After grouping, mice other than the placebo group were modeled, and each group received 21 days of various stimuli, including: inverting 24 hours, fasted 24 hours, water-forbidden 24 hours, inclined 24 hours, wet 24 hours, constraint 2 hours, ice water swimming 3 minutes, hot water swimming 3 minutes, tail clamping 1 minute, electric shock, noise 2 hours, shaking 5 minutes, peculiar smell 24 hours (different peculiar smell every week), foreign matter 24 hours (different foreign matters every week); 2 kinds of stimulation are randomly selected every day, 1-2 times of stimulation are used every week, the same stimulation cannot continuously appear, the stimulation has unpredictability, the gastric lavage administration is started after the stimulation is ended at the 2 nd week, after the administration is ended for 28 days, the emotional behavior detection is carried out after the gastric lavage treatment is ended, and the emotional behavior detection comprises a sugar water preference experiment, a forced swimming experiment and an overhead cross maze experiment. The methods and amounts of administration for each group of mice are shown in Table 1.

3. Evaluation index
(1) General state observation: the mice in the model group gradually appear in a slack and lazy state, loose fur, roughness and no luster, yellow color, arch backs, loose stool, gradually emaciated bodies, positive medicine groups, the mice in the examples 1-3 and blank groups react agilely, white fur and luster, and act sensitively, wherein the mice in the example 2 group die due to stomach filling errors. The specific status of each group of mice is shown in Table 2.

(2) Sugar water preference experiment: the adaptive training is started on the 25 th day of gastric lavage, the syrup preference test is carried out on the 28 th day, all animals are fed and fasted in a single cage, the time is 1 hour, and the syrup preference ratio of each group of mice is calculated by comparing the consumption of pure water and sucrose water before and after the test:
sugar preference = sugar water consumption/total liquid consumption x 100%
(3) Forced swimming: swimming pre-experiment was performed 24h before the formal experiment, and the mice were put into 4000mL containers with water depth of 25cm and water temperature of 25 ℃ and were swimed for 10min. Carrying out formal experiments on the following day 9:00, putting the mice into the same environment as the pre-swimming, and swimming for 6min; the mice showed passive floating, only the small movements of the limbs remained floating, and struggling behavior disappeared for 3s to be regarded as a stationary state, and the accumulated stationary time of the forced swimming of the mice within 5min after calculation.
(4) Novel inhibition of ingestion: before the start of the experiment, each group of mice was fasted for 24 hours, and the open field experiment box was prepared: a wood box without top is 60cm multiplied by 80cm, the inner four walls of the wood box are coated with black, the box bottom is divided into 8 multiplied by 8 equal parts of small squares by using paint, food is placed at the most center of the box bottom, and the time when the mice are placed in the wood box until the food is found, namely the feeding latency period is measured; note that at the beginning of the experiment, the mouse head was placed towards the wall of the box and the time for the mouse to hold up the food with both forelimbs was observed and timed with a stopwatch.

From the above data, compared with the blank group, the sugar water preference value of the model group before modeling has no significant difference, but the sugar water preference value after treatment is significantly reduced, and the total immobility time and ingestion latency time of forced swimming are significantly increased, thus indicating that modeling is successful; the ratio of sugar water preference values to model groups of the positive drug group and the examples 1-3 is larger, wherein the treatment effect of the example 2 is equivalent to that of the positive drug; the immobility time and the ingestion latency time of forced swimming are obviously shortened compared with those of a model group, wherein the shortening effect of the ingestion latency time is equivalent to that of a positive medicine, the shortening effect of the immobility time of the forced swimming is slightly lower than that of the positive medicine, so that each group of the examples has the treatment effect on depression, the group of the examples 2 has the best effect, and the improvement effect on partial indexes is equivalent to that of the positive medicine.
The traditional Chinese medicine composition disclosed by the invention is applied to treatment of depression, has positive effects on improvement of clinical symptoms and rehabilitation process of diseases, has higher safety and has clinical popularization value.
The foregoing is only a preferred embodiment of the present invention, but the scope of the present invention is not limited thereto, and any person skilled in the art, who is within the scope of the present invention, should make equivalent substitutions or modifications according to the technical scheme of the present invention and the inventive concept thereof, and should be covered by the scope of the present invention.

Claims (10)

1. The traditional Chinese medicine composition for treating liver and kidney deficiency type depression is characterized by comprising the following components in parts by weight:
2-5 parts of uncaria;
3-6 parts of clerodendranthus spicatus.
2. The traditional Chinese medicine composition for treating liver and kidney deficiency type depression according to claim 1 is characterized by being prepared from the following components in parts by weight: 3 parts of uncaria and 4 parts of clerodendranthus spicatus.
3. The traditional Chinese medicine composition for treating liver and kidney deficiency type depression according to claim 1, wherein the extracts of the raw materials and a pharmaceutically acceptable carrier are prepared into granules, powder, tablets, capsules, pills, oral liquid or mixture.
4. A method for preparing a traditional Chinese medicine composition for treating liver and kidney deficiency type depression according to any one of claims 1 to 3, which is characterized by comprising the following steps:
(1) Crushing: weighing the uncaria alba and the clerodendranthus spicatus according to the weight parts, respectively crushing, and sieving with a 30-60-mesh sieve to obtain uncaria alba powder and clerodendranthus spicatus powder;
(2) Alcohol extraction: extracting the white uncaria powder obtained in the step (1) with ethanol for 2 times, combining the 2 times of extracting solutions, concentrating until no alcohol smell exists, and obtaining white uncaria concentrated solution and dregs for later use;
(3) Acid extraction: mixing the white uncaria dreg obtained in the step (2) with the clerodendranthus spicatus powder obtained in the step (1) to obtain mixed powder, extracting for 2 times by using acid water, mixing the 2 times of extracting solutions, adding alkali to adjust the pH value, standing, filtering, and collecting precipitate;
(4) Combining: and (3) respectively drying the white hooked uncaria concentrated solution obtained in the step (2) and the precipitate obtained in the step (3), grinding into powder, and uniformly mixing to obtain the traditional Chinese medicine composition for treating liver and kidney deficiency type depression.
5. The method according to claim 4, wherein in the step (2), the concentration of ethanol is 55% to 75%.
6. The preparation method according to claim 4, wherein in the step (2), the weight ratio of the white uncaria powder to the ethanol is 1: 10-12, and the single extraction time is 2-4 h.
7. The process according to claim 4, wherein in the step (3), the acid water is a hydrochloric acid solution having a pH of 5 to 6.
8. The process according to claim 4, wherein in the step (3), the alkali is an aqueous ammonia solution, and the pH of the solution is adjusted to 7.8 to 8.5 by adding alkali.
9. The method according to claim 4, wherein in the step (3), the weight ratio of the mixed powder to the acid water is 1: 12-16, and the single extraction time is 1.5-3.5 h.
10. Use of a Chinese medicinal composition according to any one of claims 1 to 3 for the preparation of a medicament for the treatment of depression due to deficiency of the liver and kidney.
