Abstract
The invention relates to an application of paenibacillus polymyxa in preventing and treating ginger stem base rot, wherein the strain is paenibacillus polymyxa (Paenibacillus polymyx) which is preserved in China center for type culture collection (CCTCC for short), and the paenibacillus polymyxa has good inhibition effect on pathogenic bacteria of ginger stem base rot, has an anti-efficiency of more than 90 percent on ginger stem base rot diseases, can effectively reduce the incidence of ginger stem base rot, and has an effect superior to that of the existing common chemical pesticide. The paenibacillus polymyxa screened by the invention can not generate common problems of chemical pesticides such as drug resistance, pesticide residue, environmental pollution and the like after being used, is healthy to people and livestock, is environment-friendly, and meets the requirements of people on ecological agriculture.
Description
Technical Field
The invention belongs to the technical field of microorganisms, and particularly relates to application of paenibacillus polymyxa in preventing and treating ginger stem basal rot.
Background
Ginger is an important economic crop with high efficiency and high yield, has medicinal and edible value, and has the functions of relieving exterior syndrome, dispelling cold, warming middle-jiao, relieving vomiting, resolving phlegm and relieving cough. The ginger stem rot is also called ginger soft rot and ginger saprolegniasis, is a very serious fungus disease in ginger production, and severely restricts the development of ginger industry. At present, main control agents are triazolone, metalaxyl, mancozeb and the like, but a large amount of agents are used for a long time, so that certain drug resistance is generated. The quality of ginger is reduced to a certain extent, even chemical pesticide residues exceed standard, the ecological environment of a planting area is polluted, and the life and health of human beings are threatened. In order to achieve the aim of efficient pest control and sustainable development of the environment, a path for pursuing a new plant pest control strategy is developed. Biological control has been paid attention to in the aspect of disease control of ginger stem rot, biocontrol bacterial screening of Guan Shengjiang stem rot pathogenic bacteria and bacterial agents and applications thereof have been widely studied, and biocontrol strains mainly comprise bacillus subtilis, burkholderia cepacia, streptomyces, trichoderma and the like, but the application of paenibacillus polymyxa in the control of ginger stem rot is not reported.
Disclosure of Invention
In order to solve the technical problems, the invention aims to provide the application of the paenibacillus polymyxa in preventing and treating ginger stem basal rot, and the problems of pesticide residue, increased pathogenic bacteria resistance and the like caused by excessive use of chemical medicaments can be avoided.
The scheme provided by the invention is as follows:
an application of Paenibacillus polymyxa in preventing and treating ginger stem basal rot is disclosed, wherein the Paenibacillus polymyxa is preserved in China center for type culture collection (CCTCC for short), and the preservation number is CCTCC M2020361;
the method is characterized by comprising the following steps of: the diluted solution is made to live bacteria, then mixed with clay or plant ash in a mass ratio of 1:1 to prepare a medicinal preparation, and ginger seeds are soaked in the medicinal preparation for 1 minute by adopting a seed soaking method, and are sown after being dried in the air.
Further, the live bacteria concentration of the Paenibacillus polymyxa fermentation broth is 10 6 ~10 7 CFU/ml, and then 1g/ml clay or plant ash is added into the bacterial liquid to be mixed into a pharmaceutical preparation.
The invention has the beneficial effects that: the invention aims to prevent and treat ginger stem basal rot by utilizing microorganisms, so that paenibacillus polymyxa (CCTCC M2020361) capable of preventing and treating ginger stem basal rot is separated from ginger rhizosphere soil through separation screening, flat plate antagonism and seed soaking cultivation experiments. Experiments show that the paenibacillus polymyxa has good inhibition effect on pathogenic bacteria of ginger stem base rot, has the prevention effect on the ginger stem base rot disease of more than 90 percent, can effectively reduce the incidence of the ginger stem base rot, and has better effect than the existing common chemical pesticide. The paenibacillus polymyxa screened by the invention can not generate common problems of chemical pesticides such as drug resistance, pesticide residue, environmental pollution and the like after being used, is healthy to people and livestock, is environment-friendly, and meets the requirements of people on ecological agriculture.
Detailed description of the preferred embodiments
The invention will be further illustrated with reference to specific examples.
1. And (3) preparation of a microbial inoculum:
inoculating Paenibacillus polymyxa CCTCC M2020361 (Paenibacillus polymyxaL 201901) into LB culture medium, shake culturing at 28-33deg.C and 180-220rpm for 12-16 hr to obtain seed bacteria, inoculating 2% seed bacteria into modified LB culture medium, fermenting at 30deg.C and 200rpm for 48 hr to obtain fermentation broth, which is the microbial inoculum.
The Paenibacillus polymyxa is preserved in China center for type culture collection (CCTCC for short), and the preservation number is CCTCC M2020361; the culture was deposited by the collection on day 28, 7 in 2020. Address, university of chinese, armed chinese, post code 430072 telephone (027) 68754052 fax (027) 68754833E-mail cctco@whu.edu.Cn.
2. Disease prevention experiment of biocontrol bacteria
The concentration of the bacillus polymyxa CCTCC M2020361 viable bacteria in the microbial inoculum is 10 7 ~10 8 CFU/m L. Diluting the microbial inoculum with sterile water for 10 times to make the concentration of viable bacteria in the diluted solution be 10 6 ~10 7 CFU/ml, then mixing with clay or plant ash in a mass ratio of 1:1 to prepare a medicinal preparation, soaking ginger seeds in the medicinal preparation for 1 min by adopting a seed soaking method, airing and sowing. Purified water is used as a blank control group, and 1000-fold diluent of 50% carbendazim wettable powder is used as a blank control groupPositive control group, investigation after 45d sowing, calculation of emergence rate, disease index and prevention and cure effect. The results are shown in Table 1. The result shows that the control effect of the Paenibacillus polymyxa CCTCC M2020361 on the ginger stem basal rot is 90.6 percent.
TABLE 1 effects of treatments on prevention and treatment of ginger stem rot
The ginger emergence rate of different treatment modes and the control effect result (see table 1) are shown as follows: the Paenibacillus polymyxa (CCTCC M2020361) has better control effect on ginger stem rot, the control effect is 90.6%, the control effect of the 1000-time diluent of the carbendazim wettable powder is 66.1%, and the control effect of the CCTCC M2020361 medicament on ginger stem rot is obviously higher than that of the 1000-time diluent of the carbendazim wettable powder. And has obvious promoting effect on the emergence of ginger seeds.
The present invention is not limited to the above-mentioned embodiments, but any modifications, equivalents, improvements and modifications within the scope of the invention will be apparent to those skilled in the art.
