本发明属于生物材料和食品科学技术领域,具体涉及一种可食用的细胞培养肉3D支架及其制备方法和应用。
背景技术
细胞培养肉技术是近年来兴起的一项颠覆性食品制造技术,有望缓解传统畜牧业的可持续性生产压力和环境保护问题。细胞培养肉是依据动物肌肉生长修复机理,利用现代生物技术手段从动物体内分离得到的成肌细胞、干细胞等,实现动物肉组分(蛋白质、脂肪、维生素等)的体外合成,以产生具有与传统养殖肉类相同营养和感官特性的产品,从而实现对传统养殖肉等食品的替代,它不需要经过动物养殖,直接用细胞工厂化生产肉类。
目前,通常利用热引发、光引发以及辐照引发等物理辅助的方法制备水凝胶。其中热引发和光引发交联法需要在体系中额外加入引发剂或催化剂,这不仅使得水凝胶的成分和制备工艺复杂化,残留的引发剂或催化剂存在的生物毒性也会引起食品安全问题和消费者的担忧,极大的限制了水凝胶在细胞培养肉领域的应用。此外,尽管利用离子交联或静电相互作用规避了上述可能存在的安全隐患,但其较弱的作用力难以满足结构化细胞培养肉所需的机械支撑,也无法达到模拟真肉质构的目的。
发明内容
本发明的目的在于提供一种可食用的细胞培养肉3D支架及其制备方法和应用,利用辐照成胶,工艺简单,还能保证食品安全。
本发明提供了一种可食用的细胞培养肉3D支架的制备方法,包括以下步骤:将壳聚糖溶液和羧甲基纤维素钠溶液混合成均一的混合溶液,对所述混合溶液进行电子加速器辐照诱导成胶,得所述细胞培养肉3D支架。
优选的,所述混合包括在搅拌条件下,将壳聚糖溶液逐滴加入到羧甲基纤维素钠溶液中。
优选的,所述壳聚糖溶液和羧甲基纤维素钠溶液的质量比为(3~9):(1~7)。
优选的,所述壳聚糖溶液和羧甲基纤维素钠溶液均以水为溶剂,且质量百分浓度均为1~3%。
优选的,所述搅拌的转速为200~300rpm。
优选的,将壳聚糖溶液逐滴加入到羧甲基纤维素钠溶液后,还包括均质。

有益效果:本发明利用组织工程技术,通过壳聚糖和羧甲基纤维素钠复配,电子加速器辐照物理交联得到结构稳固的可食用3D支架。本发明所述可食用3D支架的制备不涉及复杂的制备工艺和可能存在的有毒交联剂,并具有良好的细胞粘附性和生物相容性,适用于骨骼肌细胞的培养,以用于生产结构化的细胞培养肉组织。
本发明使用海洋生物的副产物壳聚糖和自然界分布最广、含量最多的纤维素类作为可食用3D支架的制备材料,制备方法简单、绿色且兼顾了废弃物的高值化利用,大大降低了细胞培养肉的制作成本,并在一定程度上减少了温室气体的排放和对环境的污染,为后期商业化的工业生产奠定了实验基础。
本发明方法利用壳聚糖和羧甲基纤维素钠复配,通过电子加速器辐照成胶,无任何副产物或有毒有害成分,使得制备的细胞培养肉3D支架具有较高的食品安全性,且辐照过程具有杀菌效果,辐照后的支架可直接使用无需紫外灭菌,保障了生物安全性。

本发明提供了一种可食用的细胞培养肉3D支架的制备方法,包括以下步骤:将壳聚糖溶液和羧甲基纤维素钠溶液混合成均一的混合溶液,对所述混合溶液进行电子加速器辐照诱导成胶,得所述细胞培养肉3D支架。
本发明所述壳聚糖溶液和羧甲基纤维素钠溶液优选均以水为溶剂,且质量百分浓度均为1~3%。本发明将所述壳聚糖溶液和羧甲基纤维素钠溶液进行混合,所述混合优选包括在搅拌条件下,将壳聚糖溶液逐滴加入到羧甲基纤维素钠溶液中,所述壳聚糖溶液和羧甲基纤维素钠溶液的质量比优选为(3~9):(1~7),更优选为9:1、8:2、7:3、6:4、5:5、4:6和3:7。本发明所述搅拌的转速优选为200~300rpm,实施例中优选在磁力搅拌的条件下进行所述搅拌。
本发明将壳聚糖溶液逐滴加入到羧甲基纤维素钠溶液后,优选还包括均质,所述均质的转速优选为12000~15000rpm,均质的次数优选为3~5次,每次15秒。
本发明在得到均一的混合液后利用电子加速器进行辐照,优选采用10Mev电子加速器进行所述辐照,所述辐照的吸收剂量优选为30~50kGy。
本发明还提供了利用上述制备方法得到的可食用的细胞培养肉3D支架。
利用本发明所述制备方法制备得到的壳聚糖/羧甲基纤维素钠细胞培养肉3D支架图片如图1所示,由图1可看出细胞培养肉3D支架的外形规则圆润,具有一定厚度,结构稳固,能够为进一步的细胞增殖分化提供有力机械支撑。
本发明还提供了上述可食用的细胞培养肉3D支架在生产细胞培养肉中的应用。
本发明所述细胞培养肉3D支架具有较高的孔隙率(40%以上),在制备出15天后仍能保持较好的形态,为细胞长时间培养提供必要条件。支架孔隙率高可以构建三维的细胞培养环境,使细胞在更接近于自然情况的环境下生长和分化。此外,较高的孔隙率还可以促进培养液的渗透和流动,有利于营养物质和氧气的输送,维持细胞增殖的需求。本发明所述细胞培养肉3D支架还具有较高的硬度、胶粘性、咀嚼性和回复性,与肉的质构特性较为相似,能够为细胞的生长提供一定的机械支撑。
本发明实施例中,利用细胞培养肉3D支架对C2C12小鼠成肌细胞进行培养,培养1d后可看到3D支架黏附细胞的能力较强,具有数量最多的活细胞;培养2d后,小鼠成肌细胞(C2C12)在支架上大量增殖并开始伸展形成细长并相互连接的细胞骨架;培养5d后,小鼠成肌细胞进一步伸展延伸,体现出3D支架具有良好的生物相容性。
为了进一步说明本发明,下面结合实施例对本发明提供的一种可食用的细胞培养肉3D支架及其制备方法和应用进行详细地描述,但不能将它们理解为对本发明保护范围的限定。
实施例1
一种利用辐照成胶的可食用细胞培养肉3D支架的制备方法,包括以下步骤:
S1、将壳聚糖和羧甲基纤维素钠分别溶于水中,得到质量浓度为2%的壳聚糖溶液和羧甲基纤维素钠溶液,壳聚糖溶液和羧甲基纤维素钠溶液均通过静电相互作用形成糊状溶液。
S2、将上述步骤S1中得到的壳聚糖溶液逐滴加入到羧甲基纤维素钠溶液中,其中壳聚糖和羧甲基纤维素钠溶液的质量比为7:3,并采用均质机在12000rpm条件下均质混合,均质5次,每次15秒,得到混合溶液;
S3、将上述步骤S2中得到的混合溶液于10Mev电子加速器下进行辐照,辐照后支架的吸收剂量为30kGy,最终得到细胞培养肉3D支架。
实施例2

S3、将上述步骤S2中得到的混合溶液于10Mev电子加速器下进行辐照,辐照后支架的吸收剂量为40kGy,最终得到细胞培养肉3D支架。
实施例3
一种利用辐照成胶的可食用细胞培养肉3D支架的制备方法,包括以下步骤:
S1、将壳聚糖和羧甲基纤维素钠分别溶于水中,得到质量浓度为2%的壳聚糖溶液和羧甲基纤维素钠溶液,壳聚糖溶液和羧甲基纤维素钠溶液均通过静电相互作用形成糊状溶液。
S2、将上述步骤S1中得到的壳聚糖溶液逐滴加入到羧甲基纤维素钠溶液中,其中壳聚糖和羧甲基纤维素钠溶液的质量比为7:3,并采用均质机在12000rpm条件下均质混合,均质5次,每次15秒,得到混合溶液;

实施例4
一种利用辐照成胶的可食用细胞培养肉3D支架的制备方法,包括以下步骤:
S1、将壳聚糖和羧甲基纤维素钠分别溶于水中,得到质量浓度为2%的壳聚糖溶液和羧甲基纤维素钠溶液,壳聚糖溶液和羧甲基纤维素钠溶液均通过静电相互作用形成糊状溶液。
S2、将上述步骤S1中得到的壳聚糖溶液逐滴加入到羧甲基纤维素钠溶液中,其中壳聚糖溶液和羧甲基纤维素钠溶液的质量比为8:2,并采用均质机在12000rpm条件下均质混合,均质3次,每次15秒,得到混合溶液;
S3、将上述步骤S2中得到的混合溶液于10Mev电子加速器下进行辐照,辐照后支架的吸收剂量为40kGy,最终得到细胞培养肉3D支架。
实施例5
一种利用辐照成胶的可食用细胞培养肉3D支架的制备方法,包括以下步骤:
S1、将壳聚糖和羧甲基纤维素钠分别溶于水中,得到质量浓度为2%的壳聚糖溶液和羧甲基纤维素钠溶液,壳聚糖溶液和羧甲基纤维素钠溶液均通过静电相互作用形成糊状溶液。
S2、将上述步骤S1中得到的壳聚糖溶液逐滴加入到羧甲基纤维素钠溶液中,其中壳聚糖和羧甲基纤维素钠溶液的质量比为5:5,并采用均质机在14000rpm条件下均质混合,均质5次,每次15秒,得到混合溶液;
S3、将上述步骤S2中得到的混合溶液于10Mev电子加速器下进行辐照,辐照后支架的吸收剂量为40kGy,最终得到细胞培养肉3D支架。
实施例6
一种利用辐照成胶的可食用细胞培养肉3D支架的制备方法,包括以下步骤:
S1、将壳聚糖和羧甲基纤维素钠分别溶于水中,得到质量浓度为2%的壳聚糖溶液和羧甲基纤维素钠溶液,壳聚糖溶液和羧甲基纤维素钠溶液均通过静电相互作用形成糊状溶液。
S2、将上述步骤S1中得到的壳聚糖溶液逐滴加入到羧甲基纤维素钠溶液中,其中壳聚糖和羧甲基纤维素钠溶液的质量比为3:7,并采用均质机在13000rpm条件下均质混合,均质4次,每次15秒,得到混合溶液;
S3、将上述步骤S2中得到的混合溶液于10Mev电子加速器下进行辐照,辐照后支架的吸收剂量为40kGy,最终得到细胞培养肉3D支架。
由本发明方法制备出的壳聚糖/羧甲基纤维素钠细胞培养肉3D支架如图1所示,细胞培养肉3D支架的外形规则圆润,具有一定厚度,结构稳固,能够为进一步的细胞增殖分化提供有力机械支撑。
实验例1
细胞培养肉3D支架的性能测定实验。
对本发明实施例1~6制备的细胞培养肉3D支架的孔隙率、质量变化、质构特性进行测定。
其中测定方法如下:
孔隙率测定:使用场发射扫描电子显微镜对实施例样品表面进行扫描,使用Imagej软件对实施例孔隙率进行分析。
质量变化曲线:称量实施例初始质量(记为第0天),然后将其浸泡在DEME/F12培养基中在37℃、5%CO2环境下放置,此后分别在第1、3、5、7、9、11、13、15天取出称重。
质构特性:使用物性测定仪对不同实施例进行TPA分析。进行两次压缩,通过直径为36mm的圆柱形探针在5mm/s速度下将样品压缩至其初始高度的50%,结果以硬度、胶粘性、咀嚼性、回复性表示。
由实施例1~6制备的细胞培养肉3D支架具有较高的孔隙率,达40%以上(图2);细胞培养肉3D支架在15天后仍能保持较好的形态,为细胞长时间培养提供必要条件(图3)。支架孔隙率高可以构建三维的细胞培养环境,使细胞在更接近于自然情况的环境下生长和分化。此外,较高的孔隙率还可以促进培养液的渗透和流动,有利于营养物质和氧气的输送,维持细胞增殖的需求。实施例5制备的3D支架具有较高的硬度、胶粘性、咀嚼性和回复性,与肉的质构特性较为相似,能够为细胞的生长提供一定的机械支撑(图4)。
实验例2
细胞培养肉3D支架的成肌细胞培养实验。
对本发明实施例1~6制备的细胞培养肉3D支架进行成肌细胞培养,培养步骤如下:
1)将C2C12小鼠成肌细胞复苏离心并重悬于DMEM/F-12培养基中(10%FBS),于37℃、5%的CO2培养箱中培养至细胞汇合达到80%时,通过胰蛋白酶消化和离心获得细胞悬液。
2)将实施例1~6制备的细胞培养肉3D支架用PBS溶液洗涤三次后,加入培养基浸泡30min后弃去培养基,接种细胞后待细胞贴壁后补充培养基,在37℃、5%的CO2培养箱中培养。
3)培养1d后将支架进行活/死细胞染色,并使用荧光显微镜进行观察。
4)培养2d后将支架进行细胞骨架染色,并使用激光共聚焦显微镜进行观察,F-actin为细胞骨架。
结果如图5所示,由实施例1~6制备的细胞培养肉3D支架进行的C2C12小鼠成肌细胞培养,根据活/死细胞染色结果可以看出,培养1d后,实施例2制备的3D支架黏附细胞的能力较强,具有数量最多的活细胞。根据细胞骨架染色结果可以看出,培养2d后,小鼠成肌细胞(C2C12)在支架上大量增殖并开始伸展形成细长并相互连接的细胞骨架(图6)。根据细胞骨架染色结果可以看出,培养5d后,小鼠成肌细胞进一步伸展延伸,体现出3D支架具有良好的生物相容性(图7)。
从成肌细胞培养实验结果可以得出,除实施例5之外,其余实施例制备出的细胞培养肉3D支架均有较好的细胞增殖,且实施例2制备出的细胞培养肉3D支架具有较多的增殖数量和更好的细胞伸展。利用辐照引发交联的方法具有成本低、时间短、生产效率更高、产品质量更好和食品安全性得到保障的优点,有利于实现结构化细胞培养肉的大规模培养并推动细胞培养肉领域的进一步发展。
尽管上述实施例对本发明做出了详尽的描述,但它仅仅是本发明一部分实施例,而不是全部实施例,人们还可以根据本实施例在不经创造性前提下获得其他实施例,这些实施例都属于本发明保护范围。
Claims (10)
1.一种可食用的细胞培养肉3D支架的制备方法,其特征在于,包括以下步骤:将壳聚糖溶液和羧甲基纤维素钠溶液混合成均一的混合溶液,对所述混合溶液进行电子加速器辐照诱导成胶,得所述细胞培养肉3D支架。
2.根据权利要求1所述制备方法,其特征在于,所述混合包括在搅拌条件下,将壳聚糖溶液逐滴加入到羧甲基纤维素钠溶液中。
3.根据权利要求1或2所述制备方法,其特征在于,所述壳聚糖溶液和羧甲基纤维素钠溶液的质量比为(3~9):(1~7)。
4.根据权利要求3所述制备方法,其特征在于,所述壳聚糖溶液和羧甲基纤维素钠溶液均以水为溶剂,且质量百分浓度均为1~3%。
5.根据权利要求2所述制备方法,其特征在于,所述搅拌的转速为200~300rpm。
6.根据权利要求2所述制备方法,其特征在于,将壳聚糖溶液逐滴加入到羧甲基纤维素钠溶液后,还包括均质。
7.根据权利要求6所述制备方法,其特征在于,所述均质的转速为12000~15000rpm,均质3~5次,每次15秒。
8.根据权利要求1所述制备方法,其特征在于,采用10Mev电子加速器进行所述辐照,所述辐照的吸收剂量为30~50kGy。
9.利用权利要求1~8任一项所述制备方法得到的可食用的细胞培养肉3D支架。
10.权利要求9所述可食用的细胞培养肉3D支架在生产细胞培养肉中的应用。
