The embodiment provides a rosmarinic acid premix for relieving lung inflammation of broiler chickens induced by particles, which comprises the following raw materials in parts by weight in every 1000kg of daily ration: 613.5kg of corn, 310kg of soybean meal, 40kg of soybean oil, 16kg of stone powder, 10kg of calcium hydroxide, 2kg of salt, 1kg of lysine, 4kg of methionine, 1kg of 50% choline, 4kg of compound premix and 1kg of compound vitamin;
wherein each 1kg of the compound premix comprises the following components in parts by weight: 35g of copper sulfate pentahydrate, 150g of ferrous sulfate monohydrate, 165g of manganese sulfate monohydrate, 100g of zinc sulfate monohydrate, 32g of 1% sodium selenite (containing 0.45% of selenium), 18g of 1% potassium iodide (containing 0.76% of iodine), 390g of medical stone and 110g of 10% rosmarinic acid;
the raw materials are mixed according to the proportion, and after the compound vitamin is added, each kilogram of basic diet contains VA 12500IU, VD3 3000IU, VE 21.5mg, VK 3.7 mg, VB 1.6 mg, VB 22mg, VB 6.5 mg, VB 12.026 mg, biotin 0.04mg, folic acid 4mg, D-pantothenic acid 10mg and nicotinic acid 56mg.
To verify the feasibility of the process according to the invention, experiments were carried out on the rosmarinic acid premix described in examples 1-4, the experiments being illustrated as follows:
1. laboratory animals and groups
100 commercial AA meat hatchlings of 1 day old are selected and fed with corn-soybean meal type diet prepared according to the recommended nutrition amounts of the broilers of 1-21 days old and 22-42 days old of U.S. NRC (1994). When the chickens are raised to 14 days of age, 72 chickens with close weight are selected and randomly divided into three groups: control group, PM 2.5 Exposure group, PM 2.5 Exposure + rosmarinic acid group, 6 replicates per group, 4 broilers per replicate.
First, the exposure groups were transferred to 3 identical control rooms from 22 days old after 1 week adaptation in an artificial environment control room, and each PM was produced by a liquid aerosol generator method 2.5 Charging corresponding PM in exposure group 2.5 Concentration (1000 mug/m) 3 PM 2.5 :2 h/d); each control group adopts sterile physiological saline, and PM is monitored in real time during exposure period 2.5 Concentration, test duration, 21 days, feeding management according to the AA broiler feeding management manual.
2. Test design
All broilers were weighed on day 21 of exposure, after which 1 broiler with close weight was randomly selected for fin vein blood sampling in each repetition and supernatant was centrifuged at 3000 r/min and cryopreserved at-20 ℃. After euthanasia of broiler chickens, a small piece of lung tissue was taken, quickly frozen with liquid nitrogen, then stored at-80 ℃, and lung tissue inflammation markers were obtained by a real-time fluorescent quantitative (RT-PCR) method. The cecal content was also taken for analysis of the cecal microbiota.
Analyzing inflammatory factors in blood of broiler chickens by ELISA (enzyme-Linked immuno sorbent assay) method, wherein the inflammatory factors comprise TNF-alpha, IFN gamma, IL-1 beta, IL-6 and IL-10;
analyzing inflammatory factors in the lung of broiler chickens by adopting an RT-PCR technology, wherein the inflammatory factors comprise TNF-alpha, IFN gamma, IL-1 beta, IL-6 and IL-10; and recording the test result.
Analyzing TLR4 signal path factors in broiler lungs by adopting an RT-PCR technology, wherein the TLR4 signal path factors comprise TLR4, myD88 and NF-kB; and recording the test result.
The cecal content flora composition was analyzed using 16S rRNA sequencing technology.
3. Analysis of results:
as shown in fig. 1, the detection result of inflammatory factor in blood of broiler chicken is shown in fig. 2, the detection result of inflammatory factor in lung tissue of broiler chicken is shown in fig. 3, the detection result of TLR4 signal path in lung tissue of broiler chicken is shown in fig. 4, and the detection result of cecum flora of broiler chicken is shown in fig. 5, wherein PM group is PM 2.5 Exposure group, RA group, is feed prepared by using the compound premix raw materials added with rosmarinic acid described in examples 1-4, namely PM 2.5 Exposure + rosmarinic acid group.
According to the graph, after the rosmarinic acid premix prepared by the invention is eaten by broilers, activation of lung TLR4 signal paths is inhibited, the content of inflammatory factors is obviously reduced, intestinal flora is remodeled, the abundance of beneficial bacteria lactic acid bacteria is increased, and the abundance of salmonella is reduced, so that the rosmarinic acid premix for relieving the particulate matter-induced pulmonary inflammation of broilers and remodelling the intestinal flora has obvious effects on treating broiler pneumonia.
The foregoing description of the preferred embodiments of the invention is not intended to limit the invention to the particular embodiments disclosed, but on the contrary, the intention is to cover all modifications, equivalents, and alternatives falling within the spirit and scope of the invention.

Claims (6)

1. A rosmarinic acid premix for alleviating particulate-induced pulmonary inflammation of broilers, comprising: the material consists of the following raw materials in parts by weight: 600-620 parts of corn, 300-310 parts of bean pulp, 40-50 parts of soybean oil, 10-20 parts of stone powder, 10-20 parts of calcium hydride, 1-5 parts of salt, 1-2 parts of lysine, 1-2 parts of methionine, 1-3 parts of 50% choline, 2-4 parts of compound premix and 0.1-2.0 parts of compound vitamin;
the compound premix comprises the following components in parts by weight: 30-50g of copper sulfate pentahydrate, 120-150g of ferrous sulfate monohydrate, 150-170g of manganese sulfate monohydrate, 100-120g of zinc sulfate monohydrate, 30-50g of 1% sodium selenite, 10-20g of 1% potassium iodide, 380-400g of medical stone and 100-120g of 10% rosmarinic acid.
2. The rosmarinic acid premix for alleviating particulate-induced pulmonary inflammation of broilers of claim 1, wherein: after the compound vitamin is added, the compound vitamin meets the requirements of 12000-15000IU, 1000-3000IU, 20-22mg VE, 3 2-3.2mg VK, 1 1-3mg VB2 2-5mg VB6 2-4mg, 0.02-0.03mg VB, 0.03-0.04mg biotin, 1-3mg folic acid, 10-12mg D-pantothenic acid and 50-60mg nicotinic acid in each kilogram of basic ration.
3. The rosmarinic acid premix for alleviating particulate-induced pulmonary inflammation of broilers of claim 1, wherein: the compound premix comprises the following components in parts by weight: 30g of copper sulfate pentahydrate, 140g of ferrous sulfate monohydrate, 160g of manganese sulfate monohydrate, 120g of zinc sulfate monohydrate, 40g of 1% sodium selenite, 20g of 1% potassium iodide, 390g of medical stone and 100g of 10% rosmarinic acid.
4. The rosmarinic acid premix for alleviating particulate-induced pulmonary inflammation of broilers of claim 1, wherein: the compound premix comprises the following components in parts by weight: 40g of copper sulfate pentahydrate, 120g of ferrous sulfate monohydrate, 170g of manganese sulfate monohydrate, 120g of zinc sulfate monohydrate, 30g of 1% sodium selenite, 10g of 1% potassium iodide, 380g of medical stone and 120g of 10% rosmarinic acid.
5. The rosmarinic acid premix for alleviating particulate-induced pulmonary inflammation of broilers of claim 1, wherein: the compound premix comprises the following components in parts by weight: 50g of copper sulfate pentahydrate, 130g of ferrous sulfate monohydrate, 150g of manganese sulfate monohydrate, 110g of zinc sulfate monohydrate, 45g of 1% sodium selenite, 15g of 1% potassium iodide, 400g of medical stone and 100g of 10% rosmarinic acid.
6. The rosmarinic acid premix for alleviating particulate-induced pulmonary inflammation of broilers of claim 1, wherein: the compound premix comprises the following components in parts by weight: 35g of copper sulfate pentahydrate, 150g of ferrous sulfate monohydrate, 165g of manganese sulfate monohydrate, 100g of zinc sulfate monohydrate, 32g of 1% sodium selenite, 18g of 1% potassium iodide, 390g of medical stone and 110g of 10% rosmarinic acid.
