Abstract

背景技术
炎症是一种基本的免疫反应,使生物在遭受病原微生物感染或组织损伤期间能够存活。各种外在和内在的炎症刺激物包括但不限于各种感染性微生物(如病毒和细菌)、癌细胞或死亡细胞。尽管炎症是宿主健康免疫反应的重要组成部分,但严重的炎症会导致各种疾病,如败血症、类风湿性关节炎、急性肺损伤、慢性呼吸道疾病、炎症性肠病(IBD)、癌症、衰老,甚至死亡。其中,病毒和致病性细菌感染是引起炎症和死亡的主要原因。重症监护病房(ICU)中约50%的患者因细菌感染而发生严重败血症,感染患者的死亡率为29%。死亡原因主要归因于脂多糖(LPS)内毒素的细胞毒性作用。现有研究表明,在LPS刺激的巨噬细胞中会产生多种炎症因子,如肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β) 和白细胞介素-6(IL-6)以及高水平的细胞内活性氧(ROS)等。
免疫蛋白酶体是巨噬细胞等免疫细胞中炎症的重要介质,由炎症信号触发其表达。现有研究表明,免疫蛋白酶体不仅可以将蛋白质水解成多肽,作为抗原载入MHC-I复合物,还与巨噬细胞极化、炎性细胞因子产生、各种炎性和自身免疫性疾病以及结肠炎和结肠炎相关癌症等有关。因此,免疫蛋白酶体抑制剂已成为各种疾病的候选药物,主要用于降解过量的免疫蛋白酶体。然而,目前尚不清楚免疫蛋白酶体是否以及如何降解。
巨自噬/自噬是一种分解代谢途径,通过双层膜囊泡(自噬体)进行大量运输,最终降解溶酶体中的细胞内成分。自噬途径是一个动态过程,其功能是通过选择性降解胞质物质(如有毒蛋白质聚集体、不必要或受损的细胞器和细胞内病原体) 来维持细胞内稳态。自噬功能障碍与几种促炎性疾病有关,如类风湿性关节炎、狼疮和炎症性肠病(IBD)。自噬基因ATG16L1、NOD2、IRGM、ATG5和ATG7 的种系变异与包括克罗恩病在内的慢性炎症性疾病有关。然而,自噬限制炎症的机制目前尚不清楚。
综上所述,阐明自噬是否以及如何以免疫蛋白酶体为降解靶点,以及免疫蛋白酶体的降解在限制炎症中的作用非常重要。为了解决这个问题,本发明提供了一种泛-FGFR抑制剂在调控炎症反应中的应用,以期缓解现有难题中的至少一种。

泛-FGFR抑制剂在制备抗炎药中的应用,所述泛-FGFR抑制剂可以抑制细胞炎症,所述泛-FGFR抑制剂为(R)-(E)-2-(4-(2-(5-(1-(3,5-二氯吡啶-4- 基)乙氧基)-1H-吲唑-3基)乙烯基)-1H-吡唑-1-基)乙醇。
进一步,所述抗炎药可用于急性肺损伤炎症反应、败血症炎症反应和炎症性肠胃病。
进一步,所述泛-FGFR抑制剂通过激活细胞选择性自噬反应来促进免疫蛋白酶体降解以抑制炎症。
进一步,所述泛-FGFR抑制剂抑制FGFR活性和下游PI3K/AKT/mTOR 信号通路,导致所述选择性自噬的激活。
一种抗炎药,包含泛-FGFR抑制剂和免疫蛋白酶体抑制剂,所述泛-FGFR 抑制剂为(R)-(E)-2-(4-(2-(5-(1-(3,5-二氯吡啶-4-基)乙氧基)-1H-吲唑-3基)乙烯基)-1H-吡唑-1-基)乙醇;所述免疫蛋白酶体抑制剂为ONX-0914。

有益技术效果
在本发明中,通过激活自噬的实验验证,确定泛-FGFR(pan-FGFR)抑制剂LY2874455为一种有效的抗炎治疗应用。利用LPS诱导的巨噬细胞炎症反应作为功能筛选,进一步确定泛-FGFR抑制剂LY2874455是一种有效的细胞炎症抑制剂。本发明揭示了LY2874455对自噬的刺激促进了LPS诱导的巨噬细胞中免疫蛋白酶体的自噬降解。具体来说,LY2874455抑制AKT-mTOR轴激活了自噬,进一步引发由受体SQSTM1/p62介导的免疫蛋白酶体的降解。此外, LY2874455激活的自噬能显著抑制炎性细胞因子的产生,在本发明中,LY2874455 诱导的免疫蛋白酶体自噬降解通过LPS诱导的急性肺损伤和右旋糖酐硫酸钠(DSS)诱导的炎症性肠病的活体小鼠模型得到验证。这些实验表明,免疫蛋白酶体的选择性自噬降解是先天性炎症信号的关键调节因子,而LY2874455通过激活细胞选择性自噬反应来促进免疫蛋白酶体降解,进而抑制炎症。

巨噬细胞培养及分离
在37℃,5%CO2细胞培养箱中,利用含有10%胎牛血清的 RPMI-1640(Sigma)完全培养基对RAW264.7小鼠巨噬细胞进行培养。从6-8周的 C57BL/6J小鼠中分离原代腹腔(PM)及骨髓来源巨噬细胞(BMDM)。提前3天向6-8周龄小鼠腹腔注射3%巯基乙酸,3天后取腹水和骨髓。将从腹水中分离的细胞接种在含有RPMI1640完整培养基的细胞板中,并使其粘附4-6h。贴壁细胞是腹腔巨噬细胞。将骨髓来源的细胞接种于RPMI1640完整培养基中孵育的细胞板中,并添加M-CSF(10ng/ml,Sigma),使其粘附7天。贴壁细胞是骨髓来源巨噬细胞。
质粒构建及转染
将靶基因p62、LMP2、LMP7分别克隆到pEGFP-C1载体中。将靶基因 p62和p62△UBA分别克隆到pFlag-CMV2载体中。将靶基因LC3克隆到 pLVX-mCherry-C1中。将靶向p62序列(CCGGGTCTCTACAGATGCCAGAATCCTCGAGGATTCTGGTAGAGAGACT TTG)的sh-RNA克隆到PLKO.1载体中。将靶基因ATG7的引导RNA(gRNA) 序列(TCTGCCCACCCGCTTGACGT)克隆到PX330载体中。将敲除位点的上下游800bp的序列以及筛选基因(puro)分别克隆到pUC19载体中。根据供应商说明书,使用jetprime试剂将无内毒素质粒转染到细胞中。
活性氧检测
首先将LY2874455和/或LPS处理24小时的细胞用PBS洗涤三次,并用 10μM H2-DCFH-DA避光37℃孵育30分钟。然后用PBS洗涤细胞三次后,收集细胞,利用流式或者荧光显微镜方法进行检测。
一氧化氮检测
培养基中一氧化氮亚硝酸盐衍生物的含量依据试剂盒说明书利用Griess 反应(Beyotime)进行检测。在反应结束后,使用多功能酶标仪(Biotek)在540nm 处测量混合物的吸光度。
实时定量聚合酶链式反应
使用快速细胞/组织总RNA分离试剂盒(南京Vazyme Biotek)从细胞系、原代细胞或组织中提取总RNA。随后利用逆转录酶试剂盒(南京Vazyme)将1μ RNA反向转录成cDNA。然后说明书,使用SYBR green试剂(南京Vazyme) 将前面生成的cDNA作为模版进行实时定量聚合酶链反应(PCRs)。
蛋白酶体活性检测
利用亚基选择性荧光肽底物监测底物随时间推移的水解速率以测量单个催化亚基的催化活性。简而言之,首先使用惰性裂解缓冲液(25mM Tris PH 7.5, 100mM Nacl,5mMATP,0.2%NP40,20%甘油)制备蛋白质裂解物,并用20S 蛋白酶体测定缓冲液(20mM Tris-HCl,0.5mM EDTA,0.035%SDS,pH 8.0) 进行稀释。最后加入蛋白酶体底物在37℃下孵育2小时。底物和浓度如下: Ac-ANW-AMC(β5i,100μM)和Ac-PAL-AMC(β1i活性,100μM)。最后使用多功能酶标仪(Biotek)分别在345和445nm的激发和发射波长下测量荧光信号。
免疫印迹
用添加蛋白酶抑制剂(罗氏11697498001)的RIPA缓冲液裂解细胞或组织。根据制造商说明书,通过BCA蛋白质测定试剂盒(Sigma)对蛋白质进行定量。定量后,蛋白沉淀进行SDS-PAGE电泳。电泳结束后将蛋白质转移到PVDF 膜上,然后用5%脱脂牛奶室温封闭1小时,封闭结束后4℃下孵育一抗过夜。然后将膜与二抗在室温下孵育1h。最后利用ChemidocMP成像系统(Bio-Rad) 采集的图片。
免疫沉淀
利用0.25%TX-100,1%NP-40,50mM Tris-HCl,pH 8,150mM NaCl 和1mM EDTA的缓冲液对细胞进行裂解。调整裂解物浓度和体积,并与 GFP-Beads(Shenzhen Sports LifeTechnology Company)在4℃下孵育过夜。然后,将样品在4℃下以2000rpm离心3分钟。取沉淀,用洗涤缓冲液A[50mM tris-HCl(pH 7.5),100mM氯化钠和2mM EDTA洗涤五次。将结合蛋白收集在 RIPA缓冲液(50mM Tris,pH 7.4,150mM NaCl,0.1%SDS,1%TritonX-100 和0.5%脱氧胆酸钠)中,并将沉淀的蛋白质进行SDS-PAGE电泳。
免疫荧光
首先用4%多聚甲醛将细胞固定10分钟,然后用含0.1%TritonX100的 PBS透化10分钟,并在室温下用含有3%BSA的PBS孵育1小时。封闭后,一抗孵育,然后进行PBS洗涤并与二抗(驴抗小鼠和抗兔Alexa Fluor 488或594) 孵育。最后利用DAPI对细胞核进行染色。最后使用蔡司LSM 880进行图像采集。
动物实验
所有涉及动物的程序均按照相关准则和规定进行,并经四川大学伦理委员会批准。
急性肺损伤
将8周龄雄性小鼠随机分为3组(对照组,LPS(10mg/kg)组,LPS(10mg/kg) +LY2874455(1mg/kg)组,每组6只小鼠。为了验证LY2874455对LPS诱导的肺损伤的影响,在LPS注射(15mg/kg)的同时,用生理盐水或LY2874455(1mg/kg) 给小鼠灌胃给药。LPS注射六小时后,处死小鼠并取出肺。随后将肺组织用于定量实时荧光定量PCR测定、蛋白质印迹测定和苏木精伊红(HE)染色。
脓毒血症
将8周龄的雄性小鼠随机分为3组,每组8只小鼠。为了评估LY2874455 对小鼠存活的影响,在注射LPS前2小时灌胃给予LY2874455(1mg/kg),然后通过单次腹腔注射内毒素(LPS,35mg/kg)诱导脓毒血症。结果发现接受 LY2874455处理的动物存活率增高。
结肠炎
从第0天开始,在饮用水中添加3%葡聚糖硫酸钠(DSS;m.w.36,000-50, 000;MPBiomedicals,Solon,OH)对小鼠进行喂养。在DSS处理的第三天,小鼠接受生理盐水或LY2874455(1mg/kg)灌胃处理5天。此后,给老鼠正常饮用水。在整个实验过程中每天测量体重。然后处死小鼠并获取其肠道。最后将肠组织用于定量实时荧光定量PCR测定,蛋白质印迹测定和苏木精伊红(HE)染色。
透射电子显微镜
将处理后的巨噬细胞用0.1M磷酸盐缓冲液洗涤三次,每次15分。在 2%四氧化锇水溶液中固定1小时,然后用去离子水洗涤三次,每次15分钟。将样品用2%乙酸铀酰染色30分钟,并通过梯度乙醇(50%-100%)和100%丙酮分别脱水15分钟。之后,将样品包埋在EPON 812树脂中,分别在37℃、45℃和60℃下固化24小时。采用超薄切片机制备超薄(70nm)切片,并用2%乙酸铀酰和0.3%柠檬酸铅染色。使用Tecnai G2 Spirit透射电子显微镜(FEI公司) 采集样品的电子显微镜图像。
数据分析
本发明示出的所有数据均为来自至少三个独立重复实验的代表性结果。定量数据表示至少三个生物重复±SD的平均值。使用Prism5进行统计分析。使用具有Turkey多重比较检验的普通单向或双向方差分析,或双尾成对或不成对的t检验分析来确定统计显著性(*:P<0.05,**:P<0.01,***:P<0.001,NS:无统计差异)。
结果
泛FGFR抑制剂LY2874455有效抑制LPS刺激的巨噬细胞的炎症
本实施例在LPS处理的巨噬细胞系RAW264.7中筛选了对炎症具有潜在抑制作用的化学物质。在炎症期间,NO生成水平会增加,因此通过测量NO水平,本发明团队确定LY2874455是最有效的抗炎化学物质(展示了包括 LY2874455在内的27种化学物质)(图2A)。LY2874455((R)-(E)-2-(4- (2-(5-(1-(3,5-二氯吡啶-4-基)乙氧基)-1H-吲唑-3基)乙烯基)-1H-吡唑-1- 基)乙醇(图1A)是一种新型泛-FGFR抑制剂,本发明发现,除了抑制LPS刺激的RAW264.7巨噬细胞中NO的生成外(图1B),LY2874455也降低了LPS 刺激的RAW264.7中活性氧(ROS,通过DCFH-DA染色显示)的水平(图1C, D)。形态学上,LY2874455逆转了RAW264.7巨噬细胞从细长型(炎症状态) 到圆形(停滞型)(图1E)。LPS在RAW264.7中显著诱导促炎细胞因子水平,如肿瘤坏死因子-α(TNF-α)和白细胞介素-6(IL-6),LY2874455完全逆转了这种效应(图1F,G)。与LY2874455导致的NO生成抑制一致,通过LY2874455 处理的RAW264.7中诱导型一氧化氮合酶(iNOS)的mRNA和蛋白质水平显著降低(图1H,I)。在从小鼠(图2B-I)获得的腹腔巨噬细胞(PM)和骨髓源性巨噬细胞(BMDM)以及人类THP-1单核细胞(图2J,K)中反复观察到LY2874455 的炎症抑制作用。NF-κB信号是一种典型的促炎症调节因子,介导各种炎症细胞因子的诱导。含有IKK-α、IKK-β和IKK-γ的IκB激酶(IKK)复合物激活炎症转录因子NF-κB。IKK-β磷酸化抑制性IκBα以促进其泛素化和蛋白酶体降解,然后经典的p65/p50异源二聚体被释放到细胞核并介导NF-κB靶基因转录,其中包括各种细胞因子。LY2874455使LPS诱导的RAW264.7中IKK-β的蛋白水平减少(图1J)。在LPS刺激的RAW264.7中,p65(磷酸化)的激活形式及其向细胞核的易位也被LY2874455逆转(图1K,L)。综上所述,这些结果表明泛-FGFR 抑制剂LY2874455有效抑制LPS刺激的巨噬细胞的炎症反应。
LY2874455降低了LPS刺激的巨噬细胞中免疫蛋白酶体的蛋白质水平
为了确定LY2874455如何从分子层面来抑制炎症反应,对经LPS和 LY2874455处理的RAW264.7巨噬细胞进行了基于质谱的蛋白质组学分析(图 3A,上图)。改变蛋白质的聚集表明,LY2874455处理可下调免疫蛋白酶体特异性亚单位(图3A下,箭头所示)。免疫蛋白酶体是20s组成性蛋白酶体的一种特殊亚型,通过将20s蛋白酶体的β1、β2和β5亚基替换为不同的对应物,即β1i (低分子量多肽2,LMP2),可快速形成β2i(多催化内肽酶复合物样-1,MECL-1,也称为LMP10)和β5i(低分子量多肽2,LMP7)。在LPS刺激等炎症条件下,免疫蛋白酶体特异性亚单位β1i/LMP2、β2i/MECL-1/LMP10和β5i/LMP7取代了 20s蛋白酶体的原始β1、β2和β5亚单位,导致免疫蛋白酶体的形成(如图3B、 C所示)。通过观察免疫蛋白酶体抑制剂ONX-0914有效阻断p65的核移位,证实了免疫蛋白酶体在NF-κB途径中的功能(图4A)。免疫蛋白酶体在NF-κB途径中的功能被认为不通过降解p-IκB-α实现(图4B)。相反,免疫蛋白酶体可能通过调节促进IκB-α磷酸化的上游信号步骤在NF-κB途径中发挥作用(图4B, C)。免疫蛋白酶体在调节NF-κB途径中的确切机制尚不清楚。本发明团队通过检测NF-κB通路中的组分功能,分析了LY2874455或ONX-0914存在时的NF-κB 通路。结果表明,LY2874455和ONX-0914同样导致TRAF6和TAK1的蛋白质水平降低,而对其他成分没有影响(图4C、D、E)。这些结果表明,免疫蛋白酶体可能调节NF-κB途径中的TRAF6-TAK1通路(TRAF6-TAK1 phase)。此外, LY2874455和ONX-0914的相同作用表明,LY2874455通过免疫蛋白酶体抑制炎症途径。
本发明团队证实了蛋白质组学的结果,并揭示了LY2874455确实降低了 LPS刺激的RAW264.7巨噬细胞中免疫蛋白酶体亚单位LMP2、LMP7和LMP10 的蛋白质水平(图3D)。相反,组成性蛋白酶体亚单位的蛋白质水平(PSMB1、 PSMB2和PSMB5)在LPS刺激的RAW264.7巨噬细胞中不受LY2874455的影响(图3E)。在从小鼠分离的腹腔巨噬细胞(PM)中也获得了类似的结果(图3F,G)。此外,LPS中免疫蛋白酶体亚单位(LMP2和LMP7)的催化活性刺激RAW264.7巨噬细胞,也被LY2874455减少(图3H)。本团队证实ONX-0914能有效抑制LPS刺激的RAW264.7中的炎症反应。通过检测促炎细胞因子TNF-α和IL-6的表达来检测巨噬细胞(图3I,J)。LY2874455的额外加入并没有进一步抑制ONX-0914治疗的巨噬细胞的炎症(图3I,J)。这一结果表明,LY2874455 通过免疫蛋白酶体抑制抑制炎症,可能是通过降低免疫蛋白酶体的蛋白质水平介导的。
LY2874455激活自噬
本发明团队尝试确定LY2874455降低免疫蛋白酶体亚单位(相当于免疫蛋白酶体)蛋白质水平的机制,因为它们的mRNA水平不受LY2874455处理的影响。泛素-蛋白酶体和自噬-溶酶体系统被认为是细胞降解的两种机制。在两种细胞内降解途径(蛋白酶体途径和自噬途径)中,大底物通常是自噬降解的靶点。本团队使用蛋白酶体抑制剂MG132、自噬抑制剂氯喹(CQ)和渥曼宁(wortmannin) 阻断这两种降解途径,然后分析免疫蛋白酶体亚单位的蛋白质水平。Western blot 分析表明,MG132不能抑制LY2874455诱导的LMP2和LMP7的减少(图5A 和图6A),而LY2874455诱导的减少被CQ或wortmannin显著逆转(图5B、C 和图6B)。因此,该结果表明LY2874455诱导的免疫蛋白酶体降解比泛素-蛋白酶体系统更依赖于自噬。
这些结果也表明LY2874455可以诱导自噬。通过测量融合蛋白 mCherry-EGFP-LC3的斑点信号进行自噬通量测定。LY2874455诱导的高强度 mCherry信号和低GFP信号表明LY2874455激活自噬(图5D)。透射电镜显示用LY2874455处理的RAW264.7巨噬细胞比对照细胞中有更多的自噬体(图5E 和图6C)。p62/SQSTM1是一种自噬受体,它将底物(通常为泛素化底物)招募到自噬体中,并在自噬体与溶酶体融合后与底物一起降解。在未添加Wortmannin 的分组中,随着LY2874455添加量的增加,p62的蛋白质水平逐渐降低(图5F)。LPS刺激导致p62的积累(图5G),表明LPS阻断自噬,这与LPS降低自噬通量的观察结果一致(图5D,LPS细胞中的黄色信号)。LY2874455有效降低LPS 刺激的RAW264.7中p62的蛋白质水平(图5G)。在从小鼠分离的腹腔巨噬细胞 (PM)和骨髓源性巨噬细胞(BMDM)中,LY2874455也降低了p62的蛋白质水平(图5H,I)。这些结果证实了LY2874455在激活自噬中的作用。
FGFRs是跨膜RTK,被FGF激活形成二聚体,导致下游FRS2复合物激活,随后激活PI3K/AKT/mTOR信号通路。活化的mTOR随后磷酸化并抑制自噬启动ULK1激酶复合物,从而抑制自噬。本发明揭示了LY2874455降低了 AKT-mTOR信号的水平,示出了低水平的mTOR和AKT磷酸化(图5J)。这种LY2874455介导的对AKT-mTOR信号通路的抑制进一步表明了其激活自噬的能力。LY2874455抑制FGFR活性和下游PI3K/AKT/mTOR信号通路,导致自噬的释放和激活(如图6D所示)。
LY2874455通过自噬和选择性受体p62促进免疫蛋白酶体的降解
为了进一步验证LY2874455通过自噬促进免疫蛋白酶体降解的作用,在 RAW264.7细胞中敲除了自噬中的关键基因ATG7,导致自噬受阻、p62增加和激活型LC3(LC3-II)缺乏(图8A)。然后,分析免疫蛋白酶体亚单位LMP2和 LMP7的蛋白质水平。在ATG7敲除(ATG7-KO)的RAW264.7细胞中,与WT 细胞相比,LMP2和LMP7的蛋白质水平增加(图7A)。虽然在WT细胞中 LY2874455使LMP2和LMP7的蛋白质水平降低了,但这种效应在ATG7-KO细胞中被消除(图7B,C)。本发明团队检测了自噬体和免疫蛋白酶体的定位。结果显示自噬体标记物(LC3点)与LMP2和LMP7共定位(图7D)。总之,这些结果表明LY2874455通过自噬诱导免疫蛋白酶体的选择性捕获和降解。
一般情况下,底物选择性靶向自噬体是由受体介导的,通过进一步分析已经研究的三种受体,发现p62的行为与RAW264.7细胞中的LMP2和LMP7 相似,LPS处理增加,LY2874455进一步处理减少(图7E)。敲低RAW264.7细胞中的p62导致LMP2和LMP7蛋白水平升高(图7F和图8B)。这些结果表明 p62是免疫蛋白酶体自噬降解的受体蛋白。底物泛素化是自噬受体p62选择性靶向所必需的。事实上,LMP2和LMP7是泛素化的(图7G)。此外,LMP2和LMP7 可以与p62相互作用,这种相互作用依赖于p62的UBA结构域(泛素结合的泛素相关结构域)(图7H,I)。因此,这些结果表明p62是介导免疫蛋白酶体自噬降解的受体。
LY2874455通过自噬途径抑制炎症
本试验旨在检测LY2874455对炎症的抑制作用是否通过自噬途径发生。 LPS诱导的RAW264.7细胞中ROS水平升高(通过DCFH-DA探针显示)被 LY2874455消除,然而,这种消除被自噬抑制剂wortmannin阻断(图9A)。此外,在wortmannin处理的RAW264.7细胞(图9B、C)、小鼠分离的原代巨噬细胞(图9D、E)和骨髓来源的巨噬细胞(图10A、B)中观察到iNOS和IL-6的 mRNA水平增加。这些结果表明LY2874455通过自噬抑制炎症。在LPS和 LY2874455存在的情况下,ATG7敲除或p62敲除导致iNOS和IL-6水平升高的观察进一步证实了这一结果(图9F,G)。LY2874455对NF-κB通路的抑制作用也依赖于自噬和受体p62,正如在自噬或p62缺陷的巨噬细胞中一样,LY2874455 不能抑制NF-κB通路(图10C-F)。
加上数据显示LY2874455诱导免疫蛋白酶体的自噬降解,这些结果表明自噬缺陷细胞中的炎症增加可能是由自噬底物免疫蛋白酶体的积累引起的。然后使用免疫蛋白酶体特异性抑制剂ONX-0914治疗ATG7-KO RAW264.7细胞,然后分析诱导型一氧化氮合酶、促炎细胞因子IL-6和TNF-α的表达水平。结果表明,免疫蛋白酶体抑制剂ONX-0914可显著降低自噬缺陷细胞的炎症反应(图 9H-J)。
因此,这些结果表明LY2874455对炎症的抑制作用依赖于诱导免疫蛋白酶体的自噬降解。
LY2874455在小鼠模型中抑制肺和肠道炎症
在证明了LY2874455在细胞水平上对炎症的抑制作用后,本发明团队随后分析了LY2874455是否会影响脂多糖(LPS)诱导的急性肺损伤和败血热以及右旋糖酐硫酸钠(DSS)诱导的炎症性肠病小鼠模型。LPS刺激的小鼠肺部组织学检查显示大量的炎性细胞浸润,这是肺部炎症的典型症状(图11A)。值得注意的是,LY2874455减少了炎性细胞浸润(图11A)。LPS诱导的小鼠肺组织中高水平的促炎细胞因子IL-6也被LY2874455显著降低(图11B)。此外,还检测了LPS刺激的小鼠在接受或不接受LY2874455治疗的情况下的存活情况。对照组的存活率为100%,但LPS处理组的存活率显著下降,而LY2874455治疗组的存活率显著增加(图11C)。这些结果表明,LY2874455抑制了LPS诱导的急性肺损伤和败血症小鼠的炎症反应,并促进了LPS刺激小鼠的存活。
此外,分析LY2874455在使用DSS诱导的结肠炎模型小鼠中的潜在胃肠保护作用。结果表明,LY2874455显著改善了DSS刺激小鼠结肠长度的减少(图 11D)。为了证实LY2874455对肠道炎症和组织损伤的影响,对小鼠结肠切片进行组织学检查。DSS对肠绒毛粘膜造成严重损伤,LY2874455治疗有效地减少了组织损伤(图11E)。LY2874455治疗的小鼠结肠组织中的促炎细胞因子IL-17 和TNF-α的表达水平降低(图11F,G),这表明LY2874455对结肠炎模型小鼠的炎症有抑制作用。
综上所述,这些体内实验结果证实了LY2874455对炎症的抑制作用,以及LY2874455介导的免疫蛋白酶体更新的促进作用。
讨论
现目前,选择性自噬精确调节炎症的潜在的机制没有被完全研究透彻。在本发明中,研究团队揭示了泛-FGFR(pan-FGFR)抑制剂LY2874455在炎症抑制中的作用和机制(图12)。在LPS刺激的巨噬细胞中,免疫蛋白酶体被诱导,然后触发NF--κB介导的炎症反应,导致促炎细胞因子、ROS和NO的强烈表达和产生(图12,左图)。在LY2874455存在的情况下,自噬途径被激活,诱导自噬体靶点并捕获免疫蛋白酶体,这种效应由识别免疫蛋白酶体上的泛素信号的选择性受体p62介导。这种自噬降解降低了免疫蛋白酶体的数量,从而阻断了NF-κB的活性,并最终抑制了炎症(图12,右图)。通过上述发明,本发明实质上揭示了一种自噬抑制炎症的新机制,可能极大地促进炎症疾病新治疗方法的发展。
此外,目前尚不清楚免疫蛋白酶体的诱导和自噬降解是如何协调的。本发明假设LPS等炎症刺激可能诱导免疫蛋白酶体的表达、组装和数量增加,而 LPS抑制自噬途径,如受体p62的蛋白水平增加所示(图5F-I)。这一结果表明, LPS不仅诱导免疫蛋白酶体的产生,而且还抑制其自噬降解,从而导致免疫蛋白酶体的积累,最终导致严重的炎症。免疫蛋白酶体诱导的高水平炎症提供了有效的信号,提醒宿主有毒性刺激,从而激活宿主反应。然而,宿主受益于强烈的炎症诱导信号,同时也遭受炎症诱导的损伤,尤其是在严重/急性或慢性炎症条件下。因此,虽然中度和可控炎症之间的平衡对于维持宿主健康至关重要,但本发明提供的技术方案对控制严重的炎症具有指导意义。
在各种情况下,选择性自噬的功能是通过受体实现的,这些受体特异性识别特定类型的底物。底物的降解主要表明自噬的特定作用。自噬基质的性质因细胞类型和刺激信号而异。在本发明的实施例中,通过自噬选择性靶向免疫蛋白酶体解释了自噬对巨噬细胞炎症的抑制作用。自噬阻断诱导免疫蛋白酶体的积累,伴随着强烈的炎症反应,免疫蛋白酶体抑制剂可以完全逆转这一结果,该结论得到了证实(图9H-J)。这一结果表明,免疫蛋白酶体的自噬降解主要是自噬在炎症抑制中的作用。
如本研究所示,介导免疫蛋白酶体自噬降解的受体是p62,而不是NBR1 或Tollip。实际上存在多种自噬受体,受体的多样性可能反映了多细胞生物体自噬的特异性和精细调控。换句话说,某些受体只能在特定的细胞类型中发挥作用。明确自噬作用的细胞类型,自噬的状态以及其底物可以提高对自噬作为动态和多功能过程的了解。免疫蛋白酶体亚单位能够被多泛素化,它们依赖于p62的泛素结合结构域从而与p62的结合,这表明泛素链是自噬体靶向免疫蛋白酶体的信号。明确负责免疫蛋白酶体泛素化的泛素连接酶可以有助于明确泛素连接酶是如何直接或间接促进p62对免疫蛋白酶体的特异性识别。
综上所述,本发明的结论描述了巨噬细胞异常炎症背景下免疫蛋白酶体的选择性自噬降解(图12),并支持通过选择性自噬调节炎症的新机制。这些数据扩展了对自噬的理解,更重要的是,探索了选择性自噬和炎症之间的相互作用,这对于建立有效的炎症性疾病治疗方法是有意义的。
上面结合附图对本发明的实施例进行了描述,但是本发明并不局限于上述的具体实施方式,上述的具体实施方式仅仅是示意性的,而不是限制性的,本领域的普通技术人员在本发明的启示下,在不脱离本发明宗旨和权利要求所保护的范围情况下,还可做出很多形式,这些均属于本发明的保护之内。

1.泛-FGFR抑制剂在制备抗炎药中的应用,其特征在于,所述泛-FGFR抑制剂可以抑制细胞炎症,所述泛-FGFR抑制剂为(R)-(E)-2-(4-(2-(5-(1-(3,5-二氯吡啶-4-基)乙氧基)-1H-吲唑-3基)乙烯基)-1H-吡唑-1-基)乙醇。
2.如权利要求1所述的应用,其特征在于,所述抗炎药可用于急性肺损伤炎症反应、败血症炎症反应和炎症性肠胃病。
3.如权利要求1所述的应用,其特征在于,所述泛-FGFR抑制剂通过激活细胞选择性自噬反应来促进免疫蛋白酶体降解以抑制炎症。
4.如权利要求3所述的应用,其特征在于,所述泛-FGFR抑制剂抑制FGFR活性和下游PI3K/AKT/mTOR信号通路,导致所述选择性自噬的激活。

7.如权利要求5所述的抗炎药,其特征在于,所述抗炎药可用于急性肺损伤炎症反应、败血症炎症反应和炎症性肠胃病。
8.泛-FGFR抑制剂在制备细胞炎症检测试剂盒中的应用,其特征在于,所述试剂盒至少还包含一种脂多糖,所述泛-FGFR抑制剂可以是(R)-(E)-2-(4-(2-(5-(1-(3,5-二氯吡啶-4-基)乙氧基)-1H-吲唑-3基)乙烯基)-1H-吡唑-1-基)乙醇。
9.如权利要求8所述的应用,其特征在于,所述试剂盒可用于检测细胞的炎症水平,所述细胞包括单核巨噬细胞、腹腔巨噬细胞和/或骨髓源性巨噬细胞。
10.如权利要求9所述的应用,其特征在于,所述试剂盒可以通过检测细胞中的NO、ROS和/或促炎细胞因子的含量水平来判断细胞炎症的程度。
11.如权利要求9所述的应用,其特征在于,所述试剂盒可以通过检测细胞的形态来判断细胞是否有炎症。
