技术领域
本发明涉及基因工程技术领域,具体涉及一种II型L-天冬酰胺酶及其应用。
背景技术
II型L-天冬酰胺酶(L-asparaginase,E.C.3.5.1.1)能将天冬酰胺(Asn)催化成天冬氨酸(Asp),Asn是细胞合成糖蛋白和其他营养物质所必需的物质,经过II型L-天冬酰胺酶的治疗后,肿瘤细胞的DNA和蛋白质合成受阻,转变为营养不良状态,最终导致癌细胞凋亡。但正常细胞不受影响,它可以利用转氨酶将草酰乙酸转化为Asp,然后利用天冬酰胺合成酶(ASNS)合成Asn从而满足自身营养需求。而一些肿瘤细胞因ASNS的表达水平较低,丧失了合成Asn的能力因此只能依赖血清中的Asp。但治疗型II型L-天冬酰胺酶所面临的一个挑战是不稳定,短的血清半衰期和高的胰蛋白酶清除率。另一个挑战是天冬酰胺酶含有第二活性:谷氨酰胺酶活性,其会引发严重的副作用。因此不同来源的II型L-天冬酰胺酶可能具有不同抗原、生理、药理、血清学和药代动力学特性,其诱发副作用的倾向和治疗癌症的效果也不同。从迄今未被探索的生态位中探索新的和优良的酶,这对促进基于患者个体需求的药物和治疗方案的开发有重要意义。
发明内容
本发明的目的是提供一种II型L-天冬酰胺酶及其应用,该酶在37℃动物血清中的半衰期是现有研究中耐受时间最长的。该酶可以诱导人肝癌细胞QGY-7703和人恶性黑色毒瘤细胞A-375细胞株凋亡,且不含谷氨酰胺酶活性,在医药领域具有潜在的应用价值和应用潜力,尤其在抗癌药物开发和药物制备中具有显著的应用价值。
为了达到上述目的,本发明提供了一种II型L-天冬酰胺酶,该酶的氨基酸序列如SEQ ID NO.4所示,该酶在37℃动物血清中的半衰期为102h,在胰蛋白酶中T1/2=33min。
本发明提供的II型L-天冬酰胺酶可用于制备抗癌药物,其中,该抗癌药物可用于抑制人肝癌细胞QGY-7703或/和人恶性黑色毒瘤细胞A-375,诱导QGY-7703或/和A-375细胞凋亡。
本发明还提供了一种由上述II型L-天冬酰胺酶制备的抗癌药物,该抗癌药物可用于抑制人肝癌细胞QGY-7703或/和人恶性黑色毒瘤细胞A-375,诱导QGY-7703或/和A-375细胞凋亡。
本发明还提供了上述II型L-天冬酰胺酶的编码基因,该编码基因的核苷酸序列如SEQ ID NO.3所示。

本发明还提供了一种II型L-天冬酰胺酶的制备方法,包含如下步骤:
(1)对倭蜂猴粪便微生物提取宏基因组DNA;
(2)采用核苷酸序列如SEQ ID NO.1和SEQ ID NO.2所示的引物,以步骤(1)的基因组DNA为模板进行PCR扩增;
(3)将步骤(2)所得的扩增产物与重组载体进行连接得到重组表达载体,并将该重组表达载体转入表达菌中,得重组菌;
(4)对步骤(3)所得的重组菌进行培养,经IPTG诱导,收集菌体并超声破碎,收集上清并用Nickel-NTAAgarose纯化,即得II型L-天冬酰胺酶。
本发明的II型L-天冬酰胺酶,解决现有II型L-天冬酰胺酶不稳定、血清半衰期短等问题,具有以下优点:
本发明提供的II型L-天冬酰胺酶稳定性较好,其最适pH为8.0,该酶在pH=5.0-10.0范围内处理1h后,剩余酶活仍保持80%以上;该酶最适温度为45℃;该酶在20℃、30℃、35℃、40℃、45℃、50℃条件下耐受90min后,剩余酶活在80%以上,在20-40℃下其剩余活性仍大于100%;该酶在pH=8.0、45℃条件下,该酶的Km、和Vmax分别为5.733±0.512mM和156.2±3.056U/mg;多种离子(Li+、Hg2+、K+、Ag+、Ca2+、Cu2+、Co2+、Mn2+、Na+、Al3+、Mg2+、Zn2+、Ni2 +、Fe2+、Fe3+)、SDS、EDTA和Tween-80均能抑制重组酶的活性。该酶与其他来源的II型L-天冬酰胺酶相比具有最高比活性598.907U/mg。该酶在37℃的动物血清中的半衰期为102h,在胰蛋白酶中T1/2=33min。
目前,大多数的天冬酰胺酶存在谷氨酰胺酶活性,且被用于临床治疗的大肠杆菌天冬酰胺酶和菊氏杆菌天冬酰胺酶都存在谷氨酰胺酶活性。本发明提供的II型天冬酰胺酶NPASN10无谷氨酰胺酶活性,此性质可能会减少用药过程中所产生的副作用。本研究可能可以为制药领域提供一种性质更好的治疗酶,弥补治疗酶的缺口。
目前,II型天冬酰胺酶面临的巨大挑战是其不稳定且由于其给药途径(注射)的特殊性,可能会导致酶活性降低,因此稳定性是一个重要的指标。本发明提供的NPASN10在生理温度下,其胰蛋白酶和血清半衰期耐受是现有研究中时间最长的,很大程度上可以解决天冬酰胺酶不稳定的问题。
本发明提供的NPASN10能引起QGY-7703(人肝癌细胞)和A-375(人恶性黑色毒瘤细胞)细胞凋亡且不含谷氨酰胺酶活性,也不会引起溶血,该酶在医药领域如制药工业中具有潜在的应用价值,目前未有天冬酰胺酶对这两种细胞的细胞毒性研究。因此,本研究可以为该领域现有的研究提供补充,为未来的研究提供参考。

具体实施方式
下面将对本发明实施例中的技术方案进行清楚、完整地描述,显然,所描述的实施例仅仅是本发明一部分实施例,而不是全部的实施例。基于本发明中的实施例,本领域普通技术人员在没有做出创造性劳动前提下所获得的所有其他实施例,都属于本发明保护的范围。
本发明用到的部分试验材料和试剂如下:
1、菌株及载体:载体pEASY-E2、BL21(DE3)购自北京擎科新业生物技术有限公司。
2、基因工程操作酶类、试剂盒及其他生化试剂:限制性内切酶、DNA聚合酶、连接酶购自TaKaRa公司,质粒提取试剂盒、胶回收纯化试剂盒为美国Omega公司;其他试剂均为分析纯。
3、LB培养基:Peptone 10g,Yeast extract 5g,NaCl 10g,加蒸馏水至1000ML,pH自然(约为7);固体培养基在以上基础上加2.0%(w/v)琼脂。
说明:以下实施例中未作具体说明的分子生物学实验方法,均参照《分子克隆实验指南》(第三版)J.萨姆布鲁克一书中所列的具体方法进行,或者按照试剂盒和产品说明书进行。
实验例1II型L-天冬酰胺酶基因NPasn10的获得
1.倭蜂猴粪便微生物宏基因组II型L-天冬酰胺酶基因的筛选
在倭蜂猴粪便微生物宏基因组中,根据基因预测及功能注释得到了II型L-天冬酰胺酶的基因。基于序列同源性构建了系统发育树,根据其系统发育位置、三级结构与底物形成的BD energe、氨基酸组成及blast结果来筛选II型L-天冬酰胺酶。最终,NPasn10基因被筛选出来,待进一步克隆和表达。
2.II型L-天冬酰胺酶基因NPasn10的克隆
提取倭蜂猴粪便微生物宏基因组DNA,采用引物NPasn10-F和NPasn10-R(具体序列如下所示)以提取的基因组DNA为模板进行PCR扩增。
其中,PCR反应体系为50.0μL:宏基因组DNA 1μL,Ex Taq 10μL,RBasn10-F10.5μL,RBasn10-R10.5μL,dNTP 1μL,ddH2O补足50μL;
PCR反应扩增程序为:98℃30s;55℃30s,72℃1min,30个循环;4℃20min。PCR结果得到目的基因NPasn10,通过测序可知,该基因的核苷酸序列如SEQ ID NO.3所示。
扩增所用的引物序列如下所示(5’→3’):

实验例2II型L-天冬酰胺酶NPASN10的制备
通过常规构建方法,将实验例1中得到的NPasn10基因序列与质粒pEASY-E2连接,得到重组表达载体pEASY-E2-NPasn10,然后转化大肠杆菌BL21(DE3)获得重组大肠杆菌菌株BL21(DE3)/NPasn10。
取含有重组表达载体pEASY-E2-NPasn10的重组菌BL21(DE3)/NPasn10,以0.1%的接种量接种于LB(含100μg/mL的Amp)培养液中,37℃、180rpm下过夜培养。然后将培养的菌液以1%接种量接种到新鲜的LB(含100μg/mL的Amp)培养液中,37℃、180rpm下培养2~3h(OD600达到0.6~0.8)后,加入终浓度0.5mmol/L的IPTG诱导,于25℃、180rpm培养约20h。6000rpm离心10min,收集菌体。用适量的50mM PBS(pH=7.5)缓冲液悬浮菌体后,于冰浴条件下超声波破碎菌体。取上述的胞内浓缩的初酶液经12,000rpm、4℃离心10min后,吸取上清并用Nickel-NTAAgarose纯化目的蛋白,得到II型L-天冬酰胺酶NPASN10,其氨基酸序列如SEQ ID NO.4所示。
II型L-天冬酰胺酶NPASN10的氨基酸序列如下(SEQ ID NO.4):

对上述纯化的目的蛋白进行SDS-PAGE分析,结果如图1所示,其中,M为蛋白质Marker;泳道1为仅含有pEASY-E2载体的大肠杆菌诱导后的初酶;泳道2为未纯化的重组II型L-天冬酰胺酶;泳道3为纯化的重组II型L-天冬酰胺酶。由图1可知,重组的II型L-天冬酰胺酶在大肠杆菌中得到了表达,经Nickel-NTAAgarose纯化后为单一条带。
实验例3II型L-天冬酰胺酶NPASN10的性质测定
对实验例2中得到的II型L-天冬酰胺酶NPASN10的性质进行测定,采用纳氏试剂检测酶活力,具体如下:
混合液体系500μL:200μL 50mM Tris-Hcl缓冲液,200μL 200mM底物Asn,50μL适当稀释酶液,置于酶的最适温度的水浴锅中反应10min,随后立即加入50μL 1.5M三氯乙酸终止反应。在4℃、12000r/min条件下离心10min,取200μL上清液转移至新试管中,加入1.2mLddH2O和100μL纳氏试剂,室温静置反应10min后测定OD450。空白对照为先加入三氯乙酸终止反应,再进行后续的水浴处理。酶活单位定义:一个酶活单位(U)即在酶的最适反应条件下,每分钟水解Asn释放1μmol氨所需的酶量。
1.II型L-天冬酰胺酶的最适温度及温度稳定性测定
酶的最适温度测定:将实验例2纯化的II型L-天冬酰胺酶NPASN10在pH=8.0,于30-60℃下进行酶促反应,并测定相对酶活,其最适温度测定结果如图2所示。
酶的温度稳定性测定:将同样酶量的酶液置于设定的温度(20℃、30℃、35℃、40℃、45℃、50℃、55℃、60℃)中处理90min,每隔10分钟在pH=8.0及45℃下进行酶促反应,以未处理的酶液作为对照,并测定相对酶活,其温度稳定性测定结果如图3所示。
综合图2和图3可知,L-天冬酰胺酶的最适温度为45℃,在20、30、35、40、45、50℃条件下相对酶活保持稳定,在20℃、30℃、35℃、40℃、45℃、50℃条件下耐受90min后,剩余酶活在80%以上;在20-40℃下耐受90min后,其剩余活性仍大于100%,可知该酶的温度耐受性和稳定性较强。
2.II型L-天冬酰胺酶的最适pH和pH稳定性的测定
酶的最适pH测定:在45℃、pH=4.0-12.0的缓冲液中进行酶促反应,并测定相对酶活,结果如图4所示。
酶的pH稳定性测定:将纯化的酶液置于pH=4.0-12.0的缓冲液中,在37℃下处理1h后进行酶促反应,以未处理的酶液作为对照,并测定相对酶活,结果如图5所示。
其中,缓冲液为:0.5mol/L柠檬酸-柠檬酸钠缓冲液(pH=4.0-6.0);0.5mol/LTris-Hcl缓冲液(pH=7.0-8.0);0.5mol/L甘氨酸-氢氧化钠缓冲液(pH=9.0-12.0)。以Asn为底物,反应10min,测定纯化的L-天冬酰胺酶的酶学性质。
综合图4和图5可知,L-天冬酰胺酶的最适pH为8.0;经pH=5.0-10.0的缓冲液处理1h后,相对酶活保持80%以上。
3.不同金属离子及化学试剂对II型L-天冬酰胺酶活力影响测定
在酶促反应体系中加入终浓度为1mM金属离子和化学试剂,研究其对酶活的影响。在45℃、pH=8.0条件下,测定酶活(同样条件下以未加金属离子及化学试剂的酶促反应作为对照),结果见图6。
由图6可知,所有的金属离子和化学试剂对其活性均有抑制作用,K+、Cu2+、Na+、Ni2 +、Fe2+和Ca2+对该酶的活性有轻微抑制,而Mg2+、Li+、Fe3+、Ag+、EDTA和Tween-80几乎完全抑制该酶的活性。
4.II型L-天冬酰胺酶的动力学参数测定
动力学参数在pH=8.0、温度45℃和一级反应时间下以不同浓度的Asn为底物(2-80mM)进行测定,根据Lineweaver-Burk法计算出Km和Vmax值。经测定,在pH=8.0及温度45℃条件下该酶的Km和Vmax分别为5.733±0.512mM和156.2±3.056U/mg。
5.II型L-天冬酰胺酶的体外血清耐受测定
酶的体外血清耐受性测定:纯化的L-天冬酰胺酶(50IU,0.1mL),与2.4mL动物血清混合,用力摇匀,在30、37和40℃下进行孵育,每隔12h测定相对酶活,结果如图7所示。
由图7可知,纯化的L-天冬酰胺酶在30℃、37℃和40℃分别孵育约50h,102h和92h后,其初始活性降低了50%。
6.II型L-天冬酰胺酶的胰蛋白酶半衰期测定
酶的胰蛋白酶半衰期测定:将0.1mL纯化的天冬酰胺酶(50IU)加入2.4mL含有50IU胰蛋白酶(pH=7.5)中,反应混合液用力摇匀,在37℃下反应,每5min测定一次相对酶活,结果如图8所示。
由图8可知,纯化的L-天冬酰胺酶在37℃、50U/mL胰蛋白酶中孵育约33min后,其初始活性降低了50%。
7.II型L-天冬酰胺酶对QGY-7703和A-375细胞存活率的影响
用MTT法研究纯化的天冬酰胺酶的细胞毒性作用,将5×104个QGY-7703(人肝癌细胞)和A-375(人恶性黑色毒瘤细胞)细胞分别接种至96孔板中,待细胞贴壁后,细胞用不同浓度的天冬酰胺酶处理24h,丢弃培养基,用PBS清洗两遍,每孔加入20μL的MTT染料(5mg/mL)及180μL培养基继续培养4h后,吸弃每孔液体,加入100μL DMSO后置摇床上缓慢摇匀10min,在结晶体充分溶解后,用酶标仪在490nm处测定其光吸收值,计算细胞的存活率,结果分别如图9(QGY-7703)和图10(A-375)所示。
综合图9和图10可知,纯化的NPASN10处理的细胞的数量有明显的减少,IC50分别为5.7U/mL和6.5U/mL,与A-375相比,该酶对QGY-7703细胞表现出更好的抑制作用,可知,本发明提供的II型L-天冬酰胺酶NPASN10可应用于医药领域,尤其可用于制备抗癌药物,制备可用于治疗肝癌和恶性黑色素瘤的药物,具有潜在的应用价值。
8.II型L-天冬酰胺酶对QGY-7703和A-375细胞形态影响
在24孔板中每孔培养大约1.5×105个细胞,并用2、4、6、8和10U/mL浓度的天冬酰胺酶处理。37℃、5%CO2培养箱中培养24h,在倒置显微镜(放大100倍)下观察后拍照,其中,图11为QGY-7703细胞的显微镜镜检结果,图12为A-375细胞的显微镜镜检结果,图中的A为未经过任何处理的QGY-7703和A-375细胞,图中的B为IC50浓度下的II型L-天冬酰胺酶处理QGY-7703和A-375细胞。
综合图11和图12可知,在IC50浓度的酶处理下,细胞数量明显减少,细胞出现起泡、胀大和破裂的现象。然而,未处理的细胞未观察到形态学的变化,细胞透明度高,轮廓清晰,排列整齐。
9.II型L-天冬酰胺酶对QGY-7703和A-375细胞核的影响
为了研究天冬酰胺酶对细胞凋亡水平的影响,在24孔板中培养大约1.5×105个细胞,用不同浓度的天冬酰胺酶处理细胞,在37℃,5%CO2培养箱中培养24h,用PBS(pH 7.0)洗涤3遍,再用100μL 4%多聚甲醛将细胞固定10min,再用0.1%Triton-X-100洗3遍细胞,PBS洗涤3遍,加入100μL,1.25μg/ml 4′,6-二氨基-2-苯基吲哚二盐酸(DAPI)染色10min,在倒置荧光显微镜(放大100倍)下观察拍照,其中,图13为QGY-7703细胞经DAPI染色后的荧光显微镜镜检结果,图14为A-375细胞经DAPI染色后的荧光显微镜镜检结果,图中的A为未经过任何处理的QGY-7703和A-375细胞DAPI染色结果,图中的B为在IC50浓度下的II型L-天冬酰胺酶处理QGY-7703和A-375细胞的DAPI染色结果。
综合图13和图14可知,在IC50浓度的酶处理下,细胞明显表现出明显的细胞核缩合,染色质凝结,DNA裸露,DNA断裂,正常核结构丧失等凋亡形态学特征,核碎裂成大小不等的圆形小体,并被细胞膜所包绕。然而,在对照组细胞中,细胞有完整的细胞膜和细胞核,没有形态学改变。
10.II型L-天冬酰胺酶溶血试验
通过溶血试验研究未纯化(Crude asparaginase)及纯化的(Purifiedasparaginase)天冬酰胺酶对哺乳动物血液的溶血作用。将25μL粗品和纯化的天冬酰胺酶分别放入先前在血液琼脂板上打孔的孔中。在37℃下孵育24h后,检查平板上的透明或半透明晕带,II型L-天冬酰胺酶在平板上的溶血的情况如图15所示。
由图15可知,未纯化及纯化的天冬酰胺酶都未在血琼脂平板形成透明圈,说明从倭蜂猴粪便宏基因组来源的L-ASNase无谷氨酰胺酶活性,对红细胞无毒。
根据上述研究的结果,收集了部分现有的不同来源的II型L-天冬酰胺酶相关信息,对其酶学性质进行比较,结果见下表1,由于一些研究并不充分,部分酶的相关信息未列出,由表1可知,本发明对II型L-天冬酰胺酶的性质和功能做了相对较全的研究,且本发明提供的II型L-天冬酰胺酶NPASN10综合性能较优,且具有更广泛的应用价值。

综上可知,本发明利用宏基因组技术从倭蜂猴粪便微生物宏基因组筛选II型L-天冬酰胺酶基因,并在大肠杆菌中异源表达了II型L-天冬酰胺酶,通过对其酶学性质及抗癌特性的研究,获得了具有更高活性、更稳定且无谷氨酰胺酶活性的II型L-天冬酰胺酶NPASN10。
本发明提供的无谷氨酰胺酶活性的II型L-天冬酰胺酶可能会减少用药引起的副作用,同时不会引起溶血,极具经济价值,在制药工业应用中具有强大的应用潜力。本发明提供的II型L-天冬酰胺酶具有高的血清及胰蛋白酶半衰期,在人体生理温度及pH下有较高酶活,可应用于抗癌药物的应用和开发。本发明提供的II型L-天冬酰胺酶来自于迄今未被探索的生态位中新的和微生物菌株,这对促进基于患者个体需求的药物和治疗方案的开发有重要意义。
尽管本发明的内容已经通过上述优选实施例作了详细介绍,但应当认识到上述的描述不应被认为是对本发明的限制。在本领域技术人员阅读了上述内容后,对于本发明的多种修改和替代都将是显而易见的。因此,本发明的保护范围应由所附的权利要求来限定。

Claims (10)
1.一种II型L-天冬酰胺酶,其特征在于,该酶的氨基酸序列如SEQ ID NO.4所示。
2.如权利要求1所述的II型L-天冬酰胺酶在制备抗癌药物中的应用。
3.根据权利要求2所述的应用,其特征在于,所述的抗癌药物可抑制人肝癌细胞QGY-7703或/和人恶性黑色毒瘤细胞A-375,诱导QGY-7703或/和A-375细胞凋亡。
4.由权利要求1所述II型L-天冬酰胺酶制备的抗癌药物,其特征在于,所述的抗癌药物可用于抑制人肝癌细胞QGY-7703或/和人恶性黑色毒瘤细胞A-375,诱导QGY-7703或/和A-375细胞凋亡。
5.如权利要求1所述的II型L-天冬酰胺酶的编码基因,其特征在于,所述编码基因的核苷酸序列如SEQ ID NO.3所示。
6.包含权利要求5所述编码基因的重组表达载体。
7.根据权利要求6所述的重组表达载体,其特征在于,所述的重组表达载体选自pEASY-E2。
8.包含如权利要求5所述编码基因的重组菌。
9.根据权利要求8所述的重组菌,其特征在于,所述的重组菌选自大肠杆菌BL21(DE3)。
10.一种如权利要求1所述II型L-天冬酰胺酶的制备方法,其特征在于,包含如下步骤:
(1)对倭蜂猴粪便微生物提取宏基因组DNA;
(2)采用核苷酸序列如SEQ ID NO.1和SEQ ID NO.2所示的引物,以步骤(1)的基因组DNA为模板进行PCR扩增;
(3)将步骤(2)所得的扩增产物与重组载体进行连接得到重组表达载体,并将该重组表达载体转入表达菌中,得重组菌;
(4)对步骤(3)所得的重组菌进行培养,经IPTG诱导,收集菌体并超声破碎,收集上清并用Nickel-NTAAgarose纯化,即得II型L-天冬酰胺酶。
