Description

Antibacterial composition and application thereof
Technical Field
The invention belongs to the field of antibiosis, and particularly relates to a bacteriostatic composition and application thereof.
Background
Antibiotics are widely used for animal health and growth of infectious diseases and feed additives. Antibiotic resistance caused by overuse of antibiotics poses a threat to global health. The safe and environment-friendly antibiotic substitute is of great importance for sustainable development of pig industry. Recently, researchers have turned their eyes toward plant-derived drugs which have little side effects and are difficult to develop drug resistance.
The bamboo vinegar liquid is a brownish red transparent liquid, is a byproduct generated in the pyrolysis process of bamboo charcoal, contains more than 200 chemical components, and acetic acid is a main component (2.0% -6.1%), and phenols, ketones, alcohols and esters are secondary. Studies show that the bamboo vinegar shows good sterilization and termite killing properties due to the phenolic compounds and acetic acid content. Bamboo vinegar has the potential to replace antibiotics: coli, staphylococcus aureus and candida albicans can be killed by 1:8 dilution of bamboo vinegar. Bamboo vinegar has antibacterial activity against E.coli even at a dilution of 1:100. In addition, the bamboo vinegar has certain inhibition effect on bacteria such as pseudomonas aeruginosa, staphylococcus aureus, candida albicans and the like. According to the content of phenolic compounds, the sterilizing performance of the wood vinegar is that coconut wood vinegar > bamboo wood vinegar > eucalyptus wood vinegar in sequence. Bamboo vinegar can be used as a safe feed additive because it is safe for animals as an oral drug in the toxicological safety assessment of Sprague Dawley (SD) rats and mice.
Bidens bipinnata L.also known as Bidens bipinnata L.is an annual herb plant of Bidens bipinnata (Bidens L.) genus of the family Compositae (Asteraceae). Herba Bidentis Bipinnatae is rich in bioactive components such as polyacetylene, flavonoids, alkaloids, phenols, terpenes, volatile oils, etc. The Chinese medicine is mainly used for treating malaria, diarrhea, acute gastritis, stomachache, hiccup, acute appendicitis and the like. Researches show that the spanishneedles herb has the functions of inhibiting bacteria, reducing blood pressure and blood sugar, diminishing inflammation, easing pain, resisting tumor and the like. The alcohol extract has good inhibition effect on staphylococcus aureus, bacillus subtilis, rice sheath blight and litchi epidemic.
Herba Houttuyniae (Houttuynia cordata thunder.) is perennial herb of houttuynia genus of Saururaceae family. The main source of the fishy smell is a volatile oil called decanoyl acetaldehyde (Decanoylacetal dehyde), namely houttuynin. Herba Houttuyniae contains multiple medicinal components such as polysaccharide, alkaloid, organic acid, flavone and volatile oil, and has analgesic, antibacterial, antiviral, antiaging and antioxidant effects. The whole houttuynia cordata strain can be used as a medicine, and has the main components of inhibiting staphylococcus aureus, escherichia coli, shigella dysenteriae and the like to different degrees. The research shows that the cordate houttuynia water extract and the azithromycin have synergistic effect on antibiosis and can inhibit the formation of pseudomonas aeruginosa biomembrane. In addition, houttuynia cordata has great potential in aflatoxin prevention and control.
The polygonum hydropiper (Polygonum hydropiper Linn.) is also called as polygonum hydropiper, is an annual herb medicine and food dual-purpose plant of Polygonum of Polygonaceae, and contains various bioactive components such as flavonoid, phenols, volatile oil, tannins, fatty acid, triterpenes, glycoside, and polygonic acid. The herba polygoni hydropiperis often used as antidiarrheal, condiment, mosquito fumigant, barn insect repellent and the like in folk. Researches show that the polygonum hydropiper has the effects of resisting bacteria, diminishing inflammation, easing pain, resisting oxidation and the like, and the polygonum hydropiper ethanol extract has an inhibiting effect on staphylococcus aureus.
In summary, natural plant-derived drugs have important significance in replacing antibiotics, but generally suffer from lower drug efficacy.

The nutrient broth culture medium is divided into test tubes with 1mL of the final volume of each tube, sterilized for 20min at 120 ℃ and 0.1MPa, the sterilized culture medium is cooled, 1mL of the liquid medicine is added for full mixing in a sterile operation, the liquid medicine is continuously diluted into a No. 1-8 tube by a double continuous dilution method (volume ratio), the No. 9 tube is a positive control, the No. 10 tube is a negative control (no bacteria adding), and sterile water and 10% DMSO are used as blank controls. 100. Mu.L of bacterial liquid was added to the blank control and 1-9 tubes. After incubation at 37℃for 24h at 150rpm, the liquid in the tube was observed for clarity and recorded. All treatment groups were repeated three times, with the lowest concentration corresponding to the liquid clarified test tube, i.e. the minimum inhibitory concentration (Minimum Inhibition Concentration, MIC).

Minimum bactericidal concentration determination (MBC)
On the basis of MIC, all clear sterile growth test tubes were selected, 100. Mu.L of the culture solution was spread on nutrient agar medium, and incubated overnight at 37℃for 24h, and repeated 3 times. The minimum drug concentration corresponding to the aseptic growth treatment is the minimum bactericidal concentration (Minimum Bactericidal Concentration, MBC).
Combined bacteriostasis index (FICI)
The combined bacteriostasis index (Faction Inhibitory Concentration Index, FICI) =mic/MIC when a drug is combined alone and mic+mic/MIC when b drug is combined alone, when FICI is less than or equal to 0.5, the drugs are synergistic; FICI is more than 0.5 and less than or equal to 1, and the medicines are added; FICI is more than 1 and less than or equal to 2, and the medicines have irrelevant effects; FICI > 2, antagonism between drugs.

Figure BDA0004027063900000131
As a result, tetracycline was highly sensitive to Salmonella enteritidis and Staphylococcus aureus, and had a MIC of 7.81. Mu.g/mL, but not to E.coli, and had a MIC of 250mg/mL. The possible reason is that the Escherichia coli used in the test has developed a certain drug resistance to tetracycline, but the plant-derived drug bamboo vinegar and the traditional Chinese medicine extract are not easy to develop the drug resistance. The MIC of the bamboo vinegar to the escherichia coli K88 and salmonella enteritidis treatment group is 3.13%, the MIC of the staphylococcus aureus treatment group is 1.56%, but in the compound liquid medicine, the MIC concentration of the escherichia coli K88 and salmonella enteritidis treatment group is only 0.78-1.56%, and the MIC of the staphylococcus aureus treatment group is only 0.781%. The MIC of the six traditional Chinese medicine extracts to the escherichia coli K88 is 15.6mg/mL, the salmonella enteritidis and staphylococcus aureus treatment group is 7.81-15.6 mg/mL, but in the compound liquid medicine, the MIC of the traditional Chinese medicine extracts to the escherichia coli K88 treatment group is only 1.95-3.91 mg/mL, and the MIC of the salmonella enteritidis and staphylococcus aureus treatment group is only 1.95mg/mL. This means that the concentration of the bamboo vinegar can be reduced to 1/2-1/4 of the original concentration after the compounding, and the concentration of the traditional Chinese medicine extract can be reduced to 1/4-1/8 of the original concentration after the compounding (with synergistic effect). The FICI index shows that the treatment of the bamboo vinegar and the alcohol extract of the spanishneedles herb has a synergistic effect on the inhibition of escherichia coli, and the compounding of the bamboo vinegar and the six traditional Chinese medicine extracts has a synergistic effect on the inhibition of salmonella enteritidis. After the bamboo vinegar is compounded with six traditional Chinese medicine extracts, the effect of inhibiting bacteria is stronger than that of a single medicine, and the antibacterial capacity is greatly enhanced.
