Abstract
本发明提供了一种DNA四面体‑氨磷汀复合物及其制备方法和在辐射保护中的用途,属于生物医药技术领域。本发明DNA四面体‑氨磷汀复合物是由DNA四面体框架核酸和氨磷汀混合后形成的复合物。本发明复合物以DNA四面体为载体进行氨磷汀的协同递送。本发明DNA四面体‑氨磷汀复合物克服了氨磷汀代谢时间短的问题,并且增强了氨磷汀的辐射保护作用,取得了协同增效的辐射保护效果。本发明复合物可以在辐射环境中对DNA进行保护,同时对多种器官有保护作用,并且能提高辐射下的生存率,具有优异的辐射保护作用,在多种领域具有优异的应用前景。
Description
技术领域
本发明属于生物医药技术领域,具体涉及一种DNA四面体-氨磷汀复合物及其制备方法和在辐射保护中的用途。
背景技术
电离辐射(ionizing radiation,IR)在工业、农业和医学中被广泛应用,导致辐射暴露的风险增加。过量辐射会对机体造成严重的生理后果,甚至危及生命,如急性辐射综合征、慢性辐射综合征和癌症。目前超过一半的癌症患者在临床常规治疗中接受放射治疗。放疗作为治疗恶性肿瘤的主要方式,通过破坏癌细胞的DNA和诱导细胞凋亡来杀死癌细胞,同时也对附近的健康组织造成了不良危害。早期毒性包括皮肤和黏膜疾病;远期毒性如慢性炎症和组织纤维化,可能几个月或数年后才会出现,甚至最终引起继发性癌症。此外,下腹部或骨盆的放疗很可能影响机体生育能力。
为减少辐射损伤带来的健康危害,研究开发高效低毒的辐射保护剂具有较大的应用前景,目前研究的药物种类繁多,主要有氨巯基类药物、细胞因子类、激素类药物、氮氧自由基类、天然药物等。但这些离理想的辐射防护剂还有很大差距,且部分药物在使用后出现了严重的副作用,限制了其临床使用。其中,氨磷汀(amifostine,AMF)是一种广谱的细胞保护剂,可以抵抗电离辐射和化疗药物对DNA的破坏作用,被批准为与放疗相关的狭窄临床适应症的唯一辐射保护剂。正常组织中比肿瘤组织具有更高的碱性磷酸酶活性,可将AMF转化为WR-1065的活性形式从而发挥作用,因此AMF在正常组织中表现出选择性辐射保护作用,而不保护肿瘤。然而,AMF的消除半衰期短,很快就会被人体代谢排出体外,限制了其生物利用度和临床应用。而增加使用剂量又会引起全身毒性和诸如低血压、恶心和呕吐等并发症。因此开发高效低毒的辐射保护剂依然具有极大的应用前景和社会效益。
四面体框架核酸(tetrahedral framework nucleic acid,tFNA)又称DNA四面体(tetrahedral DNA,TDN)、四面体DNA纳米结构,它由4条单链DNA通过链间碱基互补配对形成,形态类似四面体。DNA四面体结构稳定,具有良好的生物相容性,可作为药物载体。但是DNA四面体自身的特殊结构和具有一定药用活性,使其负载小分子药物后两者相互作用如何很难预料,它们是发挥协同作用还是拮抗作用是未知的,不同结构的小分子药物效果可能会出现较大的不同。目前尚未见DNA四面体负载氨磷汀,能否使用DNA四面体负载氨磷汀,取得更好的辐射保护效果需要进一步研究。
发明内容
本发明的目的是提供一种DNA四面体-氨磷汀复合物及其制备方法和在辐射保护中的用途。
本发明提供了一种DNA四面体-氨磷汀复合物,它是由DNA四面体框架核酸和氨磷汀混合后形成的复合物。
进一步地,所述DNA四面体和氨磷汀混合时,DNA四面体和氨磷汀的摩尔比为1:160~1:1280。
进一步地,所述DNA四面体和氨磷汀的摩尔比为1:640。
进一步地,所述DNA四面体由四条DNA单链自组装合成;所述四条DNA单链的序列分别如SEQ ID NO.1、SEQ ID NO.2、SEQ ID NO.3、和SEQ ID NO.4所示。
进一步地,所述DNA四面体的合成方法包括以下步骤:将四条DNA单链加入到TM缓冲液中,95℃维持10min,4℃维持20min以上,即得。
进一步地,所述四条DNA单链为等摩尔比的四条DNA单链。
本发明还提供了一种制备前述的DNA四面体-氨磷汀复合物的方法,它包括如下步骤:
将四面体框架核酸和氨磷汀加入溶剂中混合反应,即得。
进一步地,
所述溶剂为水、PBS、DMSO中的一种或多种;
和/或,所述四面体框架核酸的浓度为200~250nM;
和/或,所述反应的温度为4~10℃;
和/或,所述反应的时间为4~6h。
本发明还提供了前述的DNA四面体-氨磷汀复合物在制备预防辐射损伤的药物中的用途。
进一步地,所述药物为预防辐射中血液系统和/或器官损伤的药物;
和/或,所述药物为预防DNA损伤的药物。
与现有技术相比,本发明的有益效果为:
(1)本发明DNA四面体-氨磷汀复合物能保护DNA免受辐射损伤;
(2)本发明DNA四面体-氨磷汀复合物能提高机体在致命辐射下的生存率;
(3)本发明DNA四面体-氨磷汀复合物能实现亚致命辐射下的多器官保护。
综上,本发明提供了一种DNA四面体-氨磷汀复合物,其以DNA四面体为载体进行氨磷汀的协同递送。本发明DNA四面体-氨磷汀复合物克服了氨磷汀代谢时间短的问题,并且增强了氨磷汀的辐射保护作用,取得了协同增效的辐射保护效果。本发明复合物可以在辐射环境中对DNA进行保护,同时对多种器官有保护作用,并且能提高辐射下的生存率,具有优异的辐射保护作用,在多种领域具有优异的应用前景。
显然,根据本发明的上述内容,按照本领域的普通技术知识和惯用手段,在不脱离本发明上述基本技术思想前提下,还可以做出其它多种形式的修改、替换或变更。

具体实施方式
本发明具体实施方式中使用的原料、设备均为已知产品,通过购买市售产品获得。
实施例1、本发明DNA四面体-氨磷汀复合物的制备和表征
本发明DNA四面体-氨磷汀复合物的合成示意图如图1所示。
1、DNA四面体(tFNAs)的制备
将四条DNA单链(S1、S2、S3、S4)溶解于TM Buffer(10mM Tris-HCl,50mM MgCl2,pH=8.0)中,四条DNA单链的终浓度均为1μM,充分混合,迅速加热至95℃保持10分钟,之后迅速降温至4℃并维持20分钟以上,即可得到DNA四面体(tFNAs)。四条DNA单链的序列如表1所示。

2、DNA四面体-氨磷汀复合物(nanosuit)的制备
氨磷汀(AMF)溶液的制备:AMF购买自MCE,直接采用ddH2O进行溶解,得到AMF溶液。
DNA四面体-氨磷汀复合物(nanosuit)的制备:将合成的DNA四面体(tFNAs)与氨磷汀溶液混合均匀,使溶液中DNA四面体的浓度为250nM,氨磷汀的浓度为160μM(溶液中DNA四面体和氨磷汀的摩尔比为1:640),混合后的溶液在4℃孵育6小时,得到DNA四面体-氨磷汀复合物(nanosuit)。
3、表征及结果
采用毛细管电泳(HPCE)、动态光散射(DLS)、透射电镜(TEM)和电荷测定对tFNAs以及nanosuit进行表征。
图2为各ssDNA、tFNAs和nanosuit的毛细管电泳图(HPCE)。图3为tFNAs和复合物nanosuit的粒径结果。图4为tFNAs和复合物nanosuit的Zeta电位结果。图5为tFNAs和复合物nanosuit的透射电镜图。
HPCE显示tFNAs大小为180bp左右,动态光散射分析出tFNAs的粒径大小为10nm左右。透射电镜检测结果表明tFNAs分布均匀并且大小约为10nm。电荷测定证明tFNAs带负电。从以上结果可知,tFNAs的合成是成功的。
HPCE可见复合物nanosuit大小约为180-200bp,粒径大小约为18nm,Zeta电位为-7.42mV。使用透射电镜观察nanosuit,镜下可见均一的特征结构。结果显示复合物nanosuit的分子大小、粒径和Zeta电位均可稳定测出,且略大于单纯tFNAs,说明nanosuit的成功合成。
实施例2、本发明DNA四面体-氨磷汀复合物的制备

2、DNA四面体-氨磷汀复合物(nanosuit)的制备
nanosuit的制备方法同实施例1,只是DNA四面体的浓度为250nM,而氨磷汀的浓度由160μM改为40μM,溶液中DNA四面体和氨磷汀的摩尔比为1:160。
实施例3、本发明DNA四面体-氨磷汀复合物的制备
1、DNA四面体(tFNAs)的制备
DNA四面体(tFNAs)的制备方法同实施例1。
2、DNA四面体-氨磷汀复合物(nanosuit)的制备

DNA四面体(tFNAs)的制备方法同实施例1。
2、DNA四面体-氨磷汀复合物(nanosuit)的制备
nanosuit的制备方法同实施例1,只是DNA四面体的浓度为250nM,而氨磷汀的浓度由160μM改为320μM,溶液中DNA四面体和氨磷汀的摩尔比为1:1280。
根据HPLC结果计算实施例1~5制备得到的复合物nanosuit的包封率。如图6A所示:随着AMF浓度的增加,包封率升高,达到峰值后下降,AMF浓度为160μM时包封率最高。为了进一步验证实施例1~5复合物nanosuit的成功合成,采用荧光分光光度计测定了Gel-red和Hochest 33342的荧光光谱。图6B证明了随着AMF浓度增加至160μM,Gel-red和Hochest 33342的荧光强度均减弱,证明了AMF与tFNAs的沟槽结合和嵌插结合。综上,选择AMF浓度160μM进行后续研究。
以下通过具体实验例证明本发明的有益效果。
实验例1、细胞实验
1、细胞类型
小鼠胚胎成纤维细胞(BALB/3T3 clone A31细胞)。
2、相关检测方法和结果
(1)检测nanosuit进入A31细胞的情况
带有Cy5(红色荧光基团)的250nM ssDNAs、250nM tFNAs和250nM nanosuit分别与等量A31细胞孵育6h和12h,不给予辐照,之后采用免疫荧光和流式细胞术检测各样品入胞情况。其中,带有Cy5的ssDNAs是在S1单链上接枝Cy5;带有Cy5的tFNAs按照实施例1所述方法制备,仅仅是将S1单链替换为前述接枝Cy5的S1;带有Cy5的nanosuit是按照实施例1所述方法制备,仅仅将tFNAs替换为前述带有Cy5的tFNAs。
免疫荧光:采用4%多聚甲醛固定细胞,鬼笔环肽(绿色)和DAPI(蓝色)分别用来染细胞骨架和细胞核。最后采用共聚焦显微镜进行观察采图。
流式细胞术:收集细胞,使用流式细胞仪检测细胞对材料的摄取情况,FlowJov10.0.7软件分析入胞数据。
免疫荧光和流式细胞术检测结果如图7所示,结果发现nanosuit在12h入胞率达到最高,显著强于ssDNAs和tFNAs。
(2)检测nanosuit对辐射损伤细胞活力的作用
通过细胞活力测试评估nanosuit在不同剂量辐射(2Gy、4Gy、6Gy、8Gy和10Gy)下对细胞存活的促进作用。
实验分组:
模型组(IR):小鼠胚胎成纤维细胞培养36h后,给予不同剂量辐照,辐照12h后进行相应检测。
实验组(tFNAs、AMF和nanosuit):小鼠胚胎成纤维细胞培养24h后分别加入250nMtFNAs、160μM AMF和250nM nanosuit进行预处理12h,然后给予不同剂量辐照,辐照12h后进行相应检测。tFNAs和nanosuit采用实施例1方法制备。
使用CCK8试剂检测,酶标仪检测吸光度,计算每组细胞活性结果。
由图8A可知:细胞经过辐照后活性显著下降,随辐照剂量增加,细胞活性下降越严重;单独使用tFNAs预处理无法对细胞进行保护;单独使用AMF预处理在一定程度上可以挽救辐射诱导的细胞死亡;但是与AMF相比,nanosuit挽救辐射诱导的细胞死亡效果显著提高,表明nanosuit对细胞具有强大的辐射保护行为。
(3)检测nanosuit的抗氧化应激作用
受辐射影响,ROS会急剧增加,引发细胞氧化应激损伤。检测nanosuit的抗氧化应激作用,方法如下:
对照组(Control):小鼠胚胎成纤维细胞培养48h。
模型组(IR):小鼠胚胎成纤维细胞培养36h后,给予6Gy辐照,辐照12h后进行相应检测。
实验组(tFNAs、AMF和nanosuit):小鼠胚胎成纤维细胞培养24h后分别加入250nMtFNAs、160μM AMF和250nM nanosuit进行预处理12h,然后给予6Gy辐照,辐照12h后进行相应检测。分别为tFNAs+IR、AMF+IR、nanosuit+IR。tFNAs和nanosuit采用实施例1方法制备。
使用ROS检测试剂盒检测细胞内ROS水平:细胞中装载DCFH-DA探针,在37℃下孵育20min。最后,使用共聚焦激光扫描显微镜进行观察采图。
活性氧测试结果(图8B)显示nanosuit预处理在一定程度上降低了IR诱导的细胞内活性氧水平,说明nanosuit的辐射保护能力与其有效的自由基清除活性有关。
(4)检测nanosuit的抗凋亡作用
细胞凋亡是放射性敏感组织中大规模细胞丢失的主要机制。具体实验分组和方法如上述(3)所示,达到时间点后,用4%多聚甲醛溶液将样品固定15-20分钟,0.5% TritonX-100打孔10分钟,5%羊血清封闭1小时,分别滴加Caspase-3(1:200)、Bcl-2(1:200)、Bax(1:350)一抗稀释液,4℃过夜;次日复温30分钟,荧光二抗避光孵育1小时,鬼笔环肽染色30分钟,DAPI染色10分钟,封片,共聚焦激光显微镜采图。
对Caspase-3、Bcl-2、Bax的荧光强度分析结果显示(图9A-9C),nanosuit预处理显著降低了Caspase-3、Bax的荧光强度,而增强了Bcl-2的表达。说明nanosuit通过调节凋亡蛋白的表达水平,对辐射诱导的细胞凋亡发挥了抑制作用。
(5)检测nanosuit对细胞内DNA的辐射保护作用
辐射诱导的细胞损伤的标志是不受控制的DNA断裂。因此,本发明检测nanosuit对细胞内DNA的辐射保护作用。具体实验分组和方法如上述(3)所示,达到时间点后,收集细胞进行彗星实验检测:①制备胶板:第一层胶(将已预热45℃的180μL 1%正常熔点琼脂糖NMA滴到同样预热的载玻片的磨砂面,迅速盖上干净的盖玻片,4℃放置10min使其凝固);第二层胶(37℃下取100μL的细胞悬液和150μL 0.8%低熔点琼脂糖LMA混匀,然后轻轻揭去盖玻片,将含细胞的LMA滴到第一层胶板上,立即盖上干净的盖玻片,4℃下固化20min);第三层胶(移开玻片,37℃将100μL 0.5%的低熔点琼脂糖迅速滴加在第二层琼脂糖上,加盖玻片,4℃下固化20min)。②裂解:移开玻片,将载玻片浸入新配制的4℃裂解液中2h。③解旋:将新配制的电泳缓冲液倒入电泳槽中,覆盖载玻片0.25cm,盖上盖子,在电泳缓冲液中放置30min。④电泳:室温下,调节缓冲液液面高低,在300mA,置于25V电泳15min。⑤洗涤和染色:电泳后,将载玻片置于平皿中,用中和缓冲液漂洗30min。呈中性后,将载玻片用50-100μL吖啶橙溶液(EB)染色10min,并用蒸馏水漂洗2次以上。⑥结果观察:使用荧光显微镜镜检。在一定条件下,DNA迁移距离(彗星尾长)和DNA含量(荧光强度)分布与DNA损伤程度呈线性相关。
彗星实验检测图像如图10A所示,DNA尾矩的定量分析结果如图10B所示。DNA尾矩的定量分析结果表明:使用单纯AMF(33.27±7.14%)或tFNAs(50.16±4.73%)预处理,对DNA损伤的保护效果与模型组相比没有显著性差异,说明使用单纯AMF或tFNAs预处理几乎无法避免细胞的DNA损伤。而使用nanosuit预处理(9.35±3.23%)的细胞DNA恢复到与未辐射DNA相当程度。说明使用nanosuit预处理对辐射导致的细胞DNA损伤有明显的保护作用,且与单纯使用AMF或tFNAs相比,发挥了协同增效的作用。
γ-H2A.X作为DNA损伤形成的产物,是DNA损伤的另一个关键生物标志物。在本研究中,与其他组相比,nanosuit预处理组的γ-H2A.X水平低于其他组(图10C)。此外,对DNAPKcs进行了免疫荧光检测以观察DNA损伤的程度,nanosuit预处理组的DNA PKcs强度比其他组更弱(图10D),表明nanosuit对细胞内DNA具有强大的辐射保护作用。
本实验例结果表明,本发明的nanosuit可通过发挥抗氧化应激和抗细胞凋亡作用,保护DNA免受辐射损伤,从而促进细胞存活。与单纯使用AMF或tFNAs相比,nanosuit发挥了协同增效的辐射保护作用。
实验例2、体内实验
1、实验动物
BALB/C小鼠(18-22g,雄性75只,雌性75只)。
2、实验分组(5组*30只)
A:Control:不给药,不进行辐照;
B:IR组:不给药,只进行辐照
C:tFNAs+IR组:给药tFNAs后进行辐照;
D:AMF+IR组:给药AMF后进行辐照;
E:nanosuit+IR组:给药nanosuit后进行辐照;
给药组为10μM tFNAs、6.4mM AMF或10μM nanosuit分别静脉注射200μL/次/d,连续给药3d后,给予6.5Gy/5Gy全身辐照。小鼠辐射保护实验设计示意图如图11所示。tFNAs和nanosuit采用实施例1方法制备。
3、相关检测方法和结果
(1)检测nanosuit对小鼠在致命辐射剂量下存活的促进作用
BALB/C小鼠经相应分组处理后,给予6.5Gy全身辐照并观察30天存活情况。使用Kaplan-Meier生存分析检测小鼠30天存活情况,结果显示tFNAs预处理后的30天生存率是0%,平均生存时间9.8天;AMF预处理后的30天生存率是20%,平均生存时间15.4天,nanosuit预处理后的30天生存率是70%,平均生存时间25.1天(图12A)。因此nanosuit有效地提高了致命辐射剂量下小鼠的存活率,降低了辐射毒性。
(2)检测nanosuit对小鼠在亚致命辐射剂量下血液系统的作用
BALB/C小鼠经相应分组处理后,给予5Gy全身辐照,到达1、7、30天时间点后麻醉小鼠,采眼球静脉血进行血液学检测。1、7和30天白细胞、红细胞、血小板的血液学参数表明nanosuit预处理组小鼠的血液系统损伤得到有效缓解,且效果显著优于单独使用AMF或tFNAs,发挥了协同增效作用:辐照后第7天白细胞计数下降最明显,而nanosuit预处理组小鼠白细胞计数是IR组的2-3倍,辐照后第30天nanosuit预处理组几乎恢复至健康水平。同时,红细胞和血小板的变化趋势与白细胞一致(图12B-12D)。
(3)检测nanosuit对小鼠在亚致命辐射剂量下骨髓的作用
BALB/C小鼠经相应分组处理后,给予5Gy全身辐照,到达1、7、30天时间点后处死小鼠,取股骨脱钙后进行石蜡包埋和切片(4μm),对组织切片进行苏木精-伊红(H&E)染色。7和30天H&E染色的股骨切片的代表性图显示辐照第7天股骨髓腔内的骨髓细胞几乎消失,髓腔内被红细胞充满;而nanosuit预处理显著减轻了辐射引起的骨髓抑制与出血,到30天时,髓腔内细胞组成几乎恢复到接近对照组的健康水平(图13A)。
从股骨中冲洗骨髓细胞,使用BM-MNCs提取试剂盒分离,检测骨髓细胞DNA含量。第1、7和30天骨髓细胞DNA含量结果显示,与健康小鼠相比,辐射后第1天和7天,骨髓细胞总DNA相对含量显著下降至0.461±0.103和0.446±0.071。与之相反,nanosuit预处理组的总DNA相对含量在第1天有一个更高的水平0.742±0.066,在第7天上升到0.905±0.103,几乎恢复到健康值(图13B),效果同样显著优于单独使用AMF或tFNAs。
(4)检测nanosuit对小鼠在亚致命辐射剂量下重要脏器的作用
BALB/C小鼠经相应分组处理后,给予5Gy全身辐照,到达1、7、30天时间点后处死小鼠,取多器官(胸腺、脾脏、肝脏、睾丸、小肠、肺、肾和卵巢)进行石蜡包埋和切片(4μm),对组织切片进行苏木精-伊红(H&E)染色。7和30天H&E染色的多器官的代表性图显示(图14~图20):辐照下胸腺和脾脏都出现了明显萎缩和结构性损伤,而nanosuit预处理可显著减轻小鼠的脾脏萎缩和胸腺萎缩;辐照下肝脏出现肝细胞变性和坏死,核富集和碎裂,而nanosuit预处理改善了肝细胞形态,ALT和AST相关肝功能检测与病理结果一致;睾丸是对辐射最敏感的组织之一,辐照小鼠生精细胞出现明显的变性和数量减少,经nanosuit预处理后,睾丸的形态基本恢复正常;小肠也是对辐射最敏感的器官之一,辐照小鼠的小肠表现为明显的病理组织学损伤,包括广泛的绒毛萎缩、排列松散、肠黏膜受损,肠隐窝细胞数量减少,但在nanosuit预处理组中小肠绒毛长而完整,隐窝无异常。其他器官包括肺、肾和卵巢在辐射下也表现出中度病变,而在nanosuit预处理后的小鼠中可以发现相应改善。这些病理结果表明,nanosuit促进了辐照下重要器官的存活。且与单纯使用AMF或tFNAs相比,nanosuit发挥了协同增效的辐射保护作用。
(5)检测nanosuit对小鼠体内SOD活力和MDA水平的改变
为了了解nanosuit在体内的保护机制,本发明检测了5Gy辐照后第7天小鼠血清、骨髓细胞、脾脏、肝脏、睾丸和小肠中的SOD活力和MDA水平。结果表明(图12E、12F、13C、13D、14B、14C、15B、15C、16B、16C、18B、18C、19B和19C)单纯辐照组SOD活性降低,MDA水平升高;相反,nanosuit预处理显著减缓了这些趋势。因此nanosuit通过恢复SOD活性和抑制MDA水平来实现自由基清除,从而保护组织器官。
本实验例结果表明,本发明的nanosuit可通过恢复SOD活性和抑制MDA水平,实现亚致命辐射下多器官保护,并促进致命辐射下机体的存活。与细胞实验一致,与单纯使用AMF或tFNAs相比,nanosuit发挥了协同增效的辐射保护作用。
综上,本发明提供了一种DNA四面体-氨磷汀复合物,其以DNA四面体为载体进行氨磷汀的协同递送。本发明DNA四面体-氨磷汀复合物克服了氨磷汀代谢时间短的问题,并且增强了氨磷汀的辐射保护作用,取得了协同增效的辐射保护效果。本发明复合物可以在辐射环境中对DNA进行保护,同时对多种器官有保护作用,并且能提高辐射下的生存率,具有优异的辐射保护作用,在多种领域具有优异的应用前景。
Claims (10)
1.一种DNA四面体-氨磷汀复合物,其特征在于:它是由DNA四面体框架核酸和氨磷汀混合后形成的复合物。
2.根据权利要求1所述的DNA四面体-氨磷汀复合物,其特征在于:所述DNA四面体和氨磷汀混合时,DNA四面体和氨磷汀的摩尔比为1:160~1:1280。
3.根据权利要求2所述的DNA四面体-氨磷汀复合物,其特征在于:所述DNA四面体和氨磷汀的摩尔比为1:640。
4.根据权利要求1~3任一项所述的DNA四面体-氨磷汀复合物,其特征在于:所述DNA四面体由四条DNA单链自组装合成;所述四条DNA单链的序列分别如SEQ ID NO.1、SEQ IDNO.2、SEQ ID NO.3、和SEQ ID NO.4所示。
5.根据权利要求4所述的DNA四面体-氨磷汀复合物,其特征在于:所述DNA四面体的合成方法包括以下步骤:将四条DNA单链加入到TM缓冲液中,95℃维持10min,4℃维持20min以上,即得。
6.根据权利要求5所述的DNA四面体-氨磷汀复合物,其特征在于:所述四条DNA单链为等摩尔比的四条DNA单链。
7.一种制备权利要求1~6任一项所述的DNA四面体-氨磷汀复合物的方法,其特征在于:它包括如下步骤:
将四面体框架核酸和氨磷汀加入溶剂中混合反应,即得。
8.根据权利要求7所述的方法,其特征在于:
所述溶剂为水、PBS、DMSO中的一种或多种;
和/或,所述四面体框架核酸的浓度为200~250nM;
和/或,所述反应的温度为4~10℃;
和/或,所述反应的时间为4~6h。
9.权利要求1~6任一项所述的DNA四面体-氨磷汀复合物在制备预防辐射损伤的药物中的用途。
10.根据权利要求9所述的用途,其特征在于:所述药物为预防辐射中血液系统和/或器官损伤的药物;
和/或,所述药物为预防DNA损伤的药物。
