The method provided by the invention is used for verifying the regulating effect of the control group cat food and the main food cat food prepared by the formulas and the corresponding processing methods in examples 1-4 on the oral cavity microorganism composition of the cat.
Materials and methods
Animals
The 20 adult cats were equally divided into two groups according to the principle of consistency of age, sex and weight, wherein the average weight of the 20 cats was 4.71kg (4.06-5.15 kg) and the average age was 4 years (3-5 years).
Feeding and raising
Before the experiment, 20 cats are uniformly fed with control cat food for 30 days, then the control group is fed with control cat food, the experiment group is fed with main food cat food, and all cats eat cat food and drink water freely during the experiment, and the experiment period is 42 days.

Sample collection
On day 0 and day 42 at the beginning of the formal experiment, microbial samples of the oral surface and gingival sulcus were collected by sampling swabs for oral microbial composition analysis.
Oral microbiological composition analysis
Microbial genomic DNA in the oral cavity sample is extracted by a genomic DNA extraction kit, V3-V4 regions of the 16S rRNA gene are amplified by using 338F and 806R primers, and the amplified sequences are summarized and sequenced on an Illumina Miseq platform. Quality control was performed on the original paired-end sequencing reads using fastp software, then the read data were pooled using flash software, and operational taxa were generated from the quality filtered and pooled sequences using UPARSE software with a 97% similarity threshold, while chimeric sequences were removed, allowing normalization of the sequence abundance for all samples. The operational classifier units (OTUs) were labeled using an RDP classifier, compared to the silva138/16s_bacteria database with a confidence threshold of 70%, and the microbial composition and differences of each sample were analyzed at different classification levels.
Results
This example uses the method of the invention to examine the effect of cat food on the alpha diversity of the oral microorganisms of cats.
As shown in fig. 5, the graph shows that there is a very significant difference between the two groups, representing P <0.001.
On experiment day 42, the Chao index and the Coverage index in the cat oral cavity microorganism alpha diversity index of the main food cat food group (CPG group) are obviously higher than the richness of the cat oral cavity microorganism alpha diversity index of the feeding control group (CON group), and the results of the embodiment show that the main food cat food can effectively improve the richness of the cat oral cavity microorganisms, and then specific differential microorganisms are further analyzed.

On experiment day 42, the cat oral clostridium (Fusobacterium) relative abundance of the staple food feeding group was significantly lower than that of the control group (P < 0.05), while the Lactobacillus (Lactobacillus) (p=0.004) and Bifidobacterium (Bifidobacterium) (p=0.004) relative abundance was significantly higher than that of the control group. The relative abundance of the bacteria of the main food cat food group Porphyromonas stomatitis (p=0.054), vibrio desulphus (p=0.042), treponema (Treponema) (p=0.031) etc. was significantly reduced compared to experimental day 0. The results of the embodiment show that the composition of the oral microorganisms of the cat can be effectively improved by feeding the main food cat food, so that the relative abundance of harmful bacteria in the oral cavity of the cat is obviously reduced, and the relative abundance of beneficial bacteria is obviously increased.
The implementation result shows that the relative abundance of the microorganisms in the oral cavity of the cat can be effectively improved by feeding the main food cat food, the relative abundance of the beneficial microorganisms in the oral cavity of the cat is improved, the relative abundance of the harmful microorganisms in the oral cavity of the cat is reduced, and the balance of the microorganisms in the oral cavity of the cat is maintained.
Example 6
Application of staple food cat food with tooth cleaning function in improving oral health of cats
With the method of the invention, verification of improving oral health effects of cats is performed on the staple food cat food prepared by the staple food cat food formula in example 1 and the corresponding staple food cat food processing method in example 3.
Materials and methods
Animals
10 adult cats (4.06-5.15 kg) with average weight of 4.71kg and average age of 4 years (3-5 years) were used to evaluate the effect of the staple food cat food on oral health improvement of cats.
Feeding and raising
10 cats were continuously fed the main food cat food for 42 days, and all cats were free to eat cat food and drink water during the experiment.

Oral health status check
On experiment day 42, the heads of each test animal were manually held in an upright position, and evaluated by a trained tester who had the nose as close as possible to the open mouth, smelled the smell in the mouth, and scored the cat oral gas in a sensory manner, and observed the distribution and area size of plaque and calculus on the cat tooth surface by plaque developer.
Results
The embodiment adopts the invention to test the effect of the staple food cat food on improving the oral health state of the cat. The results of the embodiment show that the main food cat food can effectively improve the smell of the oral cavity of the cat, reduce the dental plaque and dental calculus area, be beneficial to improving the oral health state of the cat and reduce the occurrence of oral related diseases.
