Preferably, the antibacterial composition comprises bamboo vinegar and aqueous or alcoholic extract of herba Bidentis Bipinnatae with a volume ratio of 1:1; or water extract or alcohol extract of bamboo vinegar and herba Houttuyniae at a volume ratio of 1:1; or bamboo vinegar and water or alcohol extract of herba Polygoni Hydropiperis at a volume ratio of 1:1.
Preferably, the antibacterial composition comprises 0.781% of bamboo vinegar by volume and 1.95mg/mL of sticktight alcohol extract or 1.95mg/mL of cordate houttuynia alcohol extract when the antibacterial composition is used for inhibiting escherichia coli.
Preferably, the antibacterial composition comprises 0.781% of bamboo vinegar by volume and 1.95mg/mL of herba Bidentis Bipinnatae water extract or alcohol extract when the antibacterial composition is used for inhibiting salmonella enteritidis; bamboo vinegar volume fraction 0.781% and herba Houttuyniae water extract or ethanol extract 1.95mg/mL; bamboo vinegar volume fraction 0.781% and herba Polygoni Hydropiperis water extract or ethanol extract 1.95mg/mL.
A second object of the present invention is to provide the use of the above bacteriostatic composition in the preparation of bacteriostatic drugs or feed additives.
Preferably, the antibacterial drug is a drug for inhibiting escherichia coli, salmonella enteritidis and staphylococcus aureus.
The antibacterial composition can be applied to preparation of antibacterial medicines or pig industry such as pig feed additives.
Experiments show that the treatment of the bamboo vinegar and the alcohol extract of the spanishneedles herb has a synergistic effect on the inhibition of escherichia coli, and the compounding of the bamboo vinegar and the six traditional Chinese medicine extracts has a synergistic effect on the inhibition of salmonella enteritidis. After the bamboo vinegar is compounded with six traditional Chinese medicine extracts, the effect of inhibiting bacteria is stronger than that of a single medicine, and the antibacterial capacity is greatly enhanced. Can be used for preparing antibacterial drugs or pig feed additives in pig industry.

The present invention is illustrated below with reference to examples and comparative examples, but the scope of the present invention is not limited to the following examples only. The objects of the present invention can be achieved by one of ordinary skill in the art in light of the present disclosure.
Example 1
1 preparation of aqueous extract of Bidens pilosa
1.1 taking sticktight picked in Dawanggao new region of four-party city in Zhaoqing, guangdong as raw material, and drying at 65 ℃ until the weight is constant;
1.2 pulverizing the herba Bidentis Bipinnatae hay obtained in 1.1 and sieving with 60 mesh sieve to obtain herba Bidentis Bipinnatae dry powder;
1.3 placing 60g of the sticktight dry powder obtained in 1.2 into a beaker, adding 600mL of pure water, decocting for 0.5h, pouring out water decoction, adding 600mL of pure water again, re-decocting for 0.5h, pouring out water decoction, mixing and filtering the liquid medicine obtained by the two water decoction methods to obtain filtrate;
1.4 evaporating and concentrating the filtrate obtained in the step 1.3 to obtain a sticktight water extract;
1.5 drying the aqueous extract of herba Bidentis Bipinnatae obtained in 1.4 at 65deg.C to constant weight to obtain aqueous extract of herba Bidentis Bipinnatae.
Preparation of alcohol extract of Bidens pilosa
2.1, placing 60g of the sticktight dry powder obtained in 1.2 into a Soxhlet extractor, adding 600mL of ethanol, and carrying out reflux extraction for 2 hours to obtain a liquid medicine;
2.2 filtering the liquid medicine obtained in the step 2.1, and evaporating and concentrating the filtrate to obtain the sticktight alcohol extract;
2.3 drying the sticktight alcohol extract obtained in 2.2 to constant weight at 65 ℃ to obtain the sticktight alcohol extract.
3 preparation of herba Houttuyniae aqueous extract
3.1 taking houttuynia cordata which is purchased in the market of traditional Chinese medicines as a raw material, and drying the houttuynia cordata at 65 ℃ to constant weight;
3.2 crushing the houttuynia cordata hay obtained in 3.1 and sieving the crushed houttuynia cordata hay with a 60-mesh sieve to obtain houttuynia cordata dry powder;
3.3 placing 60g of the houttuynia cordata dry powder obtained in 3.2 into a beaker, adding 600mL of pure water, decocting for 0.5h, pouring out water decoction, adding 600mL of pure water again, re-decocting for 0.5h, pouring out water decoction, mixing and filtering the liquid medicine obtained by the two water decoction methods to obtain filtrate;
3.4 evaporating and concentrating the filtrate obtained in the step 3.3 to obtain houttuynia cordata water extract;
3.5 drying the houttuynia cordata water extract obtained in 3.4 to constant weight at 65 ℃ to obtain the houttuynia cordata water extract.
4 preparation of houttuynia cordata ethanol extract
4.1, placing 60g of the houttuynia cordata dry powder obtained in 3.2 into a Soxhlet extractor, adding 600mL of ethanol, and carrying out reflux extraction for 2 hours to obtain a liquid medicine;
4.2, filtering the liquid medicine obtained in the step 4.1, and evaporating and concentrating the filtrate to obtain cordate houttuynia alcohol extract;
4.3, drying the cordate houttuynia alcohol extract obtained in 4.2 to constant weight at 65 ℃ to obtain the cordate houttuynia alcohol extract.
Preparation of 5 Polygonum hydropiper aqueous extract
5.1, taking the polygonum hydropiper obtained in the market of traditional Chinese medicines as a raw material, and drying the polygonum hydropiper at 65 ℃ to constant weight;
5.2 crushing the dried polygonum hydropiper herb obtained in 5.1 and sieving the crushed dried polygonum hydropiper herb by a 60-mesh sieve to obtain dried polygonum hydropiper powder;
5.3, placing 60g of the dry powder of the polygonum hydropiper obtained in 5.2 into a beaker, adding 600mL of pure water, decocting for 0.5h, pouring out the water decoction, adding 600mL of pure water again, re-decocting for 0.5h, pouring out the water decoction, mixing and filtering the liquid medicine obtained by the two water decoction methods to obtain filtrate;
5.4, evaporating and concentrating the filtrate obtained in the step 5.3 to obtain a water extract of the polygonum hydropiper;
5.5 drying the water extract of the polygonum hydropiper obtained in 5.4 to constant weight at 65 ℃ to obtain the water extract of the polygonum hydropiper.
Preparation of 6-polygonum hydropiper ethanol extract
6.1, placing 60g of the polygonum hydropiper dry powder obtained in the step 6.2 into a Soxhlet extractor, adding 600mL of ethanol, and carrying out reflux extraction for 2 hours to obtain a liquid medicine;
6.2, filtering the liquid medicine obtained in the step 6.1, and evaporating and concentrating the filtrate to obtain a polygonum hydropiper alcohol extract;
6.3, drying the polygonum hydropiper ethanol extract obtained in 6.2 to constant weight at 65 ℃ to obtain the polygonum hydropiper ethanol extract.
Example 2
1 preparation of unilateral liquid medicine
1.1 accurately weighing 1g of herba Bidentis Bipinnatae water extract, and dissolving in 4mL of pure water to obtain herba Bidentis Bipinnatae water extract with concentration of 250 mg/mL;
1.2 accurately weighing 1g of houttuynia cordata aqueous extract, and dissolving the extract in 4mL of pure water to obtain houttuynia cordata aqueous extract with the concentration of 250 mg/mL;
1.3 accurately weighing 1g of the water extract of the polygonum hydropiper, and dissolving the water extract of the polygonum hydropiper into 4mL of pure water to obtain a water extract of the polygonum hydropiper with the concentration of 250 mg/mL;
1.4 accurately weighing 1g of herba Bidentis Bipinnatae ethanol extract, and dissolving in 4mL of 10% dimethyl sulfoxide (DMSO) solution to obtain herba Bidentis Bipinnatae ethanol extract with concentration of 250 mg/mL;
1.5 accurately weighing 1g of houttuynia cordata ethanol extract, and dissolving the extract into 4mL of 10% DMSO solution with volume fraction to obtain houttuynia cordata ethanol extract with concentration of 250 mg/mL;
1.6 accurately weighing 1g of the polygonum hydropiper ethanol extract, and dissolving the extract into 4mL of 10% DMSO solution with volume fraction to obtain a polygonum hydropiper ethanol extract with the concentration of 250mg/mL.
2 preparation of compound liquid medicine
2.1 fully and uniformly mixing 1mL of the spanishneedles herb water extract obtained in the step 1 with 1mL of bamboo vinegar (purchased from Seiscpart Wu Tailang bamboo charcoal limited company, bamboo vinegar stock solution, the same applies below) to obtain compound liquid medicine 1;
2.2, fully and uniformly mixing 1mL of the cordate houttuynia water extract obtained in the step 1 with 1mL of bamboo vinegar to obtain compound liquid medicine 2;
2.3, fully and uniformly mixing 1mL of the polygonum hydropiper water extract obtained in the step 1 with 1mL of bamboo vinegar to obtain compound liquid medicine 3;
2.4 fully and uniformly mixing 1mL of the spanishneedles herb ethanol extract obtained in the step 1 with 1mL of bamboo vinegar to obtain compound liquid medicine 4;
2.5 fully and uniformly mixing 1mL of cordate houttuynia ethanol extract obtained in the step 1 with 1mL of bamboo vinegar to obtain compound liquid medicine 5;
2.6 fully and uniformly mixing 1mL of the polygonum hydropiper ethanol extract obtained in the step 1 with 1mL of bamboo vinegar to obtain compound liquid medicine 6.
And (3) strain:

Culture medium:

Liquid medicine: bamboo vinegar, herba Bidentis Bipinnatae water extract, herba Houttuyniae water extract, herba Polygoni Hydropiperis water extract, herba Bidentis Bipinnatae ethanol extract, herba Houttuyniae ethanol extract, herba Polygoni Hydropiperis ethanol extract, compound liquid medicine 1, compound liquid medicine 2, compound liquid medicine 3, compound liquid medicine 4, compound liquid medicine 5, and compound liquid medicine 6
The experimental steps are as follows:
activation of strain and preparation of strain liquid
(1) The three strains are respectively inoculated in nutrient broth culture medium, cultured for 16h under the condition of 80r/min in a constant temperature shaking table at 37 ℃ and are recovered.
(2) Selecting resuscitated bacterial liquid, dividing and streaking on a nutrient agar plate, culturing at 37 ℃ for 16h, selecting single bacterial colony, culturing in nutrient broth for 16h, preparing bacterial suspensions with different dilution concentrations by 10 times of sterile physiological saline, and preparing 1×10 bacterial colony by colony counting method 6 The CFU/mL bacterial suspension is ready for use.
Minimum Inhibitory Concentration (MIC) -broth culture method
The nutrient broth culture medium is divided into test tubes with 1mL of the final volume of each tube, sterilized for 20min at 120 ℃ and 0.1MPa, the sterilized culture medium is cooled, 1mL of the liquid medicine is added for full mixing in a sterile operation, the liquid medicine is continuously diluted into a No. 1-8 tube by a double continuous dilution method (volume ratio), the No. 9 tube is a positive control, the No. 10 tube is a negative control (no bacteria adding), and sterile water and 10% DMSO are used as blank controls. 100. Mu.L of bacterial liquid was added to the blank control and 1-9 tubes. After incubation at 37℃for 24h at 150rpm, the liquid in the tube was observed for clarity and recorded. Minimum bactericidal concentration determination (MBC)
On the basis of MIC, all clear sterile growth test tubes were selected, 100. Mu.L of the culture solution was spread on nutrient agar medium, and incubated overnight at 37℃for 24h, and repeated 3 times. The minimum drug concentration corresponding to the aseptic growth treatment is the minimum bactericidal concentration (Minimum Bactericidal Concentration, MBC).
Combined bacteriostasis index (FICI)
The combined bacteriostasis index (Faction Inhibitory Concentration Index, FICI) =mic/MIC when a drug is combined alone and mic+mic/MIC when b drug is combined alone, when FICI is less than or equal to 0.5, the drugs are synergistic; FICI is more than 0.5 and less than or equal to 1, and the medicines are added; FICI is more than 1 and less than or equal to 2, and the medicines have irrelevant effects; FICI > 2, antagonism between drugs.

As a result, tetracycline was highly sensitive to Salmonella enteritidis and Staphylococcus aureus, and had a MIC of 7.81. Mu.g/mL, but not to E.coli, and had a MIC of 250mg/mL. The possible reason is that the Escherichia coli used in the test has developed a certain drug resistance to tetracycline, but the plant-derived drug bamboo vinegar and the traditional Chinese medicine extract are not easy to develop the drug resistance. The MIC of the bamboo vinegar to the escherichia coli K88 and salmonella enteritidis treatment group is 3.13%, the MIC of the staphylococcus aureus treatment group is 1.56%, but in the compound liquid medicine, the MIC concentration of the escherichia coli K88 and salmonella enteritidis treatment group is only 0.78-1.56%, and the MIC of the staphylococcus aureus treatment group is only 0.781%. The MIC of the six traditional Chinese medicine extracts to the escherichia coli K88 is 15.6mg/mL, the salmonella enteritidis and staphylococcus aureus treatment group is 7.81-15.6 mg/mL, but in the compound liquid medicine, the MIC of the traditional Chinese medicine extracts to the escherichia coli K88 treatment group is only 1.95-3.91 mg/mL, and the MIC of the salmonella enteritidis and staphylococcus aureus treatment group is only 1.95mg/mL. This means that the concentration of the bamboo vinegar can be reduced to 1/2-1/4 of the original concentration after the compounding, and the concentration of the traditional Chinese medicine extract can be reduced to 1/4-1/8 of the original concentration after the compounding (with synergistic effect). The FICI index shows that the treatment of the bamboo vinegar and the alcohol extract of the spanishneedles herb has a synergistic effect on the inhibition of escherichia coli, and the compounding of the bamboo vinegar and the six traditional Chinese medicine extracts has a synergistic effect on the inhibition of salmonella enteritidis. After the bamboo vinegar is compounded with six traditional Chinese medicine extracts, the effect of inhibiting bacteria is stronger than that of a single medicine, and the antibacterial capacity is greatly enhanced.
Claims (10)
1. An antibacterial composition is characterized by comprising bamboo vinegar and herba Bidentis Bipinnatae extract, or bamboo vinegar and herba Houttuyniae extract, or bamboo vinegar and herba Polygoni Hydropiperis extract.
2. The bacteriostatic composition according to claim 1, wherein the extract of sticktight is an aqueous extract or an alcoholic extract of sticktight; the herba Houttuyniae extract is herba Houttuyniae water extract or herba Houttuyniae alcohol extract; the herba Polygoni Hydropiperis extract is water extract or ethanol extract of herba Polygoni Hydropiperis.
3. The bacteriostatic composition according to claim 1, wherein the bacteriostatic composition comprises an aqueous or alcoholic extract of bamboo vinegar and sticktight, an aqueous or alcoholic extract of bamboo vinegar and houttuynia cordata, or an aqueous or alcoholic extract of bamboo vinegar and polygonum hydropiper.
4. The bacteriostatic composition according to claim 3, wherein the aqueous extract is obtained by drying and pulverizing herba Bidentis Bipinnatae, herba Houttuyniae or herba Polygoni Hydropiperis, adding into water according to a ratio of 1:10g/ml, decocting to obtain a liquid medicine, mixing and filtering the liquid medicine obtained by the water decoction method to obtain a filtrate, evaporating and concentrating the filtrate to obtain an aqueous extract, drying at 65 ℃ to constant weight to obtain an aqueous extract, and dissolving the aqueous extract in water according to a ratio of 1g/4ml to obtain the aqueous extract of herba Bidentis Bipinnatae, herba Houttuyniae or herba Polygoni Hydropiperis respectively.
5. The bacteriostatic composition according to claim 3, wherein the alcoholic extract is obtained by drying and pulverizing herba Bidentis Bipinnatae, herba Houttuyniae or herba Polygoni Hydropiperis, adding into ethanol according to a ratio of 1:10g/ml, reflux-extracting for 2h to obtain a liquid medicine, evaporating and concentrating filtrate after filtering the liquid medicine to obtain an alcoholic extract, drying the alcoholic extract at 65 ℃ until constant weight to obtain an alcoholic extract, and dissolving the alcoholic extract into a dimethyl sulfoxide solution with a volume fraction of 10% according to a ratio of 1g/4ml to obtain the alcoholic extract of herba Bidentis Bipinnatae, herba Houttuyniae or herba Polygoni Caespitosi respectively.
6. The bacteriostatic composition according to claim 4 or 5, wherein said bacteriostatic composition comprises bamboo vinegar and aqueous or alcoholic extract of sticktight in a volume ratio of 1:1; or water extract or alcohol extract of bamboo vinegar and herba Houttuyniae at a volume ratio of 1:1;
or bamboo vinegar and water or alcohol extract of herba Polygoni Hydropiperis at a volume ratio of 1:1.
7. The bacteriostatic composition according to claim 5, wherein the bacteriostatic composition comprises 0.781% by volume of bamboo vinegar and 1.95mg/mL of alcohol extract of sticktight or 1.95mg/mL of alcohol extract of houttuynia cordata when the bacteriostatic composition is a bacteriostatic composition for inhibiting escherichia coli.
8. The bacteriostatic composition according to claim 4 or 5, wherein when it is a bacteriostatic composition for inhibiting salmonella enteritidis, said bacteriostatic composition comprises 0.781% by volume of bamboo vinegar and 1.95mg/mL of aqueous or alcoholic extract of sticktight; bamboo vinegar volume fraction 0.781% and herba Houttuyniae water extract or ethanol extract 1.95mg/mL;
bamboo vinegar volume fraction 0.781% and herba Polygoni Hydropiperis water extract or ethanol extract 1.95mg/mL.
9. Use of a bacteriostatic composition according to any one of claims 1-8 for the preparation of a bacteriostatic drug or feed additive.
