并轻轻转动,使细胞分散均匀,置37℃5% CO2及饱和湿度的细胞培养箱中培养。
(3)培养48h至培养皿中出现肉眼可见的克隆时,终止培养,弃去上清液,用PBS小心浸洗2次,每孔加入含0.5%结晶紫的甲醇1mL,染色30min;弃甲醇,用水清洗残留的甲醇;即可观察到细胞克隆;显微镜下观察,细胞数>50个才计为一个有效克隆。
结果如图16所示。结果显示,与空白对照组(vector)相比,利用siRNA对MIR210HG表达水平进行抑制后能够显著抑制前列腺癌细胞集落形成,差异具有统计学意义。
进一步地,采用corning公司transwell migration的8μm孔径的小室及24孔板进行细胞迁移实验,具体步骤如下:
(1)将小室放置于24孔板中,每孔加入1mL基础培养基润湿。
(2)将对数生长期的转染有si-1、si-3以及空白载体(si-NC)的PC-3和22Rv1细胞利用胰酶消化后,基础培养基重悬成细胞悬液,并计数,调整细胞密度为5×105个/mL。
(3)将小室内及24孔板里的基础培养基吸取并弃去,取200μL细胞悬液加入transwell小室上室,24孔培养板下室内加入600μL完全培养基(基础培养基+10%胎牛血清)。
(4)培养板置于37℃的CO2培养箱中,继续培养14小时。
(5)取出小室,PBS淋洗2遍,在24孔板内4%多聚甲醛固定20min,结晶紫溶液染色15min。
(6)用棉签小心擦去小室微孔膜上层内的细胞,倒置显微镜下拍照。
结果如图17所示。结果显示,与空白对照组(si-NC)相比,在利用抑制剂对MIR210HG的表达水平进行抑制或能够显著抑制前列腺癌细胞迁移和侵袭,差异具有统计学意义。
前述实验在体外验证了通过抑制MIR210HG的表达能够有效抑制前列腺癌细胞增殖、克隆形成、迁移以及侵袭等功能。为了进一步明确MIR210HG对于前列腺癌的功能,进一步通过体内实验加以验证,具体步骤如下:
(1)实验前一天,将已经分装好的Matrigel基质胶从-20℃提前放入4℃冰箱过夜,使其从固体状态融化为液体状态;
(2)根据MIR210HG基因编码区序列,利用si-1构建稳定敲低MIR210HG表达的慢病毒,将其感染22Rv1细胞,构建成稳定敲低MIR210HG表达的前列腺癌细胞株(si-1),对照组细胞采用空白载体(si-NC)进行构建;
(3)取10只4周龄Balb/c-nu小鼠,随机分为2组,记为组1-组2,分别于背部皮下注射肿瘤细胞;其中组1小鼠皮下注射稳定敲低MIR210HG表达的22Rv1细胞株(si-1),组2小鼠皮下转染有空白载体的22Rv1细胞株(si-NC)作为对照;
(3)每日观察小鼠生长及精神状况,待成瘤后,定期测量各组小鼠的肿瘤大小,计算肿瘤体积,其中肿瘤体积采用如下公式计算:Volume(mm3)=Length(mm)×Width2(mm2)/2。
(4)在注射细胞后32天处死小鼠,剥离各组小鼠肿瘤并拍照。
实验结果如图18-19所示。结果显示,与阴性对照组(si-NC,组2)相比较,在对MIR210HG进行抑制后(si-1,组1),无论是肿瘤的生长速度还是肿瘤的大小明显得到有效的控制;表明MIR210HG能够促进裸鼠前列腺癌细胞异种移植瘤生长,当抑制MIR210HG的表达时,则能够显著抑制肿瘤的生长。
综合上述结果可知,MIR210HG可作为前列腺癌的危险预测因子,其高表达可促进前列腺癌的进展,且通过对MIR210HG的表达水平进行抑制,则能够显著抑制前列腺癌的增殖、转移等活动功能。
实施例4
为了深入研究MIR210HG的功能,通过ChIRP实验来明确可能与MIR210HG结合的miRNA,本实用采用市售的ChIRP试剂盒(gzscbio),并按照其说明书开展检测,具体步骤如下:首先将PC-3细胞于室温下用1%的甲醛进行交联处理10min;随后使用裂解缓冲液将前列腺癌细胞进行裂解,并进行超声处理以获得RNA片段;进一步地,将细胞裂解物和用于与MIR210HG杂交的生物标记探针共同孵育;随之加入链霉亲和素以分离探针结合复合物;最后对分离物进行洗涤后进对提取获得的RNA进行测序处理。
结果如图20-21所示。结果显示,与input组相比,MIR210HG组显著富集了85个miRNA。基于MIR210HG和这85个miRNA,随后在TCGA-PRAD数据集中利用R包“GDCRNATools”(version 1.18.0)来探究其潜在的ceRNA网络。根据纳入标准(p<0.05和|correlationcoefficients|>0.4),在获取的所有LncRNA-miRNA-mRNA对及其p值和相关性系数结果中鉴定出了由77个分子(其中包括1个LncRNA、17个miRNA和59个mRNA)组成的ceRNA网络,然后使用“Cytoscape”软件(version 3.9.1)进行可视化。网格中不同的节点形状表示不同的基因类型:其中方形代表LncRNA,三角形代表miRNAs,圆形代表mRNAs。进一步地,对MIR210HG相关的功能通路进行了研究。结果显示,在MIR210HG高表达组中主要富集了TNFAsignaling via NFKB、inflammatory response、allograft rejection、complement、KRASsignaling和UV response等通路。而MIR210HG低表达组则主要与E2F targets、MYCtargets、oxidative phosphorylation、DNA repair和interferon alpha等通路相关。
接下来对MIR210HG进行了泛癌分析。结果表明,KIRC、CESC、LUSC和UCEC(UterineCorpus Endometrial Carcinoma)等肿瘤中MIR210HG的表达量相对较高,而LIHC、KICH、UVM和THCA(Thyroid carcinoma)等肿瘤中MIR210HG的表达量较低(参见图22)。通过Kaplan-Meier生存分析表明,MIR210HG高表达组患者的DSS、OS和PFI均显著缩短(p<0.05)(参见图23)。单因素Cox分析结果表明,在GBM、THYM(Thymoma)、LIHC、KICH、CESC、ACC、PCPG(Pheochromocytoma and Paraganglioma)和SARC(Sarcoma)中,MIR210HG是PFI的危险因素(参见图24)。
由上述结果可知,MIR210HG在前列腺癌组织或细胞中高表达,从而可以明确MIR210HG能够用于前列腺癌疾病的提前诊断。可以通过检测患者体内MIR210HG表达水平,对其预后进行合理预测,同时选择个性化治疗方案,以改善临床治疗效果。除此以外,MIR210HG的高表达的无论是对于细胞增殖、迁移还是侵袭性能均有明显的促进作用,即MIR210HG属于一个特异性的前列腺癌促癌因子;而当抑制MIR210HG的表达时,则能够有效降低前列腺癌细胞的各项功能活性。由此可以明确通过抑制MIR210HG表达能够对前列腺癌细胞产生显著的抑制作用,进而对前列腺癌的发生、发展、转移等起到显著的抑制活性,MIR210HG可作为前列腺癌无创诊断、预后判断及精准靶向治疗的新靶标。同时,本发明对MIR210HG相关作用信号通路进行了深入分析,并开展了一系列泛癌研究,对于后续研究中相关目标靶点的获取、关联性疾病的治疗等提供新的思路,且有助于为后续的药物研发、临床治疗等提供明确的方向,具有极高的社会价值和市场应用前景。

1.MIR210HG抑制剂在制备用于预防和/或治疗前列腺癌的药物中的应用。
2.根据权利要求1所述的应用,其特征在于,所述MIR210HG抑制剂选自基于MIR210HG基因设计的siRNA。
3.检测MIR210HG表达水平的试剂在制备用于前列腺癌辅助诊断和/或预后评估的产品中的应用。
4.根据权利要求3所述的应用,其特征在于,所述检测MIR210HG表达水平的试剂包括用于检测MIR210HG基因表达水平的引物对。
5.一种用于前列腺癌辅助诊断和/或预后评估的试剂盒,其特征在于,包括用于检测MIR210HG基因表达水平的引物对。
6.根据权利要求5所述的试剂盒,其特征在于,所述试剂盒还包括用于检测除MIR210HG以外的基因表达水平的引物对。
7.根据权利要求6所述的试剂盒,其特征在于,所述除MIR210HG以外的基因选自COL1A1、AL158071.5、SNHG8、AL390719.2、LINC00920、PCA3、LINC01088、AF165147.1中的一种或多种。
8.一种用于前列腺癌辅助诊断和/或预后评估的芯片,其特征在于,包括固相载体,以及固定在所述固相载体上的寡核苷酸探针;所述寡核苷酸探针包括对MIR210HG表达水平进行特异性检测的引物对。
9.根据权利要求8所述的芯片,其特征在于,所述芯片还包括用于对除MI R210HG以外的基因表达水平进行特异性检测的引物对。
10.根据权利要求9所述的芯片,其特征在于,所述除MIR210HG以外的基因选自COL1A1、AL158071.5、SNHG8、AL390719.2、LINC00920、PCA3、LINC01088、AF165147.1中的一种或多种。
