Abstract

氯离子通道是生物体内一种重要的阴离子通道,它们在细胞膜和胞内细胞器质膜的跨膜转运中起着特定的生物学功能。ClC型氯离子通道是一类分布广泛的阴离子通道,参与细胞内囊泡的酸化、肌肉细胞的电压稳定性等多种生理过程,其基因突变或功能异常都可导致多种疾病的发生。ClC-7是ClC型氯离子通道的重要成员,它主要位于后期内涵体、溶酶体和破骨细胞的竖起膜。CLCN7基因是ClC-7的编码基因,已有研究表明,在小鼠和人体内,CLCN7基因突变会导致骨质和溶酶体贮积病。目前,对于ClC-7的研究仅限于此,其在肿瘤上的研究还有待完善。
有鉴于此,特提出本发明。
发明内容
本发明的第一目的在于提供CLCN7在制备癌症诊断产品中的应用,以解决上述问题中的至少一种。
本发明的第二目的在于提供一种癌症诊断试剂盒。
本发明的第三目的在于提供一种CLCN7抑制剂在制备癌症治疗产品中的应用。
本发明的第四目的在于提供一种癌症治疗药物。
第一方面,本发明提供了一种CLCN7在制备癌症诊断产品中的应用;
所述CLCN7包括CLCN7基因转录的RNA或者CLCN7蛋白(即CLC-7)。
作为进一步技术方案,所述癌症包括肝癌、皮肤癌、脑胶质瘤或B淋巴细胞癌中的至少一种。
第二方面,本发明提供了一种癌症诊断试剂盒,所述试剂盒包括识别CLCN7基因转录RNA或者CLCN7蛋白的标记物。
作为进一步技术方案,所述标记物包括如下a)或b)中的至少一种:
a)结合所述RNA的引物;
b)结合所述CLCN7蛋白的抗体。
作为进一步技术方案,所述引物的核酸序列如SEQ ID NO.1和SEQ ID NO.2所示。
第三方面,本发明提供了一种CLCN7抑制剂在制备癌症治疗产品中的应用;
所述CLCN7抑制剂包括CLCN7的反义核酸或者小干扰核酸。
作为进一步技术方案,所述CLCN7抑制剂含有如SEQ ID NO.3、SEQ ID NO.4或SEQID NO.5所示的核酸序列。

与现有技术相比,本发明具有如下有益效果:
本发明提供了CLCN7在制备癌症诊断产品中的应用,其中CLCN7包括CLCN7基因转录的RNA或者CLCN7蛋白。经发明人研究发现,与健康组织相比,CLCN7基因在肝癌、皮肤癌、脑胶质瘤或B淋巴细胞癌组织中高表达,提示通过对CLCN7基因或者CLCN7蛋白表达量的检测能够实现对癌症患者的诊断,为癌症的诊断提供新的靶点和思路。
本发明提供了CLCN7抑制剂在制备癌症治疗产品中的应用,其中,CLCN7抑制剂含有如SEQ ID NO.3、SEQ ID NO.4或SEQ ID NO.5所示的核酸序列。经发明人研究发现,CLCN7抑制剂通过抑制CLCN7基因的表达,能够降低癌细胞的增殖迁移能力和成瘤能力,提示CLCN7抑制剂具有肿瘤抑制作用,能够用于癌症治疗药物的制备。

本发明中,CLCN7抑制剂是指对CLCN7的转录或者翻译存在抑制作用的抑制剂,例如可以为根据CLCN7的序列而设计得到的反义核酸或者小干扰核酸等。
经发明人研究发现,CLCN7抑制剂通过抑制CLCN7基因的表达,能够降低癌细胞的增殖迁移能力和成瘤能力,提示CLCN7抑制剂具有肿瘤抑制作用,能够用于癌症治疗药物的制备。
在一些优选的实施方式中,所述CLCN7抑制剂含有如SEQ ID NO.3、SEQ ID NO.4或SEQ ID NO.5所示的核酸序列:

需要说明的是,在本发明中,CLCN7抑制剂含有如SEQ ID NO.3所示的核酸序列,指的是CLCN7抑制剂除去SEQ ID NO.3、SEQ ID NO.4或SEQ ID NO.5所示的核苷酸序列外,还可以含有其他功能序列,如标签序列或连接序列等。
在一些优选的实施方式中,所述癌症包括肝癌、皮肤癌、脑胶质瘤或B淋巴细胞癌中的至少一种。
第四方面,本发明提供了一种癌症治疗药物,所述药物包括如下Ⅰ)-Ⅳ)中的一种或多种:
Ⅰ)CLCN7抑制剂;
Ⅱ)含有CLCN7抑制剂的编码序列的重组载体;
Ⅲ)含有CLCN7抑制剂的编码序列的重组病毒;
Ⅳ)含有CLCN7抑制剂的编码序列的重组病毒载体;
所述CLCN7抑制剂包括但不限于含有如SEQ ID NO.3、SEQ ID NO.4或SEQ ID NO.5所示的核酸序列,本领域技术人员可以根据CLCN7的基因序列采用常规手段设计得到,经发明人研究发现,当CLCN7抑制剂含有如SEQ ID NO.3、SEQ ID NO.4或SEQ ID NO.5所示的核酸序列时,效果更好。
需要说明的是,本发明提供的治疗癌症的药物,可以包括CLCN7抑制剂、含有CLCN7抑制剂的编码序列的重组病毒、含有CLCN7抑制剂的编码序列的重组病毒或含有CLCN7抑制剂的编码序列的重组病毒载体中的一种或两种或以上,例如仅包括CLCN7抑制剂,或者仅包括含有CLCN7抑制剂的编码序列的重组载体,或者包括CLCN7抑制剂和含有CLCN7抑制剂的编码序列的重组病毒载体,或者包括含有CLCN7抑制剂的编码序列的重组载体、含有CLCN7抑制剂的编码序列的重组病毒和含有CLCN7抑制剂的编码序列的重组病毒载体,或者同时包括CLCN7抑制剂、含有CLCN7抑制剂的编码序列的重组病毒、含有CLCN7抑制剂的编码序列的重组病毒和含有CLCN7抑制剂的编码序列的重组病毒载体。
在一些优选的实施方式中,所述药物还包括药学上可接受的载剂;
优选地,所述载剂包括壳聚糖、胆固醇、脂质体和纳米颗粒中的一种或几种。
上述载剂均能够有效包裹CLCN7抑制剂、含有CLCN7抑制剂的编码序列的重组病毒、含有CLCN7抑制剂的编码序列的重组病毒或含有CLCN7抑制剂的编码序列的重组病毒载体中的一种或多种,并携上述活性成分进入体内并释放,以起到给药治疗的目的。优选地,通过对上述载体进行进一步修饰,如连接结合位点等,还能够进一步起到靶向给药的作用。
在一些优选的实施方式中,所述药物的剂型包括口服制剂或注射制剂。
当口服用药时,上述药物可制成任意口服可接受的制剂形式,例如可以为,但不限于片剂、胶囊剂、颗粒剂、丸剂、糖浆剂、口服溶液剂、口服混悬剂或口服乳剂。
其中,片剂使用的载体一般包括乳糖和玉米淀粉,另外也可加入润滑剂如硬脂酸镁。胶囊剂使用的稀释剂一般包括乳糖和干燥玉米淀粉。口服混悬剂则通常是将活性成分与适宜的乳化剂和悬浮剂混合使用。
当以注射的形式给药时,上述药物可制成任意注射可接受的制剂形式,例如可以为,但不限于注射液或粉针剂。
其中,可使用的载体和溶剂包括水、林格氏溶液和等渗氯化钠溶液。另外,灭菌的非挥发油也可用作溶剂或悬浮介质,如单甘油酯或二甘油酯。
下面通过具体的实施例和对比例进一步说明本发明,但是,应当理解为,这些实施例仅仅是用于更详细地说明之用,而不应理解为用于以任何形式限制本发明。
实施例1离子通道肝癌标志物筛选与确认
(i)从GDC(https://portal.gdc.cancer.gov/)下载TCGA数据库中426例肝癌患者的RNA-seq测序数据及临床资料(其中50例患者包括癌组织及癌旁组织,其余的376例仅有癌组织);-
(ii)使用R软件的加权基因共表达WGCNA包对TCGA肝癌的mRNA数据进行分析,通过计算各基因间的Pearson相关系数选择合适的软阈值β。采用一步法构建的基因表达无尺度网络网络,将邻接矩阵转化为拓扑重叠矩阵TOM,利用动态树切割的层次聚类(Hierarchical Clustering)生成基因的层次聚类树。通过与临床特征信息进行关联,计算基因显著性(GS)和模块显著性(MS)去衡量基因与临床信息的显著性,得到与特定临床特征相关性高的基因模块。通过‘edgR’包计算TCGA中癌和癌旁组织的差异表达基因(DifferentExpression Genes,DEGs),使用R包’UpSetR’和‘venn’绘制三种不同来源的肝癌测序数据之间的共有基因。使用TCGA数据库癌症样本的临床随访信息绘制CLCN7基因在多种癌系的Kaplan-Meier生存分析曲线,结果如图1中的A和B所示。结果表明,CLCN7基因在肝癌组织中显著高表达。
(iii)分别提取Hepg2、HUH-7、SMCC-7721细胞系和L02、SQG7721细胞的RNA,采用荧光定量PCR实验检测HCC细胞系(Hepg2、HUH-7、SMCC-7721细胞系)中较正常人源肝细胞(L02、SQG7721细胞)中CLCN7基因的表达情况,结果如图2中的A所示。结果显示,CLCN7基因在肝癌细胞系中高表达。
CLCN7引物序列如下:
上游:CCACGTTCACCCTGAATTTTGT(SEQ ID NO.1);
下游:AAACCTTCCGAAGTTGATGAGG(SEQ ID NO.2)。
分别提取Hepg2、HUH-7、SMCC-7721细胞系和L02、SQG7721细胞的蛋白,以CLCN7蛋白抗体和actin抗体,采用蛋白质印迹法(western blot)实验检测HCC细胞系(Hepg2、HUH-7、SMCC-7721细胞系)中较正常人源肝细胞(L02、SQG7721细胞)中CLCN7蛋白的表达情况,结果如图2中的B所示。结果显示,CLCN7蛋白在肝癌细胞系中高表达。
结合上述研究结果可以得出,CLCN7高表达情况与肝癌不良预后密切相关。
实施例2功能性研究
构建瞬时敲低CLCN7基因HepG2细胞系(CLCN7-sh,即向HepG2细胞中转染含有如SEQ ID NO.3所示序列的质粒)及不作处理的对照组(Control),构建瞬时过表达CLCN7基因的LO2细胞系(CLCN7-oe)及不作处理对照组(Control),在37°、5%浓度CO2培养箱中培养48小室。对上面的细胞采用细胞克隆形成实验和划痕实验检测研究CLCN7通道对HCC细胞增殖、侵袭和迁移的影响。结果如图3中的A和C所示。结果表明,CLCN7的敲低显著抑制肝癌细胞的增殖能力和迁移能力。
构建肝癌细胞系(Hepg2)敲低CLCN7基因(CLCN7-sh)和阴性对照(Hepg2,即不含目的基因片段的载体)、空白对照组细胞系(CLCN7-nc,即只用转染试剂处理),在96孔板中加入的等量的细胞个数,培养48小时后,进行MTT实验测量活细胞个数,结果如图3中的B所示。证明了在Hepg2细胞系中敲低了CLCN7通道的表达,降低的了Hepg2细胞的增殖;构建正常肝细胞系(L02)过表达CLCN7基因(CLCN7-oe)和阴性对照(CLCN7-nc)、空白对照组细胞系(L02),在96孔板中加入的等量的细胞个数,培养48小时后,进行MTT实验测量活细胞个数,结果如图3中的B所示。证明了在L02细胞系中,过表达CLCN7基因可以增强细胞增殖。
体内成瘤实验:构建CLCN7基因敲除的稳转肝癌细胞系Hepg2(shCLCN7)和对照组(Scramble,引入空载体的细胞系),构建过表达CLCN7基因L02稳转细胞系(oeCLCN7)和对照组(Scramble,引入空载体的细胞系),四种细胞系分别以1*10^6/个细胞,注射到免疫缺陷小鼠腋下,进行异位稙瘤实验,每组设置6只重复(n=6)。记录23天内小鼠瘤子的体积(图4中的A)。第23天处死、刨瘤,并记录肿瘤重量(图4中的B)和肿瘤照片(图4中的C)。结果显示,抑制CLCN7可以降低肝癌细胞的成瘤能力。
实施例3检测CLCN7在其他癌种中的潜在作用
参照实施例1中的(i)和(ii),从TCGA数据库和GEO数据库(GSE99807)下载乳腺癌和癌旁组织、皮肤癌和癌旁组织、脑胶质瘤和癌旁组织以及B淋巴细胞癌和癌旁组织的RNA-seq测序数据。分别进行生物信息学分析,评估CLCN7作为乳腺癌、皮肤癌、脑胶质瘤和B淋巴细胞癌的预后诊断和标志物的潜在性。结果如图5中的A、B和C所示。
结果显示,CLCN7在皮肤癌中显著性高表达且与不良预后相关(图5中的A)、在脑胶质瘤中显著性高表达且与不良预后相关(图5中的B)、在B淋巴癌中显著性高表达且与不良预后相关(图5中的C),但是和乳腺癌的发生无明显关系。上述结果表明,CLCN7基因除了肝癌外,也具有作为潜在的皮肤癌、脑胶质瘤和B淋巴细胞癌的生物标志物的可能性。
验证CLCN7基因在其他癌种中标志物的作用:对皮肤癌细胞(SK-MEL-28、A375)和正常皮肤细胞(HACAT)进行验证CLCN7的表达情况,荧光定量PCR结果显示CLCN7的mRNA水平在SK-MEL-28中,显著性高表达(图6中的A)。敲低SK-MEL-28细胞的CLCN7基因(shCLCN-7),同时设置不做处理的对照组(Scramble),MTT结果表明,敲低CLCN7基因可以显著性抑制SK-MEL-28的增殖能力(图6中的B所示,其中,shCLCN-7分别指构建得到的两个CLCN7基因敲低的SK-MEL-28细胞,柱状图中,左边的shCLCN-7代表转染的含有如SEQ ID NO.3所示序列的质粒,右边的shCLCN-7代表转染的含有如SEQ ID NO.5所示序列的质粒)。
最后应说明的是:以上各实施例仅用以说明本发明的技术方案,而非对其限制;尽管参照前述各实施例对本发明进行了详细的说明,本领域的普通技术人员应当理解:其依然可以对前述各实施例所记载的技术方案进行修改,或者对其中部分或者全部技术特征进行等同替换;而这些修改或者替换,并不使相应技术方案的本质脱离本发明各实施例技术方案的范围。

2.根据权利要求1所述的应用,其特征在于,所述癌症包括肝癌、皮肤癌、脑胶质瘤或B淋巴细胞癌中的至少一种。
3.一种癌症诊断试剂盒,其特征在于,所述试剂盒包括识别CLCN7基因转录RNA或者CLCN7蛋白的标记物。
4.根据权利要求3所述的癌症诊断试剂盒,其特征在于,所述标记物包括如下a)或b)中的至少一种:
a)结合所述RNA的引物;
b)结合所述CLCN7蛋白的抗体。
5.根据权利要求4所述的癌症诊断试剂盒,其特征在于,所述引物的核酸序列如SEQ IDNO.1和SEQ ID NO.2所示。
6.CLCN7抑制剂在制备癌症治疗产品中的应用;
所述CLCN7抑制剂包括CLCN7的反义核酸或者小干扰核酸。
7.根据权利要求6所述的应用,其特征在于,所述CLCN7抑制剂含有如SEQ ID NO.3、SEQID NO.4或SEQ ID NO.5所示的核酸序列。
8.根据权利要求6所述的应用,其特征在于,所述癌症包括肝癌、皮肤癌、脑胶质瘤或B淋巴细胞癌中的至少一种。
9.一种癌症治疗药物,其特征在于,所述药物包括如下Ⅰ)-Ⅳ)中的一种或多种:
Ⅰ)CLCN7抑制剂;
Ⅱ)含有CLCN7抑制剂的编码序列的重组载体;
Ⅲ)含有CLCN7抑制剂的编码序列的重组病毒;
Ⅳ)含有CLCN7抑制剂的编码序列的重组病毒载体;
所述CLCN7抑制剂含有如SEQ ID NO.3、SEQ ID NO.4或SEQ ID NO.5所示的核酸序列。
