Abstract
本发明公开了一种预防口腔疾病的抑菌口腔喷雾及其制备方法,各组分按质量百分比计,包括,抗菌剂0.05wt%‑1wt%;抗氧化剂0.01wt%‑0.06wt%;甜味剂2wt%‑18wt%;保湿剂1wt%‑18wt%;pH缓冲剂0.12wt%‑4wt%;食用香精0.2wt%‑0.6wt%;矫味剂0.05wt%‑1wt%;防腐剂0.01‑0.05wt%;增溶剂0.01wt%‑2.5wt%,余量为纯化水;所述抗菌剂为异硫氰酸苄酯纳米乳和铜离子化合物的复合物;采用异硫氰酸苄酯纳米乳(BITC)和铜离子进行复配,从而具有更显著的抑菌性,安全可靠,无刺激性,有效的提高了稳定性和耐药性差、且无毒副作用。

技术领域
本发明属于生物医药技术领域,具体涉及了一种预防口腔疾病的抑菌口腔喷雾及其制备方法。
背景技术
口腔内的微生物群是人体内最多样化的微生物群之一,已知的微生物多达750种,包括细菌、真菌、病毒等。随着对口腔疾病的深入研究,研究者们发现龋齿、牙周病等困扰人们生活的口腔常见疾病都与口腔内有害细菌的滋生有关。诱发龋齿的病原微生物主要是变形链球菌、齿龈链球菌、放线菌等。这些细菌可以在口腔中形成牙菌斑,不断分泌酸性代谢物,从而导致龋齿的发生和进一步发展。此外,口腔内还有其他一些微生物可能也会对龋齿的形成起到一定的作用,如经常食用高糖和高脂食物,可能会促进龋齿的形成。由细菌污染引起的口腔疾病可能对全身性疾病的病程产生负面影响,导致较高的发病率和死亡率。因此,维持口腔中微生物群体的平衡,防止有害细菌在口腔中大量繁殖对维持口腔健康具有重要的作用。目前针对口腔病原微生物对口腔带来的疾病和危害,主要是通过抑制微生物的生长和清除牙菌斑来达到控制感染和炎症的目的。例如,口腔抗菌药物和口腔清洁用品等。其中口腔抗菌喷雾剂由于方便快捷、作用迅速、剂量准确、便于携带等优点备受消费者欢迎。
目前预防龋齿、牙周炎等口腔疾病的口腔喷剂主要包括氟化物、氨基酸和酵素三类。这些口腔喷剂在预防龋齿、牙周炎等口腔疾病方面有一定的效果,但是这些应用化学合成药物的口腔抗菌喷雾剂在使用时常常存在稳定性差、耐药性差、毒副作用等问题,致使寻找天然安全有效的抗菌药物成为口腔疾病预防的当务之急。因此植物提取物作为一种天然活性成分,由于低毒及良好的抑菌活性受到研究者的青睐。虽然目前专利中已有采用了檀香精油、蒜鳞茎提取物天然等植物成分制成的口腔喷雾剂,可以暂时抑制有害细菌,但是单个抑菌成分呈现的抑菌效果和持久性仍有待提高。
发明内容
针对现有技术中存在的问题,本发明提供一种预防口腔疾病的抑菌口腔喷雾及其制备方法,采用异硫氰酸苄酯(BITC)纳米乳和铜离子化合物进行复配,从而具有更显著的抑菌性,安全可靠,无刺激性,有效的提高了稳定性和耐药性差、且无毒副作用。
本发明是通过以下技术方案来实现:
一种预防口腔疾病的抑菌口腔喷雾,各组分按质量百分比计,包括,
抗菌剂0.05wt%-1wt%;抗氧化剂0.01wt%-0.06wt%;甜味剂2wt%-18wt%;保湿剂1wt%-18wt%;pH缓冲剂0.12wt%-4wt%;食用香精0.2wt%-0.6wt%;矫味剂0.05wt%-1wt%;防腐剂0.01-0.05wt%;增溶剂0.01wt%-2.5wt%;氟化钠0.1-0.25wt%,余量为纯化水;所述抗菌剂为异硫氰酸苄酯纳米乳和铜离子化合物的复合物;所述异硫氰酸苄酯纳米乳和铜离子化合物的质量比为1:1。
优选的,所述抗氧化剂包括十二烷醇酯、棓酸丁酯和棓酸丙酯中的至少一种;所述铜离子化合物包括硫酸铜、醋酸亚铜、氯化铜和乙酸铜中的任意一种。
优选的,所述甜味剂包括木糖醇、蔗糖和阿斯巴甜中的至少一种。
优选的,所述保湿剂包括壳聚糖、甘油和山梨糖醇中的至少一种。
优选的,所述异硫氰酸苄酯纳米乳通过25wt%的吐温80、8wt%的无水乙醇、2wt%的乙酸乙酯、2wt%的异硫氰酸苄酯和63wt%纯净水混合采用自发乳化法制备得到。
优选的,所述PH缓冲剂包括柠檬酸、磷酸氢钠、碳酸氢钠、柠檬酸钠和碳酸钠中的至少一种。
优选的,所述食用香精包括薄荷香精、苹果香精、柠檬香精和西柚香精中的至少一种。
优选的,所述矫味剂包括柠檬酸、柠檬酸钠和苹果酸中的至少一种。
优选的,所述防腐剂包括抗坏血酸、羟苯甲酯和苯甲酸钠中的至少一种。

一种预防口腔疾病的抑菌口腔喷雾的制备方法,包括:
S1:按照配比称取BITC和食用香精溶于增溶剂中,搅拌5~10min,制成混合溶液A;
S2:称取铜离子化合物、矫味剂、甜味剂、pH缓冲剂、氟化钠,抗氧化剂、保湿剂溶于水中,搅拌5~10min,使完全溶解,制成混合溶液B;
S3:将混合溶液A、混合溶液B混合,搅拌5~20min,使分散均匀,制成混合溶液C,并加入蒸馏水定容至100ml,搅拌20~40min,得到预防口腔疾病的抑菌口腔喷雾半成品;
S4:将预防口腔疾病的抑菌口腔喷雾半成品用有机系滤膜过滤,无菌灌装后制成预防口腔疾病的抑菌口腔喷雾成品。
与现有技术相比,本发明具有以下有益的技术效果:
本发明提供的一种预防口腔疾病的抑菌口腔喷雾,添加了一种新型复合抗菌成分异硫氰酸苄酯(BITC)纳米乳和硫酸铜的1:1复合物,其抗菌原理与抗生素不同,通过诱导细菌膜完整性的丧失来控制细菌的感染,而达到杀菌效果,不产生耐药性,且经研究认证,安全可靠。当BITC纳米乳与铜离子结合时,它们的抗菌效果增强,同时降低细菌耐药性发展的可能性。具有多方面的协同机制,包括,BITC纳米乳与铜离子的复合物会促进细胞膜破坏,BITC纳米乳可以破坏细菌膜的稳定,促进铜离子进入细胞;铜离子进一步破坏细胞膜,导致细胞内容物泄漏增加,并加速细菌死亡。其次,铜离子可以增强细菌细胞内ROS的生成,当与同样诱导氧化应激的BITC纳米乳结合时,ROS水平变得更高,对细胞成分造成更广泛的损伤,并增加杀菌作用。此外,BITC纳米乳与铜离子的复合物可靶向多种细胞过程,BITC纳米乳和铜离子可靶向细菌的不同细胞过程,这种组合可能会影响各种代谢途径,从而产生更广泛的抗菌活性,并降低细菌耐药性发展的可能性。
进一步,本发明的口腔喷雾剂中加入了氟化钠,它与牙釉质相互作用,形成一层保护层,有助于防止蛀牙。
进一步,本发明提供一种口腔喷雾剂可以直接喷入口腔,不需要水或其他液体辅助,使用非常方便快捷,气味清新,未添加乙醇,无刺激性,喷雾剂的喷头可以通过清洗来保持卫生,使用方便卫生。

实施例1:
一种预防口腔疾病的抑菌口腔喷雾,口腔喷雾由下述重量百分数的组分组成:BITC 0.15wt%;硫酸铜0.15wt%;十二烷醇酯0.2wt%;木糖醇12wt%;阿斯巴甜1wt%;壳聚糖5wt%;柠檬酸0.1wt%;柠檬酸钠0.2wt%;柠檬香精0.03wt%;抗坏血酸0.03wt%;氢化蓖麻油0.03wt%;氟化钠0.2wt%,余量为去离子水。
一种预防口腔疾病的抑菌口腔喷雾的制备方法,包括如下步骤:
S1:将柠檬香精和BITC溶于氢化蓖麻油中,搅拌10min,得到混合溶液A;
S2:称取硫酸铜、柠檬酸、木糖醇、阿斯巴甜、柠檬酸钠、抗坏血酸、十二烷醇酯、壳聚糖、氟化钠溶于水中,搅拌10min,使完全溶解,得混合溶液B;
S3:将混合溶液A、混合溶液B混合,搅拌5~20min,使分散均匀,制成混合溶液C,并加入蒸馏水定容至100ml,搅拌20~40min,得到预防口腔疾病的抑菌口腔喷雾半成品。
S4:将预防口腔疾病的抑菌口腔喷雾半成品用有机系滤膜过滤,无菌灌装后制成预防口腔疾病的抑菌口腔喷雾成品。
其中,采用自发乳化法制备异硫氰酸苄酯纳米乳:在容器中连续搅拌下添加25wt%的吐温80、8wt%的无水乙醇、2wt%的乙酸乙酯、2wt%的异硫氰酸苄酯和63wt%纯净水,制备获得异硫氰酸苄酯纳米乳。
氟化钠:氟化钠是一种众所周知的矿物质,因其对牙齿健康的显著影响而被广泛用于口腔护理产品。它与牙釉质相互作用,形成一层保护层,有助于防止蛀牙。优点:预防蛀牙:氟化钠能增强牙釉质,使其更能抵抗细菌和含糖食物的酸性侵蚀,从而降低患蛀牙的风险。再矿化:氟化物促进弱化牙釉质的再矿化,逆转早期蛀牙。安全且被广泛接受:氟化钠在口腔护理产品中已经使用了几十年,当以适当的浓度使用时,被认为是安全有效的。
实施例2:
一种预防口腔疾病的抑菌口腔喷雾,口腔喷雾由下述重量百分数的组分组成:BITC0.15wt%;硫酸铜0.15wt%;棓酸丁酯0.2wt%;木糖醇12wt%;蔗糖1wt%;甘油3wt%;苹果酸0.3wt%;柠檬酸钠0.2wt%;苹果香精0.02wt%;苯甲酸钠0.02wt%;PEG401.5wt%;氟化钠0.1wt%,余量为去离子水。
一种预防口腔疾病的抑菌口腔喷雾的制备方法,包括如下步骤:
S1:将苹果香精和BITC溶于氢化蓖麻油中,搅拌10min,得到混合溶液A;
S2:称取硫酸铜、苹果酸、木糖醇、蔗糖、柠檬酸钠、苯甲酸钠、PEG40、甘油、氟化钠溶于水中,搅拌10min,使完全溶解,得混合溶液B;
S3:将混合溶液A、混合溶液B混合,搅拌20min,使分散均匀,制成混合溶液C,并加入蒸馏水定容至100ml,搅拌20min,得到预防口腔疾病的抑菌口腔喷雾半成品。
S4:将预防口腔疾病的抑菌口腔喷雾半成品用有机系滤膜过滤,无菌灌装后制成预防口腔疾病的抑菌口腔喷雾成品。
实施例3:
一种预防口腔疾病的抑菌口腔喷雾,口腔喷雾由下述重量百分数的组分组成:BITC0.15wt%;硫酸铜0.15wt%;棓酸丙酯0.2wt%;木糖醇12wt%;阿斯巴甜1wt%;甘油4wt%;碳酸钠0.1wt%;柠檬酸钠0.2wt%;薄荷香精0.04wt%;羟苯甲酯0.03wt%;吐温-800.03wt%;氟化钠0.15wt%,余量为去离子水。
S1:将薄荷香精和BITC和硫酸铜的1:1复合物溶于氢化蓖麻油中,搅拌10min,得到混合溶液A;
S2:称取木糖醇、阿斯巴甜、碳酸钠、柠檬酸钠、羟苯甲酯、吐温-80、棓酸丙酯、氟化钠,溶于水中,搅拌10min,使完全溶解,得混合溶液B;
S3:将混合溶液A、混合溶液B混合,搅拌20min,使分散均匀,制成混合溶液C,并加入蒸馏水定容至100ml,搅拌40min,得到预防口腔疾病的抑菌口腔喷雾半成品。
S4:将预防口腔疾病的抑菌口腔喷雾半成品用有机系滤膜过滤,无菌灌装后制成预防口腔疾病的抑菌口腔喷雾成品。
实施例4
实施例4与实施例1的不同之处在于,铜离子化合物为醋酸亚铜。
实施例5
实施例5与实施例1的不同之处在于,铜离子化合物为氯化铜。

空白对照组为:不含BITC纳米乳与铜离子复合物,十二烷醇酯0.2wt%;木糖醇12wt%;阿斯巴甜1wt%;壳聚糖5wt%;柠檬酸0.1wt%;柠檬酸钠0.2wt%;柠檬香精0.03wt%;抗坏血酸0.03wt%;氢化蓖麻油0.03wt%;氟化钠0.1wt%,余量为去离子水。
实验例口腔喷雾1-6的抑菌活性测定:挑取S.aureusATCC25923和E.coliMG1655单克隆于无菌的TSB和LB液体培养基中,并在37℃、200r/min的条件下进行过夜培养,然后按照体积比200∶1(TSB液体培养基∶菌液)进行转接,待其OD600=0.8~1.0时,用相应液体培养基稀释菌液至OD600=0.1,并在此基础上稀释100倍,最后分别吸取5μL加至含有195μL菌液稀释液的96孔板中(200μL体系),12h药物处理后,滴板实验测定细菌菌落总数,进一步探究实例1-6对金黄色葡萄球菌的抑菌效果。

抑菌实验结果如图1所示,从图中可以看出,实施例1-6中的口腔喷雾对金黄色葡萄球菌和大肠杆菌均有很好的抑菌效果,抑菌率均大于99.9wt%。特别是选择硫酸铜的实例1-3能够抑制几乎100wt%的金黄色葡萄球菌和大肠杆菌的生长。这表明了实例1-6具有明显的抑菌能力。
实施例7:
一种预防口腔疾病的抑菌口腔喷雾,口腔喷雾由下述重量百分数的组分组成:BITC0.5wt%;硫酸铜0.5wt%;十二烷醇酯0.01wt%和棓酸丁酯0.01wt%;蔗糖17wt%;阿斯巴甜1wt%;甘油1wt%;碳酸氢钠0.12wt%;柠檬酸钠0.05wt%;西柚香精0.2wt%;羟苯甲酯0.01wt%;吐温-80的2.5wt%;余量为去离子水。
一种预防口腔疾病的抑菌口腔喷雾的制备方法,包括如下步骤:
S1:将西柚香精和BITC溶于吐温-80中,搅拌15min,得到混合溶液A;
S2:称取硫酸铜、十二烷醇酯和棓酸丁酯、蔗糖、阿斯巴甜、甘油、碳酸氢钠、柠檬酸钠、西羟苯甲酯溶于水中,搅拌10min,使完全溶解,得混合溶液B;
S3:将混合溶液A、混合溶液B混合,搅拌10min,使分散均匀,制成混合溶液C,并加入蒸馏水定容至100ml,搅拌30min,得到预防口腔疾病的抑菌口腔喷雾半成品。
S4:将预防口腔疾病的抑菌口腔喷雾半成品用有机系滤膜过滤,无菌灌装后制成预防口腔疾病的抑菌口腔喷雾成品。
实施例8:
一种预防口腔疾病的抑菌口腔喷雾,口腔喷雾由下述重量百分数的组分组成:BITC0.025wt%;硫酸铜0.025wt%;棓酸丙酯0.03wt%和棓酸丁酯0.03wt%;蔗糖1wt%和木糖醇1wt%;山梨糖醇9wt%和甘油9wt%;碳酸钠2wt%和柠檬酸钠2wt%;薄荷香精0.6wt%;苯甲酸钠0.05wt%;PEG40的0.1wt%;余量为去离子水。
一种预防口腔疾病的抑菌口腔喷雾的制备方法,包括如下步骤:
S1:将薄荷香精和BITC溶于PEG40中,搅拌15min,得到混合溶液A;
S2:称取硫酸铜、棓酸丙酯和棓酸丁酯、蔗糖、木糖醇、山梨糖醇、甘油、碳酸钠、柠檬酸钠、苯甲酸钠溶于水中,搅拌10min,使完全溶解,得混合溶液B;
S3:将混合溶液A、混合溶液B混合,搅拌10min,使分散均匀,制成混合溶液C,并加入蒸馏水定容至100ml,搅拌30min,得到预防口腔疾病的抑菌口腔喷雾半成品。
S4:将预防口腔疾病的抑菌口腔喷雾半成品用有机系滤膜过滤,无菌灌装后制成预防口腔疾病的抑菌口腔喷雾成品。
实施例9:
一种预防口腔疾病的抑菌口腔喷雾,口腔喷雾由下述重量百分数的组分组成:BITC0.04wt%;硫酸铜0.04wt%;棓酸丙酯0.02wt%和棓酸丁酯0.01wt%;蔗糖4wt%和木糖醇6wt%;山梨糖醇5wt%和甘油8wt%;苹果酸0.12wt%;柠檬酸钠0.1wt%;苹果香精0.4wt%;苯甲酸钠0.01wt%和羟苯甲酯0.03wt%;PEG40的0.01wt%;余量为去离子水。
一种预防口腔疾病的抑菌口腔喷雾的制备方法,包括如下步骤:
S1:将苹果香精和BITC溶于PEG40中,搅拌15min,得到混合溶液A;
S2:称取硫酸铜、棓酸丙酯和棓酸丁酯、蔗糖、木糖醇、山梨糖醇、甘油、苹果酸、柠檬酸钠、苯甲酸钠、羟苯甲酯溶于水中,搅拌10min,使完全溶解,得混合溶液B;
S3:将混合溶液A、混合溶液B混合,搅拌10min,使分散均匀,制成混合溶液C,并加入蒸馏水定容至100ml,搅拌30min,得到预防口腔疾病的抑菌口腔喷雾半成品。
S4:将预防口腔疾病的抑菌口腔喷雾半成品用有机系滤膜过滤,无菌灌装后制成预防口腔疾病的抑菌口腔喷雾成品。

抗菌剂0.05wt%-1wt%;抗氧化剂0.01wt%-0.06wt%;甜味剂2wt%-18wt%;保湿剂1wt%-18wt%;pH缓冲剂0.12wt%-4wt%;食用香精0.2wt%-0.6wt%;矫味剂0.05wt%-1wt%;防腐剂0.01-0.05wt%;增溶剂0.01wt%-2.5wt%;氟化钠0.1-0.25wt%,余量为纯化水;所述抗菌剂为异硫氰酸苄酯纳米乳和铜离子化合物的复合物;所述异硫氰酸苄酯纳米乳和铜离子化合物的质量比为1:1。
2.根据权利要求1所述的一种预防口腔疾病的抑菌口腔喷雾,其特征在于,所述抗氧化剂包括十二烷醇酯、棓酸丁酯和棓酸丙酯中的至少一种;所述铜离子化合物包括硫酸铜、醋酸亚铜、氯化铜和乙酸铜中的任意一种。
3.根据权利要求1所述的一种预防口腔疾病的抑菌口腔喷雾,其特征在于,所述甜味剂包括木糖醇、蔗糖和阿斯巴甜中的至少一种;所述保湿剂包括壳聚糖、甘油和山梨糖醇中的至少一种。
4.根据权利要求1所述的一种预防口腔疾病的抑菌口腔喷雾,其特征在于,所述异硫氰酸苄酯纳米乳通过25wt%的吐温80、8wt%的无水乙醇、2wt%的乙酸乙酯、2wt%的异硫氰酸苄酯和63wt%纯净水混合采用自发乳化法制备得到。
5.根据权利要求1所述的一种预防口腔疾病的抑菌口腔喷雾,其特征在于,所述pH缓冲剂包括柠檬酸、碳酸氢钠、磷酸氢钠、柠檬酸钠和碳酸钠中的至少一种。
6.根据权利要求1所述的一种预防口腔疾病的抑菌口腔喷雾,其特征在于,所述食用香精包括薄荷香精、苹果香精、柠檬香精和西柚香精中的至少一种。
7.根据权利要求1所述的一种预防口腔疾病的抑菌口腔喷雾,其特征在于,所述矫味剂包括柠檬酸、柠檬酸钠和苹果酸中的至少一种。
8.根据权利要求1所述的一种预防口腔疾病的抑菌口腔喷雾,其特征在于,所述防腐剂包括抗坏血酸、羟苯甲酯和苯甲酸钠中的至少一种。
9.根据权利要求1所述的一种预防口腔疾病的抑菌口腔喷雾,其特征在于,所述增溶剂包括氢化蓖麻油、吐温-80和PEG40中的一种。
10.一种预防口腔疾病的抑菌口腔喷雾的制备方法,基于权利要求1-9任意一项所述的预防口腔疾病的抑菌口腔喷雾,其特征在于,包括:
S1:按照配比称取异硫氰酸苄酯纳米乳和食用香精溶于增溶剂中,搅拌5~10min,制成混合溶液A;
S2:称取铜离子化合物、矫味剂、甜味剂、pH缓冲剂、氟化钠,抗氧化剂、保湿剂溶于水中,搅拌5~10min,使完全溶解,制成混合溶液B;
S3:将混合溶液A、混合溶液B混合,搅拌5~20min,使分散均匀,制成混合溶液C,并加入纯化水定容至100ml,搅拌20~40min,得到预防口腔疾病的抑菌口腔喷雾半成品;
S4:将预防口腔疾病的抑菌口腔喷雾半成品用有机系滤膜过滤,无菌灌装后制成预防口腔疾病的抑菌口腔喷雾成品。
